| 2 | PCR analysis of Yq microdeletions in infertile males, a study from South India显示文摘Aim:To estimate the frequency of microdeletions in the long arm of Y-chromosome of 20 infertile males from South India.Methods:Polymerase chain reaction (PCR) amplification using Y-specific STS of azoospermia factor (AZF) regions i.e.,SY 84 for AZFa,SY 127 for AZFb and SY 254 for AZFc.Results:Of the 20 infertile subjects 3(15%),one azoospermic and two oligozoospermic,showed microdeletions in the AZF region of Y-chromosome.Conclusion:The frequency of deletions involving AZF region of the Y-chromosome is 15% in azoospermic and severely oligozoospermic infertile men.PCR amplification of AZF locus is useful for the diagnosis of microdeletions in the Y-chromosome. | S. | 3 | Genotoxic and tumorigenic pyrrolizidine alkaloids in Chinese herbal plants显示文摘Pyrrolizidine alkaloids are a class of hepatotoxic and tumorigenic compounds detected in Chinese herbal plants, contaminated foods, and dietary supplements. In this review, the sources, toxicity, genotoxicity, tumorigenicity, and the metabolic pathways, particular the activation pathways leading to hepatotoxicity and tumorigenicity, of pyrrolizidine alkaloids are briefly discussed, with a focus on the most recent important findings concerning the genotoxic mechanism by which riddelliine liver tumors. This mechanism involves the formation of 6,7-dihydro-7-hydroxy-1-hydroxymethyl-5H-pyrrolizine (DHP)-derived DNA adducts and may be general to most carcinogenic pyrrolizidine alkaloids. | P.P. Fu Q. | 5 | Hesperidin as a preventive resistance agent in MCF-7 breast cancer cells line resistance to doxorubicin显示文摘Objective:To evaluate of hesperidin to overcome resistance of doxorubicin in MCF-7 resistant doxorubicin cells(MCF-7/Dox)in cytotoxicity apoptosis and P-glycoprotein(Pgp)expression in combination with doxorubicin.Methods:The cytotoxic properties.50%inhibition concentration(IC_(50))and its combination with doxorubicin in MCF-7 cell lines resistant to doxorubicin(MCF-7/Dox)cells were determined using MTT assay.Apoptosis induction was examined by double staining assay using ethidium bromide-acridine orange.Immunocytochemistry assay was performed to determine the level and localization of Pgp.Results:Single treatment of hesperidin showed cytotoxic activity on MCF-7/Dox cells with IC_(50)value of 11μmol/L.Thus,combination treatment from hesperidin and doxorubicin showed addictive and antagonist effect(CI>1.0).Hesperidin did not increase the apoptotic induction,but decreased the Pgp expressions level when combined with doxorubicin in low concentration.Conclusions:Hesperidin has cytotoxic effect on MCF-7/Dox cells with IC_(50)of 11μmol/L.Hesperidin did not increased the apoptotic induction combined with doxorubicin.Cochemotherapy application of doxorubicin and hesperidin on MCF-7/Dox cells showed synergism effect through inhibition of Pgp expression. | Rifki Febriansah Dyaningtyas Dewi P.P.
