2Relationship between types of urban forest and PM_(2.5) capture at three growth stages of leaves显示文摘Particulate matter diameter ≤ 2.5 μm(PM2.5) causes direct harm to human health. Finding forms of urban forest systems that with the ability to reduce the amount of particulate matter in air effectively is the aim of this study. Five commonly cultivated kinds of urban forest types were studied in Beijing city at three stages of leaf growth. Results show that the urban forest system is capable of storing and capturing dust from the air. The types of shrubs and broadleaf trees that have the ability to capture PM2.5from the air are most effective when leaves have fully developed. In the leafless season, the conifer and mixed tree types are the most effective in removing dust from the air. For all kinds of forest types and stages of leaf growth, the PM2.5concentration is highest in the morning but lower in the afternoon and evening. 5A high-fiber diet synergizes with Prevotella copri and exacerbates rheumatoid arthritis显示文摘Both preclinical and established rheumatoid arthritis(RA)patients display alterations in the gut microbiome.Prevotella spp.are preferentially enriched in a subset of RA patients.Here,we isolated a Prevotella strain,P.copri RA,from the feces of RA patients and showed that colonization of P.copri RA exacerbated arthritis in a collagen-induced arthritis(CIA)model.With the presence of P.copri RA colonization,a high-fiber diet exacerbated arthritis via microbial alterations and intestinal inflammation.Colonization of P.copri together with a high-fiber diet enabled the digestion of complex fiber,which led to the overproduction of organic acids,including fumarate,succinate and short-chain fatty acids.Succinate promoted proinflammatory responses in macrophages,and supplementation with succinate exacerbated arthritis in the CIA model.Our findings highlight the importance of dysbiosis when evaluating the effects of dietary interventions on RA pathogenesis and provide new insight into dietary interventions or microbiome modifications to improve RA management.Lingjuan Jiang Mengmeng Shang Shengnan Yu Yudong Liu Hui Zhang Yangzhong Zhou Min Wang Tingting Wang Hui Li Zhihua Liu Xuan Zhang 2022Cellular & Molecular Immunology2022,19,12:6
6Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 7Metformin activates chaperone-mediated autophagy and improves disease pathologies in an Alzheimer disease mouse model显示文摘Chaperone-mediated autophagy(CMA)is a lysosome-dependent selective degradation pathway implicated in the pathogenesis of cancer and neurodegenerative diseases.However,the mechanisms that regulate CMA are not fully understood.Here,using unbiased drug screening approaches,we discover Metformin,a drug that is commonly the first medication prescribed for type 2 diabetes,can induce CMA.We delineate the mechanism of CMA induction by Metformin to be via activation of TAK1-IKKα/β signaling that leads to phosphorylation of Ser85 of the key mediator of CMA,Hsc70,and its activation.Notably,we find that amyloid-beta precursor protein(APP)is a CMA substrate and that it binds to Hsc70 in an IKKα/β-dependent manner.The inhibition of CMA-mediated degradation of APP enhances its cytotoxicity.Importantly,we find that in the APP/PS1 mouse model of Alzheimer's disease(AD),activation of CMA by Hsc70 overexpression or Metformin potently reduces the accumulated brain Aβplaque levels and reverses the molecular and behavioral AD phenotypes.Our study elucidates a novel mechanism of CMA regulation via Metformin-TAK1-IKKα/β-Hsc70 signaling and suggests Metformin as a new activator of CMA for diseases,such as AD,where such therapeutic intervention could be beneficial.Xiaoyan Xu Yaqin Sun Xufeng Cen Bing Shan Qingwei Zhao Tingxue Xie Zhe Wang Tingjun Hou Yu Xue Mengmeng Zhang Di Peng Qiming Sun Cong Yi Ayaz Najafov Hongguang Xia 2021Protein & Cell2021,12,10:5

10Ag@Au core/shell triangular nanoplates with dual enzyme-like properties for the colorimetric sensing of glucose显示文摘Due to the serious harm of diabetes to human health,development of sensitive assays for glucose level is of high significance for early prevention and treatment of diabetes.Currently,most conventional enzyme-based glucose sensors suffer from high cost and low stability due to the inherent defects of natural enzymes.Herein,we develop a pure nanozyme-based glucose detection method using Ag@Au core/shell triangular nanoplates(TNPs),which combines glucose oxidase(GOD)-and horseradish peroxidase(HRP)-like activities of the Au shell and inherent plasmonic properties of Ag TNPs.The sensing mechanism is based on the fact that the Au shell possessed GOD-like activity,enabling the oxidation of glucose to produce H2O2,which can further etch the silver core,leading to the decrease of absorbance at 800 nm and the color change from blue to colorless.Compared with the previous nanozymes-based glucose sensors,our method avoids the use of enzymes and organic chromogenic agent.Moreover,the stability of the Ag@Au core/shell TNPs is much better than that of Ag TNPs due to the protection by the coating of the Au shell.This method was successfully applied to the detection of urine samples from patients with diabetes,indicating its practical applicability for real sample analysis.Ao Liu Mengmeng Li Jingxiang Wang Fan Feng Yu Zhang Zhiwei Qiu Yuzhu Chen Benjamin Edem Meteku Congying Wen Zifeng Yan Jingbin Zeng 2020Chinese Chemical Letters2020,31,5:3
11Effects of Technological Parameters on Carbothermal Reduction and Nitridation of Zircon显示文摘Carbothermal reduction and nitridation(CRN)of zircon(ZrSiO4)permits obtaining different composites of oxides and nitrides such as ZrO2-Si2N2O and ZrN-Si3N4.The effects of technological parameters(carbon source,reaction temperature,and carbon content)on the reaction rate and product phase composition of CRN of zircon were investigated by TGA and XRD.The results show that:(1)carbon source is an important factor for a rapid reaction,and activated carbon is chosen as the carbon source considering the expect products and reaction rate;(2)reaction temperature has vital effect on reaction rate and product.In case of carbon content above 20%,the zircon phase and m-ZrO2 phase decrease with increasing temperature,while the Zr7N8O4 phase increases firstly and then decreases,and the ZrN phase increases continually;(3)different carbon contents result in different reaction products.The higher the carbon content,the lower the starting temperature for the CRN of zircon.YU Renhong FU Yadong ZHANG Mengmeng WANG Lizhi HONG Quanquan ZHANG Ruohua 2011China's Refractories2011,20,1:3
12Emerging Organic Contaminants in Chinese Surface Water:Identification of Priority Pollutants显示文摘The occurrence and impacts of emerging organic contaminants(EOCs)in the aquatic environment have gained widespread attention over the past two decades.Due to large number of potential contaminants,monitoring campaigns,treatment plants,and proposed regulations should preferentially focus on specific pollutants with the highest potential for ecological and human health effects.In the present study,a multi-criteria screening approach based on hazard and exposure potentials was developed for prioritization of 405 unregulated EOCs already present in Chinese surface water.Hazard potential,exposure potential,and risk quotients for ecological and human health effects were quantitatively analyzed and used to screen contaminants.The hazard potential was defined by contaminant persistence,bioaccumulation,ecotoxicity,and human health effects;similarly,the exposure potential was a function of contaminant concentration and detection frequency.In total,123 compounds passed the preselection process,which involved a priority index equal to the normalized hazard potential multiplied by the normalized exposure potential.Based on the prioritization scheme,11 compounds were identified as top-priority,and 37 chemicals were defined as high-priority.The results obtained by the priority index were compared with four other prioritization schemes based on exposure potential,hazard potential,or risk quotients for ecological effects or human health.The priority index effectively captured and integrated the results from the more simplistic prioritization schemes.Based on identified data gaps,four uncertainty categories were classified to recommend:①regular monitoring,derivation of environmental quality standards,and development of control strategies;②increased monitoring;③fortified hazard assessment;and④increased efforts to collect occurrence and toxicity data.Overall,20 pollutants were recommended as priority EOCs.The prioritized list of contaminants provides the necessary information for authoritative regulations to monitor,control,evaluate,and manage the risks of environmentally-relevant EOCs in Chinese surface water.Mengmeng Zhong Tielong Wang Wenxing Zhao Jun Huang Bin Wang Lee Blaney Qingwei Bu Gang Yu 2022Engineering2022,8,4:3
13Structure-activity relationship optimization for lassa virus fusion inhibitors targeting the transmembrane domain of GP2显示文摘Dear Editor,Lassa virus(LASV)belongs to the Mammarenavirus genus,Arenaviridae family.Arenaviruses are classified into two main groups-Old World(OW)and New World(NW)-based on virus genetics,serology,antigenic properties and geographical relationships.The OW LASV and Lujo virus(LUJV),as well as NW Jurn'n virus(JUNV),Machupo virus(MACV),Guanarito virus(GTOV),Sabia virus(SABV)and Chapare virus(CHAPV),are known to cause severe hemorrhagic fever and are listed as biosafety level 4(BSL-4)agents.The arenavirus glycoprotein complex(GPC)contains three subunits-the retained stable-signal peptide(SSP),the receptor-binding subunit GP1,and the membrane fusion subunit GP2(Lenz et al.,2001).Notably,the proximate external membrane region and TM of GP2,together with the ectodomain loop and TMs of SSP,form an SSP-GP2 interface,playing essential roles in regulating membrane fusion,and providing targets for distinct fusion inhibitors(Larson et al.,2008;Lee et al.,2008;York et al.,2008;York and Nunberg,2009;Thomas et al.,2011;Burgeson et al.,2013a;Shankar et al.,2016;Wang et al.,2016;Wang et al.,2018).Among these inhibitors,ST-161 is LASV specific(Burgeson et al.,2013a).In this study,we conducted structure-activity relationship(SAR)optimization of ST-161.As a result,21 derivatives with IC50 values<1 pmol/L are presented in Table S1.Hit compounds 21,29 and 57 exhibiting robust inhibition of the LASV pseudotype virus(LASVpv,VSV backbone enveloped by LASV GPC with single cycle infection)entry with IC50 values lower than 0.2 nmol/L(Figs.1A and S1),as well as hit compound 72 with an ester bond instead of acylhydrazone,were further investigated.To test whether the four hit compounds inhibit LASV entry by blocking the GPC-mediated membrane fusion,the inhibition effects of these compounds against LASV GPC mediated fusion were quantitatively determined by dual-luciferase assay(Thomas et al.,2011;Wang et al.,2018).Guangshun Zhang Junyuan Cao Yan Cai Yang Liu Yanli Li Peilin Wang Xiaoying Jia Mengmeng Zhang Gengfu Xiao Yu Guo Wei Wang 2019Protein & Cell2019,10,2:3
14On-demand assembly of polymeric nanoparticles for longer-blood-circulation and disassembly in tumor for boosting sonodynamic therapy显示文摘Sonodynamic therapy (SDT) is one of the promising strategies for tumor therapy, but its application is usually hindered by fast clearance in blood-circulation, abnormal tumor microenvironment, and inefficient generation of reactive oxygen species. To solve these problems, we proposed an on-demand assembly-disassembly strategy, where the assembly is favorable for longer-blood-circulation and then the disassembly in tumor is favorable for boosting SDT. Hematoporphyrin monomethyl ether (HMME) as the model of organic sonosensitizers were conjugated with hyaluronic acid (HA). Then HA-HMME was mixed with catalase (CAT) and assembled into polymeric nanoparticles (CAT@HA-HMME NPs) with size of ~80 nm. CAT@HA-HMME NPs exhibit good biocompatibility and a longer blood half-time (t1/2 = 4.17 h) which is obviously longer than that (~0.82 h) of HMME molecules. After HA receptor-mediated endocytosis of cancer cells, CAT@HA-HMME NPs can be cleaved by endogenous hyaluronidase, resulting in the on-demand disassembly in tumor to release HA-HMME molecules and CAT. The CAT catalyzes the endogenous H_(2)O_(2) into O_(2) to relieve the hypoxic microenvironment, and the released HA-HMME exhibits a higher ROS generation ability, greatly boosting SDT for the inhibition of tumor growth. 15Settling behavior of spherical particles in vertical annulus:Experimental study and model development显示文摘Coiled tubing(CT)drilling technology offers significant advantages in terms of cost and efficiency for exploitations of unconventional oil and gas resources.However,the development of CT drilling technol-ogy is restricted by cuttings accumulation in the wellbore due to non-rotation of the drill string and limited circulating capacity.Cuttings cleaning becomes more difficult with the wall resistance of pipe-wellbore annulus on the cutting transport.Accurate description of particle transport process in the pipe-wellbore annulus is,therefore,important for improving the wellbore cleanliness.In this study,high-speed cam-era is used to record and analyze the settling process of particles in the transparent annulus filled with power-law fluids.A total of 540 tests were carried out,involving dimensionless diameters of 0.10-0.95 and particle Reynolds Numbers of 0.01-12.97,revealing the effect of the dimensionless diameter and particle Reynolds number on the annulus wall effect,and the wall factor model with an average relative error of2.75%was established.In addition,a dimensionless parameter,Archimedes number,independent of the settling velocity,was introduced to establish an explicit model of the settling velocity of spherical particles in the vertical annulus,with the average relative error of 7.89%.Finally,a calculation example was provided to show how to use the explicit model of settling velocity in annulus.The results of this study are expected to provide guidance for field engineers to improve the wellbore cleanliness of coiled tubing drilling.Zhaopeng Zhu Mengmeng Zhou Xianzhi Song Shuo Zhu Gensheng Li Zhengming Xu Xuezhe Yao Buwen Yu 2022Particuology2022,20,9:2
16Curcumin alleviates TGF-β1-induced fibrosis in NRK-49F cells via suppression of miR-21 expression,and regulation of the TGF-β1/smad3 signaling pathway显示文摘Objective:Overexpression of microRNA-21(miR-21)has been well-recognized during fibrosis in a wide range of tissue types through regulation of fibrogenesis via diverse pathways.Curcumin exhibits antifibrotic effects in various organs.We and others have previously demonstrated that curcumin has positive effects on the protection against fibrosis.However,the effect of curcumin on miR-21 expression has not been reported.Methods:In this study,transforming growth factor-b1(TGF-b1)-stimulated rat renal fibroblast cells(NRK-49F)were transfected with miR-21 inhibitors,then the expression of Smad3,a-smooth muscle actin(a-SMA),type I collagen(COL1A1),and type III collagen(COL3A1)was determined using quantitative real-time-PCR(qRT-PCR)and western blot analysis.The effect of curcumin on production of the extracellular matrix was evaluated using immunofluorescence,qRT-PCR,and western blot analysis.Results:TGF-b1 stimulation upregulated the expression of miR-21 and induced fibrogenesis in NRK-49F cells.Transfection with miR-21 inhibitors selectively decreased smad3 activity,and noticeably reduced the expression of a-SMA,COL1A1,and COL3A1.Curcumin treatment significantly inhibited the expressions of smad3,a-SMA,COL1A1,COL3A1,as well as miR-21 expression in NRK-49F cells in a dosedependent manner.Conclusion:Curcumin exerted its anti-fibrotic effects by targeting the TGF-b1/smad3 signaling pathway and suppressing miR-21 expression,thereby provides novel insight in the protective effects of curcumin against fibrosis in various organs.Lan Zhang Yi Liu Xiting Wang Cheng Li Mengmeng Shen Liming Di Shuangshuang He Yu Li 2020Journal of Traditional Chinese Medical Sciences2020,7,1:2
17Effect of ZrO_2-nitrides Composite Powder Addition on Oxidation Resistance of Al_2O_3-C Refractories显示文摘Oxidation of carbon is the main problem of Al2O3-C refractories.ZrO2-nitrides composite powder was synthesized through carbothermal reduction and nitridation (CRN) of zircon.The effect of ZrO2-nitrides composite powder addition on oxidation resistance of the Al2O3-C refractories was investigated by measuring the thickness of oxidation layer.Phase compositions of the Al2O3-C refractories before and after oxidation were investigated by X-ray diffraction (XRD).Results show that the oxidation resistance of the Al2O3-C refractories can be obviously improved by adding the synthesized ZrO2-nitrides composite powder.The formation of mullite and zircon in the oxidation layer results in the densification of oxidation layer,which prevents oxygen diffusion and improves the oxidation resistance of the Al2O3-C refractories.ZHANG Mengmeng YU Renhong ZHOU Ningsheng 2011China's Refractories2011,20,3:2
18A neglected GABAergic astrocyte:Calcium dynamics and involvement in seizure activity显示文摘Gamma-aminobutyric acid(GABA) contributes substantially to neurocognitive function as an important inhibitory neurotransmitter in the human cerebral cortex. However, the pathophysiology of disorders such as epilepsy are not well understood, since GABA agonists are not quite effective in treating epilepsy. Knowledge of the mechanism of action of GABA would contribute to review previously proposed anti-epileptic processes by GABA agonists. In this study based on recent experiments on GABAergic astrocytes, we developed a modified GABAergic astrocyte model, and successfully simulated a long-lasting Ca^(2+) oscillation in astrocytes after 0.5-s stimulation of GABAergic transmission. We then incorporated this GABAergic astrocyte model into a classical Ullah-Schiff seizure model and surprisingly found that this GABAergic astrocyte model functions to hinder the anti-epileptic action of GABA agonists, thereby explaining their low efficiency in previous experiments. 20Selective coordination and localized polarization in graphene quantum dots: Detection of fluoride anions using ultra-low-field NMR relaxometry显示文摘The development of ultra-sensitive methods for detecting anions is limited by their low charge to radius ratios, microenvironment sensitivity, and p H sensitivity. In this paper, a magnetic sensor is devised that exploits the controllable and selective coordination that occurs between a magnetic graphene quantum dot(GQD) and fluoride anion(F–). The sensor is used to measure the change in relaxation time of aqueous solutions of magnetic GQDs in the presence of F–using ultra-low-field(118 μT) nuclear magnetic resonance relaxometry. The method was optimized to produce a limit of detection of 10 nmol/L and then applied to quantitatively detect F–in domestic water samples. More importantly, the key factors responsible for the change in relaxation time of the magnetic GQDs in the presence of F–are revealed to be the selective coordination that occurs between the GQDs and F–as well as the localized polarization of the water protons.
