3Lean heart: Role of leptin in cardiac hypertrophy and metabolism显示文摘Leptin is an adipokine that has been linked with the cardiovascular complications resulting from obesity such as hypertension and heart disease. Obese patients have high levels of circulating leptin due to increased fat mass. Clinical and population studies have correlated high levels of circulating leptin with the development of cardiac hypertrophy in obesity. Leptin has also been demonstrated to increase the growth of cultured cardiomyocytes. However, several animal studies of obese leptin deficient mice have not supported a role for leptin in promoting cardiac hypertrophy so the role of leptin in this pathological process remains unclear. Leptin is also an important hormone in the regulation of cardiac metabolism where it supports oxidation of glucose and fatty acids. In addition, leptin plays a critical role in protecting the heart from excess lipid accumulation and the formation of toxic lipids in obesity a condition known as cardiac lipotoxicity. 4渗透泵强化对流释放给药治疗恶性脑胶质瘤的裸小鼠模型显示文摘目的探寻渗透泵强化对流释放给药治疗脑肿瘤的祼小鼠模型,为新药的临床前期应用提供良好的实验动物模型。方法 14只祼小鼠基底节区种植4×105个人胶质瘤U87细胞,并安置渗透泵强化对流释放给药系统。裸小鼠按体重和活体荧光信号强度分为二组:治疗组(n=7)的泵内注入100μl(1μg)的DTATEGF,对照组(n=7)泵内注入100μl(1μg)的无关毒素DT。观察动物的生长特征,Kaplan-Meier生存曲线分析和脑内肿瘤的病理学特征。结果实验动物没有与渗透泵及外科手术相关的病残和病死发生。动物对渗透泵给药系统耐受好,无抓搔及渗透泵脱位,伤口无感染及裂开等情况。Kaplan-Meier生存曲线显示治疗组动物的中位生存期较对照组明显延长,差异有显著性。(治疗组126天vs.对照组68天,P=0.0002)。治疗组中有两只小鼠生存期超过180天。病理学检测祼小鼠脑内种植生长的肿瘤与人类胶质母细胞瘤的病理特征极为相似。结论该祼小鼠实验动物模型简便、实用、可靠,适用于新的药物包括单克隆抗体、靶向小分子、肿瘤疫苗等通过强化对流投递治疗脑内恶性肿瘤的临床前期应用。黄军 龙小艳 李波 Yan Michael Li Walter A Hall 刘定阳 刘庆 王君宇 袁贤瑞 2012国际神经病学神经外科学杂志2012,39,4:3

9脑肿瘤靶向毒素治疗的现状和展望显示文摘靶向毒素,亦称为免疫毒素或细胞毒素,是一种肿瘤细胞表面特异性抗原或受体相结合的细胞分子。如与EG-FR、IL-13受体等相结合的细胞分子,其毒素成份可杀死肿瘤细胞。目前研究的毒素多为以白喉毒素、假单胞杆菌外毒素等为母基合成的毒素。双重特异性靶向毒素能同时靶向肿瘤细胞和/或其新生血管上的两个过表达受体的毒素,如DTAT13靶向uPAR和IL-13受体,DTATEGF靶向EGFR和uPAR,其较单一靶向毒素具有更高的特异性和更强的毒性,同时免疫源性亦大为降低。寻找特异性的肿瘤细胞表面标志物、降低毒素的免疫源性及研究实体肿瘤毒素的投递方式是靶向毒素治疗脑肿瘤的研究方向。黄军 Yan Michael Li Walter A Hall 2011国际神经病学神经外科学杂志2011,38,5:2
