1Molecular immune pathogenesis and diagnosis of COVID-19显示文摘Coronavirus disease 2019(COVID-19)is a kind of viral pneumonia which is caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).The emergence of SARS-CoV-2 has been marked as the third introduction of a highly pathogenic coronavirus into the human population after the severe acute respiratory syndrome coronavirus(SARS-CoV)and the Middle East respiratory syndrome coro-navirus(MERS-CoV)in the twenty-first century.In this minireview,we provide a brief introduction of the general features of SARS-CoV-2 and discuss current knowledge of molecular immune pathogenesis,diagnosis and treatment of COVID-19 on the base of the present understanding of SARS-CoV and MERS-CoV infections,which may be helpful in offering novel insights and potential therapeutic targets for combating the SARS-CoV-2 infection.Xiaowei Li Manman Geng Yizhao Peng Liesu Meng Shemin Lu 2020Journal of Pharmaceutical Analysis2020,10,2:31
2Sleep Deprivation Selectively Down-Regulates Astrocytic 5-HT2B Receptors and Triggers Depressive-Like Behaviors via Stimulating P2X7 Receptors in Mice显示文摘Chronic loss of sleep damages health and disturbs the quality of life.Long-lasting sleep deprivation(SD)as well as sleep abnormalities are substantial risk factors for major depressive disorder,although the underlying mechanisms are not clear.Here,we showed that chronic SD in mice promotes a gradual elevation of extracellular ATP,which activates astroglial P2X7 receptors(P2X7Rs).Activated P2X7Rs,in turn,selectively down-regulated the expression of 5-HT2B receptors(5-HT2BRs)in astrocytes.Stimulation of P2X7Rs induced by SD selectively suppressed the phosphorylation of AKT and FoxO3 a in astrocytes,but not in neurons.The overexpression of FoxO3a in astrocytes inhibited the expression of 5-HT2BRs.Down-regulation of 5-HT2BsRs instigated by SD suppressed the activation of STAT3 and relieved the inhibition of Ca2+-dependent phospholipase A2.This latter cascade promoted the release of arachidonic acid and prostaglandin E2.The depression-like behaviors induced by SD were alleviated in P2X7R-KO mice.Our study reveals the mechanism underlying chronic SD-induced depression-like behaviors and suggests 5-HT2BRs as a key target for exploring therapeutic strategies aimed at the depression evoked by sleep disorders.Maosheng Xia Zexiong Li Shuai Li Shanshan Liang Xiaowei Li Beina Chen Manman Zhang Chengyi Dong Alexei Verkhratsky Dawei Guan Baoman Li 2020Neuroscience Bulletin2020,36,11:10

6Pharmacokinetics of H002, a novel S1PR_1 modulator, and its metabolites in rat blood using liquid chromatography–tandem mass spectrometry显示文摘A rapid and sensitive liquid chromatography–tandem mass spectrometry(LC–MS/MS) method was developed and validated for the simultaneous determination of H002 and its phosphorylated metabolite, H002-P and hydroxylated metabolite H002-M, in rat blood. H001, an analogue of H002, was used as the internal standard.Blood samples were prepared by simple protein precipitation. The analytes and internal standard were separated on a Zorbax SB-C18 column with a gradient mobile phase consisting of methanol and water containing 0.1% formic acid at a flow rate of 0.2 mL /min with an operating temperature of 20 1C. The detection was performed on a triple quadrupole tandem mass spectrometer with positive electrospray ionization in multiple-reaction monitoring mode.Linear detection responses were obtained from 0.2–100 ng/mL for H002 and H002-M, while 0.5–100 ng/mL for H002-P. The intra- and inter-day precision(RSD%) was within 11.76%, with the accuracy(RE%) ranging from –9.84% to 9.12%. 8Stimulus-Responsive Room Temperature Phosphorescence Materials:Internal Mechanism,Design Strategy,and Potential Application显示文摘CONSPECTUS:Room temperature phosphorescence(RTP)materials,which could respond to external stimuli,such as force,heat,light,electric filed,etc.,have drawn increasing attention for their broad application prospects,especially in the fields of anticounterfeiting,sensors,data storage,and so on.In comparison with the traditional fluorescence ones,RTP materials show much longer emission lifetimes,which can be even caught by the naked eye.Thus,the change in emission lifetime under an external stimulus for RTP materials can be also a potential monitoring parameter,in addition to emission color and intensity.In other words,the number of visual monitoring parameters could increase from two to three in RTP materials,which would greatly facilitate their practical applications.Until now,RTP materials have been typically limited to metal-containing inorganic materials,particularly rare-earth phosphors.Their emissions are governed by the slow liberation of trapped charge carriers from isolated traps of impurities,defects,or ions through thermal stimulation with low luminescence efficiency.However,these materials suffer from some intrinsic disadvantages,including high cost,potential toxicity,and instability in aqueous environments.In order to solve these problems,the purely organic RTP materials should be a good choice.However,these kinds of materials are really scarce now,especially for the ones with stimulus response characteristic.To develop purely organic RTP materials with a stimulus response effect,we and other scientists have tried a lot.Luckily,some progresses have been made.In this Account,we present our recent progress on the stimulus-responsive room temperature phosphorescence of organic materials,mainly focusing on the internal mechanism and potential applications.First,the fundamental knowledge is described to illustrate the importance and main principles of the stimulus-responsive RTP effect.Then,some typical stimulus-responsive RTP materials based on different internal mechanisms are discussed.Mainly,two kinds of stimulus-responsive RTP materials were introduced,namely,single-component and multicomponent ones.Correspondingly,their dynamic change of the RTP property under external stimulus occurred based on the distinct internal mechanisms.For single-component materials,the changes in molecular structure,packing,or conformation,have played a significant role in their corresponding stimulus-responsive RTP effect.As for multicomponent materials,the changed oxygen concentration in matrix and intermolecular distance between different components were found more during the stimulus-responsive RTP process.Accordingly,different potential applications were explored based on the different stimulus-responsive RTP processes.With the classification of stimulus-responsive RTP materials based on different internal mechanisms,the corresponding design strategy could be well proposed,thus guiding the further development of this research field.Jie Yang Manman Fang Zhen Li 2021Accounts of Materials Research2021,2,8:3
9Mechanisms of traditional Chinese medicines in the treatment of allergic rhinitis using a network biology approach显示文摘Objective:To systematically explore the effect and mechanism of traditional Chinese herbs in the treatment of allergic rhinitis.Methods:Magnolia flower,xanthium fruit,astragalus root,paniculate cynanchum,scutellaria root,cicada molting,stephania root,small centipeda herb,and licorice root as traditional Chinese herbs are widely used to treat allergic rhinitis.With multiple database integration and entity grammar systems,a molecular interaction network of traditional Chinese herbs for treating allergic rhinitis was established.Bioinformatics approaches were adopted to integrate relevant data and biological information by multiple databases.Molecular interaction network of Chinese herbs for treating allergic rhinitis was constructed based on entity grammar systems model.The network was then used for elucidating the antiallergic rhinitis mechanism of the Chinese herbs on the molecular level.The subnetworks were also extracted to explicitly display the pathway and targets where effective components acted on.Results:Traditional Chinese herbs could influence various pathological aspects in allergic rhinitis including the production of pro-inflammatory substances,such as histamine and cytokine interleukin-4(IL-4),interleukin-13(IL-13),lowering immunoglobulin E(IgE)level or blocking antigen binding,altering the biological processes of IgE,modulating the balance of T helper(Th)or other cells by cell proliferation and differentiation,mediating cellecell signaling and second-messenger-mediated signaling,stabilizing the cell membrane,and affecting regulation of cellular defense response.Conclusion:The research theoretically confirms the mechanism of anti-allergic rhinitis by traditional Chinese herbs,provides important fundamental research information for treatment of allergic rhinitis,and may serve as a reference for new drug development and effective ingredients of compatibility.Li Du Xiaotong Ye Manman Li Huihui Wang Baixia Zhang Rao Zheng Yun Wang 2021Journal of Traditional Chinese Medical Sciences2021,8,1:3

13Analysis of the Winter Cloud-to-Ground Lightning Activity and Its Synoptic Background in China during 2010-20显示文摘Cloud-to-ground(CG)lightning data and the ECMWF ERA-Interim reanalysis dataset are analyzed to gain insight into the spatiotemporal distribution and synoptic background of winter-season CG flashes between December 2010 and February 2020 in China.We identify three Winter Lightning Frequent Areas(WLAs):the southwest side of the Yunnan-Guizhou Plateau(WLA1),the east side of the Yunnan-Guizhou Plateau(WLA2),and the Poyang Lake Plain(WLA3).The CG lightning flashes most frequently occur at local midnight and have a monthly peak in February.The CG lightning in WLA1 is mostly generated in non-frontal weather;however,the lightning in WLA2 and WLA3 mostly occurs in frontal systems.The frontal circulation situation is divided into four typical types:transversal trough after high pressure,low vortex,confrontational convergence,and asymptotic convergence.In all typical weather patterns,the lightning occurs downstream of a 500 hPa trough and is accompanied by a southwesterly low-level jet.The convective parameters of winter thunderstorms differ greatly from those of summer thunderstorms.The maximum convective available potential energy(MCAPE)and K-index(KI)are more useful metrics than convective available potential energy(CAPE)and Showalter index(SI)during winter.This study further deepens the understanding of the distribution characteristics of winter CG lightning in China,which motivates further research to improve the ability of winter thunderstorm prediction.Manman MA Xiaogang HUANG Jianfang FEI Chi ZHANG Chao LI Xiaoping CHENG 2022Advances in Atmospheric Sciences2022,39,6:2
14Characterization of microbial communities in sediments of the South Yellow Sea显示文摘Illumina sequencing and quantitative PCR(qPCR)based on the 16 S ribosomal RNA(rRNA)gene were conducted to characterize the vertical distribution of bacterial and archaeal communities in the sediments of two sites from the South Yellow Sea.Both bacterial and archaeal communities showed a clear stratified distribution with sediment depth.The microbial communities in the upper layers were distinct from those in the deeper layers;the relative abundances of sequences of Thaumarchaeota,Gammaproteobacteria,and Actinobacteria were higher in the upper than in the deeper sediments,whereas the sequences of Bathyarchaeia,Lokiarchaeota,Euryarchaeota,Chloroflexi,and Deltaproteobacteria were relatively more abundant in the deeper sediments.Sediment depth and total organic carbon(TOC)can significantly influence both the bacterial and archaeal communities.Furthermore,bacterial and archaeal groups potentially involved in nitrogen,sulfur,and methane metabolism were detected in both sites.In our study,both ammonia-oxidizing bacteria(Nitrospira)and ammonia-oxidizing archaea(Candidatus Nitrosopumilus)were responsible for ammonia oxidization.Additionally,sulfur-reducing bacteria SEEP-SRB1 forming consortia with anaerobic methane-oxidizing archaea ANME-2 a-2 b were capable of anaerobic methane oxidation(AOM)in the 3400-02 sediment samples.Ye CHEN Siqi LI Xiaoqing XU Manman MA Tiezhu MI Yu ZHEN Zhigang YU 2021Journal of Oceanology and Limnology2021,39,3:2

17Detection of Ratoon Stunting Disease in Virus-free Seedcane via Real-time Fluorescence Quantitative PCR显示文摘This study was to develop the real-time fluorescence quantitative PCR technique for detecting the ratoon stunting disease (RSD) in virus-free seedcane seedlings. Healthy tissue culture seedlings were obtained from six plants of sugarcane ROC22, which had been confirmed RSD-positive by detecting the sugarcane juice, by employing the sugarcane seedlings production protocol. Real-time fluorescence quantitative PCR was used to detect RSD pathogens in tissue culture samples. The results showed that target fragment of RSD pathogens was not found in all 10 samples in real-time fluorescence quantitative PCR, with the Ct values of 37-39. 20Genipin improves lipid metabolism and sperm parameters in obese mice via regulation of miR-132 expression显示文摘Obesity has now surpassed malnutrition and infectious diseases as the most significant contributor to health problems worldwide.In particular,obesity is associated with several metabolic disorders,including hyperlipidemia,hepatic steatosis,and subfertility.Genipin(GNP),the aglycone of geniposide,is isolated from the extract of the traditional Chinese medicine Gardenia jasminoides Ellis and has been used in traditional oriental medicine against several inflammation-driven diseases.However,the effect and molecular mechanism of GNP on obesityassociated dyslipidemia and sperm dysfunction still need to be explored.In this study,we detect the effects of GNp on hyperlipidemia,hepatic lipid accumulation and sperm function using a high-fat diet(HFD)-induced obese mouse model.We find that obese mice treated with GNP show an improvement in body weight,serum triglyceride levels,serum hormone levels,serum inflammatory cytokines,hepatic steatosis and sperm function.At the molecular level,HFD/GNP diversely regulates the expression of miR-132 in a tissue-specific manner.miR-132 further targets and regulates the expression of SREBP-1c in liver cells,as well as the expressions of SREBP-1c and StAR in Leydig cells in the testis,thus modifying lipogenesis and steroidogenesis,respectively.Collectively,our data demonstrate that GNP shows a broad effect on the improvement of HFD-induced metabolic disorder and sperm dysfunction in male mice by tissue-specific regulation of miR-132.Our findings reveal the function GNP in ameliorating hepatic lipid metabolism and sperm function and suggest that this compound is a versatile drug to treat metabolic disorders.Li Wangi Ge Chen Shuyao Wu YihuaXul Chenxi Guo Manman Wang Tingming Liang Zhigang Guo Hong-Jie Di Zhigang Hu 2022Acta Biochimica et Biophysica Sinica2022,54,9:1
