1Somatic Embryogenesis Receptor Kinases Control Root Development Mainly via Brassinosteroid-Independent Actions in Arabidopsis thaliana显示文摘Brassinosteroids(BRs),a group of plant steroidal hormones,play critical roles in many aspects of plant growth and development.Previous studies showed that BRI1-mediated BR signaling regulates cell division and differentiation during Arabidopsis root development via interplaying with auxin and other phytohormones.Arabidopsis somatic embryogenesis receptor-like kinases(SERKs),as co-receptors of BRI1,were found to play a fundamental role in an early activation step of BR signaling pathway.Here we report a novel function of SERKs in regulating Arabidopsis root development.Genetic analyses indicated that SERKs control root growth mainly via a BR-independent pathway.Although BR signaling pathway is completely disrupted in the serk1 bak1 bkk1 triple mutant,the root growth of the triple mutant is much severely damaged than the BR deficiency or signaling null mutants.More detailed analyses indicated that the triple mutant exhibited drastically reduced expression of a number of genes critical to polar auxin transport,cell cycle,endodermis development and root meristem differentiation,which were not observed in null BR biosynthesis mutant cpd and null BR signaling mutant bri1-701.Junbo Du Hongju Yin Shasha Zhang Zhuoyun Wei Baolin Zhao Jinghua Zhang Xiaoping Gou Honghui Lin Jia Li 2012Journal of Integrative Plant Biology2012,54,6:22

3Identification and fine mapping of two blast resistance genes in rice cultivar 93-11显示文摘Rice blast, caused by Magnaporthe oryzae, is a major disease of rice almost worldwide. The Chinese indica cultivar 93-11 is resistant to numerous isolates of the blast fungus in China, and can be used as broad-spectrum resistance resource, particularly in japonica rice breeding programs. In this study, we identified and mapped two blast resistance genes, Pi60(t) and Pi61(t), in cv. 93-11 using F2 and F3 populations derived from a cross between the susceptible cv. Lijiangxintuanheigu(LTH) and resistant cv. 93-11 and inoculated with M. oryzae isolates from different geographic origins. Pi60(t) was delimited to a 274 kb region on the short arm of chromosome 11, flanked by InDel markers K1-4 and E12 and cosegregated with InDel markers B1 and Y10. Pi61(t) was mapped to a 200 kb region on the short arm(near the centromere) of chromosome 12, flanked by InDel markers M2 and S29 and cosegregating with InDel marker M9. In the 274 kb region of Pi60(t), 93-11 contains six NBS-LRR genes including the two Pia/ PiCO39 alleles(BGIOSGA034263 and BGIOSGA035032) which are quite close to the two Pia/ PiCO39 alleles(SasRGA4 and SasRGA5) in Sasanishiki and CO39, with only nine amino acids differing in the protein sequences of BGIOSGA035032 and SasRGA5. In the 200 kb region of Pi61(t), 93-11 contains four NBS-LRR genes, all of which show high identities in protein sequence with their corresponding NBS-LRR alleles in susceptible cv. Nipponbare. Comparison of the response spectra and physical positions between the target genes and other R genes in the same chromosome regions indicated that Pi60(t) could be Pia/PiCO39 or its allele, whereas Pi61(t) appears to be different from Pita, Pita-2, Pi19(t), Pi39(t) and Pi42(t) in the same R gene cluster. 4Assessment of genetic diversity in the forest musk deer (Moschus berezovskii) using microsatellite and AFLP markers显示文摘Microsatellite genotyping and amplified fragment length polymorphism (AFLP) techniques are often utilized in studies of conservation genetics of endangered animals. To select a more effective marker system for conserving the endangered forest musk deer, we used microsatellite and AFLP markers to estimate levels of genetic diversity of two populations, the pure mother Jinfengshan (JFS) group and the offspring Baisha (BS) group with introduction of new blood. It was expected that JFS would pos- sess significantly higher genetic variability than BS if it is assumed that no new deer were introduced into BS. The results revealed that the 15 microsatellite markers produced 141 alleles and the 22 AFLP primer combinations yielded 908 polymorphic bands, showing that AFLPs had higher resolving power than microsatellites. Nonetheless, compared with other deer, both marker systems indicated that the forest musk deer contains a relatively high level of genetic variation. The 15 microsatellite loci indicated a similar level of genetic diversity was present in the JFS and BS populations, seemingly suggesting that the new blood obviously elevated the level of genetic diversity in BS. However, 22 suites of AFLP markers yielded a significantly higher level of genetic variation in JFS than that in BS, in contrast to the diversity pattern of microsatellites. In view of the difference in resolving power of these two marker systems, we conclude that gene introgression was limited in the BS population, as revealed by the AFLP markers. 5Donor-Derived CD19-Targeted T Cell Infusion Eliminates B Cell Acute Lymphoblastic Leukemia Minimal Residual Disease with No Response to Donor Lymphocytes after Allogeneic Hematopoietic Stem Cell Transplantation显示文摘Leukemia relapse is still the leading cause of treatment failure after allogeneic hematopoietic stem cell transplantation (allo-HSCT) for B cell acute lymphoblastic leukemia (B-ALL). Relapsed patients with BALL after allo-HSCT have a very short median survival. Minimal residual disease (MRD) is predictive of forthcoming hematological relapse after hematopoietic stem cell transplantation (HSCT);furthermore, eliminating MRD effectively prevents relapse. Donor lymphoblastic infusion (DLI) is the main established approach to treat B-ALL with MRD after allo-HSCT. However, about one-third of patients with MRD are non-responsive to DLI and their prognosis worsens. Although donor-derived cluster of differentiation (CD)19-directed chimeric antigen receptor-modified (CAR) T cells (CART19s) can potentially cure leukemia, the efficiency and safety of infusions with these cells have not yet been investigated in patients with MRD after HSCT. Between September 2014 and February 2018, six patients each received one or more infusions of CART19s from HSCT donors. Five (83.33%) achieved MRD-negative remission, and one case was not responsive to the administration of CAR T cells. Three of the six patients are currently alive without leukemia. No patient developed acute graft-versus-host disease (aGVHD), and no patient died of cytokine release syndrome. 6“Exploring petroleum inside source kitchen”: Shale oil and gas in Sichuan Basin显示文摘The Sichuan Basin is rich in shale oil and gas resources,with favorable geological conditions that the other shale reservoirs in China cannot match.Thus,the basin is an ideal option for fully'exploring petroleum inside source kitchen'with respect to onshore shale oil and gas in China.This paper analyzes the characteristics of shale oil and gas resources in the United States and China,and points out that maturity plays an important role in controlling shale oil and gas composition.US shale oil and gas exhibit high proportions of light hydrocarbon and wet gas,whereas Chinese marine and transitional shale gas is mainly dry gas and continental shale oil is generally heavy.A comprehensive geological study of shale oil and gas in the Sichuan Basin reveals findings with respect to the following three aspects.First,there are multiple sets of organic-rich shale reservoirs of three types in the basin,such as the Cambrian Qiongzhusi Formation and Ordovician Wufeng Formation-Silurian Longmaxi Formation marine shale,Permian Longtan Formation transitional shale,Triassic Xujiahe Formation lake-swamp shale,and Jurassic lacustrine shale.Marine shale gas enrichment is mainly controlled by four elements:Deep-water shelf facies,moderate thermal evolution,calcium-rich and silicon-rich rock association,and closed roof/floor.Second,the'sweet section'is generally characterized by high total organic carbon,high gas content,large porosity,high brittle minerals content,high formation pressure,and the presence of lamellation/bedding and natural microfractures.Moreover,the'sweet area'is generally characterized by very thick organic-rich shale,moderate thermal evolution,good preservation conditions,and shallow burial depth,which are exemplified by the shale oil and gas in the Wufeng-Longmaxi Formation,Longtan Formation,and Daanzhai Member of the Ziliujing Formation.Third,the marine,transitional,and continental shale oil and gas resources in the Sichuan Basin account for 50%,25%,and 30%of the respective types of shale oil and gas geological resources in China,with great potential to become the cradle of the shale oil and gas industrial revolution in China.Following the'Conventional Daqing-Oil'(i.e.,the Daqing oilfield in the Songliao Basin)and the'Western Daqing-Oil&Gas'(i.e.,the Changqing oilfield in the Ordos Basin),the Southwest oil and gas field in the Sichuan Basin is expected to be built into a'Sichuan-Chongqing Daqing-Gas'in China.Caineng ZOU Zhi YANG Shasha SUN Qun ZHAO Wenhua BAI Honglin LIU Songqi PAN Songtao WU Yilin YUAN 2020Science China Earth Sciences2020,63,7:6

8NEDD9 promotes cancer stemness by recruiting myeloid-derived suppressor cells via CXCL8 in esophageal squamous cell carcinoma显示文摘Objective:Esophageal squamous cell carcinoma(ESCC)has high morbidity and mortality rates worldwide.Cancer stem cells(CSCs)may cause tumor initiation,metastasis,and recurrence and are also responsible for chemotherapy and radiotherapy failures.Myeloid-derived suppressor cells(MDSCs),in contrast,are known to be involved in mediating immunosuppression.Here,we aimed to investigate the mechanisms of interaction of CSCs and MDSCs in the tumor microenvironment.Methods:ESCC tissues and cell lines were evaluated.Neural precursor cell expressed,developmentally downregulated 9(NEDD9)was knocked down and overexpressed by lentiviral transfection.Quantitative PCR,Western blot,immunohistochemistry,cell invasion,flow cytometry,cell sorting,multiplex chemokine profiling,and tumor growth analyses were performed.Results:Microarray analysis revealed 10 upregulated genes in esophageal CSCs.Only NEDD9 was upregulated in CSCs using the sphere-forming method.NEDD9 expression was correlated with tumor invasion(P=0.0218),differentiation(P=0.0153),and poor prognosis(P=0.0373).Additionally,NEDD9 was required to maintain the stem-like phenotype.Screening of chemokine expression in ESCC cells with NEDD9 overexpression and knockdown showed that NEDD9 regulated C-X-C motif chemokine ligand 8(CXCL8)expression via the ERK pathway.CXCL8 mediated the recruitment of MDSCs induced by NEDD9 in vitro and in vivo.MDSCs promoted the stemness of ESCC cells through NEDD9 via the Notch pathway.Conclusions:As a marker of ESCC,NEDD9 maintained the stemness of ESCC cells and regulated CXCL8 through the ERK pathway to recruit MDSCs into the tumor,suggesting NEDD9 as a therapeutic target and novel prognostic marker for ESCC.Dongli Yue Shasha Liu Tengfei Zhang Yong Wang Guohui Qin Xinfeng Chen Huanyu Zhang Dong Wang Lan Huang Feng Wang Liping Wang Song Zhao Yi Zhang 2021Cancer Biology & Medicine2021,18,3:4
9Effect of coal slurry on the corrosion of coal-mine equipment显示文摘The corrosion of coal mine equipment immersed in coal slurry is addressed. The corrosion of low carbon steel samples immersed in coal slurries of different concentrations (80, 130, and 180 g/L) prepared from coals of different rank (long-flame coal, meager lean coal, and anthracite) and different granularity (0.25-0.5 mm, 0.074-0.25 mm, and less than 0.074 mm particle size) was studied by the electrochemical method of polarization curve measurement, controlled potential sweeping, and continuous scanning. The results show that the corrosion rate in an anthracite slurry, where the coal has high coalification, is far greater than corrosion in a long-flame or a meager lean coal slurry. Furthermore the corrosion current, polarization current, and corrosion rate of low carbon steel become larger, and the polarizability becomes smaller, as the coal particle size decreases. 11Mn-Co oxide decorated on Cu nanowires as efficient catalysts for catalytic oxidation of toluene显示文摘Mn-Co mixed oxides were electrodeposited on Cu nanowires generated on Cu foam(CF)and used for effectively catalytic oxidation of toluene.The physical and chemical properties of the prepared catalysts were characterized by SEM,TEM,XRD,H_(2)-TPR,O_(2)-TPD and XPS.It is found that the Mn-Co mixed metal oxides were uniformly coated on the Cu nanowires by the electrochemical method,whose Mn/Co ratio can be tuned by adjusting the molar ratio of Mn/Co in the initial solution for the electrodeposition.The intimate contact between Mn and Co nanocrystals was found by HRTEM,which is important for realizing synergetic effects on improving catalytic activity.Meanwhile,the formation of the active surface oxygen species and the increase of the active species of Mn^(4+)and Co^(3+)were considered to make significant contribution to the catalytic oxidation of toluene.Mn-Co mixed metal oxide catalysts exhibited higher performance than the single metal oxide,and especially 0.10Mn-0.01Co/CF catalyst with the Mn/Co molar ratio of 10:1 in the initial solution for the electrodeposition achieved the highest catalytic activity with a low toluene conversion temperature(T_(90%))of 251℃,and displayed excellent catalytic stability even in the presence of water vapor.It is expected that such a simply-electrodeposited mixed metal oxides based catalysts could be applied for the oxidation of volatile organic compounds(VOCs)in a practical process.Jing Wang Peifen Wang Akihiro Yoshid Qiang Zhao Shasha Li Xiaogang Hao Abuliti Abudula Guangwen Xu Guoqing Guan 2020Carbon Resources Conversion2020,3,1:4

13Efficacy and safety of high-dose esomeprazole–amoxicillin dual therapy for Helicobacter pylori rescue treatment:a multicenter,prospective,randomized,controlled trial显示文摘Background:High-dose dual therapy(HDDT)with proton pump inhibitors(PPIs)and amoxicillin has attracted widespread attention due to its favorable efficacy in eradicating Helicobacter pylori(H.pylori).This study aimed to compare the efficacy and safety of high-dose PPI-amoxicillin dual therapy and bismuth-containing quadruple therapy for H.pylori rescue treatment.Methods:This was a prospective,randomized,multicenter,non-inferiority trial.Patients recruited from eight centers who had failed previous treatment were randomly(1:1)allocated to two eradication groups:HDDT(esomeprazole 40 mg and amoxicillin 1000 mg three times daily;theHDDTgroup)and bismuth-containing quadruple therapy(esomeprazole 40 mg,bismuth potassium citrate 220 mg,and furazolidone 100 mg twice daily,combined with tetracycline 500 mg three times daily;the tetracycline,furazolidone,esomeprazole,and bismuth[TFEB]group)for 14 days.The primary endpoint was the H.pylori eradication rate.The secondary endpoints were adverse effects,symptom improvement rates,and patient compliance.Results:A total of 658 patients who met the criteria were enrolled in this study.The HDDT group achieved eradication rates of 75.4%(248/329),81.0%(248/306),and 81.3%(248/305)asdetermined by the intention-to-treat(ITT),modified intention-totreat(MITT),and per-protocol(PP)analyses,respectively.The eradication rates were similar to those in the TFEB group:78.1%(257/329),84.2%(257/305),and 85.1%(257/302).The lower 95%confidence interval boundary(9.19%in the ITT analysis,9.21%in the MITT analysis,and9.73%in the PP analysis)was greater than the predefined non-inferiority margin of10%,establishing a non-inferiority of the HDDT group vs.the TFEB group.The incidence of adverse events in the HDDT group was significantly lower than that in the TFEB group(11.1%vs.26.8%,P<0.001).Symptom improvement rates and patients’compliance were similar between the two groups.Conclusions:Fourteen-day HDDT is non-inferior to bismuth-containing quadruple therapy,with fewer adverse effects and good treatment compliance,suggesting HDDT as an alternative for H.pylori rescue treatment in the local region.Trial registration:Clinicaltrials.gov,NCT04678492.Hanxin Bi Xingxing Chen Yuxin Chen Xin Zhao Shasha Wang Jiehong Wang Ting Lyu Shuang Han Tao Lin Mingquan Li Donghong Yuan Junye Liu Yongquan Shi 2022Chinese Medical Journal2022,,14:3
14A comparison of zooplankton assemblages in Nansi Lake and Hongze Lake,potential influences of the East Route of the South-to-North Water Transfer Project,China显示文摘Nansi Lake and Hongze Lake are both water storage lakes along the East Route of the South-to-North Water Transfer project(ESNT).Frequent changes in hydrologic properties are responsible factors for controlling the zooplankton community assemblages in both lakes,so we studied the possible influence of water transfer and environmental factors on zooplankton community structure and abundance.Zooplankton assemblages were investigated seasonally for one year in both lakes;a total of 133 and 122 zooplankton taxa were identified in Nansi Lake and Hongze Lake,respectively.The most dominant rotifer species were littoral,e.g.,Keratella tecta,Keratella valga and Lecane lunaris in Nansi Lake and Brachionus angularis,Brachionus forficula and Polyarthra vulgaris in Hongze Lake.Comparatively,Nansi Lake had a higher Shannon-Wiener diversity index value(5.13),while Hongze Lake had a higher species richness index(4.21).The average number of zooplankton across seasons in Nansi Lake(protozoa:774±63 ind./L,rotifers:4817±212 ind./L,cladocerans:896±14 ind./L,copepod:435±42 ind./L)was comparatively lower than Hongze Lake(protozoa:1238±63 ind./L,rotifers:6576±112 ind./L,cladocerans:1013±20 ind./L,copepod:534±25 ind./L).Canonical correspondence analysis identified differing environmental gradients that were most responsible for influencing zooplankton communities in the two lakes(Hongze Lake:NH4-N,total nitrogen,transparency and pH;Nansi:pH,temperature and total phosphorus).Frequent changes related to water transfer in lakes favoured the diversity of rotifers and protozoa communities.Zooplankton habitat preference,changes in community structure and opportunistic peaks and extinction of certain taxa were also observed in the study lakes.Kandathil Radhakrishnan DIVYA Shasha ZHAO Yushun CHEN Fei CHENG Lei ZHANG Jiao QIN Thundiparambil Sathrajith ARUNJITH V.Bjorn SCHMIDT Songguang XIE 2021Journal of Oceanology and Limnology2021,39,2:3

Because of their advantages in reflecting the overall biological effects or functions of drugs and providing visual and intuitive results, some biotechnological analysis methods have been gradually applied to pharmaceutical analysis from raw material to manufacturing and final product analysis,including DNA super-barcoding, DNA-based rapid detection, multiplex ligation-dependent probe amplification, hyperspectral imaging combined with artificial intelligence, 3D biologically printed organoids,omics-based artificial intelligence, microfluidic chips, organ-on-a-chip, signal transduction pathwayrelated reporter gene assays, and the zebrafish thrombosis model. 16Analysis of impacting factors on polarimetric SAR oil spill detection显示文摘Polarimetric synthetic aperture radar(SAR) oil spill detection parameters conformity coefficient(μ), Muller matrix parameters(|C|, B_0), the eigenvalues of simplified coherency matrix(λ_(nos)) and the influence of SAR observing parameters, ocean environment and noise level are investigated. Radarsat-2 data are used to make systematic analysis of polarimetric parameters for different incidences, wind speeds, noise levels and the ocean phenomena(oil slick and look likes). The influence of the SAR observing parameters, the ocean environment and the noise level on the typical polarimetric SAR parameter conformity coefficient has been analyzed. The results indicate that conformity coefficient cannot be simply used for oil spill detection, which represents the image signal to the noise level to some extent. When the signals are below the noise level for the oil slick and the look likes, the conformity coefficients are negative; while the signals above the noise level corresponds to positive conformity coefficients. For dark patches(low wind and biogenic slick) with the signal below the noise,polarization features such as conformity coefficient cannot separate them with oil slick. For the signal above the noise, the oil slick, the look likes(low wind and biogenic slick) and clean sea all have positive conformity coefficients, among which, the oil slick has the smallest conformity coefficient, the look likes the second, and the clean sea the largest value. For polarimetric SAR data oil spill detection, the noise plays a significant role. 20The histone methyltransferase SUVR2 promotes DSB repair via chromatin remodeling and liquidliquid phase separation显示文摘Maintaining genomic integrity and stability is particularly important for stem cells,which are at the top of the cell lineage origin.Here,we discovered that the plant-specific histone methyltransferase SUVR2 maintains the genome integrity of the root tip stem cells through chromatin remodeling and liquid-liquid phase separation(LLPS)when facing DNA double-strand breaks(DSBs).The histone methyltransferase SUVR2(MtSUVR2)has histone methyltransferase activity and catalyzes the conversion of histone H3 lysine 9 monomethylation(H3K9me1)to H3K9me2/3 in vitro and in Medicago truncatula.Under DNA damage,the proportion of heterochromatin decreased and the level of DSB damage marker y-H2AX increased in suvr2 mutants,indicating that MtSUVR2 promotes the compaction of the chromatin structure through H3K9 methylation modification to protect DNA from damage.Interestingly,MtSUVR2 was induced by DSBs to phase separate and form droplets to localize at the damage sites,and this was confirmed by immunofluorescence and fluorescence recovery after photobleaching experiments.The IDR1 and lowcomplexity domain regions of MtSUVR2 determined its phase separation in the nucleus,whereas the IDR2 region determined the interaction with the homologous recombinase MtRAD51.Furthermore,we found that MtSUVR2 drove the phase separation of MtRAD51 to form'DNA repair bodies,'which could enhance the stability of MtRAD51 proteins to facilitate error-free homologous recombination repair of stem cells.Taken together,our study reveals that chromatin remodeling-associated proteins participate in DNA repair through LLPS.Qianwen Liu Peng Liu Tuo Ji Lihua Zheng Chen Shen Shasha Ran Jinling Liu Yafei Zhao Yiding Niu Tao Wang Jiangli Dong 2022Molecular Plant2022,15,7:1
