1Expression of Overlapping PreS1 Fragment Recombinant Proteins for the Determination of Immunogenic Domains in HBsAg PreS1 Region显示文摘In this study, eight preS1 fragments overlapped in preS1 (21–119) region of HBV adr subtype, i.e. preS1 (21–47), preS1 (34–59), preS1 (48–70), preS1 (60–85), preS1 (71–94), preS1 (86–109), preS1 (95–119) and preS1 (21–119), were cloned by PCR, and expressed as GST fusion proteins. These GST- preS1 fusion proteins were highly expressed in soluble form in E. coli, and about 50 to 90 mg soluble fusion proteins were purified from 1 L culture. Using these fusion proteins, the immunogenic domains in preS1 (21– 119) region were identified by Western blot analysis and competitive ELISA. The results showed that the immunogenic domains mainly existed in preS1 (21–59) in N-terminus and preS1 (95–109) in C-terminus, and more importantly, a major immunogenic domain preS1 (34–59), which has much stronger immunogenicity, was identified. It was also supported by the predictions of secondary structure and immunological property in the preS1 (21–119) region. 2Exotic origin of the Chinese continental shelf: new insights into the tectonic evolution of the western Pacific and eastern China since the Mesozoic显示文摘The effect of paleo-Pacific subduction on the geological evolution of the western Pacific and continental China is likely complex. Nevertheless, our analysis of the distribution of Mesozoic granitoids in the eastern continental China in space and time has led us to an interesting conclusion: The basement of the continental shelf beneath East and South China Seas may actually be of exotic origin geologically unrelated to the continental lithosphere of eastern China. By accepting the notion that the Jurassic–Cretaceous granitoids in the region are genetically associated with western Pacific subduction and the concept that subduction may cease to continue only if the trench is being jammed, then the termination of the granitoidmagmatism throughout the vast region at *88 ± 2 Ma manifests the likelihood of ‘‘sudden'', or shortly beforehand(*100 Ma), trench jam of the Mesozoic western Pacific subduction. Trench jam happens if the incoming‘‘plate'' or portion of the plate contains a sizeable mass that is too buoyant to subduct. The best candidate for such a buoyant and unsubductable mass is either an oceanic plateau or a micro-continent. We hypothesize that the basement of the Chinese continental shelf represents such an exotic, buoyant and unsubductable mass, rather than seaward extension of the continental lithosphere of eastern China. The locus of the jammed trench(i.e., the suture) is predictably located on the shelf in the vicinity of, and parallel to, the arc-curved coastal line of the southeast continental China. It is not straightforward to locate the locus in the northern section of the East China Sea shelf because of the more recent(\20 Ma) tectonic re-organization associated with the opening of the Sea of Japan. We predict that the trench jam at *100 Ma led to the re-orientation of the Pacific plate motion in the course of NNW direction as inferred from the age-progressive Emperor Seamount Chain of Hawaiian hotspot origin(its oldest unsubdued Meiji and Detroit seamounts are *82 Ma),making the boundary between the Pacific plate and the newly accreted plate of eastern Asia a transform fault at the location east of the continental shelf of exotic origin. This explains the apparent *40 Myr magmatic gap from *88to *50 Ma prior to present-day western Pacific subduction initiation. We propose that basement penetration drilling on well-chosen sites is needed to test the hypothesis in order to reveal the true nature of the Chinese continental shelf basement. This testing becomes critical and cannot longer be neglected in order to genuinely understand the tectonic evolution of the western Pacific and its effect on the geology of eastern China since the Mesozoic, includingthe cratonic lithosphere thinning, related magmatism/mineralization, and the mechanism of the subsequent South China Sea opening, while also offering novel perspectives on aspects of the plate tectonics theory. We also suggest the importance of future plate tectonic reconstruction of the western Pacific to consider the nature and histories of the Chinese continental shelf of exotic origin as well as the probable transform plate boundary from *100 to*50 Ma. 3Late Cretaceous-Cenozoic exhumation history of Tiantangzhai region of Dabieshan Orogen: Constraints from (U-Th)/He and fission track analysis显示文摘Integrated fission track and (U-Th)/He analysis is carried out on 6 apatite and 6 zircon samples from a near vertical section in The Tiantangzhai region at the core of the present Dabieshan orogen. The result shows that the region experienced cooling/exhumation during the Late Cretaceous and Early Tertiary period. Age-elevation relationships for different dating systems and different minerals suggest a pulse of rapid exhumation at ~110 Ma before present, preserved in the structurally highest samples. At lower elevations, ages begin to decrease with decreasing elevation, suggesting lower exhumation rates since 90 Ma. Two periods of different exhumation rates are identified since 90 Ma. 4Suppression of bcl-2 Gene by RNA Interference Increases Chemosensitivity to Cisplatin in Nasopharyngeal Carcinoma Cell Line CNE1显示文摘To explore the effect of suppressing BCL-2 expression using RNA interference (RNAi) technique in nasopharyngeal carcinoma cell line CNE 1. CNE 1 cell lines stably expressing shRNAs targeted bcl-2 and GL3 gene were established and gene expression inhibition was assessed by Western blotting analysis. The effect of suppressing bcl-2 by RNAi on cell growth was studied, the apoptosis induction and the sensitization of CNE 1 cells to cisplatin were quantified by MTT assay and flow cytometry. The results showed that: stable transfection of CNE 1 cells with vectors expressing shRNAs against bcl-2 decreased the expression of BCL-2 protein; suppression of BCL-2 expression did not affect cell proliferation but could increase the chemosensitivity to cisplatin in CNE 1 cells. 5Proteomic Comparison of Two-Dimensional Gel Electrophoresis Profiles from Human Lung Squamous Carcinoma and Normal Bronchial Epithelial Tissues显示文摘Differential proteome profiles of human lung squamous carcinoma tissue compared to paired tumor-adjacent normal bronchial epithelial tissue were established and analyzed by means of immobilized pH gradient-based two-dimensional polyacrylamide gel electrophoresis (2-D PAGE) and matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS). The results showed that well-resolved, reproducible 2-DE patterns of human lung squamous carcinoma and adjacent normal bronchial epithelial tissues were obtained under the condition of 0.75-ug protein-load. The average deviation of spot position was 0.733+0.101 mm in IEF direction, and 0.925+0.207 mm in SDS-PAGE direction. For tumor tissue, a total of 1241±88 spots were detected, 987±65 spots were matched with an average matching rate of 79.5%. For control, a total of 1190+72 spots were detected, and 875±48 spots were matched with an average matching rate of 73.5%. A total of 864±34 spots were matched between tumors and controls.Forty-three differential proteins were characterized: some proteins were related to oncogenes, and others involved in the regulation of cell cycle and signal transduction. 8Effect of Yiqibushenhuoxue decoction on chronic obstructive pulmonary disease measured by St. George's respiratory disease questionnaire scores and forced expiratory volume显示文摘OBJECTIVE: To investigate the effects of Yiqibushenhuoxue decoction on stable chronic obstructive pulmonary disease(COPD) by observing its influences on patients' quality of life and airway inflammation.METHODS: Seventy patients with stable COPD were randomly divided into a treatment group(n=35) treated with Yiqibushenhuoxue decoction plus Seretide and a control group(n=35) treated with Seretide only.The dosage of Yiqibushenhuoxue decoction was 100 mL each time, twice a day, and the dosage of Seretide was salmeterol 50 μg/fluticasone 250 μg twice a day. Both groups were treated for 12 weeks. Before and after the treatment, St George's respiratory disease questionnaire(SGRQ)scores,forced expiratory volume,and forced expiratory volume in 1 second/forced vital capacity(FEV1/FVC)were measured.RESULTS:The SGRQ scores in both groups were significantly lower than those before treatment(P<0.05). After treatment, the total SGRQ scores and each subscore in the treatment group were significantly lower than those in the control group(P<0.05). The percentage of the predicted FEV1% and FEV1/FVC were higher in both groups, but no statistical differences were detected from before to after the treatment or between the two groups(P>0.05).CONCLUSION: Yiqibushenhuoxue decoction could significantly decrease the SGRQ scores in patients with stable COPD, which suggests that it is able to improve patient symptoms.Jianqiu Liu Fengli Gao Zhuying Li 2014Journal of Traditional Chinese Medicine2014,34,4:8
9Identification of the immunogenic domains in HBsAg preS1 region using overlapping preS1 fragment fusion proteins显示文摘AIM: The incorporation of hepatitis B virus (HBV) preS1 region into epitope-based vaccines against HBV has been accepted widely, but the incorporate site and size of preS1 sequence is controversial. Therefore our purpose was to further investigate its immunogenic domains for the epitopebased hepatitis B vaccine design.METHODS: Eight GST fusion proteins containing overlapping preS1 fragments in preS1 (21-119) region were expressed in E.coli. Using these purified fusion proteins, the immunogenic domains in preS1 region were identified in detail in mice and humans by Western blot analysis and ELISA.RESULTS: The results in mice showed that the immunogenic domains mainly existed in preS1 (21-59) and preS1 (95-109). Similarly, these fragments had strong immunogenicity in humans; whereas the other parts except for preS1 (60-70) also had some immunogenicity.More importantly, a major immunogenic domain, preS1 (34-59), which has much stronger immunogenicity, was identified. Additionally, the antibodies against some preS1 fragments, especially preS1 (34-59), were speculated to be virus-neutralizing.CONCLUSION: Eight GST fusion proteins containing overlapping preS1 fragments were prepared successfully. They were used for the study on the immunogenic domains in preS1 (21-119) region. The preS1 (34-59) fragments were the major immunogenic domains in the preS1 region, and the antibodies against these fragments were speculated to be virus-neutralizing. Therefore, the incorporation of preS1 (34-59) fragments into epitopebased HBV vaccines may be efficient for enhancement of immune response. 10Wide-steering-angle high-resolution optical phased array显示文摘Optical phased array(OPA)technology is considered a promising solution for solid-state beam steering to supersede the traditional mechanical beam steering.As a key component of the LIDAR system for long-range detection,OPAs featuring a wide steering angle and high resolution without beam aliasing are highly desired.However,a wide steering range requires a waveguide pitch less than half of the wavelength,which is easily subjected to cross talk.Besides,high resolution requires a large aperture,and it is normally achieved by a high count number of waveguides,which complicates the control system.To solve the mentioned issues,we design two high-performance 128-channel OPAs fabricated on a multilayered SiN-on-SOI platform.Attributed to the nonuniform antenna pitch,only 128 waveguides are used to achieve a 4 mm wide aperture.Besides,by virtue of innovative dual-level silicon nitride(Si_(3)N_(4))waveguide grating antennas,the fishbone antenna OPA achieves a 100°×19.4°field of view(FOV)with divergence of 0.021°×0.029°,and the chain antenna OPA realizes a 140°×19.23°FOV with divergence of 0.021°×0.1°.To our best knowledge,140°is the widest lateral steering range in two-dimensional OPA,and 0.029°is the smallest longitudinal divergence.Finally,we embed the OPA into a frequency-modulated continuous-wave system to achieve 100 m distance measurement.The reflected signal from 100 m distance is well detected with 26 dBm input transmitter power,which proves that OPA serves as a promising candidate for transceiving optical signal in a LIDAR system.YINGZHI LI BAISONG CHEN QUANXIN NA QIJIE XIE MIN TAO LANXUAN ZHANG ZIHAO ZHI YUXUAN LI XIAOBIN LIU XIANSHU LUO GUOQIANG LO FENGLI GAO XUEYAN LI JUNFENG SONG 2021Photonics Research2021,9,12:7

12Stress field modeling of northwestern South China Sea since 5.3 Ma and its tectonic significance显示文摘Tectonically,the northwestern South China Sea(SCS)is located at the junction between three micro-plates,i.e.,the Indochina,South China and Zhongsha-Xisha micro-plates,and involves three basins,i.e.,the Yinggehai Basin,the Qiongdongnan Basin and Xisha Trough in the east,and the Zhongjiannan Basin in the south.Since the Pliocene(5.3 Ma),the Yinggehai Basin has experienced repeated accelerating subsidence,high thermal fluid,and widely developing mud-rich overpressure chambers,abundant mud diapers and crust-mantle mixed CO2.While a large central canyon was developed in the Qiongdongnan Basin,new rift occurred in the Xisha Trough.These characteristics demonstrate a single tectonic unit for the northwestern SCS,for which we have undertaken stress field modeling to understand its plate deformations and sedimentary responses.Our results demonstrate that an extension tectonic event occurred after 5.3 Ma in the Yinggehai-Qiongdongnan-Xisha trough area,which is characterized by thinner crust(<16 000 m),half-graben or graben structural style and thicker sedimentary sequences(>3 500 m).A new rift system subsequently was developed in this area;this event was mainly driven by the combined effects of different movement velocity and direction of the three micro-plates,and the far-field effect of the continental collision between the Indian Plate and the Tibetan Plateau,and subduction of the Pacific Plate underneath the Eurasian Plate.YANG Fengli ZHOU Zuyi ZHANG Na LIU Ning NI Bin 2013Acta Oceanologica Sinica2013,32,12:5
13Chemical composition,energy and amino acid digestibility in double-low rapeseed meal fed to growing pigs显示文摘Background:The nutritional value of rapeseed meal may be variable due to the variation of its chemical composition.And a precise understanding of the nutritional value of an ingredient is beneficial for the accurate diet formulation and reduction of feed costs.This study was conducted to determine the chemical composition,digestible energy(DE) and metabolizable energy(ME) content,and apparent ileal digestibility(AID) and standardized ileal digestibility(SID) of amino acids(AA) for growing pigs.Thirteen solvent-extracted double-low rapeseed meal(DLRSM) samples were obtained from the main double-low rapeseed producing areas in China.Methods:The DE and ME contents of the 13 DLRSM samples were measured in growing pigs(six pigs per DLRSM sample,average initial body weight(BW) = 48.3 kg).The AID and SID of AA of 10 DLRSM samples were determined in 12 crossbred barrows(average initial BW = 35.3 kg) by using two 6x6 Latin square designs.Each Latin square comprised one N-free diet and 5 DLRSM test diets.Results:The chemical composition of DLRSM varied among samples,and the coefficient of variation was greater than 10%for ether extract(EE),neutral detergent fiber(NDF),acid detergent fiber(ADF),calcium(Ca),and total glucosinolates.The AA content of DLRSM varied among samples especially for lysine(Lys) and methionine(Met).On a dry matter(DM) basis,the apparent total tract digestibility(ATTD) of gross energy(GE),the DE and ME and the ME:DE ratio of DLRSM averaged 62.39%,2862 kcal/kg and 2723 kcal/kg,and 94.95%,respectively.The mean value of SID of Lys was 70.52% which varied from 66.54-76.54%.The SID of crude protein(CP),Met,and threonine(Thr) averaged 72.81%,82.41%,and 69.76%,respectively.Conclusions:There was great variability in chemical composition especially in the concentration of EE,NDF and ADF,but no significant differences in energy content of the DLRSM samples were observed.In addition,the AID and SID of all AA were relatively similar among DLRSM samples except for that of Lys.Peili Li Fengli Wang Fei Wu Jinrong Wang Ling Liu Changhua Lai 2015Journal of Animal Science and Biotechnology2015,6,4:5

15Determination of the Observation Area for Metallographic Microstructure of GH4033 Superalloy in a Uniaxially Thermo-Mechanical Compression Experiment显示文摘Based on two-dimensional(2D) rigid-plastic finite element(FE) method, the optimum observation area for metallographic microstructure and the influence of the height–diameter ratio on which were analyzed for the cylindrical samples with the diameter of 8 mm and different heights from 8 to 16 mm in nonuniform compressive experiments. It is shown that the representative metallographic observation area relevant to the applied deformation condition is located at about 0.835 of the radius from the center of the sample to the solder joint between the sample and the thermocouples. At the same time, the microstructure in that area is more appropriate as the height of the sample is 12 mm. 16Scalar-matrix-structured ghost imaging显示文摘The features of the characteristic matrix used in linear intensity correlation reconstruction methods are directly related to the quality of ghost imaging. In order to suppress the noise caused by the off-diagonal elements in the characteristic matrix, we propose a reconstruction method for ghost imaging called scalar-matrix-structured ghost imaging(SMGI). The characteristic matrix is made to approximate a scalar matrix by modifying the measurement matrix. 17Experimental Investigation on the Hydrodynamic Characteristics of Fluidized Bed Particle Solar Receiver with Gas-Solid Countercurrent Flow Pattern显示文摘To design a particle solar receiver(PSR),a vital energy conversion system,is still a bottleneck for researchers.This study presents a novel PSR based on countercurrent fluidized bed(CCFB)technology,named CCFB receiver.In this design,downward-moving particles are subjected to the action of an up-flow gas to reduce the falling speed and enhance the radial disturbance,and hence increase the residence time of particles and improve the heat transfer.A cold-mold visual experimental setup is established.The influence factors are investigated experimentally,including the superficial gas velocity,solid flux,aeration gas,particle size and transport tube diameter.The results indicate that the maximum solid holdup can exceed 9%or so with fine particles of diameter d_(p)=113.5 μm and a tube diameter of 40 mm.It is proved that the CCFB can operate stably and adjust the solid flux rapidly.The results of this study provide a new structure for PSRs in the concentrated solar power field and could fill the research insufficiency in the gas-solid counterflow field.JIANG Kaijun WANG Fengli KONG Yanqiang XU Chao DU Xiaoze 2021Journal of Thermal Science2021,30,6:4
18Preparation and Primary Application of Monoclonal Antibodies against a Novel Ribosome-inactivating Protein Moschatin from Pumpkin Seeds显示文摘Plant ribosome-inactivating proteins (RIPs) have multiple biological functions, and have beenwidely used in the studies on biomedical and agronomic applications. Moschatin is a novel single-chain RIPrecently purified from pumpkin seeds, and it has been successfully applied to construct the immunotoxin thatcan selectively kill the cultured human melanoma cells. Six stable strains of hybridomas (2H8, 4A8, 5B6,6F8, 4H10 and 6C2) that can secrete high specific monoclonal antibodies against Moschatin have beensuccessfully prepared using hybridoma technique. 19Petrogenesis and tectonic implications of the Triassic rhyolites in the East Kunlun Orogenic Belt, northern Tibetan Plateau显示文摘The East Kunlun Orogenic Belt(EKOB),which is in the northern part of the Greater Tibetan Plateau,contains voluminous Late Triassic intermediate-felsic volcanic rocks.In the east end of the EKOB,we identified highly differentiated peralkaline-like Xiangride rhyolites(~209 Ma)that differ from the widespread andesitic-rhyolitic Elashan volcanics(~232–225 Ma)in terms of their field occurrences and mineral assemblages.The older,more common calc-alkaline felsic Elashan volcanics may have originated from partial melting of the underthrust Paleo-Tethys oceanic crust under amphibolite facies conditions associated with continental collision.The felsic Elashan volcanics and syn-collisional granitoids of the EKOB are different products of the same magmatic event related to continental collision.The Xiangride rhyolites are characterized by elevated abundances of high field strength elements,especially the very high Nb and Ta contents,the very low Ba,Sr,Eu,P,and Ti contents;and the variably high ^(87)Sr/^(86)Sr ratios(up to 0.96),exhibiting remarkable similarities to the characteristic peralkaline rhyolites.The primitive magmas parental to the Xiangride rhyolites were most likely alkali basaltic magmas that underwent protracted fractional crystallization with continental crust contamination.The rock associations from the early granitoids and calc-alkaline volcanic rocks to the late alkaline basaltic dikes and peralkaline-like rhyolites in the Triassic provide important information about the tectonic evolution of the EKOB from syn-collisional to post-collisional.We infer that the transition from collisional compression to postcollisional extension occurred at about 220 Ma.Fengli Shao Yaoling Niu Juanjuan Kong Yi Liu Guodong Wang Yu Zhang 2021Geoscience Frontiers2021,12,6:3
