1Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. 2De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. 3Progress and perspective of the cathode/electrolyte interface construction in all-solid-state lithium batteries显示文摘Security risks of flammability and explosion represent major problems with the use of conventional lithium rechargeable batteries using a liquid electrolyte.The application of solid-state electrolytes could effectively help to avoid these safety concerns.However,integrating the solid-state electrolytes into the all-solid-state lithium batteries is still a huge challenge mainly due to the high interfacial resistance present in the entire battery,especially at the interface between the cathode and the solid-state electrolyte pellet and the interfaces inside the cathode.Herein,recent progress made from investigations of cathode/solid-state electrolyte interfacial behaviors including the contact problem,the interlayer diffusion issue,the space-charge layer effect,and electrochemical compatibility is presented according to the classification of oxide-,sulfide-,and polymer-based solid-state electrolytes.We also propose strategies for the construction of ideal next-generation cathode/solid-state electrolyte interfaces with high room-temperature ionic conductivity,stable interfacial contact during long cycling,free formation of the space-charge region,and good compatibility with high-voltage cathodes.Shiming Su Jiabin Ma Liang Zhao Kui Lin Qidong Li Shasha Lv Feiyu Kang Yan-Bing He 2021Carbon Energy2021,3,6:3
4RECQL4 regulates DNA damage response and redox homeostasis in esophageal cancer显示文摘Objective:RECQL4(a member of the RECQ helicase family)upregulation has been reported to be associated with tumor progression in several malignancies.However,whether RECQL4 sustains esophageal squamous cell carcinoma(ESCC)has not been elucidated.In this study,we determined the functional role for RECQL4 in ESCC progression.Methods:RECQL4 expression in clinical samples of ESCC was examined by immunohistochemistry.Cell proliferation,cellular senescence,the epithelial-mesenchymal transition(EMT),DNA damage,and reactive oxygen species in ESCC cell lines with RECQL4 depletion or overexpression were analyzed.The levels of proteins involved in the DNA damage response(DDR),cell cycle progression,survival,and the EMT were determined by Western blot analyses.Results:RECQL4 was highly expressed in tumor tissues when compared to adjacent non-tumor tissues in ESCC(P<0.001)and positively correlated with poor differentiation(P=0.011),enhanced invasion(P=0.033),and metastasis(P=0.048).RECQL4 was positively associated with proliferation and migration in ESCC cells.Depletion of RECQL4 also inhibited growth of tumor xenografts in vivo.RECQL4 depletion induced G0/G1 phase arrest and cellular senescence.Importantly,the levels of DNA damage and reactive oxygen species were increased when RECQL4 was depleted.DDR,as measured by the activation of ATM,ATR,CHK1,and CHK2,was impaired.RECQL4 was also shown to promote the activation of AKT,ERK,and NF-k B in ESCC cells.Conclusions:The results indicated that RECQL4 was highly expressed in ESCC and played critical roles in the regulation of DDR,redox homeostasis,and cell survival.Guosheng Lyu Peng Su Xiaohe Hao Shiming Chen Shuai Ren Zixiao Zhao Yaoqin Gong Qiao Liu Changshun Shao 2021Cancer Biology & Medicine2021,18,1:2

10Potential factors and mechanism of particulate matters explosive increase induced by free radicals oxidation显示文摘Atmospheric particulate pollution in China has attracted much public attention.Occasionally, the particle number concentration increases sharply in a short time period,which is defined as a 'particulate matter explosive increase'. Heavy particulate matter pollution not only reduces visibility but also has an adverse effect on human health. Hence,there is an urgent need to discover the causes of particulate matter explosive increase.During this campaign, the particle number concentration and free radicals were measured at a tall building on the campus of Lanzhou University of Technology. Additionally, we examined a series of chemicals to reproduce the observed particulate matter explosive increase in a smog chamber to determine its potential factors. Then, we analyzed the mechanism of particulate matter explosive increase in the presence of free radicals. We found that, among the potential inorganic and organic sources analyzed, a mixture of organic and SO_2 in the research region had a major effect on particulate matter explosive increase. 13Stability of Fe-As composites formed with As(Ⅴ)and aged ferrihydrite显示文摘During the aging process,ferrihydrite was transformed into mineral mixtures composed of different proportions of ferrihydrite,goethite,lepidocrocite and hematite.Such a transformation may affect the fixed ability of arsenic.In this study,the stability of Fe-As composites formed with As(Ⅴ)and the minerals aged for 0,1,4,10 and 30 days of ferrihydrite were systematically examined,and the effects of molar of ratios Fe/As were also clarified using kinetic methods combined with multiple spectroscopic techniques.The results indicated that As(Ⅴ)was rapidly adsorbed on minerals during the initial polymerization process,which delayed both the ferrihydrite conversion and the hematite formation.When the Fe/As molar ratio was 1.875 and 5.66,the As(Ⅴ)adsorbed by ferrihydrite began to release after 6 hr and 12 hr,respectively.The corresponding release amounts of As(Ⅴ)were 0.55 g/L and 0.07 g/L,and the adsorption rates were 92.43%and 97.50%at 60 days,respectively.However,the As(Ⅴ)adsorbed by the transformation products aged for 30 days of ferrihydrite began to release after adsorbed 30 days.The corresponding release amounts of As(Ⅴ)were 0.25 g/L and 0.03 g/L,and the adsorption rates were 84.23%and 92.18%after adsorbed 60 days,for the Fe/As=1.875 and 5.66,respectively.Overall,the combination of As(Ⅴ)with ferrihydrite and aged products transformed from a thermodynamically metastable phase to a dynamically stable state within a certain duration.Moreover,the aging process of ferrihydrite reduced the sorption ability of arsenate by iron(hydr)oxide but enhanced the stability of the Fe-As composites.Zhonglan Yang Lingyu Bai Shiming Su Yanan Wang Cuixia Wu Xibai Zeng Benhua Sun 2021Journal of Environmental Sciences2021,33,2:0

15Photoactive materials based on semiconducting nanocarbons——A challenge opening new possibilities for photocatalysis显示文摘This perspective paper introduces the concept that nanocarbons and related materials such as carbon dots are an interesting intrinsic photocatalytic semiconducting material, and not only a modifier of the existing(semiconducting) materials to prepare hybrid materials. The semiconducting properties of the nanocarbons, and the possibility to have the band gap within the visible-light region through defect band engineering, introduction of light heteroatoms and control/manipulation of the curvature or surface functionalization are discussed. These materials are conceptually different from the 'classical' semiconducting photocatalysts, because semiconductor domains with tuneable characteristics are embedded in a conductive carbon matrix, with the presence of various functional groups(as C=O groups) enhancing charge separation by trapping electrons. These nanocarbons open a range of new possibilities for photocatalysis both for energetic and environmental applications.
