| 1 | pH-dependent leaching behaviour and other performance properties of cement-treated mixed contaminated soil显示文摘Portland cement has been widely used for stabilisation/solidification(S/S) treatment of contaminated soils.However,there is a dearth of literature on pH-dependent leaching of contaminants from cement-treated soils.This study investigates the leachability of Cu,Pb,Ni,Zn and total petroleum hydrocarbons(TPH) from a mixed contaminated soil.A sandy soil was spiked with 3000 mg/kg each of Cd,Cu,Pb,Ni and Zn,and 10,000 mg/kg of diesel,and treated with ordinary Portland cement(CEM I).Four different binder dosages,5%,10%,15% and 20%(m/m) and different water contents ranging from 13%-19% dry weight were used in order to find a safe operating envelope for the treatment process.The pH-dependent leaching behaviour of the treated soil was monitored over an 84-day period using a 3-point acid neutralisation capacity(ANC) test.The monolithic leaching test was also conducted.Geotechnical properties such as unconfined compressive strength(UCS),hydraulic conductivity and porosity were assessed over time.The treated soils recorded lower leachate concentrations of Ni and Zn compared to the untreated soil at the same pH depending on binder dosage.The binder had problems with Pb stabilisation and TPH leachability was independent of pH and binder dosage.The hydraulic conductivity of the mixes was generally of the order,10-8 m/sec,while the porosity ranged from 26%-44%.The results of selected performance properties are compared with regulatory limits and the range of operating variables that lead to acceptable performance described. | Reginald B. | 4 | Role for urinary biomarkers in diagnosis of acute rejection in the transplanted kidney显示文摘Despite the introduction of potent immunosuppressive medications within recent decades, acute rejection still accounts for up to 12% of all graft losses, and is generally associated with an increased risk of late graft failure. Current detection of acute rejection relies on frequent monitoring of the serum creatinine followed by a diagnostic renal biopsy. This strategy is flawed since an alteration in the serum creatinine is a late clinical event and significant irreversible histologic damage has often already occurred. Furthermore, biopsies are invasive procedures that carry their own inherent risk. The discovery of non-invasive urinary biomarkers to help diagnose acute rejection has been the subject of a significant amount of investigation. We review the literature on urinary biomarkers here, focusing on specific markers perforin and granzyme B m RNAs, FOXP3 m RNA, CXCL9/CXCL10 and mi RNAs. | 8 | MicroRNA profile in neosquamous esophageal mucosa following ablation of Barrett's esophagus显示文摘AIM To investigate the micro RNA expression profile in esophageal neosquamous epithelium from patients who had undergone ablation of Barrett's esophagus.METHODS High throughput screening using Taq Man~ Array Human Micro RNA quantitative PCR was used to determine expression levels of 754 micro RNAs in distal esophageal mucosa(1 cm above the gastro-esophageal junction) from 16 patients who had undergone ablation of non-dysplastic Barrett's esophagus using argon plasma coagulation vs pretreatment mucosa, posttreatment proximal normal non-treated esophageal mucosa, and esophageal mucosal biopsies from 10 controls without Barrett's esophagus. Biopsies of squamous mucosa were also taken from 5 cm above the pre-ablation squamo-columnar junction. Predicted m RNA target pathway analysis was used to investigate the functional involvement of differentially expressed micro RNAs.RESULTS Forty-four micro RNAs were differentially expressed between control squamous mucosa vs post-ablation neosquamous mucosa. Nineteen micro RNAs were differentially expressed between post-ablation neosquamous and post-ablation squamous mucosa obtained from the more proximal non-treated esophageal segment. Twelve microRNAs were differentially expressed in both neosquamous vs matched proximal squamous mucosa and neosquamous vs squamous mucosa from healthy patients. Nine micro RNAs(mi R-424-5p, mi R-127-3p, mi R-98-5p, mi R-187-3p, mi R-495-3p, mi R-34c-5p, mi R-223-5p, mi R-539-5p, mi R-376a-3p, mi R-409-3p) were expressed at higher levels in post-ablation neosquamous mucosa than in matched proximal squamous and healthy squamous mucosa. These micro RNAs were also more highly expressed in Barrett's esophagus mucosa than matched proximal squamous and squamous mucosa from controls. Target prediction and pathway analysis suggests that these micro RNAs may be involved in the regulation of cell survival signalling pathways. Three micro RNAs(mi R-187-3p, mi R-135b-5p and mi R-31-5p) were expressed at higher levels in postablation neosquamous mucosa than in matched proximal squamous and healthy squamous mucosa. These mi RNAs were expressed at similar levels in preablation Barrett's esophagus mucosa, matched proximal squamous and squamous mucosa from controls. | 10 | Implications of seismic pounding on the longitudinal response of multi-span bridges-an analytical perspective显示文摘Seismic pounding between adjacent frames in multiple-frame bridges and girder ends in multi-span simply supported bridges has been commonly observed in several recent earthquakes. The consequences of pounding include damage to piers, abutments, shear keys, bearings and restrainers, and possible collapse of deck spans. This paper investigates pounding in bridges from an analytical perspective. A simplified nonlinear model of a multiple-frame bridge is developed including the effects of inelastic frame action and nonlinear hinge behavior, to study the seismic response to longitudinal ground motion. Pounding is implemented using the contact force-based Kelvin model, as well as the momentum-based stereomechanical approach, Parameter studies are conducted to determine the effects of frame period ratio, column hysteretic behavior, energy dissipation during impact and near source ground motions on the pounding response of the bridge. The results indicate that pounding is most critical for highly out-of-phase frames and is not significant for frame period ratios greater than 0.7. Impact models without energy dissipation overestimate the displacement and acceleration amplifications due to impact, especially for elastic behavior of the frames. Representation of stiffness degradation in bridge columns is essential in capturing the accurate response of pounding frames subjected to far field ground motion. | 13 | Overexpression of fibrinogen-like protein 2 protects against T cell-induced colitis显示文摘AIM To determine the effect of overexpression of fibrinogenlike protein 2(FGL2) on regulatory T cell(Treg) and effector T(Teff) cell function on T cell-induced colitis in Rag1-/-mice.METHODS Treg and Teff cells from fgl2-/-, fgl2+/+, and fgl2 Tg mice were purified by FACS. They were studied in vitro for immunosuppressive activity and cell proliferation and in vivo for their effects on the development and prevention of T cell-induced colitis in Rag1-/-mice. RESULTS In vitro, fgl2 Tg Treg had enhanced immunosuppressive activity, and fgl2 Tg Teff had reduced proliferation to alloantigen stimulation. Transfer of Teff from C57Bl/6J mice(fgl2+/+) into Rag1-/-mice produced both clinical and histologic colitis with dense infiltrates of CD3+ T cells, crypt abscesses and loss of goblet cells. Fgl2 Tg Treg prevented the development of T cell-induced colitis, whereas fgl2+/+ and fgl2-/-Treg were only partially protective. In mice that received fgl2 Tg Treg, the ratio of Foxp3+ to CD3+ cells was increased both in the colon and in mesenteric lymph nodes, and Teff cell proliferation as determined by staining with Ki67 was reduced. Teff cells from fgl2 Tg mice did not produce colitis. CONCLUSION Here we show that fgl2 Tg Teff are hypoproliferative and do not induce colitis. We further demonstrate that fgl2 Tg Treg prevent colitis in contrast to fgl2+/+ Treg, which were only partially protective.
