1Schistosoma japonicum infection induces macrophage polarization显示文摘The role of macrophages(Mφ)as the first line of host defense is well accepted.These cells play a central role in orchestrating crucial functions during schistosomal infection.Thus,understanding the functional diversity of these cells in the process of infection as well as the mechanisms underlying these events is crucial for developing disease control strategies.In this study,we adopted a Mφpolarization recognition system.Ml macrophage was characterized by expressing CD16/32,IL-12 and iNOS.M2 macrophage was characterized by expressing CD206,IL-10 and arg-1.In vivo(mouse peritoneal macrophages of different infection stages were obtained)and in vitro(different 5.japonicum antigens were used to stimulate RAW264.7)were characterized by using the above mentioned system.NCA and ACA stimulated RAW264.7 express significantly higher levels of IL-12 while significantly higher levels of IL-10 were detected after soluble egg antigen(SEA)stimulation.The results showed that dramatic changes of antigen in the microenvironment before and after egg production led to macrophage polarization.Furthermore,through TLR blocking experiments,the TLR4 signaling pathway was found to play a role in the process of macrophage polarization toward Ml.Our data suggest that macrophage polarization during S.japonicum infection had significant effects on host immune responses to S.japonicum.Jingwei Xu Hao Zhang Lin Chen Donghui Zhang Minjun Ji Haiwei Wu Guanling Wu 2014The Journal of Biomedical Research2014,28,4:20

4Nanopolyphenol rejuvenates microglial surveillance of multiple misfolded proteins through metabolic reprogramming显示文摘Microglial surveillance plays an essential role in clearing misfolded proteins such as amyloid-beta,tau,andα-synuclein aggregates in neurodegenerative diseases.However,due to the complex structure and ambiguous pathogenic species of the misfolded proteins,a universal approach to remove the misfolded proteins remains unavailable.Here,we found that a polyphenol,α-mangostin,reprogrammed metabolism in the disease-associated microglia through shifting glycolysis to oxidative phosphorylation,which holistically rejuvenated microglial surveillance capacity to enhance microglial phagocytosis and autophagy-mediated degradation of multiple misfolded proteins.Nanoformulation ofα-mangostin efficiently deliveredα-mangostin to microglia,relieved the reactive status and rejuvenated the misfolded-proteins clearance capacity of microglia,which thus impressively relieved the neuropathological changes in both Alzheimer’s disease and Parkinson’s disease model mice.These findings provide direct evidences for the concept of rejuvenating microglial surveillance of multiple misfolded proteins through metabolic reprogramming,and demonstrate nanoformulatedα-mangostin as a potential and universal therapy against neurodegenerative diseases.Dayuan Wang Xiao Gu Xinyi Ma Jun Chen Qizhi Zhang Zhihua Yu Juan Li Meng Hu Xiaofang Tan Yuyun Tang Jianrong Xu Minjun Xu Qingxiang Song Huahua Song Gan Jiang Zaiming Tang Xiaoling Gao Hongzhuan Chen 2023Acta Pharmaceutica Sinica B2023,13,2:1
5Size control and electronic manipulation of Ru catalyst over B,N codoped carbon network for high-performance hydrogen evolution reaction显示文摘Exploring highly efficient Pt-free catalysts for hydrogen evolution reaction(HER)is of great importance for hydrogen(H2)production.Herein,a novel HER electrocatalyst having abundant ultra-small(2–3 nm)Ru electronically confined by a B,N codoped polar carbon surface(Ru/(B-N)-PC)was constructed.The Ru/(B-N)-PC catalyst exhibits a low overpotential of 15 mV at the current density of 10 mA·cm^(−2),a low Tafel slope of 22.6 mV·dec^(−1),superior durability,which outperforms the benchmark Pt/C catalyst.Both experimental characterizations and theory calculations suggest that an electron communication established between B,N co-doped carbon surface and ultra-small Ru nanoparticles with electrons transferred from N atoms to Ru and backtransferred from Ru to B atoms,which exerts a moderate electronic modification of Ru.This,in turn,affords a modest H adsorption energy and a lower H2O dissociation barrier,leading to the high-performance hydrogen evolution reaction.The work provides meaningful insight into the size control and electronic modulation of Ru catalyst for intrinsic HER activity improvement.Shuai Xu Mang Niu Guowei Zhao Shujun Ming Xingyun Li Qilong Zhu Liang-Xin Ding Minjun Kim Asma A.Alothman Mohammed Sheikh Saleh Mushab Yusuke Yamauchi 2023Nano Research2023,16,5:1

8Modulating autophagic flux via ROS-responsive targeted micelles to restore neuronal proteostasis in Alzheimer’s disease显示文摘Compromised autophagy and defective lysosomal clearance significantly contribute to impaired neuronal proteostasis,which represents a hallmark of Alzheimer’s disease(AD)and other age-related neurodegenerative disorders.Growing evidence has implicated that modulating autophagic flux,instead of inducing autophagosome formation alone,would be more reliable to rescue neuronal proteostasis.Concurrently,selectively enhancing drug concentrations in the leision areas,instead of the whole brain,will maximize therapeutic efficacy while reduing non-selective autophagy induction.Herein,we design a ROS-responsive targeted micelle system(TT-NM/Rapa)to enhance the delivery efficiency of rapamycin to neurons in AD lesions guided by the fusion peptide TPL,and facilitate its intracellular release via ROS-mediated disassembly of micelles,thereby maximizing autophagic flux modulating efficacy of rapamycin in neurons.Consequently,it promotes the efficient clearance of intracellular neurotoxic proteins,β-amyloid and hyperphosphorylated tau proteins,and ameliorates memory defects and neuronal damage in 3×Tg-AD transgenic mice.Our studies demonstrate a promising strategy to restore autophagic flux and improve neuronal proteostasis by rationally-engineered nano-systems for delaying the progression of AD.Shuting Xu Peng Yang Kang Qian Yixian Li Qian Guo Pengzhen Wang Ran Meng Jing Wu Jinxu Cao Yunlong Cheng Minjun Xu Qizhi Zhang 2022Bioactive Materials2022,7,5:1

12Multidimensional autophagy nano-regulator boosts Alzheimer's disease treatment by improving both extra/intraneuronal homeostasis显示文摘Intraneuronal dysproteostasis and extraneuronal microenvironmental abnormalities in Alzheimer’s disease(AD)collectively culminate in neuronal deterioration.In the context of AD,autophagy dysfunction,a multi-link obstacle involving autophagy downregulation and lysosome defects in neurons/microglia is highly implicated in intra/extraneuronal pathological processes.Therefore,multidimensional autophagy regulation strategies co-manipulating“autophagy induction”and“lysosome degradation”in dual targets(neuron and microglia)are more reliable for AD treatment.Accordingly,we designed an RP-1 peptide-modified reactive oxygen species(ROS)-responsive micelles(RT-NM)loading rapamycin or gypenoside XVII.Guided by RP-1 peptide,the ligand of receptor for advanced glycation end products(RAGE),RT-NM efficiently targeted neurons and microglia in AD-affected region.This nanocombination therapy activated the whole autophagy-lysosome pathway by autophagy induction(rapamycin)and lysosome improvement(gypenoside XVII),thus enhancing autophagic degradation of neurotoxic aggregates and inflammasomes,and promoting Aβ phagocytosis.Resultantly,it decreased aberrant protein burden,alleviated neuroinflammation,and eventually ameliorated memory defects in 3×Tg-AD transgenic mice.Our research developed a multidimensional autophagy nano-regulator to boost the efficacy of autophagy-centered AD therapy.Yixian Li Peng Yang Ran Meng Shuting Xu Lingling Zhou Kang Qian Pengzhen Wang Yunlong Cheng Dongyu Sheng Minjun Xu Tianying Wang Jing Wu Jinxu Cao Qizhi Zhang 2024Acta Pharmaceutica Sinica B2024,14,3:0
