1Transcriptional activation and phosphorylation of OsCNGC9 confer enhanced chilling tolerance in rice显示文摘Low temperature is a major environmental factor that limits plant growth and productivity.Although transient elevation of cytoplasmic calcium has long been recognized as a critical signal for plant cold tolerance,the calcium channels responsible for this process have remained largely elusive.Here we report that OsCNGC9,a cyclic nucleotide-gated channel,positively regulates chilling tolerance by mediating cytoplasmic calcium elevation in rice(Oryza sativa).We showed that the loss-of-function mutant of OsCNGC9 is defective in cold-induced calcium influx and more sensitive to prolonged cold treatment,whereas OsCNGC9 overexpression confers enhanced cold tolerance.Mechanistically,we demonstrated that in response to chilling stress,OsSAPK8,a homolog of Arabidopsis thaliana OST1,phosphorylates and activates OsCNGC9 to trigger Ca2+influx.Moreover,we found that the transcription of OsCNGC9 is activated by a rice dehydration-responsive element-binding transcription factor,OsDREB1A.Taken together,our results suggest that OsCNGC9 enhances chilling tolerance in rice through regulating cold-induced calcium influx and cytoplasmic calcium elevation.Jiachang Wang Yulong Ren Xi Liu Sheng Luo Xiao Zhang Xin Liu Qibing Lin Shanshan Zhu Hua Wan Yang Yang Yu Zhang Bin Lei Chunlei Zhou Tian Pan Yongfei Wang Mingming Wu Ruonan jing Yang Xu Meng Han Fuqing Wu Cailin Lei Xiuping Guo Zhijun Cheng Xiaoming Zheng Yihua Wang Zhigang Zhao Ling Jiang Xin Zhang Yong-Fei Wang Haiyang Wang Jianmin Wan 2021Molecular Plant2021,14,2:10
2Curcumenol triggered ferroptosis in lung cancer cells via lncRNA H19/miR-19b-3p/FTH1 axis显示文摘Curcumenol,an effective ingredient of Wenyujin,has been reported that exerted its antitumor potential in a few cancer types.However,the effect and molecular mechanism of curcumenol in lung cancer are largely unknown.Here,we found that curcumenol induced cell death and suppressed cell proliferation in lung cancer cells.Next,we demonstrated that ferroptosis was the predominant method that contributed to curcumenol-induced cell death of lung cancer in vitro and vivo for the first time.Subsequently,using RNA sequencing,we found that the long non-coding RNA H19(lncRNA H19)was significantly downregulated in lung cancer cells treated with curcumenol,when compared to untreated controls.Overexpression of lncRNA H19 eliminated the anticancer effect of curcumenol,while lncRNA H19 knockdown promoted ferroptosis induced by curcumenol treatment.Mechanistically,we showed that lncRNA H19 functioned as a competing endogenous RNA to bind to miR-19b-3p,thereby enhanced the transcription activity of its endogenous target,ferritin heavy chain 1(FTH1),a marker of ferroptosis.In conclusion,our data show that the natural product curcumenol exerted its antitumor effects on lung cancer by triggering ferroptosis,and the lncRNA H19/miR-19b-3p/FTH1 axis plays an essential role in curcumenol-induced ferroptotic cell death.Therefore,our findings will hopefully provide a valuable drug for treating lung cancer patients.Ruonan Zhang Ting Pan Yu Xiang Mingming Zhang Han Xie Zimao Liang Bi Chen Cong Xu Jing Wang Xingxing Huang Qianru Zhu Ziming Zhao Quan Gao Chengyong Wen Wencheng Liu Weirui Ma Jiao Feng Xueni Sun Ting Duan Elaine Lai-Han Leung Tian Xie Qibiao Wu Xinbing Sui 2022Bioactive Materials2022,7,7:8
3High levels of circulating GM-CSF^(+) CD4^(+) T cells are predictive of poor outcomes in sepsis patients: a prospective cohort study显示文摘Granulocyte colony-stimulating factor(GM-CSF),produced by CD4^(+)T cells,has recently been implicated in the pathogenesis of inflammatory diseases,such as multiple sclerosis and juvenile arthritis.However,the role of GM-CSF-producing CD4^(+)T cells in sepsis remains unknown.This study reports peripheral changes in GM-CSF-producing CD4^(+)T cells in septic patients and the possible underlying mechanism by which GM-CSF influences the outcome of sepsis.Forty-three septic patients,20 SIRS patients,and 20 healthy controls were enrolled in this study and followed for 28 days to assess mortality.We measured the peripheral frequency of GM-CSF^(+)CD4^(+)T cells and recorded their associated relationship with disease progression.Our data demonstrated that peripheral GM-CSF-producing CD4^(+)T cells were significantly higher in septic patients than in both SIRS patients and healthy controls.These cells exhibit a memory phenotype and impaired IFN-γ-secreting capacity in sepsis patients.Using a receiver operating curve analysis with 8.01%as a cut-off point,the percentage of GM-CSF^(+)CD4^(+)T cells could predict the outcome of septic patients.Combined with the increase in GM-CSF-producing CD4^(+)T cells,inflammatory cytokines IL-1βand IL-6 were also upregulated.Using an in vitro neutrophil model,we found that GM-CSF inhibited C3aR expression,while inducing IL-8 production.Furthermore,this effect was transferrable in plasma from sepsis patients and was attenuated by inhibition of GM-CSF using an anti-GM-CSF antibody.These results indicate that GM-CSF-producing CD4^(+)T cells may serve as a marker of sepsis severity.Thus,targeting GM-CSF overproduction may benefit sepsis patients.Huihuang Huang Siyu Wang Tianjun Jiang Rong Fan Zheng Zhang Jinsong Mu Ke Li Yonggang Wang Lei Jin Fang Lin Jie Xia Lijian Sun Biao Xu Chengcheng Ji Jing Chen Juan Chang Bo Tu Bing Song Chao Zhang Fu-Sheng Wang Ruonan Xu 2019Cellular & Molecular Immunology2019,16,6:6
4Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants,are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. 5The incubation period of COVID-19: a global meta-analysis of 53 studies and a Chinese observation study of 11 545 patients显示文摘Background:The incubation period is a crucial index of epidemiology in understanding the spread of the emerging Coronavirus disease 2019(COVID-19).In this study,we aimed to describe the incubation period of COVID-19 globally and in the mainland of China.Methods:The searched studies were published from December 1,2019 to May 26,2021 in CNKI,Wanfang,PubMed,and Embase databases.A random-efect model was used to pool the mean incubation period.Meta-regression was used to explore the sources of heterogeneity.Meanwhile,we collected 11545 patients in the mainland of China outside Hubei from January 19,2020 to September 21,2020.The incubation period ftted with the Log-normal model by the coarseDataTools package.Results:A total of 3235 articles were searched,53 of which were included in the meta-analysis.The pooled mean incubation period of COVID-19 was 6.0 days(95%confdence interval[CI]5.6–6.5)globally,6.5 days(95%CI 6.1–6.9)in the mainland of China,and 4.6 days(95%CI 4.1–5.1)outside the mainland of China(P=0.006).The incubation period varied with age(P=0.005).Meanwhile,in 11545 patients,the mean incubation period was 7.1 days(95%CI 7.0–7.2),which was similar to the fnding in our meta-analysis.Conclusions:For COVID-19,the mean incubation period was 6.0 days globally but near 7.0 days in the mainland of China,which will help identify the time of infection and make disease control decisions.Furthermore,attention should also be paid to the region-or age-specifc incubation period.Cheng Cheng DongDong Zhang Dejian Dang Juan Geng Peiyu Zhu Mingzhu Yuan Ruonan Liang Haiyan Yang Yuefei Jin Jing Xie Shuaiyin Chen Guangcai Duan 2021Infectious Diseases of Poverty2021,10,5:1
6Advanced carbon materials with different spatial dimensions for supercapacitors显示文摘Supercapacitors(SCs)have attracted extensive attention due to their ultrahigh power density,fast charging/discharging rate,excellent electrochemical stability and environmental friendliness.Currently,the main commercial electrode materials for SCs are carbon materials in term of low cost,excellent conductivity,large specific surface area and good electrochemical stability.Recently,various dimensional carbon materials including zero dimensional(0D)carbon materials(nanosphere,dot etc.),1D carbon materials(nanotube,nanofiber etc.),2D carbon materials(nanosheet)as well as 3D carbon materials have been developed for SCs.Carbon materials with different spatial dimensions have their unique properties when used as the electrode materials for SCs.In this review,recent advances in the fabrication of different dimensional carbons for SCs are summarized.Several key issues for enhancing the electrochemical properties of carbon-based SCs and some mutual relationships among various influence parameters are reviewed,and challenges and perspectives in this field are also discussed.Xiaoliang Wu Ruonan Liu Jing Zhao Zhuangjun Fan 2021Nano Materials Science2021,3,3:1
7TBBPA rather than its main derivatives enhanced growth of endometrial cancer via p53 ubiquitination显示文摘Tetrabromobisphenol A(TBBPA)and its derivatives widely exist in various environments and biota.Although the available data indicate that TBBPA exposure is highly associated with the increased incidence of endometrial cancer(EC),the effects of TBBPA and its main derivatives on EC proliferation and the involved crucial mechanism remain unclear.The present study aimed to investigate the effects of TBBPA and its derivatives under environmental concentrations on the proliferation of EC,and the crucial mechanism on the progression of EC caused by bromine fame retardants exposure.In this research,TBBPA and two of the most common TBBPA derivatives including TBBPA bis(2-hydroxyethyl ether)(TBBPABHEE)and TBBPA bis(dibromopropyl ether)(TBBPA-BDBPE)were screened for their capacities in induced EC proliferation and explored the related mechanism by in vitro cell culture model and in vivo mice model.Under environmental concentrations,TBBPA promoted the proliferation of EC,the main derivatives of TBBPA(TBBPA-BHEE and TBBPA-BDBPE)did not present the similar facilitation effects.The ubiquitination degradation of p53 was crucial in TBBPA induced EC proliferation,which resulted in the increase of downstream cell cycle and decrease of apoptosis.The further molecular docking result suggested the high affinity between TBBPA and ubiquitinated proteasome.This finding revealed the effects of TBBPA and its derivatives on EC proliferation,thus providing novel insights into the underlying mechanisms of TBBPA-caused EC.Hongyan Yu Jingxu Zhang Jing Liu Ruonan Pan Yu Wang Xiaoting Jin Rifat Zubair Ahmed Yuxin Zheng 2024Journal of Environmental Sciences2024,,3:0

9Excipient-free porphyrin/SN-38 based nanotheranostics for drug delivery and cell imaging显示文摘Nanotheranostics with comprehensive diagnostic and therapeutic capabilities show exciting cancer treatment potentials.Here,we develop an excipient-free drug delivery system for cancer diagnosis as well as therapy,in which a near infra-red photosensitizer and a chemotherapeutic drug can be self-delivered without any carriers.The building block of the drug delivery system was synthesized by covalently conjugating four anticancer drugs(7-ethyl-10-hydroxy-camptothecin,SN-38)with a photosensitizer(porphyrin)via hydrolyzable ester linkage,which endows the drug delivery system with 100%active pharmaceutical ingredients,excellent imaging,and therapeutic functionalities.The conjugates can readily self-assemble into nanosheets(PS NSs)and remain stable for at least 20 days in aqueous solution.In PS NSs,fluorescence resonance energy transfer(FRET)dominates the fluorescence of SN-38 and enables to monitor the drug release fluorescentiy.The PS NSs also show excellent anticancer activity in vitro,due to the increased cell uptake with the synergistic effect of photodynamic therapy and chemotherapy.Ye Yuan Ruonan Bo Di Jing Zhao Ma Zhongling Wang Tzu-yin Lin Lijie Dong Xiangdong Xue Yuanpei Li 2020Nano Research2020,13,2:0

11Young Leaf White Stripe encodes a P-type PPR protein required for chloroplast development显示文摘Pentatricopeptide repeat(PPR) proteins function in post-transcriptional regulation of organellar gene expression. Although several PPR proteins are known to function in chloroplast development in rice(Oryza sativa), the detailed molecular functions of many PPR proteins remain unclear.Here, we characterized a rice young leaf white stripe(ylws) mutant, which has defective chloroplast development during early seedling growth.Map-based cloning revealed that YLWS encodes a novel P-type chloroplast-targeted PPR protein with 11 PPR motifs. Further expression analyses showed that many nuclear-and plastid-encoded genes in the ylws mutant were significantly changed at the RNA and protein levels. The ylws mutant was impaired in chloroplast ribosome biogenesis and chloroplast development under low-temperature conditions. The ylws mutation causes defects in the splicing of atpF, ndhA, rpl2,and rps12, and editing of ndhA, ndhB, and rps14transcripts. YLWS directly binds to specific sites in the atpF, ndhA, and rpl2 pre-mRNAs.
