1全日制放牧BMY牛的活体采卵-体外胚胎生产-胚胎移植研究显示文摘为建立BMY牛的活体采卵(OPU)-体外胚胎生产(IVP)-胚胎移植(ET)快速扩繁技术体系,从人工草地全放牧牛群中选择健康的经产空怀BMY牛作为供体(n=14)集中OPU一次;卵母细胞经TCM-199体外成熟培养(IVM)、Fert-TALP液体外受精(IVF),受精卵用合成输卵管液(SOF)体外培养(IVC)生产胚胎;BMY牛受体(n=81)用CUE-MATETM孕酮阴道栓+EB+FSH+PG方法同期发情处理,对黄体合格的移植OPU-IVP来源鲜胚。研究结果表明:①OPU头均获卵母细胞23.50枚,获10枚以上卵母细胞的供体占85.7%;IVP的平均囊胚数和可用胚数分别为5.21和4.00枚;OPU前8~9d卵巢上有黄体(CL)的供体,头均获卵母细胞数及IVP的囊胚数、可移植胚数、囊胚率和可移植胚率分别比对照组高7.28枚(P<0.05)、7.29枚(P<0.05)、6.00枚(P<0.05)、27.3%(P<0.01)和23.2%(P<0.01);②受体黄体合格率69.1%(56/81),胚胎移植妊娠率37.5%(21/56),产犊率35.7%(20/56);③受体在胚胎移植时注射GnRH,其妊娠率比对照组高16.2%(P>0.05)。结果显示,BMY母牛具有良好的OPU潜力,但个体间差异较大;可通过严格供体选择及改善IVP技术等措施,实现OPU-IVP效率最大化。CUE-MATETM+EB+FSH+PG同期发情处理方法适于在牛规模化胚胎移植(ET)和人工授精(AI)中应用。综合应用OPU、IVP、同期发情和ET技术是加快优良BMY牛扩繁的重要途径之一。和占星 王喆 张继才 王安奎 金显栋 袁希平 Joaquim M.Garcia Marina R.de Lima 何永珍 黄必志 2013畜牧兽医学报2013,44,1:4

3An alternative,zeaxanthin epoxidase-independent abscisic acid biosynthetic pathway in plants显示文摘Abscisic acid(ABA)is an important carotenoid-derived phytohormone that plays essential roles in plant response to biotic and abiotic stresses as well as in various physiological and developmental processes.In Arabidopsis,ABA biosynthesis starts with the epoxidation of zeaxanthin by the ABA DEFICIENT 1(ABA1)enzyme,leading to epoxycarotenoids;e.g.,violaxanthin.The oxidative cleavage of 9-cis-epoxycaro-tenoids,a key regulatory step catalyzed by 9-C/S-EPOXYCAROTENOID DIOXYGENASE,forms xanthoxin,which is converted in further rea.ctions mediated by ABA DEFICIENT 2(ABA2),ABA DEFICIENT 3(ABA3),and ABSCISIC ALDEHYDE OXIDASE 3(AAO3)into ABA.By combining genetic and biochemical approaches,we unravel here an ABA1-independent ABA biosynthetic pathway starting upstream of zeaxanthin.We iden-tified the carotenoid cleavage products(i.e.,apocarotenoids,β-apo-11-carotenal,9-cis-β-apo-11-carotenal,3-OH-β-apo-11-carotenal,and 9-cis-3-OH-β-apo-11-carotenal)as intermediates of this ABA1-independent ABA biosynthetic pathway.Using labeled compounds,we showed thatβ-apo-11-carotenal,9-cis-β-apo-11-carotenal,and 3-OH-β-apo-11-carotenal are successively converted into 9-cis-3-OH-β-apo-11-carotenal,xanthoxin,and finally into ABA in both Arabidopsis and rice.When applied to Arabidopsis,theseβ-apo-11-carotenoids exert ABA biological functions,such as maintaining seed dormancy and inducing the expression of ABA-responsive genes.Moreover,the transcdptomic analysis revealed a high overlap of differentially expressed genes regulated byβ-apo-11-carotenoids and ABA,suggesting thatβ-apo-11-carot-enoids exert ABA-independent regulatory activities.Taken together,our study identifies a biological function for the common plant metabolites,β-apo-11-carotenoids,extends our knowledge about ABA biosynthesis,and provides new insights into plant apocarotenoid metabolic networks.Kun-Peng Jia Jianing Mi Shawkat Ali Hajime Ohyanagi Juan C.Moreno Abdugaffor Ablazov Aparna Balakrishna Lamis Berqdar Alessia Fiore Gianfranco Diretto Claudio Martínez Angel R.de Lera Takashi Gojobori Salim Al-Babili 2022Molecular Plant2022,15,1:2

5Time-dependent measurement of high-power laser light reflection by low-Z foam plasma显示文摘Porous materials have many applications for laser–matter interaction experiments related to inertial confinement fusion.Obtaining new knowledge about the properties of the laser-produced plasma of porous media is a challenging task.In this work,we report,for the first time to the best of our knowledge,the time-dependent measurement of the reflected light of a terawatt laser pulse from the laser-produced plasma of low-Z foam material of overcritical density.The experiments have been performed with the ABC laser,with targets constituted by foam of overcritical density and by solid media of the same chemical composition.We implemented in the MULTI-FM code a model for the light reflection to reproduce and interpret the experimental results.Using the simulations together with the experimental results,we indicate a criterion for estimating the homogenization time of the laser-produced plasma,whose measurement is challenging with direct diagnostic techniques and still not achieved.M.Cipriani S.Yu.Gus’kov F.Consoli R.De Angelis A.A.Rupasov P.Andreoli G.Cristofari G.Di Giorgio M.Salvadori 2021High Power Laser Science and Engineering2021,9,3:2
6Preliminary focus on the mechanical and antibacterial activity of a PMMA-based bone cement loaded with gold nanoparticles显示文摘In total knee arthroplasty(TKA)and total hip replacement(THR)the restoration of the normal joint function represents a fundamental feature.A prosthetic joint must be able to provide motions and to transmit functional loads.As reported in the literature,the stress distribution may be altered in bones after the implantation of a total joint prosthesis.Some scientific works have also correlated uncemented TKA to a progressive decrease of bone density below the tibial component.Antibiotic-loaded bone cements are commonly employed in conjunction with systemic antibiotics to treat infections.Furthermore,nanoparticles with antimicrobial activity have been widely analysed.Accordingly,the current research was focused on a preliminary analysis of the mechanical and antibacterial activity of a PMMA-based bone cement loaded with gold nanoparticles.The obtained results demonstrated that nanocomposite cements with a specific concentration of gold nanoparticles improved the punching performance and antibacterial activity.However,critical aspects were found in the optimization of the nanocomposite bone cement.T.Russo A.Gloria R.De Santis U.D'Amora G.Balato A.Vollaro O.Oliviero G.Improta M.Triassi L.Ambrosio 2017Bioactive Materials2017,2,3:2

8Surface functionalization of acrylic based photocrosslinkable resin for 3D printing applications显示文摘The limited number of resins,available for stereolithography applications,is one of the key drivers in research applied to rapid prototyping.In this work an acrylic photocrosslinkable resin based on methyl methacrylate(MMA),butyl methacrylate(BMA)and poly(ethylene glycol)dimethacrylate(PEGDA)was developed with different composition and characterized in terms of mechanical,thermal and biological behaviour.Two different systems have been developed using different amount of reagent.The influence of every components have been evaluated on the final characteristic of the resin in order to optimize the final composition for applications in bone tissue engineering.The crosslinked materials showed good mechanical properties and thermal stabilities and moreover cytotoxicity test confirms good biocompatibility with no cytotoxic effect on cells metabolism.Moreover two different treatments have been proposed,using fetal bovine serum(FBS)and methanol(MeOH),in order to improve cell recognition of the surfaces.Samples threatened with MeOH allow cell adhesion and survival,promoting spreading,elongation and fusion of C2C12 muscle myoblast cells.A.Ronca F.Maiullari M.Milan V.Pace A.Gloria R.Rizzi R.De Santis L.Ambrosio 2017Bioactive Materials2017,2,3:1

15神经干/祖细胞移植对脑损伤导致的胶质细胞活化有调节作用并有助于移植细胞与宿主胶质细胞间缝隙连接的建立显示文摘移植神经干/祖细胞有助于受损脑组织形态和功能的恢复。移植部位的微环境及移植细胞与宿主细胞间的通讯在神经干/祖细胞移植的神经保护机制中有重要意义。笔者既往研究显示,对中枢轴索损伤动物进行神经干/祖细胞移植有助于恢复损伤神经元的电活动及突触联系,并可增加神经营养因子的释放。本研究拟观察移植的神经干/祖细胞与宿主胶质细胞间的解剖关系,以探讨神经干/祖细胞移植的神经保护作用的可能机制。从新生大鼠的脑室下区提取神经干/祖细胞移植到内侧纵束横断伤模型大鼠脑内。移植后8周取脑进行相关检测。免疫组化检测结果显示,与未移植神经干/祖细胞的大鼠相比,移植了神经干/祖细胞的大鼠脑内更多的小胶质细胞被激活。移植的神经干/祖细胞聚集在激活的小胶质细胞和星形胶质细胞附近。缝隙连接蛋白CX-43表达在损伤部位的神经干/祖细胞和胶质细胞上,且较多的存在于移植细胞和胶质细胞相连接的部位。移植的神经干/祖细胞和宿主胶质细胞及宿主小胶质细胞间均形成了缝隙连接,但移植的神经干/祖细胞和宿主胶质细胞间的缝隙连接较多。本研究结果显示,神经干/祖细胞移植对脑损伤导致的胶质细胞活化有调节作用,并可能通过在移植细胞和宿主细胞间建立缝隙连接来调节细胞间的通讯,这可能是细胞移植的神经保护作用的机制之一。Rocío Talaverón Esperanza R.Matarredona Rosa R.de la Cruz David Macías 2014神经损伤与功能重建2014,9,2:1

19Time-of-flight methodologies with large-area diamond detectors for ion characterization in laser-driven experiments显示文摘The time-of-flight technique coupled with semiconductor detectors is a powerful instrument to provide real-time characterization of ions accelerated because of laser-matter interactions.Nevertheless,the presence of strong electromagnetic pulses(EMPs)generated during the interactions can severely hinder its employment.For this reason,the diagnostic system must be designed to have high EMP shielding.Here we present a new advanced prototype of detector,developed at ENEA-Centro Ricerche Frascati(Italy),with a large-area(15 mm×15 mm)polycrystalline diamond sensor having 150 μm thickness.The tailored detector design and testing ensure high sensitivity and,thanks to the fast temporal response,high-energy resolution of the reconstructed ion spectrum.The detector was offline calibrated and then successfully tested during an experimental campaign carried out at the PHELIX laser facility(E_(L)~100 J,τ_(L)=750 fs,I_(L)(1-2.5)×10^(19)W/cm^(2))at GSI(Germany).The high rejection to EMP fields was demonstrated and suitable calibrated spectra of the accelerated protons were obtained.M.Salvadori G.Di Giorgio M.Cipriani M.Sciscio C.Verona P.L.Andreoli G.Cristofari R.De Angelis M.Pillon N.E.Andreev P.Antici N.G.Borisenko D.Giulietti M.Migliorati O.Rosmej S.Zahter F.Consoli 2022High Power Laser Science and Engineering2022,10,1:1
