| 1 | Cytotoxic and antibacterial substances against multi-drug resistant pathogens from marine sponge symbiont:Citrinin,a secondary metabolite of Penicillium sp.显示文摘Objective:To Isolate,purify,characterize,and evaluate the bioaclive compounds from the sponge-derived fungus Penicillium sp.FF001 and to elucidate its structure.Methods:The fungal strain FF001 with an interesting bioactivity profile was isolated from a marine Fijian sponge Melophlus sp.Based on conidiophores aggregation,conidia development and mycelia morphological characteristics,the isolate FF001 was classically identified as a Penicillium sp.The bioactive compound was identified using various spectral analysis of UV,high resolution electrospray ionization mass spectra,1H and 13C NMR spectral data.Further minimum inhibitory concentrations(MICs)assay and brine shrimp cytotoxicity assay were also carried out to evaluate the biological properties of the purified compound.Results:Bioassay guided fractionation of the EtOAc extract of a static culture of this Penicillium sp.by different chromatographic methods led the isolation of an antibacterial,anticryptococcal and cytotoxic active compound,which was identified as citrinin(1).Further,citrinin(1)is reported for its potent antibacterial activity against methicillin-resistant Staphylococcus aureus(S.aureus),rifampicin-resistant 5.aureus,wild type S.aureus and vancomycin-resistant Enterococcus faecium showed MICs of 3.90,0.97,1.95 and7.81μg/mL,respectively.Further citrinin(1)displayed significant activity against the pathogenic yeast Cryptococcus neoformans(MIC 3.90μg/mL),and exhibited cytotoxicity against brine shrimp larvae LD_(50)of 96μg/mL.Conclusions:Citrinin(1)is reported from sponge associated Penicillium sp.from this study and for its strong antibacterial activity against multi-drug resistant human pathogens including cytotoxicity against brine shrimp larvae,which indicated that sponge associated Penicillium spp.are promising sources of natural bioactive metabolites. | Ramesh Subramani Rohitesh Kumar Pritesh Prasad William Aalbersberg | 2013 | Asian Pacific Journal of Tropical Biomedicine2013,3,4: | 6 |
| 2 | Flexible high energy density zinc-ion batteries enabled by binder-free MnO_(2)/reduced graphene oxide electrode显示文摘We demonstrate a rechargeable zinc-ion battery with high energy density and cyclability using MnO_(2)and reduced graphene oxide(MnO_(2)/rGO)electrode.The flexible and binder free electrode,with high MnO_(2)mass ratio(80 wt%of MnO_(2)),is fabricated using vacuum filtration without any additional additives other than rGO.Compared to batteries with conventional MnO_(2)electrodes,the Zn-MnO_(2)/rGO battery shows a significant enhanced capacity(332.2 mAh g^(-1)at 0.3 A g^(-1)),improved rate capability(172.3 mAh g^(-1)at 6 A g^(-1))and cyclability.The capacity retention remains 96%after 500 charge/discharge cycles at 6 A g^(-1).The high MnO_(2)mass ratio makes MnO_(2)/rGO electrode advantageous when the capacity is normalized to the whole electrode,particularly at high rates.The calculated gravimetric energy density of Zn-MnO_(2)/rGO battery is 33.17Wh kg^(-1),which is comparable to the existing commercial lead-acid batteries(30-40Wh kg^(-1)).Furthermore,the discharge profile and capacity of our Zn-MnO_(2)/rGO battery shows no deterioration during bending test,indicating good flexibility.As a result,zinc-ion battery is believed to be a promising technology for powering next generation flexible electronics. | Yuan Huang Jiuwei Liu Qiyao Huang Zijian Zheng Pritesh Hiralal Fulin Zheng Dilek Ozgit Sikai Su Shuming Chen Ping-Heng Tan Shengdong Zhang Hang Zhou | 2018 | npj Flexible Electronics2018,2,1: | 4 |
| 3 | Development of a sensitive and rapid method for quantitation of (S)-(-)- and (R)-(+)-metoprolol in human plasma by chiral LC–ESI–MS/MS显示文摘A selective, sensitive and high throughput liquid chromatography-tandem mass spectrometry(LC–ESI–MS/MS) method has been developed for separation and quantification of metoprolol enantiomers on a chiral Lux Amylose-2(250 mm 4.6 mm, 5 mm) column. Solid phase extraction of(S)-()- and(R)-(t)-metoprolol and rac-metoprolol-d6 as an internal standard(IS)was achieved on Lichrosep DVB HL cartridges employing 200 mL human plasma. Both the analytes were chromatographically separated with a resolution factor of 2.24 using 15 mM ammonium acetate in water, pH 5.0 and 0.1%(v/v) diethyl amine in acetonitrile(50:50, v/v) as the mobile phase within 7.0 min. The precursor-product ion transitions for the enantiomers and IS were monitored in the multiple reaction monitoring and positive ionization mode. The method was validated over the concentration range of 0.500–500 ng/mL for both the enantiomers. Matrix effect was assessed by post-column analyte infusion experiment and the mean extraction recovery was greater than 94.0% for both the enantiomers at all quality control levels. The stability of analytes was evaluated in plasma and whole blood under different storage conditions. The method was successfully applied to a clinical study in 14 healthy volunteers after oral administration of200 mg metoprolol tablet under fasting conditions. | 4 | Characterization of colloidal arsenic at two abandoned gold mine sites in Nova Scotia, Canada, using asymmetric flow-field flow fractionation-inductively coupled plasma mass spectrometry显示文摘Asymmetric flow-field flow fractionation-inductively-coupled plasma-mass spectrometry was used to determine whether colloidal arsenic(As) exists in soil pore water and soil extract samples at two arsenic-contaminated abandoned gold mines(Montague and Goldenville, Nova Scotia). Colloidal arsenic was found in 12 out of the 80 collected samples(= 15%), and was primarily associated with iron(Fe) in the encountered colloids. The molar Fe/As ratios indicate that the colloids in some samples appeared to be discrete iron–arsenic minerals, whereas in other samples, they were more consistent with As-rich iron(oxy)hydroxides. Up to three discrete size fractions of colloidal As were encountered in the samples, with mean colloid diameters between 6 and 14 nm. | 6 | Antifertility effect of chronically administered Tabernaemontana divaricata leaf extract on male rats显示文摘Objective:To investigate the antifertility effect of chronically administered Tabernaemontana divarkala(T.divaricata) leaf extract on male rats.Methods:The effect of 50%ethanol extract of T.divaricata leaves on reproduction was studied on male rats.The study was divided into four groups of five animals each.The first groups(1) received vehicle alone to serve as control. The second,third and fourth groups(Ⅱ,ⅡandⅣ) of animals were administered the leaf extract daily at 50 mg/kg body weight,p.o.,100 nig/kg body weight,p.o.,and 200 mg/kg body weight, p.o.,respectively,for a period of 60 days.Results:Significant decreases in the weight of testes, epididymis,seminal vesicle and ventral prostate were observed.A dose related reduction in the testicular sperm count,epididymal sperm count and motility,number of fertile male,ratio between delivered and inseminated females and numbers of pups were observed.The testis showed a clear correlation between the dose and severity of lesions of seminiferous epithelium. In general,the seminiferous tubules appear reduced in size with a frequently filled eosinophilic material.Spermatogenesis arrested at the secondary spermatocyte stage.Pachytene spermatocytes were undergoing degeneration.Disorganisation and sloughing of immature germ cell were visible. Leydinf cells were atrophied.No morphological changes were observed in Sertoli cells.Significant reduction in serum concentration of luteinizing hormone and testosterone were observed.No distinct change in serum FSH concentration was recorded.The final body weights of all groups were elevated markedly.No alterations were recorded in any hematologiocal parameters. | 13 | Withania somnifera (Ashwagandha):a Novel Source of L-asparaginase显示文摘Different parts of plant species belonging to Solanaceae and Fabaceae families were screened for L-asparaginase enzyme (E.C.3.5.1.1.). Among 34 plant species screened for L-asparaginase enzyme, Withania somnifera L. was identified as a potential source of the enzyme on the basis of high specific activity of the enzyme. The enzyme was purified and characterized from W. somnifera, a popular medicinal plant in South East Asia and Southern Europe. Purification was carried out by a combination of protein precipitation with ammonium sulfate as well as Sephadex-gel filtration. The purified enzyme is a homodimer, with a molecular mass of 72 ± 0.5 kDa as estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresisand size exclusion chromatography. The enzyme has a pH optimum of 8.5 and an optimum temperature of 37-C. The Km value for the enzyme is 6.1 × 10-2 mmol/L. After liquid-liquid extraction with n-hexane, ergocalciferol was derivatized by reacting with 4-phenyl-1,2,4-triazoline-3,5-dione(PTAD), a strong dienophile based on Diels-Alder reaction.Ergocalciferol and its deuterated internal standard, ergocalciferol-d6, were analyzed on X Select CSH C_(18)(100 mm×4.6 mm, 2.5 μm) column using acetonitrile and 0.1%(v/v) formic acid in water containing 0.14% methylamine within 6.0 min under gradient elution mode. Tandem mass spectrometry in positive ionization mode was used to quantify ergocalciferol by multiple reaction monitoring(MRM). Entire data processing was done using Watson LIMS^(TM) software which provided excellent data integrity and high throughput with improved operational efficiency. The major advantage of this method includes higher sensitivity(0.10 ng/mL),superior extraction efficiency(≥83%) and small sample volume(100 μL) for processing. The method was linear in the concentration range of 0.10–100 ng/mL for ergocalciferol. The intra-batch and inter-batch accuracy and precision(% CV) values varied from 97.3% to 109.0% and 1.01% to 5.16%, respectively.
