| 1 | Is it time to replace propranolol with carvedilol for portal hypertension?显示文摘Beta-adrenergic receptor antagonists(β-blockers) have been well established for use in portal hypertension for more than three decades. Different Non-selective β-blockers like propranolol, nadolol, timolol, atenolol, metoprolol and carvedilol have been in clinical practice in patients with cirrhosis. Carvedilol has proven 2-4 times more potent than propranolol as a beta-receptor blocker in trials conducted testing its efficacy for heart failure. Whether the same effect extends to its potency in the reduction of portal venous pressures is a topic of on-going debate. The aim of this review is to compare the hemodynamic and clinical effects of carvedilol with propranolol, and attempt assess whether carvedilol can be used instead of propranolol in patients with cirrhosis. Carvedilol is a promising agent among the beta blockers of recent time that has shown significant effects in portal hypertension hemodynamics. It has also demonstrated an effective profile in its clinical application specifically for the prevention of variceal bleeding. Carvedilol has more potent desired physiological effects when compared to Propranolol. However, it is uncertain at the present juncture whether the improvement in hemodynamics also translates into a decreased rate of disease progression and complications when compared to propranolol. | 3 | DNNGP, a deep neural network-based method for genomic prediction using multi-omics data in plants显示文摘Genomic prediction is an effective way to accelerate the rate of agronomic trait improvement in plants.Traditional methods typically use linear regression models with clear assumptions;such methods are unable to capture the complex relationships between genotypes and phenotypes.Non-linear models(e.g.,deep neural networks)have been proposed as a superior alternative to linear models because they can capture complex non-additive effects.Here we introduce a deep learning(DL)method,deep neural network genomic prediction(DNNGP),for integration of multi-omics data in plants.We trained DNNGP on four datasets and compared its performance with methods built with five classic models:genomic best linear unbiased prediction(GBLUP);two methods based on a machine learning(ML)framework,light gradient boosting machine(LightGBM)and support vector regression(SVR);and two methods based on a DL framework,deep learning genomic selection(DeepGS)and deep learning genome-wide association study(DLGWAS).DNNGP is novel in five ways.First,it can be applied to a variety of omics data to predict phenotypes.Second,the multilayered hierarchical structure of DNNGP dynamically learns features from raw data,avoiding overfitting and improving the convergence rate using a batch normalization layer and early stopping and rectified linear activation(rectified linear unit)functions.Third,when small datasets were used,DNNGP produced results that are competitive with results from the other five methods,showing greater prediction accuracy than the other methods when large-scale breeding data were used.Fourth,the computation time required by DNNGP was comparable with that of commonly used methods,up to 10 times faster than DeepGS.Fifth,hyperparameters can easily be batch tuned on a local machine.Compared with GBLUP,LightGBM,SVR,DeepGS and DLGWAS,DNNGP is superior to these existing widely used genomic selection(GS)methods.Moreover,DNNGP can generate robust assessments from diverse datasets,including omics data,and quickly incorporate complex and large datasets into usable models,making it a promising and practical approach for straightforward integration into existing GS platforms. | Kelin Wang Muhammad Ali Abid Awais Rasheed Jose Crossa Sarah Hearne Huihui Li | 2023 | Molecular Plant2023,16,1: | 3 |
| 4 | Why more research needs to be done on echinococcosis in Pakistan显示文摘Background:Echinococcosis has a worldwide geographical distribution with endemic foci on every inhabited continent.Due to the frequent outbreaks in different parts of Pakistan in the recent past,echinococcosis is being described as a neglected tropical disease and is considered one of the most neglected parasitic diseases in the country.In endemic regions,predominantly settings with limited resources,there are high numbers of echinococcosis patients,as these communities do not have access to appropriate treatment.In Pakistan,there are limited reports on echinococcosis.The disease is prevalent in human and livestock,but this has not been sufficiently explored yet.Pakistan is an agricultural country and due to the disease’s zoonotic mode of transmission,there is a dire need of future research on it.The present paper is an effort to highlight the importance of echinococcosis in Pakistan.Discussion:There is a dire need for future research on echinococcosis in Pakistan as very few investigations had been carried out on this topic thus far.The prevalence of the disease in neighbouring countries highlights that Pakistan might be at severe risk of this zoonotic infection and further supports the need for more research.In Pakistan,the majority of the population lives in rural areas with limited acess to proper hygienic/sanitary facilities.These conditions favour the outbreak of diseases such as echinococcosis.The limited available data could result in higher outbreaks in the future,and thus cause the already weak healthcare system to overburden.The country has a meagre annual budget for health,which is being spent on known infections such as polio,dengue fever and hepatic viral infections.A proper surveillance system for echinococcosis is required across the country as treatment is usually expensive,complicated and may require extensive surgery and/or prolonged drug therapy.Development of new/novel drugs and other treatment modalities receives very little,if any,attention.Prognostic awareness programmes against this infection involve deworming of the infected animals,improved food inspection and slaughterhouse hygiene,and public education campaigns.Conclusion:Future research on echinococcosis is anticipated to demonstrate whether the epidemiology,diagnosis and recombinant vaccines/antibodies relating to echinococcosis can meet the quality standards(purity,potency,safety and efficacy)defined by the World Health Organization.Research work should be carried out on the epidemiology and serodiagnosis of echinocossis in the different areas of Pakistan,which will be useful for the proper eradication of echinococcosis in this region.The health department should implement awareness-raising campaigns for the general public in order to reduce the burden of disease. | Haroon Ahmed Shahzad Ali Muhammad Sohail Afzal Abid Ali Khan Hamid Raza Zaheer Hussain Shah Sami Simsek | 2017 | Infectious Diseases of Poverty2017,6,1: | 2 |
| 5 | Development of bis-thiobarbiturates as successful urease inhibitors and their molecular modeling studies显示文摘Bis-thiobarbiturate derivatives 1–15 have been synthesized, characterized by1 HNMR and EI-MS and screened for urease inhibition. All compounds showed various degree of urease inhibitory activity with IC_(50) values ranging 7.45 0.12 74.24 0.81 mmol/L while the standard thiourea behaved normally(IC_(50) = 21.10 0.12). Compounds 1(IC_(50) = 7.45 0.12 μmol/L), 9(IC_(50) = 18.17 1.03 μmol/L) and 13(IC_(50) = 8.61 0.45 μmol/L) showed excellent urease inhibitory activity in the series. Molecular modeling studies were performed to understand the binding site with the bimetallic nickel center of the enzyme.Structure-activity relationship has also been established for these compounds. | 7 | Genetic variation in LBL1 contributes to depth of leaf blades lobes between cotton subspecies, Gossypium barbadense and Gossypium hirsutum显示文摘Leaf is a essential part of the plants for photosynthetic activities which mainly economize the resources for boll heath. Significant variations of leaf shapes across the Gossypium sp. considerably influence the infiltration of sunlight for photosynthesis. To understand the genetic variants and molecular processes underlying for cotton leaf shape, we used F_2 population derived from upland cotton genotype P30 A(shallow-lobed leaf) and sea-island cotton genotype ISR(deeplobed leaf) to map leaf deep lobed phenotype controlling genes LBL1 and LBL2. Genetic analysis and localization results have unmasked the position and interaction between both loci of LBL1 and LBL2, and revealed the co-dominance impact of the genes in regulating depth of leaf blades lobes in cotton. LBL1 had been described as a main gene and member of transcription factor family leucine zipper(HD-ZIPI) from a class I homologous domain factor Gorai.002 G244000. The qRT-PCR results elaborated the continuous change in expression level of LBL1 at different growth stages and leaf parts of cotton. Higher expression level was observed in mature large leaves followed by medium and young leaves respectively. For further confirmation, plants were tested from hormonal induction treatments, which explained that LBL1 expression was influenced by hormonal signaling. | 11 | Call Census,Habitat Suitability Modeling,and Local Communities’Perceptions for the Conservation of a Globally Threatened Avian Flagship Species显示文摘The Western tragopan(Tragopan melanocephalus)is recorded in the IUCN Red List of 2017 as a vulnerable bird species in Pakistan.This study provides valuable information for the conservation of Western tragopan,which is a globally threatened avian flagship species in Pakistan.This study was conducted to investigate and resolve the conservation challenges surrounding the species in two major habitat zones-Salkhala Game Reserve and Machiara National Park.The study was implemented in May-June 2020 for population density using call count data.Also,questionnaire was used for local residents’perceptions and habitat suitability modeling map was generated using the MaxEnt model based on previously recorded occurrence points as well as recorded in the survey.A total of 26 western Tragopans were identified by call count during the sampling period.Moreover,about 77.3%cited Western Tragopan they more likely to hunt while the remaining 22.6%locals recorded other pheasant.About 45.3%of this hunting for fun but only 22%for economic values and only 3%of people consider hunting as a part of their culture.Results of modeling habitat suitability of the Western tragopan showed that the species suitable habitats are small and patchy in Pakistan.We found that the Normalized Difference Vegetation Index(NDVI)with 40.6 percent contribution was the most important variable in shaping the species distribution.Our model identified some new suitable patches which can be the target of future field monitoring for finding new populations of the species and future conservation planning. | Abid Ali Iftikhar Uz Zaman Abbas Khan Masoud Yousefi Zahid Ali Muhammad Numan Khan | 2022 | Journal of Zoological Research2022,4,1: | 0 |

| 14 | Industrial Automation Information Analogy for Smart Grid Security显示文摘Industrial automation or assembly automation is a strictly monitored environment,in which changes occur at a good speed.There are many types of entities in the focusing environment,and the data generated by these devices is huge.In addition,because the robustness is achieved by sensing redundant data,the data becomes larger.The data generating device,whether it is a sensing device or a physical device,streams the data to a higher-level deception device for calculation,so that it can be driven and configured according to the updated conditions.With the emergence of the Industry 4.0 concept that includes a variety of automation technologies,various data is generated through numerous devices.Therefore,the data generated for industrial automation requires unique Information Architecture(IA).IA should be able to satisfy hard real-time constraints to spontaneously change the environment and the instantaneous configuration of all participants.To understand its applicability,we used an example smart grid analogy.The smart grid system needs an IA to fulfill the communication requirements to report the hard real-time changes in the power immediately following the system.In addition,in a smart grid system,it needs to report changes on either side of the system,i.e.,consumers and suppliers configure and reconfigure the system according to the changes.In this article,we propose an analogy of a physical phenomenon.A point charge is used as a data generating device,the streamline of electric flux is used as a data flow,and the charge distribution on a closed surface is used as a configuration.Finally,the intensity changes are used in the physical process,e.g.,the smart grid.This analogy is explained by metaphors,and the structural mapping framework is used for its theoretical proof.The proposed analogy provides a theoretical basis for the development of such information architectures that can represent data flows,definition changes(deterministic and non-deterministic),events,and instantaneous configuration definitions of entities in the system.The proposed analogy provides a mechanism to perform calculations during communication, using a simpleconcept on the closed surface to integrate two-layer cyber-physical systems(computation, communication, and physical process). | 18 | Giardiasis in patients with dyspeptic symptoms显示文摘AIM: To investigate the prevalence of giardiasis in patients with dyspeptic symptoms.METHODS: Clinical records of consecutive patients who attended Gastroenterology Department at Aga Khan University Hospital from January 2000 to June 2003 and had esophagogastroduodenoscopy (EGD) with duodenal biopsies and international classification of diseases 9th revision with clinical modifications (ICD-9-CM) coded with giardiasis were studied.RESULTS: Two hundred and twenty patients fulfilled the above criteria. There were 44% (96/220) patients who were giardiasis positive, 72% (69/96) of them were males and 28% (27/96) of them were females.There were 65% (81/124) males and 35% (43/124)females who were giardiasis negative.
