| 1 | Schistosoma japonicum infection induces macrophage polarization显示文摘The role of macrophages(Mφ)as the first line of host defense is well accepted.These cells play a central role in orchestrating crucial functions during schistosomal infection.Thus,understanding the functional diversity of these cells in the process of infection as well as the mechanisms underlying these events is crucial for developing disease control strategies.In this study,we adopted a Mφpolarization recognition system.Ml macrophage was characterized by expressing CD16/32,IL-12 and iNOS.M2 macrophage was characterized by expressing CD206,IL-10 and arg-1.In vivo(mouse peritoneal macrophages of different infection stages were obtained)and in vitro(different 5.japonicum antigens were used to stimulate RAW264.7)were characterized by using the above mentioned system.NCA and ACA stimulated RAW264.7 express significantly higher levels of IL-12 while significantly higher levels of IL-10 were detected after soluble egg antigen(SEA)stimulation.The results showed that dramatic changes of antigen in the microenvironment before and after egg production led to macrophage polarization.Furthermore,through TLR blocking experiments,the TLR4 signaling pathway was found to play a role in the process of macrophage polarization toward Ml.Our data suggest that macrophage polarization during S.japonicum infection had significant effects on host immune responses to S.japonicum. | Jingwei Xu Hao Zhang Lin Chen Donghui Zhang Minjun Ji Haiwei Wu Guanling Wu | 2014 | The Journal of Biomedical Research2014,28,4: | 20 |

| 3 | Xeno-repopulation of Fah^(-/-)Nod/Scid mice livers by human hepatocytes显示文摘Functional human hepatocytes xenografted into the liver of mice can be used as a model system to study pharmacokinetics,infection of hepatitis viruses,and the efficacy of hepatitis vaccines.Significant levels of liver xeno-repopulation have been reported in Fah-/-Rag2-/-Il2rg-/-mice.However,the high mortality and low breeding rate of this model may hinder its application.A new model,termed Fah-/-Nod/Scid mice,which combines the advantages of liver repopulation in Fah-/-mice with the ease of xenotransplantation in Nod/Scid mice was obtained by gradual cross-breeding.Fah-/-Nod/Scid mice were easily maintained in breeding colonies and in adult animal care facilities.FK506 treatment combined with gradual withdrawal of NTBC before cell transplantation ensured that Fah-/-Nod/Scid mice were susceptible to liver xeno-repopulation by human hepatocytes;the proportion of engrafted human hepatocytes reached 33.6%.The function of the expanded human hepatocytes within the chimeric liver was confirmed by weight curve analysis,the expression of characteristic proteins,and the biochemical analysis of liver function.These results show that Fah-/-Nod/Scid mice are an ideal humanized liver mouse model with many useful applications. | SU BaoLiang LIU ChangCheng XIANG Dao ZHANG HaiBin YUAN SiMing WANG MinJun CHEN Fei ZHU HaiYing HE ZhiYing WANG Xin HU YiPing | 2011 | Science China(Life Sciences)2011,54,3: | 5 |

| 5 | A specific aggregation-induced emission-conjugated polymer enables visual monitoring of osteogenic differentiation显示文摘Osteogenic differentiation is the basis of bone growth and repair related to many diseases,in which evaluating the degree and ability of osteogenic transformation is quite important and highly desirable.However,fixing or stopping the growth of cells is required for conventional methods to monitor osteogenic differentiation,which cannot realize the full investigation of the dynamic process.Herein,a new anion conjugated polymer featuring aggregation-induced emission(AIE)characteristics is developed with excellent solubility for in-situ monitoring the process of osteogenic differentiation.This novel polymer can bind with osteogenic differentiated cells,and the intracellular fluorescence increases gradually with the enhancement of osteogenic differentiation.Moreover,it possesses good biosafety with negligible effect on cell activity and osteogenic differentiation,which cannot be realized by the typical method of Alizarin Red S staining.Further study shows that the polymer crosses the cell membrane through endocytosis and enriches in lysosomes,whereas no obvious fluorescence is detected with other cells,including non-differentiated osteoblast cells,under the same conditions,demonstrating the high selectivity.This is the first fluorescent probe with excellent specificity to realize real-time observation of the process of osteogenic differentiation.Therefore,PTB-EDTA shows great promise in the study of osteogenic differentiation and related applications. | Zhenyu Zheng Taotao Zhou Rong Hu Minjun Huang Xiang Ao Jun Chu Tao Jiang Anjun Qin Zhongmin Zhang | 2020 | Bioactive Materials2020,5,4: | 3 |
| 6 | IFN-inducible p47 GTPases display differential responses to Schistosoma japonicum acute infection显示文摘Interferon gamma induced GTPase(IGTP)(also named lrgm3)and interferon gamma inducible protein 47(IRG-47)(also named lIrgd)are interferon(IFN)-inducible p47 GTPases that have been shown to regulate host resistance to intracellular pathogens.Little knowledge has been known about the role of p47 GTPases in host responses against extracellular pathogens.To investigate possible roles of IGTP and IRG-47 in the course of Schistosoma japonicum infection,IGTP and IRG-47 knockout and wild-type(WT)mice were challenged with cercariae of S.japonicum,and host responses were analyzed.At the acute stage of S.japonicum infection,mice that lacked IGTP displayed similar parasite burden and pathological damage to WT mice.Importantly,S.japonicum-infected lRG-47-deficient mice,in contrast to IGTP-deficient mice and WT mice,showed significantly reduced worms and lower egg-burden,but intense granulomatous reaction evoked by schistosome eggs in peripheral parts of liver lobes.In addition,upregulation of inflammation-related gene expression was observed in the spleen of IRG-47-deficient mice using oligonucleotide microarrays,in which multiple pathways of cytokine-cytokine receptor interaction,T-cell receptor signaling,complement,coagulation cascades and cell adhesion molecules were highlighted.Taken together,these data suggest that IGTP and IRG-47 might have distinct features that were differentially required for resistance to S.japonicum. | Xuesong Chen Xiaotang Du Meijuan Zhang Donghui Zhang Minjun Ji Guanling Wu | 2010 | Cellular & Molecular Immunology2010,7,1: | 2 |

| 10 | A Frame Breaking Based Hybrid Algorithm for UHF RFIDAnti-Collision显示文摘Multi-tag collision imposes a vital detrimental effect on reading performanceof an RFID system. In order to ameliorate such collision problem and to improve thereading performance, this paper proposes an efficient tag identification algorithm termedas the Enhanced Adaptive Tree Slotted Aloha (EATSA). The key novelty of EATSA is toidentify the tags using grouping strategy. Specifically, the whole tag set is divided intogroups by a frame of size F. In cases multiple tags fall into a group, the tags of the groupare recognized by the improved binary splitting (IBS) method whereas the rest tags arewaiting in the pipeline. In addition, an early observation mechanism is introduced toupdate the frame size to an optimum value fitting the number of tags. | 18 | Insights on evolution of virulence and resistance from the whole-genome analysis of a predominant methicillin-resistant Staphylococcus aureus clone sequence type 239 in China显示文摘Earlier, we reported that ST239 was the15-year predominant methicillin-resistant Staphylococcus aureus(MRSA) clone in China. In this study, MRSA strain CN79 belonging to ST239 and isolated from blood was used to determine the whole genome sequence. Comparative genomics analysis was done between MRSA CN79and 25 sequenced S. aureus in the NCBI GenBank database. A total of 2,734 protein-encoding genes were identified in the MRSA CN79 genome, which carries 11antibiotic resistance genes and 65 virulence genes. Two prophages phiCN79A and phiNM3-like were found on the MRSA CN79 genome. MRSA CN79 carries 30 specific genes that are absent from the 25 sequenced S. aureus genomes. Most of them were prophage-related genes.Several antibiotic resistance genes, such as b-lactamase and ABC-type multidrug transport system gene, were located on the genomic island mSab. The antibiotic resistance genes, such as tet(M), ermA1, and blaZ, were also located on different transposons. The virulence genes sea,map, hlb, and sak are located on phiNM3-like prophage and the exotoxin genes are carried on the genomic island mSaa. | 19 | Ultraviolet-attenuated cercariae ofSchistosoma japonicum fail to effectively induce a Th1 response in spite of up-regulating expression of cytotoxicity-related genes in C57BL/6 mice显示文摘Objective:To better understand the reason that Schistosoma japonicum(S.japonicum)ultraviolet(UV)-radiated cercariae could not induce high level of protection in C57BL/6 mice.Methods:Microarray technology was performed to investigate the gene transcription profile in skin draining lymph nodes(sdLNs)at 1 w after exposure to attenuated cercariae(AC)or normal cercariae(NC)of S.japonicum in C57BL/6 mice.The expressions of some representative genes were further confirmed by real-time PCR.Subsequently,the expressions of Th1/Th2 cytokine genes,cytotoxicity-related genes,as well as co-stimulator genes in spleens from AC-vaccinated and NC- infected mice were analyzed by real-time PCR at w 3 and 6 post-exposure.Results:The gene expressions of Th1 cytokines,including interferon-γ(IFN-γ),interleukin(IL)-12 and tumor necrosis factor-α(TNF-α)in the sdLNs were significantly lower in AC-vaccinated mice than in NC-infected mice.Furthermore,the gene expressions of Th1-and Th2-cytokines,including IFN-γ,IL-12,TNF-α,IL-4 and IL-10,in the spleens from AC-vaccinated mice showed little changes at w 3 and 6 post-vaccination.In addition,cytotoxicity-related molecules including granzyme A,granzyme B,granzyme K,perforin 1 and Fas L were up-regulated from the early stage of vaccination,and peaked at the 3rd w after vaccination with UV-AC.Conclusion:UV-AC of S.japonicum could not effectively induce a Th1 response in C57BL/6 mice,which may be an explanation for the low protection against parasite challenge,and the role played by up-regulated expression of cytotoxicity-related genes in mice needs to be further investigated. | Meijuan Zhang Fang Tian Yanan Gao Minjun Ji Guanling Wu | 2010 | The Journal of Biomedical Research2010,24,4: | 0 |
