1AXL is a candidate receptor for SARS-CoV-2 that promotes infection of pulmonary and bronchial epithelial cells显示文摘The current coronavirus disease 2019(COVID-19)pandemic presents a global public health challenge.The viral pathogen responsible,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),binds to the host receptor ACE2 through its spike(S)glycoprotein,which mediates membrane fusion and viral entry.Although the role of ACE2 as a receptor for SARS-CoV-2 is clear,studies have shown that ACE2 expression is extremely low in various human tissues,especially in the respiratory tract.Thus,other host receptors and/or co-receptors that promote the entry of SARS-CoV-2 into cells of the respiratory system may exist.In this study,we found that the tyrosine-protein kinase receptor UFO(AXL)specifically interacts with the N-terminal domain of SARS-CoV-2 S.Using both a SARS-CoV-2 virus pseudotype and authentic SARS-CoV-2,we found that overexpression of AXL in HEK293T cells promotes SARS-CoV-2 entry as efficiently as overexpression of ACE2,while knocking out AXL significantly reduces SARS-CoV-2 infection in HI 299 pulmonary cells and in human primary lung epithelial cells.Soluble human recombinant AXL blocks SARS-CoV-2 infection in cells expressing high levels of AXL.The AXL expression level is well correlated with SARS-CoV-2 S level in bronchoalveolar lavage fluid cells from COVID-19 patients.Taken together,our findings suggest that AXL is a novel candidate receptor for SARS-CoV-2 which may play an important role in promoting viral infection of the human respiratory system and indicate that it is a potential target for future clinical intervention strategies.Shuai Wang Zongyang Qiu Yingnan Hou Xiya Deng Wei Xu Tingting Zheng Peihan Wu Shaofang Xie Weixiang Bian Chong Zhang Zewei Sun Kunpeng Liu Chao Shan Aifu Lin Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Jing Huang Xu Li 2021Cell Research2021,31,2:39

3Ten cities cross-sectional questionnaire survey of children asthma and other allergies in China显示文摘Asthma,rhinitis and eczema(allergic or non-allergic)have increased throughout the world during the last decades,especially among children.Changes in the indoor environment are suspected to be important causes.China has experienced a dramatic change in indoor environmental exposures during the past two decades.However,such changes and their associations with children’s asthma and other health aspects have not been thoroughly studied.China,Children,Homes,Health(CCHH),Phase I,was a cross-sectional questionnaire survey of 48219 children 1–8 years old in 10 Chinese cities during 2010–2012.The questionnaire includes the International Study of Asthma and Allergies in Childhood(ISAAC)core health questions and additional questions regarding housing,life habits and outdoor environment.In health analyses,children aged 3–6 years old were included.The prevalences of doctor diagnosed asthma varied from 1.7%to 9.8%(mean 6.8%),a large increase from 0.91%in 1999 and 1.50%in2000.The prevalence of wheeze,rhinitis and atopic eczema(last 12 months)varied from 13.9%to 23.7%,24.0%to 50.8%and4.8%to 15.8%,respectively.Taiyuan had the lowest prevalences of all illnesses and Shanghai the highest,except for wheezewhere the highest value was for Urumqi.We found(1)no obvious association between disease prevalences and ambient PM10concentrations and(2)higher prevalences of disease in humid climates with hot summers and cold winters,but with no centrally heated buildings.Associations between the diseases and economic status as indexed by Gross Domestic Product(GDP)requires further study.ZHANG YinPing LI BaiZhan HUANG Chen YANG Xu QIAN Hua DENG QiHong ZHAO ZhuoHui LI AnGui ZHAO JiaNing ZHANG Xin QU Fang HU Yu YANG Qin WANG Juan ZHANG Ming WANG Fang ZHENG XiaoHong LU Chan LIU ZhiJian SUN YueXia MO JinHan ZHAO YiLi LIU Wei WANG TingTing NORBCK Dan BORNEHAG Carl-Gustaf SUNDELL Jan 2013Chinese Science Bulletin2013,58,34:22
4Role of secondary aerosols in haze formation in summer in the Megacity Beijing显示文摘A field experiment from 18 August to 8 September 2006 in Beijing, China, was carried out. A hazy day was defined as visibility < 10 km and RH(relative humidity) < 90%. Four haze episodes, which accounted for ~ 60% of the time during the whole campaign, were characterized by increases of SNA(sulfate, nitrate, and ammonium) and SOA(secondary organic aerosol) concentrations. The average values with standard deviation of SO2-+4, NO-3, NH4 and SOA were 49.8(± 31.6), 31.4(±22.3), 25.8(±16.6) and 8.9(±4.1) μg/m3, respectively, during the haze episodes, which were 4.3, 3.4, 4.1, and 1.7 times those in the non-haze days. The SO2-4,NO-3, NH+4, and SOA accounted for 15.8%, 8.8%, 7.3%, and 6.0% of the total mass concentration of PM10 during the non-haze days. The respective contributions of SNA species to PM10 rose to about27.2%, 15.9%, and 13.9% during the haze days, while the contributions of SOA maintained the same level with a slight decrease to about 4.9%. The observed mass concentrations of SNA and SOA increased with the increase of PM10 mass concentration, however, the rate of increase of SNA was much faster than that of the SOA. The SOR(sulfur oxidation ratio) and NOR(nitrogen oxidation ratio) increased from non-haze days to hazy days, and increased with the increase of RH. 8The influence of stage at diagnosis and molecular subtype on breast cancer patient survival: a hospital.based multi.center study显示文摘Background: Stage at diagnosis and molecular subtype are important clinical factors associated with breast cancer patient survival. However, subgroup survival data from a large study sample are limited in China.To estimate the survival differences among patients with different stages and various subtypes of breast cancer, we conducted a hospital-based multi-center study on breast cancer in Beijing, China.Methods: All resident patients diagnosed with primary, invasive breast cancer between January 1,2006 and December 31,2010 from four selected hospitals in Beijing were included and followed up until December 31,2015. Hospitalbased data of stage at diagnosis, hormone receptor status, and selected clinical characteristics, including body mass index(BMI), menopausal status, histological grade, and histological type, were collected from the medical records of the study subjects. Overall survival(OS) and cancer-specific survival(CSS) were estimated. Cox proportional hazards models were employed to evaluate the associations of stage at diagnosis and molecular subtype with patient survival.Results: The 5-year OS and CSS rates for all patients were 89.4% and 90.3%. Survival varied by stage and molecular subtype. The 5-year OS rates for patients with stage I, Ⅱ, Ⅲ, and IV diseases were 96.5%, 91.6%, 74.8%, and 40.7%,respectively, and the corresponding estimates of 5-year CSS rates were 97.1%, 92.6%, 75.6%, and 42.7%, respectively.The 5-year OS rates for patients with luminal A, luminal B, HER2, and triple-negative subtypes of breast cancer were92.6%, 88.4%, 83.6%, and 82.9%, respectively, and the corresponding estimates of 5-year CSS rates were 93.2%, 89.1 %,85.4%, and 83.5%, respectively. Multivariate analysis showed that stage at diagnosis and molecular subtype were important prognostic factors for breast cancer.Conclusions: Survival of breast cancer patients varied significantly by stage and molecular subtype. Cancer screening is encouraged for the early detection and early diagnosis of breast cancer. 9Structure, evolution and regional tectonic implications of the Queshan metamorphic core complex in eastern Jiaodong Peninsula of China显示文摘The Queshan MCC is an important example of a crustal extensional structure in the eastern Jiaodong Peninsula along the southeastern margin of the NCC in the Early Cretaceous. The MCC is a typical Cordilleran-type core complex with a three-layered structure:(1) the upper plate is constituted by the Cretaceous supradetachment basin and Paleoproterozoic basement;(2) the lower plate comprises the Neoarchean high-grade metamorphic complexes and late Mesozoic granitic intrusions; and(3) the two plates are separated by a master detachment fault. A series of late NEN-oriented brittle faults superimposed on and destructed the early MCC. Petrology, geometry, kinematics, macro- and micro-structures and quartz c-axis fabrics imply that the MCC has a progressive exhumation history from middle-lower to subsurface level(via middle-upper crustal level) under the nearly WNW-ESE regional extensional regime. We present structural and geochronological evidence to constrain the exhumation of the Queshan MCC from ca. 135 to 113 Ma. Based on the comprehensive analysis of the different patterns of extensional structures in the Jiaodong and Liaodong Peninsula, we have defined the Jiao-Liao Early Cretaceou extensional province and further divided the crustal extension of it into two stages: the first stage was the intense flow of the middle-lower crust and the second stage was the extension of the middle-upper crust. 11Inhibition of SIRT6 in prostate cancer reduces cell viability and increases sensitivity to chemotherapeutics显示文摘SI RT6 is an important histone modifying protein that regulates DNA repair,telomere maintenance,energy me-tabolism,and target gene expression.Recently SIRT6 has been identifi ed as a tumor suppressor and is down-regulated in certain cancer types,but not in other can-cers.From deposited gene profi ling studies we found that SIRT6 was overexpressed in prostate tumors,compared with normal or paratumor prostate tissues.Tissue micro-array studies confi rmed the higher levels of SIRT6 in both prostate tumor tissues and prostate cancer cells than in their normal counterparts.Knockdown of SIRT6 in human prostate cancer cells led to sub-G1 phase arrest of cell cy-cle,increased apoptosis,elevated DNA damage level and decrease in BCL2 gene expression.Moreover,SIRT6-de-fi ciency reduced cell viability and enhanced chemothera-peutics sensitivity.Taken together,this study provides the fi rst evidence of SIRT6 overexpression in human prostate cancer,and SIRT6 regulation could be exploited for pros-tate cancer therapy.Yewei Liu Qian Reuben Xie Boshi Wang Jiaxiang Shao Tingting Zhang Tengyuan Liu Gang Huang Weiliang Xia 2013Protein & Cell2013,4,9:17
12Mitochondrial DNA variation analysis suggests extreme low genetic diversity in Guizhou snub-nosed monkeys (Rhinopithecus brelichi)显示文摘Guizhou snub-nosed monkey (Rhinopithecus brelichi) is a unique, endangered primate in China, mainly distributed in Fanjing Mountain National Natural Reserve, Guizhou Province, in an area of 275 km2. Recently, habitat loss and fragmentation have caused population isolation. To assess genetic diversity within this species and its population structure, we sequenced 400 bp of the hypervariable I segment from the mitochondrial DNA control region for 128 individuals. Only one haplotype was identified from these individuals. 13Mouse macrophage specific knockout of SIRT1 influences macrophage polarization and promotes angiotensin Ⅱ-induced abdominal aortic aneurysm formation显示文摘Abdominal aortic aneurysm(AAA) is a vascular degenerative disease. Macrophage polarization and the balance between classically activated macrophages(M1) and alternatively activated macrophages(M2)are crucial for AAA pathogenesis. The present study aims to investigate the roles of macrophage SIRT1 in AAA formation and macrophage polarization. We found that in mouse peritoneal macrophages, SIRT1 expression was decreased after M1 stimulation, but was enhanced after M2 stimulation. Results from SIRT1^(flox/flox) mice and macrophage specific SIRT1 knockout mice with treatment of angiotensin II(Ang II)for 4 weeks showed that macrophage specific deficiency of SIRT1 increased the incidence of AAA and exacerbated the severity, including more severe aneurysm types, enlarged diameter of the aneurysm and increased degradation of elastin. In mouse aortas, SIRT1 deficiency increased the proinflammatory M1 molecule inducible nitric oxide synthase(i NOS), and decreased M2 molecules such as arginase 1(Arg1) and mannose receptor(MR). Furthermore, in peritoneal macrophages, SIRT1 deficiency increased the expression of M1 inflammatory molecules, but decreased the expression of M2 molecules. Overexpression of SIRT1 had the opposite effects. 14Chemical and optical properties of aerosols and their interrelationship in winter in the megacity Shanghai of China显示文摘A field campaign on air quality was carried out in Shanghai in winter of 2012. The concentrations of NO, NO2, NOx, SO2, CO, and PM2.5increased during haze formation. The average masses of SO42-, NO3-and NH4+were 10.3, 11.7 and 6.7 μg/m3 during the haze episodes, which exceeded the average(9.2, 7.9, and 3.4 μg/m3) of these components in the non-haze days. The mean values for the aerosol scattering coefficient(b sp), aerosol absorption coefficient(b ap) and single scattering albedo(SSA) were 288.7, 27.7 and0.91 Mm-1, respectively. A bi-peak distribution was observed for the mass concentrations of CO, NO, NO2, and NOx. More sulfate was produced during daytime than that in the evening due to photochemical reactions. The mass concentration of NH4+achieved a small peak at noontime. NO3-showed lower concentrations in the afternoon and higher concentrations in the early morning. There were obvious bi-peak diurnal patterns for b sp and b ap as well as SSA. b sp and b ap showed a positive correlation with PM2.5mass concentration.(NH4)2SO4, NH4NO3, organic mass, elemental carbon and coarse mass accounted for 21.7%, 19.3%, 31.0%, 9.3% and 12.3% of the total extinction coefficient during non-haze days, and 25.6%, 24.3%, 30.1%, 8.1% and 8.2% during hazy days. Organic matter was the largest contributor to light extinction. 15Arginine, N-carbamylglutamate,and glutamine exert protective effects against oxidative stress in rat intestine显示文摘The objective of the current study is to evaluate the effects of dietary supplementation with arginine(ARG), N-carbamylglutamate(NCG), and glutamine(GLN) on rat intestinal morphology and antioxidant status under oxidative stress. Rats were fed for 30 d with one of the following iso-nitrogenous diets:basal diet(BD), BD plus 1% ARG, BD plus 0.1% NCG, and BD plus 1% GLN. On day 28, half of the rats fed BD were intraperitoneally injected with 12 mg/kg body weight of diquat(DT; i.e., the DT group) and the other half was intraperitoneally injected with sterile solution(i,e., the control group). The other diet groups were intraperitoneally injected with 12 mg/kg body weight of DT(i,e., DT + 1% GLN [DT + GLN],DT + 1% ARG [DT + ARG], and DT + 0.1% NCG [DT + NCG]). Rat jejunum samples obtained at 48 h after DT injection were analyzed. Results showed that DT significantly decreased catalase(CAT) activity and glutathione(GSH) content by 58.25% and 56.57%, respectively, and elevated malondialdehyde(MDA)content and crypt depth(CD) by 19.39% and 22.13%, respectively, in the jejunum(P < 0.05, relative to the control group). Compared with the DT group, the DT + GLN group exhibited significantly improved villus height(VH), villus width(VW), villus surface area(VSA), CD and total antioxidant capacity(T-AOC) activity(P < 0.05); the DT + ARG group exhibited significantly increased the ratio of VH to CD(H:D) and TAOC activity(P < 0.05); the DT + GLN, DT + ARG and DT + NCG groups exhibited significantly enhanced CAT activity and GSH content as well as decreased MDA content(P < 0.05). Moreover, VH, VW, VSA, CD and GSH content in the DT + GLN group were higher whereas MDA content was lower compared with the corresponding values observed in both the DT + ARG and the DT + NCG groups(P < 0.05). The H:D ratio in the DT + ARG group significantly increased compared with that in the DT + NCG and DT + GLN groups(P < 0.05). 17Comprehensive analysis of NAC transcription factors in diploid Gossypium: sequence conservation and expression analysis uncover their roles during fiber development显示文摘Determining how function evolves following gene duplication is necessary for understanding gene expansion.Transcription factors(TFs)are a class of proteins that regulate gene expression by binding to specific cis-acting elements in the promoters of target genes,subsequently activating or repressing their transcription.In the present study,we systematically examined the functional diversification of the NAC transcription factor(NAC-TFs)family by analyzing their chromosomal location,structure,phylogeny,and expression pattern in Gossypium raimondii(Gr)and G.arboreum(Ga).The 145 and 141 NAC genes identified in the Gr and Ga genomes,respectively,were annotated and divided into 18 subfamilies,which showed distinct divergence in gene structure and expression patterns during fiber development.In addition,when the functional parameters were examined,clear divergence was observed within tandem clusters,which suggested that subfunctionalization had occurred among duplicate genes.The expression patterns of homologous gene pairs also changed,suggestive of the diversification of gene function during the evolution of diploid cotton.These findings provide insights into the mechanisms underlying the functional differentiation of duplicated NAC-TFs genes in two diploid cotton species.Haihong Shang Zhongna Wang Changsong Zou Zhen Zhang Weijie Li Junwen Li Yuzhen Shi Wankui Gong Tingting Chen Aiying Liu Juwu Gong Qun Ge Youlu Yuan 2016Science China(Life Sciences)2016,59,2:12
18Lightweight and High-Performance Microwave Absorber Based on 2D WS2-RGO Heterostructures显示文摘Two-dimensional(2D)nanomaterials are categorized as a new class of microwave absorption(MA)materials owing to their high specific surface area and peculiar electronic properties.In this study,2D WS2-reduced graphene oxide(WS2-rGO)heterostructure nanosheets were synthesized via a facile hydrothermal process;moreover,their dielectric and MA properties were reported for the first time.Remarkably,the maximum reflection loss(RL)of the sample-wax composites containing 40 wt% WS2-rGO was-41.5 dB at a thickness of 2.7 mm;furthermore,the bandwidth where RL<-10 dB can reach up to 13.62 GHz(4.38-18 GHz).Synergistic mechanisms derived from the interfacial dielectric coupling and multiple-interface scattering after hybridization of WS2 with rGO were discussed to explain the drastically enhanced microwave absorption performance.The results indicate these lightweight WS2-rGO nanosheets to be potential materials for practical electromagnetic wave-absorbing applications.Deqing Zhang Tingting Liu Junye Cheng Qi Cao Guangping Zheng Shuang Liang Hao Wang MaoSheng Cao 2019Nano-Micro Letters2019,11,3:11
19The Coix Genome Provides Insights into Panicoideae Evolution and Papery Hull Domestication显示文摘Coix is a grass crop domesticated as early as the Neolithic era.It is still widely cultivated for both highly nutritional food and medicinal use.However,the genetic study and breeding of this crop are hindered by the lack of a sequenced genome.Here,we report de novo sequencing and assembly of the 1619-Mb genome of Coix,and annotation of 75.39%repeats and 39629 protein-coding genes.Comparative genomics analysis showed that Coix is more closely related to sorghum than maize,but intriguingly only Coix and maize had a recent genome duplication event,which was not detected in sorghum.We further constructed a genetic map and mapped several important traits,especially the strength of hull.Selection of papery hull(thin:easy dehulling)from the stony hull(thick:difficult dehulling)in wild progenitors was a key step in Coix domestication.The papery hull makes seed easier to process and germinate.Anatomic and global transcriptome analysis revealed that the papery hull is a result of inhibition of cell division and wall biogenesis.We also successfully demonstrated that seed hull pressure resistance is controlled by two major quantitative trait loci(QTLs),which are associated with hull thickness and color,respectively.The two QTLs were further fine mapped within intervals of 250 kb and 146 kb,respectively.These resources provide a platform for evolutionary studies and will facilitate molecular breeding of this important crop.Chao Guo Yanan Wang Aiguo Yang Jun He Chaowen Xiao Shanhua Lv Fengming Han Yibing Yuan Yuan Yuan Xiaolong Dong Juan Guo YawenYang-Hailan Liu Ningzhi Zuo Yaxi Hu Kangxu Zhao Zhengbo Jiang Xing Wang Tingting Jiang Yaou Sherf Moju Cao Yuan Wang Zhaobo Long Tingzhao Rong Luqi Huang Shufeng Zhou 2020Molecular Plant2020,13,2:11
