1Xeno-repopulation of Fah^(-/-)Nod/Scid mice livers by human hepatocytes显示文摘Functional human hepatocytes xenografted into the liver of mice can be used as a model system to study pharmacokinetics,infection of hepatitis viruses,and the efficacy of hepatitis vaccines.Significant levels of liver xeno-repopulation have been reported in Fah-/-Rag2-/-Il2rg-/-mice.However,the high mortality and low breeding rate of this model may hinder its application.A new model,termed Fah-/-Nod/Scid mice,which combines the advantages of liver repopulation in Fah-/-mice with the ease of xenotransplantation in Nod/Scid mice was obtained by gradual cross-breeding.Fah-/-Nod/Scid mice were easily maintained in breeding colonies and in adult animal care facilities.FK506 treatment combined with gradual withdrawal of NTBC before cell transplantation ensured that Fah-/-Nod/Scid mice were susceptible to liver xeno-repopulation by human hepatocytes;the proportion of engrafted human hepatocytes reached 33.6%.The function of the expanded human hepatocytes within the chimeric liver was confirmed by weight curve analysis,the expression of characteristic proteins,and the biochemical analysis of liver function.These results show that Fah-/-Nod/Scid mice are an ideal humanized liver mouse model with many useful applications.SU BaoLiang LIU ChangCheng XIANG Dao ZHANG HaiBin YUAN SiMing WANG MinJun CHEN Fei ZHU HaiYing HE ZhiYing WANG Xin HU YiPing 2011Science China(Life Sciences)2011,54,3:5
2Roadway layout for recycling residual coal pillar in room-and-pillar mining of thick coal seam显示文摘In the context of a room-and-pillar mining gob in Shanxi province in China,this paper numerically investigates the stress distribution and deformation rules of roadway surrounding rocks at various locations of residual coal pillars in room-and-pillar mining gobs using software FLAC3 D.It is found that the concentrated stress beneath coal pillars distributes in a shape of ellipse.A reasonable roadway layout is then proposed.In this design,it is indicated that roadways should be designed to avoid the supporting zones of pillars with increasing compression and take into account the roof falling and crushing in the upper gob.According to the surrounding rock deformation characteristics and mining roadway locations as well as the supporting principles of timely support,rock reinforcing,piecewise management and suiting local conditions,a new asymmetric shield supporting plan is proposed.The field surveying results show that this supporting plan can effectively control the roadway rock deformation,thus guarantee the safe and smooth construction of roadways.Jin Gan Wang Lianguo Zhang Jihua Hu Minjun Duan Ning 2015International Journal of Mining Science and Technology2015,25,5:2
3Nanopolyphenol rejuvenates microglial surveillance of multiple misfolded proteins through metabolic reprogramming显示文摘Microglial surveillance plays an essential role in clearing misfolded proteins such as amyloid-beta,tau,andα-synuclein aggregates in neurodegenerative diseases.However,due to the complex structure and ambiguous pathogenic species of the misfolded proteins,a universal approach to remove the misfolded proteins remains unavailable.Here,we found that a polyphenol,α-mangostin,reprogrammed metabolism in the disease-associated microglia through shifting glycolysis to oxidative phosphorylation,which holistically rejuvenated microglial surveillance capacity to enhance microglial phagocytosis and autophagy-mediated degradation of multiple misfolded proteins.Nanoformulation ofα-mangostin efficiently deliveredα-mangostin to microglia,relieved the reactive status and rejuvenated the misfolded-proteins clearance capacity of microglia,which thus impressively relieved the neuropathological changes in both Alzheimer’s disease and Parkinson’s disease model mice.These findings provide direct evidences for the concept of rejuvenating microglial surveillance of multiple misfolded proteins through metabolic reprogramming,and demonstrate nanoformulatedα-mangostin as a potential and universal therapy against neurodegenerative diseases.Dayuan Wang Xiao Gu Xinyi Ma Jun Chen Qizhi Zhang Zhihua Yu Juan Li Meng Hu Xiaofang Tan Yuyun Tang Jianrong Xu Minjun Xu Qingxiang Song Huahua Song Gan Jiang Zaiming Tang Xiaoling Gao Hongzhuan Chen 2023Acta Pharmaceutica Sinica B2023,13,2:1

5A Frame Breaking Based Hybrid Algorithm for UHF RFIDAnti-Collision显示文摘Multi-tag collision imposes a vital detrimental effect on reading performanceof an RFID system. In order to ameliorate such collision problem and to improve thereading performance, this paper proposes an efficient tag identification algorithm termedas the Enhanced Adaptive Tree Slotted Aloha (EATSA). The key novelty of EATSA is toidentify the tags using grouping strategy. Specifically, the whole tag set is divided intogroups by a frame of size F. In cases multiple tags fall into a group, the tags of the groupare recognized by the improved binary splitting (IBS) method whereas the rest tags arewaiting in the pipeline. In addition, an early observation mechanism is introduced toupdate the frame size to an optimum value fitting the number of tags. 7Modulating autophagic flux via ROS-responsive targeted micelles to restore neuronal proteostasis in Alzheimer’s disease显示文摘Compromised autophagy and defective lysosomal clearance significantly contribute to impaired neuronal proteostasis,which represents a hallmark of Alzheimer’s disease(AD)and other age-related neurodegenerative disorders.Growing evidence has implicated that modulating autophagic flux,instead of inducing autophagosome formation alone,would be more reliable to rescue neuronal proteostasis.Concurrently,selectively enhancing drug concentrations in the leision areas,instead of the whole brain,will maximize therapeutic efficacy while reduing non-selective autophagy induction.Herein,we design a ROS-responsive targeted micelle system(TT-NM/Rapa)to enhance the delivery efficiency of rapamycin to neurons in AD lesions guided by the fusion peptide TPL,and facilitate its intracellular release via ROS-mediated disassembly of micelles,thereby maximizing autophagic flux modulating efficacy of rapamycin in neurons.Consequently,it promotes the efficient clearance of intracellular neurotoxic proteins,β-amyloid and hyperphosphorylated tau proteins,and ameliorates memory defects and neuronal damage in 3×Tg-AD transgenic mice.Our studies demonstrate a promising strategy to restore autophagic flux and improve neuronal proteostasis by rationally-engineered nano-systems for delaying the progression of AD.Shuting Xu Peng Yang Kang Qian Yixian Li Qian Guo Pengzhen Wang Ran Meng Jing Wu Jinxu Cao Yunlong Cheng Minjun Xu Qizhi Zhang 2022Bioactive Materials2022,7,5:1

10Impaired SERCA2a phosphorylation causes diabetic cardiomyopathy through impinging on cardiac contractility and precursor protein processing显示文摘Diabetic cardiomyopathy(DCM)is currently a progressive and nonstoppable complication in type 2 diabetic patients.Metabolic insults and insulin resistance are involved in its pathogenesis;however,the underlying mechanisms are still not clearly understood.Here we show that calcium dysregulation can be both a cause and a consequence of cardiac insulin resistance that leads to DCM.A western diet induces the development of DCM through at least three phases in mice,among which an early phase depends on impaired Thr^(484)-phosphorylation of sarcoplasmic/endoplasmic reticulum calcium ATPase 2a(SERCA2a)elicited by insulin resistance.Mutation of SERCA2a-Thr^(484) to a nonphosphorylatable alanine delays calcium re-uptake into the sarcoplasmic reticulum in the cardiomyocytes and decreases cardiac function at the baseline.Importantly,this mutation blunts the early phase of DCM,but has no effect on disease progression in the following phases.Interestingly,impairment of sarcoplasmic reticulum calcium re-uptake caused by the SERCA2a-Thr^(484) mutation inhibited processing of insulin receptor precursor through FURIN convertase,resulting in cardiac insulin resistance.Collectively,these data reveal a bidirectional relationship between insulin resistance and impairment of calcium homeostasis,which may underlie the early pathogenesis of DCM.Our findings have therapeutic implications for early intervention of DCM.Chao Quan Sangsang Zhu Ruizhen Wang Jiamou Chen Qiaoli Chen Min Li Shu Su Qian Du Minjun Liu Hong-Yu Wang Shuai Chen 2022Life Metabolism2022,1,1:0

12Insights on evolution of virulence and resistance from the whole-genome analysis of a predominant methicillin-resistant Staphylococcus aureus clone sequence type 239 in China显示文摘Earlier, we reported that ST239 was the15-year predominant methicillin-resistant Staphylococcus aureus(MRSA) clone in China. In this study, MRSA strain CN79 belonging to ST239 and isolated from blood was used to determine the whole genome sequence. Comparative genomics analysis was done between MRSA CN79and 25 sequenced S. aureus in the NCBI GenBank database. A total of 2,734 protein-encoding genes were identified in the MRSA CN79 genome, which carries 11antibiotic resistance genes and 65 virulence genes. Two prophages phiCN79A and phiNM3-like were found on the MRSA CN79 genome. MRSA CN79 carries 30 specific genes that are absent from the 25 sequenced S. aureus genomes. Most of them were prophage-related genes.Several antibiotic resistance genes, such as b-lactamase and ABC-type multidrug transport system gene, were located on the genomic island mSab. The antibiotic resistance genes, such as tet(M), ermA1, and blaZ, were also located on different transposons. The virulence genes sea,map, hlb, and sak are located on phiNM3-like prophage and the exotoxin genes are carried on the genomic island mSaa. 14Multidimensional autophagy nano-regulator boosts Alzheimer's disease treatment by improving both extra/intraneuronal homeostasis显示文摘Intraneuronal dysproteostasis and extraneuronal microenvironmental abnormalities in Alzheimer’s disease(AD)collectively culminate in neuronal deterioration.In the context of AD,autophagy dysfunction,a multi-link obstacle involving autophagy downregulation and lysosome defects in neurons/microglia is highly implicated in intra/extraneuronal pathological processes.Therefore,multidimensional autophagy regulation strategies co-manipulating“autophagy induction”and“lysosome degradation”in dual targets(neuron and microglia)are more reliable for AD treatment.Accordingly,we designed an RP-1 peptide-modified reactive oxygen species(ROS)-responsive micelles(RT-NM)loading rapamycin or gypenoside XVII.Guided by RP-1 peptide,the ligand of receptor for advanced glycation end products(RAGE),RT-NM efficiently targeted neurons and microglia in AD-affected region.This nanocombination therapy activated the whole autophagy-lysosome pathway by autophagy induction(rapamycin)and lysosome improvement(gypenoside XVII),thus enhancing autophagic degradation of neurotoxic aggregates and inflammasomes,and promoting Aβ phagocytosis.Resultantly,it decreased aberrant protein burden,alleviated neuroinflammation,and eventually ameliorated memory defects in 3×Tg-AD transgenic mice.Our research developed a multidimensional autophagy nano-regulator to boost the efficacy of autophagy-centered AD therapy.Yixian Li Peng Yang Ran Meng Shuting Xu Lingling Zhou Kang Qian Pengzhen Wang Yunlong Cheng Dongyu Sheng Minjun Xu Tianying Wang Jing Wu Jinxu Cao Qizhi Zhang 2024Acta Pharmaceutica Sinica B2024,14,3:0
15Income Growth,Urbanization,Changing Life Style and Energy Requirements in China显示文摘This paper aims to estimate the effects of changing life style and consumption demands driven by income growth and urbanization on increase of energy requirements in China, and es-timate the impacts of improvement in household consumption on mitigating energy requirements towards 2020, based on input-out-put analysis and scenarios simulation approach. The result shows that energy requirement per capita has increased by 159% for urban residents and 147% for rural residents from 1995 to 2004. Growth in household consumption driven by income growth and urbanization may induce a successive increase in energy require-ments in future. Per capita energy requirements of urban residents will increase by 240% during 2002-2015 and 330% during 2002-2020. Urbanization might lead to 0.75 billion ton of increment of energy requirements in 2020. About 45%-48% of total energy requirements in China might be a consequence of residents' life styles and the economic activities to support consumption demands in 2020. Under low-carbon life style scenario, per capita energy requirements of urban residents may decline to 97% in 2015 and 92% in 2020 in contrast with baseline scenario. 16Cilia regulate meiotic recombination in zebrafish显示文摘Meiosis is essential for evolution and genetic diversity in almost all sexual eukaryotic organisms.The mechanisms of meiotic recombination,such as synapsis,have been extensively investigated.However,it is still unclear whether signals from the cytoplasm or even from outside of the cell can regulate the meiosis process.Cilia are microtubule-based structures that protrude from the cell surface and function as signaling hubs to sense extracellular signals.Here,we reported an unexpected and critical role of cilia during meiotic recombination.During gametogenesis of zebrafish,cilia were specifically present in the prophase stages of both primary spermatocytes and primary oocytes.By developing a germ cell-specific CRISPR/Cas9 system,we demonstrated that germ cell-specific depletion of ciliary genes resulted in compromised double-strand break repair,reduced crossover formation,and increased germ cell apoptosis.Our study reveals a previously undiscovered role for cilia during meiosis and suggests that extracellular signals may regulate meiotic recombination via this particular organelle.Haibo Xie Xiaosi Wang Minjun Jin Lanqin Li Junwen Zhu Yunsi Kang Zhe Chen Yonghua Sun Chengtian Zhao 2022Journal of Molecular Cell Biology2022,14,7:0
