| 1 | Anti-inflammatory pathways and alcoholic liver disease: Role of an adiponectin/interleukin-10/heme oxygenase-1 pathway显示文摘The development of alcoholic liver disease (ALD) is a complex process involving both the parenchymal and non-parenchymal cells in the liver. Enhanced inflammation in the liver during ethanol exposure is an important contributor to injury. Kupffer cells, the resident macrophages in liver, are particularly critical to the onset of ethanol-induced liver injury. Chronic ethanol exposure sensitizes Kupffer cells to activation by lipopolysaccharide via Toll-like receptor 4. This sensitization enhances production of inflammatory mediators, such as tumor necrosis factor-α and reactive oxygen species, that contribute to hepatocyte dysfunction, necrosis, apoptosis, and fibrosis. Impaired resolution of the inflammatory process probably also contributes to ALD. The resolution of inflammation is an active, highly coordinated response that can potentially be manipulated via therapeutic interventions to treat chronic inflammatory diseases. Recent studies have identif ied an adiponectin/interleukin-10/heme oxygenase-1 (HO-1) pathway that is profoundly effective in dampening the enhanced activation of innate immune responses in primary cultures of Kupffer cells, as well as in an in vivo mouse model of chronic ethanol feeding. Importantly, induction of HO-1 also reduces ethanol-induced hepatocellular apoptosis in this in vivo model. | 3 | Critical role of OX40 signaling in the TCR-independent phase of human and murine thymic Treg generation显示文摘Regulatory T cells(Tregs)play a pivotal role in immune-tolerance,and loss of Treg function can lead to the development of autoimmunity.Natural Tregs generated in the thymus substantially contribute to the Treg pool in the periphery,where they suppress self-reactive effector T cells(Teff)responses.Recently,we showed that OX40L(TNFSF4)is able to drive selective proliferation of peripheral Tregs independent of canonical antigen presentation(CAP-independent)in the presence of low-dose IL-2.Therefore,we hypothesized that OX40 signaling might be integral to the TCR-independent phase of murine and human thymic Treg(tTreg)development.Development of tTregs is a two-step process:Strong T-cell receptor(TCR)signals in combination with co-signals from the TNFRSF members facilitate tTreg precursor selection,followed by a TCR-independent phase of tTreg development in which their maturation is driven by IL-2.Therefore,we investigated whether OX40 signaling could also play a critical role in the TCR-independent phase of tTreg development.OX40−/−mice had significantly reduced numbers of CD25−Foxp3low tTreg precursors and CD25+Foxp3+mature tTregs,while OX40L treatment of WT mice induced significant proliferation of these cell subsets.Relative to tTeff cells,OX40 was expressed at higher levels in both murine and human tTreg precursors and mature tTregs.In ex vivo cultures,OX40L increased tTreg maturation and induced CAP-independent proliferation of both murine and human tTregs,which was mediated through the activation of AKT-mTOR signaling.These novel findings show an evolutionarily conserved role for OX40 signaling in tTreg development and proliferation,and might enable the development of novel strategies to increase Tregs and suppress autoimmunity. | Prabhakaran Kumar Alejandra Marinelarena Divya Raghunathan Vandhana K Ragothaman Shikha Saini Palash Bhattacharya Jilao Fan Alan L Epstein Ajay V Maker Bellur S Prabhakar | 2019 | Cellular & Molecular Immunology2019,16,2: | 4 |
| 4 | Application of artificial intelligence to forecast hydrocarbon production from shales显示文摘Artificial intelligence(AI)methods and applications have recently gained a great deal of attention in many areas,including fields of mathematics,neuroscience,economics,engineering,linguistics,gaming,and many others.This is due to the surge of innovative and sophisticated AI techniques applications to highly complex problems as well as the powerful new developments in high speed computing.Various applications of AI in everyday life include machine learning,pattern recognition,robotics,data processing and analysis,etc.The oil and gas industry is not behind either,in fact,AI techniques have recently been applied to estimate PVT properties,optimize production,predict recoverable hydrocarbons,optimize well placement using pattern recognition,optimize hydraulic fracture design,and to aid in reservoir characterization efforts.In this study,three different AI models are trained and used to forecast hydrocarbon production from hydraulically fractured wells.Two vastly used artificial intelligence methods,namely the Least Square Support Vector Machine(LSSVM)and the Artificial Neural Networks(ANN),are compared to a traditional curve fitting method known as Response Surface Model(RSM)using second order polynomial equations to determine production from shales.The objective of this work is to further explore the potential of AI in the oil and gas industry.Eight parameters are considered as input factors to build the model:reservoir permeability,initial dissolved gas-oil ratio,rock compressibility,gas relative permeability,slope of gas oil ratio,initial reservoir pressure,flowing bottom hole pressure,and hydraulic fracture spacing.The range of values used for these parameters resemble real field scenarios from prolific shale plays such as the Eagle Ford,Bakken,and the Niobrara in the United States.Production data consists of oil recovery factor and produced gas-oil ratio(GOR)generated from a generic hydraulically fractured reservoir model using a commercial simulator.The Box-Behnken experiment design was used to minimize the number of simulations for this study.Five time-based models(for production periods of 90 days,1 year,5 years,10 years,and 15 years)and one rate-based model(when oil rate drops to 5 bbl/day/fracture)were considered.Particle Swarm Optimization(PSO)routine is used in all three surrogate models to obtain the associated model parameters.Models were trained using 80%of all data generated through simulation while 20%was used for testing of the models.All models were evaluated by measuring the goodness of fit through the coefficient of determination(R2)and the Normalized Root Mean Square Error(NRMSE).Results show that RSM and LSSVM have very accurate oil recovery forecasting capabilities while LSSVM shows the best performance for complex GOR behavior.Furthermore,all surrogate models are shown to serve as reliable proxy reservoir models useful for fast fluid recovery forecasts and sensitivity analyses. | Palash Panja Raul Velasco Manas Pathak Milind Deo | 2018 | Petroleum2018,4,1: | 4 |
| 5 | Comparative metabolomic analysis of wild type and mads3 mutant rice anthers显示文摘Rice(Oryza sativa L.) MADS3 transcription factor regulates the homeostasis of reactive oxygen species(ROS)during late anther development,and one MADS3 mutant,mads3-4,has defective anther walls,aborted microspores and complete male sterility.Here,we report the untargeted metabolomic analysis of both wild type and mads3-4 mature anthers.Mutation of MADS3 led to an unbalanced redox status and caused oxidative stress that damages lipid,protein,and DNA.To cope with oxidative stress in mads3-4 anthers,soluble sugars were mobilized and carbohydrate metabolism was shifted to amino acid and nucleic acid metabolism to provide substrates for the biosynthesis of antioxidant proteins and the repair of DNA.Mutation of MADS3 also affected other aspects of rice anther development such as secondary metabolites associated with cuticle,cell wall,and auxin metabolism.Many of the discovered metabolic changes in mads3-4 anthers were corroborated with changes of expression levels of corresponding metabolic pathway genes.Altogether,this comparative metabolomic analysis indicated that MADS3 gene affects rice anther development far beyond the ROS homeostasis regulation. | Guorun Qu Sheng Quan Palash Mondol Jie Xu Dabing Zhang Jianxin Shi | 2014 | Journal of Integrative Plant Biology2014,56,9: | 3 |
