| 1 | SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality. | Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li | 2019 | Horticulture Research2019,6,1: | 39 |
| 2 | MicroTom Metabolic Network: Rewiring Tomato Metabolic Regulatory Network throughout the Growth Cycle显示文摘Tomato(Solanum lycopersicum)is a major horticultural crop worldwide and has emerged as a preeminent model for metabolic research.Although many research efforts have focused on the analysis of metabolite differences between varieties and species,the dynamics of metabolic changes during the tomato growth cycle and the regulatory networks that underlie these changes are poorly understood.In this study,we integrated high-resolution spatio-temporal metabolome and transcriptome data to systematically explore the metabolic landscape across 20 major tomato tissues and growth stages.In the resulting MicroTom Metabolic Network,the 540 detected metabolites and their co-expressed genes could be divided into 10 distinct clusters based on their biological functions.Using this dataset,we constructed a global map of the major metabolic changes that occur throughout the tomato growth cycle and dissected the underlying regulatory network.In addition to verifying previously well-established regulatory networks for important metabolites,we identified novel transcription factors that regulate the biosynthesis of important secondary metabolites such as steroidal glycoalkaloids and flavonoids.Our findings provide insights into spatiotemporal changes in tomato metabolism and generate a valuable resource for the study of metabolic regulatory processes in model plants. | Yan Li Yang Chen Lu Zhou Shengjie You Heng Deng Ya Chen Saleh Alseekh Yong Yuan Rao Fu Zixin Zhang Dan Su Alisdair R.Fernie Mondher Bouzayen Tao Ma Mingchun Liu Yang Zhang | 2020 | Molecular Plant2020,13,8: | 10 |

| 5 | Clinical management and associated costs for moderate and severe Alzheimer’s disease in urban China: a Delphi panel study显示文摘Background:Healthcare resource utilisation for Alzheimer’s disease(AD)in China is not well understood.This Delphi panel study aimed to describe the clinical management pathways for moderate and severe AD patients in urban China and to define the amount and cost of healthcare resources used.Methods:A panel of 11 experts was recruited from urban China to participate in two rounds of preparatory interviews.In the first round,9 physicians specialised in dementia gave a qualitative description of the clinical management of AD patients.In the second round,2 hospital administrators were asked about the cost of AD management and care.Results from the interviews were discussed by the experts in a Delphi panel meeting,where consensus was reached on quantitative aspects of AD management,including the rate of healthcare resource utilisation,the respective unit costs and caregiving time.Results:Interviewees reported that mild AD is under-recognised in China;most patients are diagnosed with moderate to severe AD.Loss of independence and agitation/aggression are the main drivers for healthcare resource utilisation and contribute to a heavier caregiver burden.It was estimated that 70%moderate AD patients are independent/non-aggressive at the time of diagnosis,15%are independent/aggressive,10%are dependent/non-aggressive,and 5%are dependent/aggressive.Dependent/aggressive AD patients are more likely to be hospitalised(70–90%)than accepted in a nursing home(0–20%),while the opposite is true for dependent/non-aggressive patients(5–35%for hospitalisation vs.80%for nursing home).Independent AD patients require 1–3 hours/day of caregiver time,while dependent patients can require up to 12–15 hours/day.Experts agreed that AD complicates the management of age-related comorbidities,found in 70–80%of all AD patients,increasing the frequency and cost of hospitalisation.Conclusions:The Delphi panel approach was an efficient method of gathering data about the amount of healthcare resources used and associated costs for moderate and severe AD patients in urban China.The results of this study provide a useful source of information for decision makers to improve future healthcare policies and resource planning,as well as to perform economic evaluations of AD therapies. | Xin Yu Shengdi Chen Xiaochun Chen Jianjun Jia Chunhou Li Cong Liu Mondher Toumi Dominique Milea | 2015 | Translational Neurodegeneration2015,4,1: | 2 |
