| 1 | Targeting LIF/LIFR signaling in cancer显示文摘Leukemia inhibitory factor (LIF), and its receptor (LIFR), are commonly over-expressed in many solid cancers and recent studies have implicated LIF/LIFR axis as a promising clinical target for cancer therapy. LIF/LIFR activate oncogenic signaling pathways including JAK/STAT3 as immediate effectors and MAPK, AKT, mTOR further downstream. LIF/LIFR signaling plays a key role in tumor growth, progression, metastasis, stemness and therapy resistance. Many solid cancers show overexpression of LIF and autocrine stimulation of the LIF/LIFR axis;these are associated with a poorer relapse-free survival. LIF/LIFR signaling also plays a role in modulating multiple immune cell types present in tumor micro environment (TME). Recently, two targeted agents that target LIF (humanized anti-LIF antibody, MSC-1) and LIFR inhibitor (EC359) were under development. Both agents showed effectivity in preclinical models and clinical trials using MSC-1 antibody are in progress. | 2 | Enhancement in antinociceptive and anti-inflammatory effects of tramadol by transdermal proniosome gel显示文摘Oral therapy of tramadol,an opiate analgesic,undergoes extensive hepatic metabolism and requires frequent administration.Transdermal therapy by virtue can overcome these issues and can improve the efficacy and reduce abuse liability of tramadol.The aim of this research was to investigate the possibility of transdermal delivery of tramadol by formulating proniosome gel and evaluate its therapeutic potential in vivo.The effect of formulation composition as well as amount of drug on physicochemical characteristics of prepared proniosomes were examined.Best proniosome gel(F4)was selected and evaluated for drug release,stability and transdermal efficacy by ex vivo and in vivo experiments.The vesicles demonstrated optimal properties including spherical shape,nanosize with good entrapment efficiency,adequate zeta potential,higher stability and greater transdermal flux.The amorphization and dispersion of tramadol in the aqueous core of proniosome vesicles was confirmed by differential scanning calorimeter.Release profile of F4 was distinct(P<0.001)from control and displayed steady and prolonged tramadol release by Fickian diffusion.Transdermal therapy of F4 showed prominent reduction of induced twitches(P<0.005)in mice and edema(P<0.05)in rats,as compared to oral tramadol.The improvement in clinical efficacy of tramadol in transdermal therapy is correlated with the pharmacokinetic data observed.In conclusion,the observed improvement in antinociceptive and anti-inflammatory effects from proniosome carriers signifies its potential to be a suitable alternative to oral therapy of tramadol with greater efficacy. | Jigar Shah Anroop B.Nair Hiral Shah Shery Jacob Tamer M.Shehata Mohamed Aly Morsy | 2020 | Asian Journal of Pharmaceutical Sciences2020,15,6: | 3 |

| 4 | Enhanced cavitation erosion resistance of a friction stir processed high entropy alloy显示文摘Friction stir processing of an Al0.1CoCrFeNi high entropy alloy(HEA)was performed at controlled cooling conditions(ambient and liquid submerged).Microstructural and mechanical characterization of the processed and as-cast HEAs was evaluated using electron backscatter diffraction,micro-hardness testing and nanoindentation.HEA under the submerged cooling condition showed elongated grains(10μm)with fine equiaxed grains(2μm)along the boundary compared to the coarser grain(~2 mm)of as-cast HEA.The hardness showed remarkable improvements with four(submerged cooling condition)and three(ambient cooling condition)times that of as-cast HEA(HV^150).The enhanced hardness is attributed to the significant grain refinement in the processed HEAs.Cavitation erosion behavior was observed for samples using an ultrasonication method.All of the HEAs showed better cavitation erosion resistance than the stainless steel 316L.The sample processed under a submerged liquid condition showed approximately 20 and 2 times greater erosion resistance than stainless steel 316L and ascast HEA,respectively.The enhanced erosion resistances of the processed HEAs correlate to their increased hardness,resistance to plasticity,and better yield strength than the as-cast HEA.The surface of the tested samples showed nucleation and pit growth,and plastic deformation of the material followed by fatigue-controlled disintegration as the primary material removal mechanism. | Rakesh B.Nair H.S.Arora Harpreet Singh Grewal | 2020 | International Journal of Minerals,Metallurgy and Materials2020,27,10: | 2 |
| 5 | Optical parameters induced by phase transformation in RF magnetron sputtered TiO_2 nanostructured thin films显示文摘Pure TiO2 thin films were deposited onto quartz substrates using a ceramic TiO2 target at an elevated substrate temperature of 573 K by RF magnetron sputtering, and an analysis of structural, optical and photoluminescence characteristics of the films upon phase transformation is reported in this paper. Structural investigations using X-ray diffraction revealed that the as-deposited film was amorphous in nature. Thermal annealing for 2 h at 873 K in air resulted in the formation of anatase phase, and a phase transformation to rutile was observed at 1073 K.An increase in grain size and an improvement in crystallinity were also observed on annealing. Rod- like rutile crystallites were observed in the SEM images of the film annealed at 1273 K. As-deposited films and films annealed up to 1073 K were highly transparent in the visible region with a transparency 480%. Optical band gap of the films decreased upon thermal annealing which is attributed to phase transformation from amorphous to anatase and then to rutile. Optical parameters such as refractive index, optical conductivity and optical dielectric constant increased with increase in annealing temperature. Since rutile is the optically active phase, the superior refractive index of the film annealed at 1073 K along with its high transparency in visible region suggests the application of this film in antireflective coatings. Photoluminescence emission of maximum intensity was observed for the film annealed at 873 K, which exhibits anatase phase. | 7 | Machine learned interatomic potentials using random features显示文摘We present a method to model interatomic interactions such as energy and forces in a computationally efficient way.The proposed model approximates the energy/forces using a linear combination of random features,thereby enabling fast parameter estimation by solving a linear least-squares problem.We discuss how random features based on stationary and non-stationary kernels can be used for energy approximation and provide results for three classes of materials,namely two-dimensional materials,metals and semiconductors.Force and energy predictions made using the proposed method are in close agreement with density functional theory calculations,with training time that is 96%lower than standard kernel models.Molecular Dynamics calculations using random features based interatomic potentials are shown to agree well with experimental and density functional theory values.Phonon frequencies as computed by random features based interatomic potentials are within 0.1%of the density functional theory results.Furthermore,the proposed random features-based potential addresses scalability issues encountered in this class of machine learning problems. | Gurjot Dhaliwal Prasanth B.Nair Chandra Veer Singh | 2022 | npj Computational Materials2022,,1: | 0 |
