| 1 | Relationship between Cognition Function and Hippocampus Structure after Long-term Microwave Exposure显示文摘Objective To analyze the effects of long-term microwave exposure on hippocampal structure and function in the rat.Methods Experiments were performed on 184 male Wistar rats(three exposure groups and a sham group).Microwaves were applied daily for 6 min over 1 month at average power densities of 2.5,5,and 10 mW/cm2.Learning and memory abilities were assessed by Morris water maze.High performance liquid chromatography was used to detect neurotransmitter concentrations in the hippocampus.Hippocampal structures were observed by histopathological analysis.Results Following long-term microwave exposure there was a significant decrease in learning and memory activity in the 7 d,14 d,and 1 m in all three microwave exposure groups.Neurotransmitter concentrations of four amino acids(glutamate,aspartic acid,glycine,and gamma-aminobutyric acid) in hippocampus were increased in the 2.5 and 5 mW/cm2 groups and decreased in the 10 mW/cm2 group.There was evidence of neuronal degeneration and enlarged perivascular spaces in the hippocampus in the microwave exposure groups.Further,mitochondria became swollen and cristae were disordered.The rough endoplasmic reticulum exhibited sacculated distension and there was a decrease in the quantity of synaptic vesicles.Conclusion These data suggest that the hippocampus can be injured by long-term microwave exposure,which might result in impairment of cognitive function due to neurotransmitter disruption. | ZHAO Li PENG Rui Yun WANG Shui Ming WANG Li Feng GAO Ya Bing DONG Ji LI Xiang SU Zhen Tao | 2012 | Biomedical and Environmental Sciences2012,25,2: | 27 |
| 2 | Role of PERK/eIF2α/CHOP Endoplasmic Reticulum Stress Pathway in Oxidized Low-density Lipoprotein Mediated Induction of Endothelial Apoptosis显示文摘Objective PERK/eI F2α/CHOP is a major signaling pathway mediating endoplasmic reticulum(ER) stress related with atherosclerosis.Oxidized LDL(ox-LDL) also induces endothelial apoptosis and plays a vital role in the initiation and progression of atherosclerosis.The present study was conducted to explore the regulatory effect of ox-LDL on PERK/e IF2α/CHOP signaling pathway in vascular endothelial cells.Methods The effects of ox-LDL on PERK and p-e IF2α protein expression of primary human umbilical vein endothelial cells(HUVECs) were investigated by Western blot analysis.PERK gene silencing and selective eI F2α phosphatase inhibitor,salubrinal were used to inhibit the process of ox-LDL induced endothelial cell apoptosis,caspase-3 activity,and CHOP mR NA level.Results Ox-LDL treatment significantly increased the expression of PERK,PERK-mediated inactivation of e IF2α phosphorylation,and the expression of CHOP,as well as the caspase-3 activity and apoptosis.The effects of ox-LDL were markedly decreased by knocking down PERK with stable transduction of lentiviral sh RNA or by selective eI F2α phosphatase inhibitor,salubrinal.Conclusion This study provides the first evidence that ox-LDL induces apoptosis in vascular endothelial cells mediated largely via the PERK/eI F2α/CHOP ER-stress pathway.It adds new insights into the molecular mechanisms underlying the pathogenesis and progression of atherosclerosis. | TAO Yong Kang YU Pu Lin BAI Yong Ping YAN Sheng Tao ZHAO Shui Ping ZHANG Guo Qiang | 2016 | Biomedical and Environmental Sciences2016,29,12: | 21 |
| 3 | Elevated Platelet Activating Factor Level in Ischemia-Related Arrhythmia and Its Electrophysiological Effect on Myocardium显示文摘Objective The mechanism through which platelet activating factor (PAF) induces cardiac electrical activity and arrhythmia is not well understood and previous studies have suggested a potential involvement of ion channels in its action. The present study was aimed to clarify the role of PAF in fatal arrhythmias following acute myocardia infarction (AMI) and the underlying mechanism. Methods (1) Blood PAF levels were measured among 72 AMI patients at the time of diagnosis with AMI and 48 h later, and their electrocardiogram (ECG) was recorded continuously. (2) Ischemia simulation and surface electrocardiogram were conducted in 20 pigs and their PAF levels were measured. (3) PAF perfusion and standard microelectrode recording were performed on guinea pig papillarymuscles. Results In both humans and pigs, elevated PAF levels were detected in AMI and simulated ischemia, respectively, and even higher PAF levels were found when fatal arrhythmias occurred. In guinea pig myocardium, PAF induced a shortening of action potential duration at 90% level of repolarization (APD 90 )under non-ischemic conditions and a more pronounced shortening under early simulated ischemic conditions. Conclusion AMI and ischemia are associated with increased PAF levels in humans and pigs, which are further raised when fatal arrhythmia follows. | 6 | Effect of PCEs with Different Functional Groups on the Performance of Cement Paste显示文摘The adsorption behaviors and dispersing properties of polycarboxylate superplasticizer(PCE) with different functional groups were systematically analyzed to reveal the theory and methods of modifying PCE molecular structures and regulating PCE performances. By substituting carboxylic groups with sulfonic groups, ester groups or acylamino groups, respectively, modified PCEs with different functional groups were synthesized. Results show that introducing low amount of ester groups or sulfonic groups into the PCE molecules has no negative effects on the fluidity of cement paste, while introducing acylamino groups into PCE molecules significantly weakens the fluidity of cement paste. At low amount(when the molar ratio of sodium methallyl sulfonate to TPEG is lower than 0.4), the rapid adsorption of sulfonic groups onto the cement particles contributes to the high dispersing performance of the sulfonic group modified PCEs. When the substitution ratio of acrylic acid by sulfonic acid is higher than 0.4, the viscosity and the yield stress of cement paste increases sharply. Redundant sulfonic groups lead to the excessive charge density of the PCE, which contributes to the inhomogeneous adsorption on the cement grains and hence results in the decline of the dispersing performance. Substitution of carboxylic group by acylamino group or ester group slightly changes the viscosity as well as the yield stress of cement paste. | 12 | Carbon and nitrogen stable isotope analyses of mammal bone fossils from the Zhongba site in the Three Gorges Reservoir region of the Yangtze River,China显示文摘Based on AMS 14 C dating data,carbon and nitrogen isotope analyses were conducted on mammal bone collagen of deer,cattle and pigs from the Zhongba site in the Three Gorges Reservoir region of the Yangtze River.These analyses were conducted to reconstruct palaeodiets of mammals,palaeoecology,palaeoenviroment and previous human activities in the study area.Results show that the collagen loss of bone did not change the in vivo isotopic composition of carbon and nitrogen stable isotopes,and most of the bone fossils were well preserved.The bone collagen of samples from deer had a mean δ 13 C of-23.1‰ and a mean δ 15 N of 4.7‰,suggesting that deer subsisted in a closed habitat and fed on branches and leaves.The bone collagen of cattle had a mean δ 13 C of-19.6‰ and a mean δ 15 N of 5.2‰,which indicates that cattle subsisted in an open habitat and fed on grasses and stems.The δ 13 C values show that both deer and cattle fed on C 3 plants and lived in the same ecosystem,but the t-test results show that deer δ 13 C and δ 15 N values were both more negative than those of cattle,indicating that they inhabited different niches.The δ 13 C and δ 15 N values of cattle partially overlapped those of deer,suggesting some competition in diets between them.The t-tests show that the δ 13 C and δ 15 N values of pigs were more positive than those of cattle and deer,which signifies that pigs occupied a higher trophic level compared to cattle and deer.The wide range of pig δ 13 C values demonstrates that pig trading had been taking place from early Neolithic Age to late Bronze Age.There were no significant differences in deer δ 13 C and δ 15 N values among different archaeological periods,making it clear that climatic,ecological and environmental conditions were kept relatively stable from 2200 to 4200 a BP.This stability may have been responsible for the extensive and complete cultural layers at the Zhongba site.The minimum number of samples required to estimate the mean δ 13 C values of deer,pigs and cattle are 8,73 and 16,respectively,and for mean δ 15 N values of deer,pigs and cattle,the minimum numbers are 4,5 and 6,respectively. | TIAN XiaoSi ZHU Cheng SUN ZhiBin SHUI Tao HUANG YunPing FLAD Rowan Kimon LI YuMei | 2011 | Chinese Science Bulletin2011,56,2: | 2 |
