1Effects of Lactobacillus plantarum on gut barrier function in experimental obstructive jaundice显示文摘AIM:To investigate the mechanisms of Lactobacillus plantarum(L.plantarum)action on gut barrier in preoperative and postoperative experimental obstructive jaundice in rats.METHODS:Forty rats were randomly divided into groups of sham-operation,bile duct ligation(BDL),BDL +L.plantarum,BDL+internal biliary drainage(IBD),and BDL+IBD+L.plantarum.Ten days after L.plantarum administration,blood and ileal samples were collected from the rats for morphological examination,and intestinal barrier function,liver function,intestinal oxidative stress and protein kinase C(PKC)activity measurement.The distribution and expression of the PKC and tight junction(TJ)proteins,such as occludin,zonula occludens-1,claudin-1,claudin-4,junction adhesion molecule-A and F-actin,were examined by confocal laser scanning microscopy,immunohistochemistry,Western blotting,real-time fluorescent quantitative polymerase chain reaction assay.RESULTS:L.plantarum administration substantially restored gut barrier,decreased enterocyte apoptosis,improved intestinal oxidative stress,promoted the activity and expression of protein kinase(BDL vs BDL+L.plantarum,0.295±0.007 vs 0.349±0.003,P<0.05;BDL+IBD vs BDL+IBD+L.plantarum,0.407±0.046 vs 0.465±0.135,P<0.05),and particularly enhanced the expression and phosphorylation of TJ proteins in the experimental obstructive jaundice(BDL vs BDL+L.plantarum,0.266±0.118 vs 0.326±0.009,P<0.05).The protective effect of L.plantarum was more prominent after internal biliary drainage(BDL+IBD vs BDL +IBD+L.plantarum,0.415±0.105 vs 0.494±0.145,P<0.05).CONCLUSION:L.plantarum can decrease intestinal epithelial cell apoptosis,reduce oxidative stress,and prevent TJ disruption in biliary obstruction by activating the PKC pathway.Yu-Kun Zhou Huan-Long Qin Ming Zhang Tong-Yi Shen Hong-Qi Chen Yan-Lei Ma Zhao-Xin Chu Peng Zhang Zhi-Hua Liu 2012World Journal of Gastroenterology2012,18,30:21
2Blood-based biomarkers for early detection of esophageal squamous cell carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a common malignant tumor of the digestive system worldwide,especially in China.Due to the lack of effective early detection methods,ESCC patients often present at an advanced stage at the time of diagnosis,which seriously affects the prognosis of patients.At present,early detection of ESCC mainly depends on invasive and expensive endoscopy and histopathological biopsy.Therefore,there is an unmet need for a noninvasive method to detect ESCC in the early stages.With the emergence of a large class of non-invasive diagnostic tools,serum tumor markers have attracted much attention because of their potential for detection of early tumors.Therefore,the identification of serum tumor markers for early detection of ESCC is undoubtedly one of the most effective ways to achieve early diagnosis and treatment of ESCC.This article reviews the recent advances in the discovery of blood-based ESCC biomarkers,and discusses the origins,clinical applications,and technical challenges of clinical validation of various types of biomarkers.Ling-Yu Chu Yu-Hui Peng Xue-Fen Weng Jian-Jun Xie Yi-Wei Xu 2020World Journal of Gastroenterology2020,26,15:14
3Scutellaria barbata D.Don Inhibits Colorectal Cancer Growth via Suppression of Wnt/β-Catenin Signaling Pathway显示文摘Objective: To investigate the effect of the ethanol extract of Scutellaria barbata D. Don(EESB) on colorectal cancer(CRC) growth and Wnt/β-catenin signaling pathway in vivo and in vitro. Methods: In vivoexperiment, CRC xenograft mouse model was constructed with injection of HT-29 cells. Following xenograft implantation, twenty mice were randomly divided into EESB-treated group(n=10) and control group(n=10) by a random number table, and were given with intra-gastric administration of 2 g/kg EESB or saline, 5 days a week for 16 days, respectively. At the end of experiment, tumors were removed and weighed by electronic scales. The proliferation biomarker Ki-67 of tumor was evaluated by immunohistochemistry(IHC) assay. In vitro study, HT-29 cells were treated with 0, 0.5, 1.5, 2.5 mg/m L EESB for 24 h. At the end of the treatment, the viability and survival of HT-29 cells were determined by methylthiazolyldiphenyl-tetrazolium bromide(MTT) assay and colony formation assay, respectively. The m RNA expression of c-Myc, Survivin and adenomatous polyposis coli(APC) was examined by reverse transcription-polymerase chain reaction(RT-PCR) both in tumor tissues of CRC xenograft mice and HT-29 cells. Protein expression of c-Myc, Survivin, APC, and β-catenin as well as β-catenin phosphorylation level were evaluated by IHC assay or Western blotting. Results: EESB significantly reduced tumor weight in CRC xenografts mice, compared with the control group(P<0.05). IHC assay showed that EESB significantly inhibited protein expression of Ki-67 in tumor tissues(P<0.05). MTT assay showed that EESB significantly reduced HT-29 cell viability in a dose-dependent manner(P<0.05). Colony formation assay showed that EESB dose-dependently decreased the survival of HT-29 cells(P<0.05). In addition, RT-PCR assay showed that EESB decreased the m RNA expression of c-Myc and Survivin and increased APC expression, both in tumor tissues of CRC xenograft mice and HT-29 cells(P<0.05). IHC assay or Western blotting showed that EESB decreased protein expression of β-catenin, c-Myc and Survivin, as well as increased APC expression and β-catenin phosphorylation in tumor tissues or HT-29 cells(P<0.05). Conclusions: EESB significantly reduced tumor growth in CRC xenografts mice, and inhibited the viability and survival of HT-29 cells. EESB could suppress the activation of the Wnt/β-catenin pathway, which might be one of the mechanisms whereby Scutellaria barbata D. 4Genetic Analysis and Primary Mapping of pms4, a Photoperiod-Sensitive Genic Male Sterility Gene in Rice (Oryza sativa)显示文摘To understand the genetic characteristics of a new photoperiod-sensitive genic male sterile line Mian 9S, some reciprocal crosses were made between Mian 9S and six indica rice materials, Yangdao 6, Luhui 602, Shuihui 527, Mianhui 725, Fuhui 838 and Yixiang 1B. Genetic analysis results suggested that the photoperiod-sensitive genic male sterility (PGMS) of Mian 9S was controlled by a single recessive nuclear gene. Thus, the F2 population derived from the cross of Yangdao 6/Mian 9S was used to map the PGMS gene in Mian 9S. By using SSR markers, the PGMS gene of Mian 9S was mapped on one side of the markers, RM6659 and RM1305, on rice chromosome 4, with the genetic distances of 3.0 cM and 3.5 cM, respectively. The gene was a novel PGMS gene and designated tentatively as pms4. 5Damage identification method of girder bridges based on finite element model updating and modal strain energy显示文摘A timely and accurate damage identification for bridge structures is essential to prevent sudden failures/collapses and other catastrophic accidents.Based on response surface model(RSM)updating and element modal strain energy(EMSE)damage index,this paper proposes a novel damage identification method for girder bridge structures.The effectiveness of the proposed damage identification method is investigated using experiments on four simply supported steel beams.With Xiabaishi Bridge,a prestressed continuous rigid frame bridge with large span,as the engineering background,the proposed damage identification method is validated by using numerical simulation to generate different bearing damage scenarios.Finally,the efficiency of the method is justified by considering its application to identifying cracking damage for a real continuous beam bridge called Xinyihe Bridge.It is concluded that the EMSE damage index is sensitive to the cracking damage and the bearing damage.The locations and levels of multiple cracking damages and bearing damages can be also identified.The results illuminate a great potential of the proposed method in identifying damages of real bridge structures.NIU Jie ZONG Zhou Hong CHU Fu Peng 2015Science China(Technological Sciences)2015,58,4:12
6Qingxuan Jiangya Decoction(清眩降压汤) Prevents Blood Pressure Elevation and Ameliorates Vascular Structural Remodeling via Modulating TGF-β 1/Smad Pathway in Spontaneously Hypertensive Rats显示文摘Objective:To elevate the effects of Qingxuan Jiangya Decoction(清眩降压汤,QXJYD)on hypertension and vascular structural remodeling(VSR)in spontaneously hypertensive rats(SHRs),and investigate the underlying mechanisms.Methods:SHRs(n=8)were given intra-gastric administration with 60 mg/kg of QXJYD or saline,daily for 8 weeks,while rats in SHR-control(n=8)and WKY(n=8)groups were received equal volumes of saline solution.Systolic blood pressures(SBP),diastolic blood pressures(DBP)and mean blood pressures(MBP)were measured once a week.The levels of angiotensinⅡ(AngⅡ),endothelin 1(ET-1)and plasma renin activity(PRA)were tested by enzyme-linked immunosorbent assay(ELISA)and radioimmunoassay,respectively.The effect of QXJYD on VSR was determined by examining the media thickness and the ex vivo contractility of thoracic aortic.The proliferation and fibrosis of vascular smooth muscle cells(VSMCs)were examined via immunohistochemical(IHC)staining for proliferating cell nuclear antigen(PCNA),collagen Ⅰ and collagen Ⅲ,respectively.The mRNA and protein expressions of transforming growth factor β1(TGF-β1),Smad3 and phosphorylation of Smad3 in thoracic aorta tissues were determined by real-time polymerase chain reaction(PCR)and Western blot assay,respectively.Results:QXJYD treatment led to a significant decrease of the elevation of blood pressure in SHRs and reduced the levels of AngⅡ,ET-1 and PRA in the serum(P<0.05).In addition,QXJYD treatment remarkably ameliorated VSR and vascular function in SHRs.Moreover,QXJYD inhibited VSMC proliferation and fibrosis by suppressing the expression of PCNA,collagen Ⅰ and collagen Ⅲ in thoracic aortic.Furthermore,QXJYD inhibited the expression of TGF-β1,Smad3 and the phosphorylation of Smad3,respectively(P<0.05).Conclusion:QXJYD reversed VSR by inhibiting VSMC proliferation and collagen deposition via regulation of TGF-β1/Smad signaling pathway,which may,in part,illuminate its anti-hypertensive activities.HE Fei CHU Jian-feng CHEN Hong-wei LIN Wei LIN Shan CHEN You-qin PENG Jun CHEN Ke-ji 2020Chinese Journal of Integrative Medicine2020,26,3:12
7Huoxin Pill(活心丸)Attenuates Cardiac Fibrosis by Suppressing TGF-β1/Smad2/3 Pathway in Isoproterenol-Induced Heart Failure Rats显示文摘Objective:To evaluate the effects of Huoxin Pill(活心丸,HXP)on cardiac fibrosis and heart failure(HF)in isoproterenol(ISO)-induced HF rats.Methods:Thirty Wistar rats were randomly divided into 5 groups including control,HF,isosorbide mononitrate(ISMN),HXP low(HXP-L),and HXP high(HXP-H)groups(n=6 for each group)according to the complete randomization method.Rats were pretreated with ISMN(5 mg/kg daily),low concentration of HXP(10 mg/kg daily)or high concentration of HXP(30 mg/kg daily)or equal volume of saline by intragastric administration for 1 week,followed by intraperitoneal injection of ISO(10 mg/kg,14 days),and continually intragastric administrated with above medicines or saline for additional 6 weeks.The effects of HXP treatment on the cardiac function,heart weight index(HWI),pathological changes,and collagen content were further assessed.Moreover,the role of HXP on activation of transforming growth factor-β1(TGF-β1)/Smads pathway was further explored using immunohistochemistry(IHC)and Westernblot assay.Results:HXP treatment significantly alleviated the decrease of ejection fraction(EF)and fractional shortening(FS),while decreased the elevation of left ventricular end-systolic volume(LVESV)in ISO-induced HF rats(P<0.05).Moreover,HXP treatment obviously attenuated the increase of HWI and serum level of creatine kinase MB(CK-MB,P<0.05),as well as pathological changes in ISO-induced HF rats.Further determination indicated that HXP treatment alleviated the elevation of collagenⅠand collagenⅢprotein expression in cardiac tissues of ISO-induced HF rats.Furthermore,HXP treatment significantly down-regulated the increase of TGF-β1 and p-Smad2/3 protein expression in cardiac tissues of HF rats(P<0.05),while did not affect the expression of total Smad2/3.Conclusions:HXP attenuated heart failure and cardiac fibrosis in ISO-induced HF rats by suppression of TGF-β1/Smad2/3 pathway.PENG Mei-zhong YANG Mei-ling SHEN A-ling ZHOU Xue-ling LU Yan LI Qi SHEN Zhi-qing HUANG Bin PENG Jun CHU Jian-feng 2021Chinese Journal of Integrative Medicine2021,27,6:10
8Donepezil attenuates vascular dementia in rats through increasing BDNF induced by reducing HDAC6 nuclear translocation显示文摘Vascular dementia(VD)is the second most common dementia disease after Alzheimer’s diseases(AD)in the world.Donepezil is used to treat mild to moderate AD,and it has been shown to treat cognitive impairment and memory deficits caused by VD.However,the action mechanism of donepezil against VD has not been clarified.In this study,a bilateral common carotid artery occlusion(BCCAO)model was established in rats to simulate the pathology of VD.Two weeks after the surgery,the rats were administered donepezil(10 mg·kg^−1·d^−1,ig)for 3 weeks,and then subjected to behavioral tests.We showed that donepezil treatment significantly improved the performance of BCCAO rats in Morris Water Mazes test and Step-down test.Furthermore,we showed that donepezil treatment significantly attenuated neurodegeneration and restored the synapse dendritic spines density in cortex and hippocampus.We revealed that donepezil treatment significantly increased BDNF expression in cortex and hippocampus.Interestingly,donepezil treatment significantly decreased nuclear translocation of HDAC6 and the binding between HDAC6 and BDNF promoter IV in cortex,but not in the hippocampus.The attenuated neurodegeneration by donepezil in cortex and hippocampus might due to the reduced ROS levels and increased phosphorylation of AMPK,whereas increased phosphorylation of AKT was only detected in cortex.In conclusion,our results demonstrate that donepezil attenuates neurodegeneration in cortex and hippocampus via increasing BDNF expression;the regulation of donepezil on HDAC6 occurred in cortex,but not in the hippocampus.This study further clarifies the pharmacological mechanism of donepezil,while also emphasizes the promising epigenetic regulation of HDAC6.Wen-xuan Jian Zhao Zhang Jia-hong Zhan Shi-feng Chu Ye Peng Ming Zhao Qi Wang Nai-hong Chen 2020Acta Pharmacologica Sinica2020,41,5:9

10Research on Low Cycle Fatigue Reliability-based Robust Design Optimization of Turbine Blade显示文摘针对涡轮叶片低周疲劳可靠性稳健设计优化问题,对叶片材料进行了高温疲劳试验,采用定量方程随机化方法处理试验数据,获得叶片材料的概率-应变-寿命曲线。采用贝塞尔曲线描述叶片型线方程,建立了涡轮叶片结构及流场的参数化模型,采用热-流-固耦合有限元法对涡轮流场和叶片进行了数值分析,得到叶片动能效率和应力应变分布特性。建立了叶片疲劳可靠性稳健设计优化模型,并采用响应面法获得叶片结构性能函数和极限状态函数,将叶片低周疲劳可靠性作为基本约束条件,采用序列二次规划优化法得到设计优化结果。研究结果表明,优化后的叶片低周疲劳可靠性以及稳健性显著提高,模型及方法正确可行,可用于涡轮叶片以及其他复杂结构的低周疲劳可靠性稳健设计优化。PENG Maolin YANG Zichun CAO Yueyun CHU Zhuli 2013中国电机工程学报2013,33,11:8
11Analysis of genetic architecture and favorable allele usage of agronomic traits in a large collection of Chinese rice accessions显示文摘Genotyping and phenotyping large natural populations provide opportunities for population genomic analysis and genome-wide association studies(GWAS). Several rice populations have been re-sequenced in the past decade;however, many major Chinese rice cultivars were not included in these studies. Here, we report large-scale genomic and phenotypic datasets for a collection mainly comprised of 1,275 rice accessions of widely planted cultivars and parental hybrid rice lines from China. The population was divided into three indica/Xian and three japonica/Geng phylogenetic subgroups that correlate strongly with their geographic or breeding origins. We acquired a total of 146 phenotypic datasets for 29 agronomic traits under multi-environments for different subpopulations. With GWAS, we identified a total of 143 significant association loci, including three newly identified candidate genes or alleles that control heading date or amylose content. Our genotypic analysis of agronomically important genes in the population revealed that many favorable alleles are underused in elite accessions, suggesting they may be used to provide improvements in future breeding efforts. 12Construction of a macromolecular structural model of Chinese lignite and analysis of its low-temperature oxidation behavior显示文摘The aim of this paper is to analyze the change in the active structure of lignite during the process of lowtemperature oxidation by constructing a molecular structure model for lignite. Using quantum computation combined with experimental results of proximate analysis, ultimate analysis, Fourier transform infrared spectroscopy(FTIR) and X-ray photoelectron spectroscopy(XPS), a structural model for the large molecular structure was constructed. By analyzing the bond lengths in the model molecule, the evolution law for the active structure of lignite was predicted for the process of low-temperature oxidation. In low-temperature oxidation,alkanes and hydroxyls are the primary active structures observed in lignite, though ether may also react. These active functional groups react with oxygen to release heat, thereby speeding up the reaction between coal and oxygen. 15Molecular Characterization and Correlation with β-lactam Resistance of Streptococcus pneumonia Isolates in Hangzhou,China显示文摘Penicillin-binding proteins(PBPs) are the target of β-lactam antibiotics(the major treatment for Streptococcus pneumoniae infections),and mutations in PBPs are considered as a primary mechanism for the development of β-lactam resistance in S.pneumoniae.This study was conducted to investigate the mutations in the PBPs of clinical S.pneumoniae isolates in Hangzhou,China,in correlation with β-lactam resistance.Results showed that 19 F was the predominant serotype(7/27) and 14 of the S.pneumoniae isolates were resistant to both penicillin G and cephalosporin.Genotyping results suggested that β-lactam-resistant isolates primarily exhibited single-site mutations in both the STMK and SRNVP motifs of pbp1 a in combination with double-site mutations in the STMK motif of pbp2 x,which might be the primary mechanisms underlying the β-lactam resistance of the isolates in this study.chu mei fen liu xiao xiang zhang shao ni huang yan ying du peng wang li fang ji lei yan jie sun ai hua 2018Biomedical and Environmental Sciences2018,31,5:7
16Bear Bile Powder Inhibits Growth of Hepatocellular Carcinoma via Suppressing STAT3 Signaling Pathway in Mice显示文摘Objective: To evaluate the inhibitory effect of bear bile powder(BBP) on hepatocellular carcinoma(HCC) growth in vivo and investigate the underlying mechanisms. Methods: A HCC xenograft mouse model was developed by producing with huh7 cells. After 5 days following xenograft implantation, ten HCC xenograft mice were given intra-gastric administration with 10 mg/(kg·d) dose of BBP or saline for 3 weeks. Tumor growth in HCC xenograft mice was evaluated by measuring the tumor weight and volume. Cell apoptosis, proliferation or tumor angiogenesis were examined via immunohistochemical(IHC) staining for transferase-mediated deoxyuridine triphosphate-biotin nick end labeling(TUNEL), proliferating cell nuclear antigen(PCNA) or cluster of differentiation 31(CD31), respectively. Phosphorylation of signal transducer and activator of transcription 3(STAT3) were determined by Western blot. The m RNA and protein expressions of Bcl-2, Bax, Cyclin D1 and Cyclin-dependent kinase 4(CDK4) in HCC tumor tissues were respectively determined by reverse transcription polymerase chain reaction(RT-PCR) and Western blot. The protein expression of vascular endothelial growth factor A(VEGF-A) in tumor tissues was examined by IHC staining. Results: BBP treatment led to a significant decrease on tumor volume and tumor weight in HCC mice(P<0.05) and had no effect on the change of body weight. In addition, BBP profoundly promoted cell apoptosis, inhibited cell proliferation and intratumoral microvessel density in HCC tumor tissues(P<0.05). Moreover, BBP treatment remarkably suppressed the STAT3 phosphorylation and modulated the expression of critical target genes including Bcl-2, Bax, Cyclin D1, CDK4 and VEGF-A in HCC mice. Neurological deficit scores, western blotting, and reverse transcription-PCR results demonstrated that electroacupuncture markedly reduced neurological deficits, decreased corpus striatum aquaporin-4 protein and mRNA expression, and relieved damage to the blood-brain barrier in a rat model of cerebral ischemia/reperfusion injury.
