1Marine Bacterial Biofilms in Bioremediation of Polycyclic Aromatic Hydrocarbons(PAHs) Under Terrestrial Condition in a Soil Microcosm显示文摘Polycyclic aromatic hydrocarbons(PAHs) in soil retain for a quite long period due to their hydrophobicity and aggregation properties. Biofilm-forming marine bacterial consortium(named as NCPR), composed of Stenotrophomonas acidaminiphila NCW702,Alcaligenes faecalis NCW402, Pseudomonas mendocina NR802, Pseudomonas aeruginosa N6P6, and Pseudomonas pseudoalcaligenes NP103, was used for the bioremediation of PAHs in a soil microcosm. Phenanthrene and pyrene were used as reference PAHs. Parameters that can affect PAH degradation, such as chemotaxis, solubility of PAHs in extracellular polymeric substances(EPS), and catechol2,3-dioxygenase(C23O) activity, were evaluated. P. aeruginosa N6P6 and P. pseudoalcaligenes NP103 showed chemotactic movement towards both the reference PAHs. The solubility of both the PAHs was increased with an increase in EPS concentration(extracted from all the 5 selected isolates). Significantly(P < 0.001) high phenanthrene(70.29%) and pyrene(55.54%) degradation was observed in the bioaugmented soil microcosm. The C23O enzyme activity was significantly(P < 0.05) higher in the bioaugmented soil microcosm with phenanthrene added at 173.26 ± 2.06 nmol min^(-1) mg^(-1) protein than with pyrene added at 61.80 ± 2.20 nmol min^(-1) mg^(-1) protein. The C23O activity and gas chromatography-mass spectrometer analyses indicated catechol pathway of phenanthrene metabolism. 2Anti-inflammatory effects of traditional mixed extract of medicinal herbs(MEMH)on monosodium urate crystal-induced gouty arthritis显示文摘Korean oriental medicine prescription is widely used for the treatment of gouty diseases. In the present study, we investigated anti-inflammatory effects of modified Korean herbal formulation, mixed extract of medicinal herbs(MEMH), and its modulatory effects on inflammatory mediators associated with gouty arthritis. Both in vitro and in vivo studies were carried out to assess the anti-inflammatory efficacy of MEMH on monosodium urate(MSU) crystals-induced gouty inflammation. MSU crystals stimulated human chondrosarcoma cell line, SW1353, and human primary chondrocytes were treated with MEMH in vitro. The expression levels of pro-inflammatory mediators and metalloproteases were analyzed. The effect of MEMH on NFκB signaling pathway in SW1353 cells was examined. Effect of MEMH on the mR NA expression level of pro-inflammatory mediators and chemotactic factor from human monocytic cell line, THP-1, was also analyzed. The probable role of MEMH in the differentiation process of osteoblast like cells, SaO S-2, after MSU treatment was also observed. To investigate the effects of MEMH in vivo, MSU crystals-induced ankle arthritic model was established. Histopathological changes in affected joints and plasma levels of pro-inflammatory mediators(IL-1β and TNFα) were recorded. MEMH inhibited NFκB signaling pathway and COX-2 protein expression in chondrocytes. MSU-induced mR NA expressions of pro-inflammatory mediators and chemotactic cytokines were suppressed by MEMH. In MSU crystals-induced ankle arthritic mouse model, administration of MEMH relieved inflammatory symptoms and decreased the plasma levels of IL-1β and TNFα. 5Role of prebiotics, probiotics, and synbiotics in management of inflammatory bowel disease: Current perspectives显示文摘Experimental evidence supports the fact that changes in the bowel microflora due to environmental or dietary factors have been investigated as implicating factors in the etiopathogenesis of inflammatory bowel disease(IBD).The amassing knowledge that the inhabited microbiome regulates the gut physiology and immune functions in IBD,has led researchers to explore the effectiveness of prebiotics,probiotics,and synbiotics in treating IBD.This therapeutic approach focuses on restoring the dynamic balance between the microflora and host defense mechanisms in the intestinal mucosa to prevent the onset and persistence of intestinal inflammation.Numerous microbial strains and carbohydrate blends,along with their combinations have been examined in experimental colitis models and clinical trials,and the results indicated that it can be an attractive therapeutic strategy for the suppression of inflammation,remission induction,and relapse prevention in IBD with minimal side effects.Several mechanisms of action of probiotics(for e.g.,Lactobacillus species,and Bifidobacterium species)have been reported such as suppression of pathogen growth by releasing certain antimicrobial mediators(lactic and hydrogen peroxide,acetic acid,and bacteriocins),immunomodulation and initiation of an immune response,enhancement of barrier activity,and suppression of human T-cell proliferation.Prebiotics such as lactulose,lactosucrose,oligofructose,and inulin have been found to induce the growth of certain types of host microflora,resulting in an enriched enteric function.These non-digestible food dietary components have been reported to exert anti-inflammatory effects by inhibiting the expression of tumor necrosis factor-α-related cytokines while augmenting interleukin-10 levels.Although proand prebiotics has established their efficacy in healthy subjects,a better understanding of the luminal ecosystem is required to determine which specific bacterial strain or combination of probiotics and prebiotics would prove to be the ideal treatment for IBD.Clinical trials,however,have given some conflicting results,requiring the necessity to cite the more profound clinical effect of these treatments on IBD remission and prevention.The purpose of this review article is to provide the most comprehensive and updated review on the utility of prebiotics,probiotics,and synbiotics in the management of active Crohn’s disease and ulcerative colitis/pouchitis.Supriya Roy Suneela Dhaneshwar 2023World Journal of Gastroenterology2023,29,14:4

8Molt and reproduction enhancement together with hemolymph ecdysteroid elevation under eyestalk ablation in the female fiddler crab,Uca triangularis(Brachyura:Decapoda)显示文摘We present the results of eyestalk extirpation experiments performed on the fiddler crab, Uca triangularis at seasons of molting and reproduction, with a view to have a better understanding of the regulatory mechanisms of these two highly energy-demanding processes. Bilateral eyestalk ablation resulted in precocious acceleration of both molting and reproduction, irrespective of the season at which each experiment was conducted. The rate of accelerated ovarian growth, however, was maximum if the eyestalk ablation was conducted during August–January, the breeding season in the wild, or in February–May(molting-reproductive season), wherein a section of the wild population would be engaged in molting and another section in breeding. The highest degree of precocious molt acceleration, on the other hand, was obtained during June–July when the population was primarily engaged in molting, but with no reproductive activity. The precocious oocyte maturation(due to de-eyestalking) was minimal in June–July. Significantly, the eyestalk ablation also resulted in a dramatic increase in the hemolymph ecdysteroid titer, revealing that a high ecdysteroid titer would have no restraining influence on vitellogenesis. No spawning was, however, observed among de-eyestalked females, even though their final oocyte size surpassed the size of the normal mature oocytes, implying that spawning is not exclusively under the control of eyestalk hormones. A comparative study performed on the vitelline components of the experimentals and the controls revealed that the precociously incorporated yolk under eyestalk ablation was biochemically impoverished. These results indicate that throughout the annual cycle, both the somatic and the reproductive growth of U. triangularis are under the influence of inhibitory principles from the eyestalks. It is also revealing that mere deprival of the inhibitory principles does not culminate into successful vitellogenesis. 12Primary pediatric mid-brain lymphoma:Report of a rare pediatric tumor in a rare location显示文摘Primary central nervous system lymphoma(PCNSL) is a rare disease in pediatric age group. A thirteen-yearold male child presented with complaints of headache for six months, vomiting and diplopia for three days. Magnetic resonance imaging of the brain showed a single lesion of 1.7 cm × 1.6 cm × 1.6 cm in the mid brain and tectum. He underwent a gross total resection of the tumor. The histopathological evaluation revealed B cell high grade non Hodgkin lymphoma. The patient was treated with High dose methotrexate and cranio spinal radiation. The patient was alive without disease 12 mo after completion of treatment. This case highlights importance of keeping PCNSL as differential in brain stem lesions of pediatric patients also.
