1糖尿病视网膜病变患病率的流行病学调查:在中国6省13473名糖尿病患者中的横断面研究显示文摘目的本研究旨在探究中国糖尿病患者中糖尿病视网膜病变(DR)和视力威胁性糖尿病视网膜病变(STDR)的患病率和严重程度。研究设计、设置与参与者本研究为横断面流行病学调查,研究对象为中国内地地区的糖尿病患者(N=13473)。研究对象为被内科医师诊断为糖尿病,并转诊到筛查门诊的糖尿病患者(其中住院患者占1/3,城市居民占1/3,农村居民占1/3)。研究对象的每只眼睛应用彩色、免散瞳和非立体眼底照相机拍摄2张眼底照片,并根据英国DR分级指南进行分级。主要结果和测量指标DR与STDR的患病率和严重程度。结果在参与本研究的13473例糖尿病患者中,共发现4591例DR和1769例STDR患者,DR的总体患病率为34.08%[95%可信区间(凹)33.28%~34.88%],其中STDR的总体患病率为13.13%(95%C112.56%~13.70%)。在所有研究对象中,12780例参与了眼底照片分级(94.86%)。年龄大于65岁的研究对象患DR或STDR的可能性较低(P〈0.001)。性别对是否患病没有显著影响(P〉0.05)。STDR患者与非STDR患者相比,视力损伤更为严重(P〈0.001)。中国北方DR和STDR的患病率高于南方(P〈0.001)。DR的患病率与糖尿病患病时间f比值比(鲫)2.63;95%C12.42~2.86;P〈0.001]以及糖尿病发病年龄(OR 0.38;95%CI为0.35~0.41;P〈0.001)密切相关。结论DR和STDR在中国内地地区的患病率偏高,并且不同地区患病率存在差异。在DR中,非增殖性DR更常见,但STDR更需要及时的诊断和治疗,特别是在经济欠发达地区。本研究强调了中国内地地区DR筛查和治疗的重要性。Liu, Yan Song, Yifan Tao, Liyuan Qiu, Weiqiang Lv, Huibin Jiang, Xiaodan Zhang, Mingzhou Li, Xuemin 2017英国医学杂志中文版2017,20,11:23
2DNA methylation-mediated repression of miR-181a/135a/302c expression promotes the microsatellite-unstable colorectal cancer development and 5-FU resistance via targeting PLAG1显示文摘Micro sate Hite instability(MSI) defines a subtype of colorectal cancer(CRC) with typical clinicopathologic characteristics. CRCs with MSI(MSI CRCs) frequently acquire accelerated carcinogenesis and 5-FU resistance, and the exact underlying mechanism remains incompletely understood. Our previous study has identified the microRNA(miRNA) expression profile in MSI CRCs. In this study, three miRNAs(miR-181 a, miR-135 a and miR-302 c) were validated by qRT-PCR to be dramatically decreased in 67 CRC samples. Proliferation and apoptosis assays demonstrated that miR-181 a/135 a/302 c function as tumor suppressors via repressing PLAG1/IGF2 signaling. Moreover, we presented compelling evidence that restoration of miR-181 a/135 a/302 c expression promoted sensitivity of MSI CRC cells to 5-FU treatment. miR-181 a/135 a/302 c exerted their effect on chemoresistance through attenuating PLAG1 expression. Notably, the hypermethylation status of MSI CRC accounts for the decrements of miR-181 a/135 a/302 c. 3Building a sequence map of the pig pan-genome from multiple de novo assemblies and Hi-C data显示文摘Pigs were domesticated independently in the Near East and China,indicating that a single reference genome from one individual is unable to represent the full spectrum of divergent sequences in pigs worldwide.Therefore,12 de novo pig assemblies from Eurasia were compared in this study to identify the missing sequences from the reference genome.As a result,72.5 Mb of nonredundant sequences(~3% of the genome)were found to be absent from the reference genome(Sscrofa11.1)and were defined as pan-sequences.Of the pan-sequences,9.0 Mb were dominant in Chinese pigs,in contrast with their low frequency in European pigs.One sequence dominant in Chinese pigs contained the complete genic region of the tazarotene-induced gene 3(TIG3)gene which is involved in fatty acid metabolism.Using flanking sequences and Hi-C based methods,27.7% of the sequences could be anchored to the reference genome.The supplementation of these sequences could contribute to the accurate interpretation of the 3D chromatin structure.A web-based pan-genome database was further provided to serve as a primary resource for exploration of genetic diversity and promote pig breeding and biomedical research.Xiaomeng Tian Ran Li Weiwei Fu Yan Li Xihong Wang Ming Li Duo Du Qianzi Tang Yudong Cai Yiming Long Yue Zhao Mingzhou Li Yu Jiang 2020Science China(Life Sciences)2020,63,5:7
4Amplification of the miR-181c/d cluster is inversely correlated with PDCD4 expression in gastric cancer显示文摘miR-181c/d is dysregulated in gastric cancer(GC).We investigated the amplification and expression of miR-181c/d and its predicted target genes in GC.Amplification of miR-181c/d was quantified by genomic real-time PCR in GC and adjacent normal tissues,as well as the levels of mature miR-181c/d was performed by real-time PCR in the same tissues.The potential target genes of miR-181c/d were predicted using bioinformatics software.Expression of one potential target gene,PDCD4,was measured by semiquantitative RT-PCR,real-time PCR,and immunohistochemistry.Next,the relationship between miR181c/d expression and PDCD4 expression was analyzed.Results indicated that the amplification and expression of miR-181c/d were significantly higher in GC than in adjacent normal tissues(primary miR-181c/d,P\0.001;miR-181c,P=0.0344;miR-181d,P=0.0153),and there was a strong correlation between mature miR-181c/d and primary miR-181c/d.Thirty-two target genes were predicted,including PDCD4 which is a known tumor suppressor gene.Expression of PDCD4 was significantly down-regulated in GC as compared to adjacent normal tissues and was inversely correlated with miR-181c/d expression in GC(miR-181c and PDCD4:R=-0.496,P=0.008;miR-181d and PDCD4:R=-0.454,P=0.003).Therefore,miR-181c/d may play a pivotal role in the pathogenesis of GC by downregulating PDCD4 expression.Yuanming Pan Rui Xing Juan An Jiantao Cui Wenmei Li Mingzhou Guo Youyong Lu 2014Chinese Science Bulletin2014,59,19:6
5Differential expression analysis and regulatory network reconstruction for genes associated with muscle growth and adipose deposition in obese and lean pigs显示文摘During the growth and development of skeletal muscle cells and adipose cells, the regulatory mechanism of micro-effect polygenes determines porcine meat quality, carcass characteristics and other relative quantitative traits. Obese and lean type pig breeds show obvi- ous differences in muscle growth and adipose deposition; however, the molecular mechanism underlying this phenotypic variation remains unknown. We used pathway-focused oligo microarray studies to examine the expression changes of 140 genes associated with muscle growth and adipose deposition in longissimus dorsi muscle at six growth stages (birth, 1, 2, 3, 4 and 5 months) of Landrace (a leaner, Western breed) and Taihu pigs (a fatty, indigenous, Chinese breed). Variance analysis (ANOVA) revealed that differences in the expression of 18 genes in Landrace pigs and three genes in Taihu pigs were very significant (FDR-adjusted permutation, P < 0.01) and di?erences for 22 genes in Landrace pigs and seven genes in Taihu pigs were significant (FDR-adjusted permutation, P < 0.05) among six growth stages. Clustering analysis revealed a high level of signi?cance (FDR-adjusted, P < 0.01) for four gene expression patterns, in which genes that strongly up-regulated were mainly associated with the positive regulation of myofiber formation and fatty acid biogenesis and genes that strongly down-regulated were mainly associated with the inhibition of cell proliferation and positive regulation of fatty acid b-oxidation. Based on a dynamic Bayesian network (DBN) model, gene regulatory networks (GRNs) were reconstructed from time-series data for each pig breed. These two GRNs initially revealed the distinct differences in physiological and biochemical aspects of muscle growth and adipose deposition between the two pig breeds; from these results, some potential key genes could be identified. Quantitative real-time RT-PCR (QRT-PCR) was used to verify the microarray data for five modulated genes, and a good correlation between the two measures of expression was observed for both two pig breeds at different growth stages (r = 0.876 ± 0.095). 6Phase Ⅱ study of apatinib in combination with oral vinorelbine in heavily pretreated HER2-negative metastatic breast cancer and clinical implications of monitoring ctDNA显示文摘Objective:Apatinib is an oral TKI targeting VEGFR-2.Single-agent apatinib treatment has been shown to produce an objective response in patients with pretreated m BC.Oral vinorelbine also holds promise as a treatment of choice in patients with m BC.This study aimed to investigate the efficacy and safety of the oral vinorelbine-apatinib combination in patients with pretreated m BC.In addition,we detected gene variants in ct DNA to explore the therapeutic implications.Methods:This study enrolled patients with HER2-negative m BC who were pretreated with anthracycline/taxanes.Patients were treated with apatinib at 500 mg/425 mg daily plus oral vinorelbine 60 mg/m2 on days 1,8,and 15 of every cycle(3 weeks).The primary endpoint was PFS.The secondary endpoints were ORR,CBR,OS,and safety.Patients eligible for ct DNA detection were evaluated before and during treatment.Results:Forty patients were enrolled.The median PFS was 5.2 months(95%CI,3.4–7.0 months),and the median OS was 17.4 months(95%CI,8.0–27.0 months).The ORR was 17.1%(6/35),and the CBR was 45.7%(16/35).The most common AEs included gastrointestinal reaction,myelosuppression,and hypertension.In 20 patients,ct DNA was detected at baseline and during treatment.A significant difference was found in PFS for undetected vs.detected baseline ct DNA(13.9 months vs.3.6 months,P=0.018).Conclusions:All-oral therapy with apatinib plus vinorelbine displayed objective efficacy in patients with heavily pretreated HER2-negative m BC,with acceptable and manageable toxicity profiles.Patients with no gene variant detected and lower variant allele frequencies in ct DNA at baseline showed longer PFS.Anjie Zhu Peng Yuan Nanlin Hu Mingzhou Li Wenmiao Wang Xue Wang Jian Yue Jiayu Wang Yang Luo Fei Ma Pin Zhang Qing Li Binghe Xu Shanbo Cao Giuseppe Lippi Yoichi Naito Mohammed A.Osman Gustavo N.Marta Gianluca Franceschini Armando Orlandi 2021Cancer Biology & Medicine2021,18,3:4
7Alterations in cecal microbiota of Jinhua piglets fostered by a Yorkshire sow显示文摘Gut microbiota play important roles in host health and diseases.Several factors,in particular antibiotics,affect the gut microbiota of pigs.Cross-fostering has been applied as a regular practice to equalize litter size,reduce pre-weaning mortality and increase body weight.However,the effect of cross-fostering on cecal microbiota is unclear.In this study,we fostered three Jinhua pigs to a Yorkshire sow.The fostered Jinhua piglets grew significantly faster than their biological siblings.To explore whether the cecal microbiota of piglets will alter during fostering,we characterized the cecal microbiota of allpiglets by examining the V3 hypervariable region of 16S rDNA.We observed altered cecal microbiota in these piglets using the Illumina HiSeq 2000 platform,and this was accompanied with an increase in growth rate after fostering.The relative abundance of Bacteroidetes in the fostered Jinhua piglets was decreased compared with their biological siblings,although still higher in comparison with their new littermates.Beta-diversity analysis also showed that the cecal microbiota of the adopted Jinhua piglets differed from their biological siblings with a shift toward their step-siblings.Our data show that cecal microbiota of piglets were altered after cross-fostering while the growth rate increased.Lingjin Xian Ying Li Zhi Jiang Jideng Ma Long Jin Lei Chen Chaowei Zhou Jie Zhang Yingkai Liu Li Zhu An-an Jiang Guoqing Tang Jian Xiao Jinyong Wang Bo Zeng Mingzhou Li Xuewei Li 2014Chinese Science Bulletin2014,59,32:4

9Genome-wide analysis reveals selection for Chinese Rongchang pigs显示文摘Livestock have undergone domestication and consequently strong selective pressure on genes or genomic regions that control desirable traits. To identify selection signatures in the genome of Chinese Rongchang pigs, we generated a total of about 170 Gb of DNA sequence data with about 6.4-fold coverage for each of six female individuals. By combining these data with the publically available genome data of 10 Asian wild boars,we identified 449 protein-coding genes with selection signatures in Rongchang pigs, which are mainly involved in growth and hormone binding, nervous system development, and drug metabolism. The accelerated evolution of these genes may contribute to the dramatic phenotypic differences between Rongchang pigs and Chinese wild boars. 10Reorganization of 3D genome architecture across wild boar and Bama pig adipose tissues显示文摘Background:A growing body of evidence has revealed that the mammalian genome is organized into hierarchical layers that are closely correlated with and may even be causally linked with variations in gene expression.Recent studies have characterized chromatin organization in various porcine tissues and cell types and compared them among species and during the early development of pigs.However,how chromatin organization differs among pig breeds is poorly understood.Results:In this study,we investigated the 3D genome organization and performed transcriptome characterization of two adipose depots(upper layer of backfat[ULB]and greater omentum[GOM])in wild boars and Bama pigs;the latter is a typical indigenous pig in China.We found that over 95%of the A/B compartments and topologically associating domains(TADs)are stable between wild boars and Bama pigs.In contrast,more than 70%of promoterenhancer interactions(PEIs)are dynamic and widespread,involving over a thousand genes.Alterations in chromatin structure are associated with changes in the expression of genes that are involved in widespread biological functions such as basic cellular functions,endocrine function,energy metabolism and the immune response.Approximately 95%and 97%of the genes associated with reorganized A/B compartments and PEIs in the two pig breeds differed between GOM and ULB,respectively.Conclusions:We reported 3D genome organization in adipose depots from different pig breeds.In a comparison of Bama pigs and wild boar,large-scale compartments and TADs were mostly conserved,while fine-scale PEIs were extensively reorganized.The chromatin architecture in these two pig breeds was reorganized in an adipose depotspecific manner.These results contribute to determining the regulatory mechanism of phenotypic differences between Bama pigs and wild boar.Jiaman Zhang Pengliang Liu Mengnan He Yujie Wang Hua Kui Long Jin Diyan Li Mingzhou Li 2022Journal of Animal Science and Biotechnology2022,13,4:2
11Incorporating transportation network modeling tools within transportation economic impact studies of disasters显示文摘Transportation system disruption due to a disaster results in 'ripple effects' throughout the entire transportation system of a metropolitan region.Many researchers have focused on the economic costs of transportation system disruptions in transportation-related industries,specifically within commerce and logistics,in the assessment of the regional economic costs.However,the foundation of an assessment of the regional economic costs of a disaster needs to include the evaluation of consumer surplus in addition to the direct cost for reconstruction of the regional transportation system.The objective of this study is to propose a method to estimate the regional consumer surplus based on indirect economic costs of a disaster on intermodal transportation systems in the context of diverting vehicles and trains.The computational methods used to assess the regional indirect economic costs sustained by the highway and railroad system can utilize readily available state departments of transportation(DOTs)and metropolitan planning organizations(MPOs) traffic models allowing prioritization of regional recovery plans after a disaster and strengthening of infrastructure before a disaster.Hurricane Katrina is one of the most devastating hurricanes in the history of the United States.Due to the significance of Hurricane Katrina,a case study is presented to evaluate consumer surplus in the Gulf Coast Region of Mississippi.Results from the case study indicate the costs of rerouting and congestion delays in the regional highway system and the rent costs of right-of-way in the regional railroad system are major factors of the indirect costs in the consumer surplus.Yi Wen Li Zhang Zhitong Huang Mingzhou Jin 2014Journal of Traffic and Transportation Engineering(English Edition)2014,1,4:2
12Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants,are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. 16Spatiotemporal dynamics of ecosystem fires and biomass burning-induced carbon emissions in China over the past two decades显示文摘Fire is a major type of disturbance that has important influences on ecosystem dynamics and carbon cycles.Yet our understanding of ecosystem fires and their carbon cycle consequences is still limited,largely due to the difficulty of large-scale fire monitoring and the complex interactions between fire,vegetation,climate,and anthropogenic factors.Here,using data from satellite-derived fire observations and ecosystem model simulations,we performed a comprehensive investigation of the spatial and temporal dynamics of China’s ecosystem fire disturbances and their carbon emissions over the past two decades(1997–2016).Satellite-derived results showed that on average about 3.47-4.53×10^(4) km^(2) of the land was burned annually during the past two decades,among which annual burned forest area was about 0.81-1.25×10^(4) km^(2),accounting for 0.33-0.51%of the forest area in China.Biomass burning emitted about 23.02 TgC per year.Compared to satellite products,simulations from the Energy Exascale Earth System Land Model(ELM)strongly overestimated China’s burned area and fire-induced carbon emissions.Annual burned area and fire-induced carbon emissions were high for boreal forest in Northeast China’s Daxing’anling region and subtropical dry forest in South Yunnan,as revealed by both the satellite product and the model simulations.Our results suggest that climate and anthropogenic factors play critical roles in controlling the spatial and seasonal distribution of China’s ecosystem fire disturbances.Our findings highlight the importance of multiple complementary approaches in assessing ecosystem fire disturbance and its carbon consequences.Further studies are required to improve the methods of observing and modelling China’s ecosystem fire disturbances,which will provide valuable information for fire management and ecosystem sustainability in an era when both human activities and the natural environment are rapidly changing.Anping Chen Rongyun Tang Jiafu Mao Chao Yue Xiran Li Mengdi Gao Xiaoying Shi Mingzhou Jin Daniel Ricciuto Sam Rabin Phillippe Ciais Shilong Piao 2020Geography and Sustainability2020,1,1:1
