2The involvement of wheat U-box E3 ubiquitin ligase TaPUB1 in salt stress tolerance显示文摘U-box E3 ubiquitin ligases play important roles in the ubiquitin/26 S proteasome machinery and in abiotic stress responses. Ta PUB1-overexpressing wheat(Triticum aestivum L.) were generated to evaluate its function in salt tolerance. These plants had more salt stress tolerance during seedling and flowering stages, whereas the Ta PUB1-RNA interference(RNAi)-mediated knock-down transgenic wheat showed more salt stress sensitivity than the wild type(WT). Ta PUB1 overexpression upregulated the expression of genes related to ion channels and increased the net root Na+efflux, but decreased the net K+efflux and H+influx, thereby maintaining a low cytosolic Na+/K+ratio, compared with the WT. However,RNAi-mediated knock-down plants showed the opposite response to salt stress. Ta PUB1 could induce the expression of some genes that improved the antioxidant capacity of plants under salt stress. Ta PUB1 also interacted with Ta MP(Triticum aestivum α-mannosidase protein), a regulator playing an important role in salt response in yeast and in plants. Thus, low cytosolic Na+/K+ratios and better antioxidant enzyme activities could be maintained in wheat with overexpression of Ta PUB1 under salt stress. 3Copy number alteration profiling facilitates differential diagnosis between ossifying fibroma and fibrous dysplasia of the jaws显示文摘Ossifying fibroma(OF)and fibrous dysplasia(FD)are two fibro-osseous lesions with overlapping clinicopathological features,making diagnosis challenging.In this study,we applied a whole-genome shallow sequencing approach to facilitate differential diagnosis via precise profiling of copy number alterations(CNAs)using minute amounts of DNA extracted from morphologically correlated microdissected tissue samples.Freshly frozen tissue specimens from OF(n=29)and FD(n=28)patients were obtained for analysis.Lesion fibrous tissues and surrounding normal tissues were obtained by laser capture microdissection(LCM),with~30–50 cells(5000–10000µm2)per sample.We found that the rate of recurrent CNAs in OF cases was much higher(44.8%,13 of 29)than that in FD cases(3.6%,1 of 28).Sixty-nine percent(9 of 13)of the CNA-containing OF cases involved segmental amplifications and deletions on Chrs 7 and 12.We also identified eight CNA-associated genes(HILPDA,CALD1,C1GALT1,MICALL2,PHF14,AIMP2,MDM2,and CDK4)with amplified expression,which was consistent with the copy number changes.We further confirmed a jaw lesion with a previous uncertain diagnosis due to its ambiguous morphological features and the absence of GNAS mutation as OF based on the typical Chr 12 amplification pattern in its CNA profile.Moreover,analysis of a set of longitudinal samples collected from an individual with a cellular lesion in suspicion of OF at the first surgery,recurrence and the latest malignant transformation revealed identical CNA patterns at the three time points,suggesting that copy number profiling can be used as an important tool to identify borderline lesions or lesions with malignant potential.Overall,CNA profiling of fibro-osseous lesions can greatly improve differential diagnosis between OF and FD and help predict disease progression.Ming Ma Lu Liu Ruirui Shi Jianyun Zhang Xiaotian Li Xuefen Li Jiaying Bai Jianbin Wang Yanyi Huang Tiejun Li 2021International Journal of Oral Science2021,13,2:3

5Optical and SAR image registration based on improved nonsubsampled wavelet transform for sea islands显示文摘Homologous feature point extraction is a key problem in the optical and synthetic aperture radar(SAR) image registration for islands. A new feature point extraction method using a threshold shrink operator and non-subsampled wavelet transform(TSO-NSWT) for optical and SAR image registration was proposed. Moreover, the matching for this proposed feature was different from the traditional feature matching strategies and was performed using a similarity measure computed from neighborhood circles in low-frequency bands. Then, a number of reliably matched couples with even distributions were obtained, which assured the accuracy of the registration. Application of the proposed algorithm to SPOT-5(multi-spectral) and YG-1(SAR) images showed that a large number of accurately matched couples could be identified. Additionally, both of the root mean square error(RMSE) patterns of the registration parameters computed based on the TSO-NSWT algorithm and traditional NSWT algorithm were analyzed and compared, which further demonstrated the effectiveness of the proposed algorithm. 8An Adaptive Physics-Informed Neural Network with Two-Stage Learning Strategy to Solve Partial Differential Equations显示文摘Physics-Informed Neural Network(PINN)represents a new approach to solve Partial Differential Equations(PDEs).PINNs aim to solve PDEs by integrating governing equations and the initial/boundary conditions(I/BCs)into a loss function.However,the imbalance of the loss function caused by parameter settings usually makes it difficult for PINNs to converge,e.g.because they fall into local optima.In other words,the presence of balanced PDE loss,initial loss and boundary loss may be critical for the convergence.In addition,existing PINNs are not able to reveal the hidden errors caused by non-convergent boundaries and conduction errors caused by the PDE near the boundaries.Overall,these problems have made PINN-based methods of limited use on practical situations.In this paper,we propose a novel physics-informed neural network,i.e.an adaptive physics-informed neural network with a two-stage training process.Our algorithm adds spatio-temporal coefficient and PDE balance parameter to the loss function,and solve PDEs using a two-stage training process:pre-training and formal training.The pre-training step ensures the convergence of boundary loss,whereas the formal training process completes the solution of PDE by balancing various loss functions.In order to verify the performance of our method,we consider the imbalanced heat conduction and Helmholtz equations often appearing in practical situations.The Klein-Gordon equation,which is widely used to compare performance,reveals that our method is able to reduce the hidden errors.Experimental results confirm that our algorithm can effectively and accurately solve models with unbalanced loss function,hidden errors and conduction errors.The codes developed in this manuscript are publicy available at http://gffzz188fe103f8f1460asbv599x0foq0w6n69.ffgz.tsg.suse.edu.cn/callmedrcom/ATPINN.Shuyan Shi Ding Liu Ruirui Ji Yuchao Han 2023Numerical Mathematics(Theory,Methods and Applications)2023,16,2:0

10Small protein Cgl2215 enhances phenolic tolerance by promoting MytA activity in Corynebacterium glutamicum显示文摘Corynebacterium glutamicum is a promising chassis microorganism for the bioconversion of lignocellulosic biomass owing to its good tolerance and degradation of the inhibitors generated in lignocellulosic pretreatments.Among the identified proteins encoded by genes within the C.glutamicum genome,nearly 400 are still functionally unknown.Based on previous transcriptome analysis,we found that the hypothetical protein gene cgl2215 was highly upregu-lated in response to phenol,ferulic acid,and vanillin stress.The cgl2215 deletion mutant was shown to be more sensitive than the parental strain to phenolic compounds as well as other environmental factors such as heat,ethanol,and oxidative stresses.Cgl2215 interacts with C.glutamicum mycoloyltransferase A(MytA)and enhances its in vitro esterase activity.Sensitivity assays of theΔmytA andΔcgl2215ΔmytA mutants in response to phenolic stress estab-lished that the role of Cgl2215 in phenolic tolerance was mediated by MytA.Furthermore,transmission electron microscopy(TEM)results showed that cgl2215 and mytA deletion both led to defects in the cell envelope structure of C.glutamicum,especially in the outer layer(OL)and electron-transparent layer(ETL).Collectively,these results indicate that Cgl2215 can enhance MytA activity and affect the cell envelope structure by directly interacting with MytA,thus playing an important role in resisting phenolic and other environmental stresses.Huawei Gu Xinwei Hao Ruirui Liu Zhenkun Shi Zehua Zhao Fu Chen Wenqiang Wang Yao Wang Xihui Shen 2022Stress Biology2022,2,1:0

13Post-modification of MOF to fabricate single-atom dispersed hollow nanocages catalysts for enhancing CO_(2)conversion显示文摘During the catalytic process,the microenvironment and surface area of the catalyst will affect the catalytic performance.Hence,an assisted organic linker coated metal-organic framework(MOF)has been applied,to form Ni/HNC(HNC represents hollow nanocage)for electrocatalytic CO_(2)reduction.Remarkably,Ni/HNC achieves superb activity with high Faradaic efficiency(FE)of 97.2%at 0.7 V vs.reversible hydrogen electrode(RHE)towards CO_(2)conversion to CO.In contrast to Ni/NPC(afforded from the naked MOF),the Ni/HNC displays higher FE and selectivity on CO rather than H_(2),owing to the large nanocage which extraordinarily facilitates CO_(2)enrichment and the active sites easily accessible.This work provides a general and feasible route to construct high-efficient electrochemical CO_(2)reduction reaction(EC-CO_(2)RR)catalysts via post-modified MOFs.Ruirui Yun Ruiming Xu Changsong Shi Beibei Zhang Tuanhui Li Lei He Tian Sheng Zheng Chen 2023Nano Research2023,16,7:0
14Transcriptome and carotenoid profiling of different varieties of Coffea arabica provides insights into fruit color formation显示文摘The processability and ultimate quality of coffee(Coffea arabica)are determined by the composition of the matured fruits.The basis of genetic variation in coffee fruit quality could be explained by studying color formation during fruit maturation.Transcriptome profiling was conducted on matured fruits of four C.arabica varieties(orange colored fruits(ORF);purple colored fruits(PF);red colored fruits(RF)and yellow colored fruits(YF))to identify key color-regulating genes,biosynthesis pathways and transcription factors implicated in fruit color formation.A total of 39,938 genes were identified in the transcriptomes of the four C.arabica varieties.In all,2745,781 and 1224 differentially expressed genes(DEGs)were detected in YF_vs_PF,YF_vs_RF and YF_vs_ORF,respectively,with 1732 DEGs conserved among the three pairwise groups.Functional annotation of the DEGs led to the detection of 28 and 82 key genes involved in the biosynthesis of carotenoids and anthocyanins,respectively.Key transcription factors bHLH,MYB,NAC,MADS,and WRKY implicated in fruit color regulation were detected.The high expression levels of gene-LOC113688784(PSY),gene-LOC113730013(b-CHY),gene-LOC113728842(CCD7),gene-LOC113689681(NCED)and gene-LOC113729473(ABA2)in YF may have accounted for the yellow coloration.The differential expression of several anthocyanin and carotenoid-specific genes in the fruits substantially account for the purple(PF),red(RF),and orange(ORF)colorations.This study provides important insights into fruit color formation and variations in C.arabica and will help to develop coffee varieties with specific color and quality traits.Faguang Hu Xiaofei Bi Hongming Liu Xingfei Fu Yanan Li Yang Yang Xiaofang Zhang Ruirui Wu Guiping Li Yulan Lv Jiaxiong Huang Xinping Luo Rui Shi 2022Plant Diversity2022,44,3:0
15Successive treatment with naltrexone induces epithelial–mesenchymal transition and facilitates the malignant biological behaviors of bladder cancer cells显示文摘Naltrexone is widely used for alleviating opioid-related side effects in cancer patients.However,the effects of naltrexone on cancer progression are controversial in the literature.The present study was carried out to investigate the effects of successive treatment with clinically relevant doses of naltrexone on the malignant biological behaviors of bladder cancer cells.The human bladder cancer T24 cells and mouse bladder cancer MB49 cells were treated with naltrexone.Cell proliferation,migration,and invasion abilities were analyzed.Morphological changes of the cells were confirmed by F-actin immunofluorescence staining.Epithelial–mesenchymal transition(EMT)-related markers and transcriptional factors,as well as activation of the phosphatidylinositol 3 kinase(PI3K)/AKT signaling pathway,were analyzed.Results showed that,compared with the control group,successive treatment with naltrexone significantly promoted the proliferation and decreased the apoptosis of bladder cancer cells,together with increase in cell migration and invasion ability.Continuous treatment with naltrexone also significantly reduced the expression of epithelial markers(E-cadherin and cytokeratin 19),increased the expression of mesenchymal markers(N-cadherin and vimentin)and EMT-inducing transcription factors(Snail and Slug),and further shifted the morphological phenotype of bladder cancer cells to a mesenchymal phenotype.The PI3K/AKT signaling pathway was activated by successive treatment with naltrexone.Notably,incubation with the specific PI3K inhibitor LY294002 together with naltrexone reversed the naltrexone-induced EMT progression.In conclusion,successive treatment with naltrexone may be favorable for the progression of bladder tumors by activating the PI3K/AKT signaling pathway and inducing EMT.Long-term exposure to naltrexone should be used cautiously in patients with bladder cancer.Xiaoqiang Wang Ruirui Zhang Tong Wu Yumiao Shi Xiao Zhou Dan Tang Weifeng Yu Edmund Cheung So Xiaodan Wu Zhiying Pan Jie Tian 2021Acta Biochimica et Biophysica Sinica2021,53,2:0
16Correlation of PCSK1 with nonalcoholic fatty liver disease in a Han Chinese population:a case-control observational study显示文摘Objective: This study aimed to investigate the association between single-nucleotide polymorphisms (SNPs) ofPCSK1 (proprotein convertase subtilisin/kexin type 1) related to obesity and nonalcoholic fatty liver disease (NAFLD).Methods: In this case-control observational study, four candidate SNPs (rs6234, rs155971, rs6232, rs3811951) ofPCSK1 were genotyped in 732 NAFLD patients and 823 healthy control participants, all of whom were of ethnic Han Chinese descent. All participants came from Shanghai, China, and joined our study during 2015 to 2016. The frequencies of each allele and genotype, paired linkage disequilibrium, and haplotype were calculated on the SHEsis platform. In addition to SHEsis, five different genetic models (codominant, dominant, recessive, overdominant, and log-additive) were employed to identify the correlation between genotype frequency and NAFLD. This study was approved by the Medical Ethics Committee of Shanghai University of Traditional Chinese Medicine (approved No. 2017LCSY069).Results: In a comparison of NAFLD patients and healthy participants, none of the fourPCSK1 SNPs were significantly correlated with the occurrence of NAFLD (P>0.05), in either genotypic or allelic distribution.
