2Breast cancer development and progression:Risk factors,cancer stem cells,signaling pathways,genomics,and molecular pathogenesis显示文摘As the most commonly occurring cancer in women worldwide,breast cancer poses a formidable public health challenge on a global scale.Breast cancer consists of a group of biologically and molecularly heterogeneous diseases originated from the breast.While the risk factors associated with this cancer varies with respect to other cancers,genetic predisposition,most notably mutations in BRCA1 or BRCA2 gene,is an important causative factor for this malignancy.Breast cancers can begin in different areas of the breast,such as the ducts,the lobules,or the tissue in between.Within the large group of diverse breast carcinomas,there are various denoted types of breast cancer based on their invasiveness relative to the primary tumor sites.It is important to distinguish between the various subtypes because they have different prognoses and treatment implications.As there are remarkable parallels between normal development and breast cancer progression at the molecular level,it has been postulated that breast cancer may be derived from mammary cancer stem cells.Normal breast development and mammary stem cells are regulated by several signaling pathways,such as estrogen receptors(ERs),HER2,and Wnt/b-catenin signaling pathways,which control stem cell proliferation,cell death,cell differentiation,and cell motility.Furthermore,emerging evidence indicates that epigenetic regulations and noncoding RNAs may play important roles in breast cancer development and may contribute to the heterogeneity and metastatic aspects of breast cancer,especially for triple-negative breast cancer.This review provides a comprehensive survey of the molecular,cellular and genetic aspects of breast cancer.Yixiao Feng Mia Spezia Shifeng Huang Chengfu Yuan Zongyue Zeng Linghuan Zhang Xiaojuan Ji Wei Liu Bo Huang Wenping Luo Bo Liu Yan Lei Scott Du Akhila Vuppalapati Hue H.Luu Rex C.Haydon Tong-Chuan He Guosheng Ren 2018Genes & Diseases2018,5,2:24
3Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
4Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 5Error Compensation of Thin Plate-shape Part with Prebending Method in Face Milling显示文摘Low weight and good toughness thin plate parts are widely used in modern industry, but its flexibility seriously impacts the machinability. Plenty of studies focus on the influence of machine tool and cutting tool on the machining errors. However, few researches focus on compensating machining errors through the fixture. In order to improve the machining accuracy of thin plate-shape part in face milling, this paper presents a novel method for compensating the surface errors by prebending the workpiece during the milling process. First, a machining error prediction model using finite element method is formulated, which simplifies the contacts between the workpiece and fixture with spring constraints. Milling forces calculated by the micro-unit cutting force model are loaded on the error prediction model to predict the machining error. The error prediction results are substituted into the given formulas to obtain the prebending clamping forces and clamping positions. Consequently, the workpiece is prebent in terms of the calculated clamping forces and positions during the face milling operation to reduce the machining error. Finally, simulation and experimental tests are carried out to validate the correctness and efficiency of the proposed error compensation method. The experimental measured flatness results show that the flatness improves by approximately 30 percent through this error compensation method. 6A Brief Introduction to BNU-HESM1.0 and Its Earth Surface Temperature Simulations显示文摘Integrated assessment models and coupled earth system models both have their limitations in understanding the interactions between human activity and the physical earth system. In this paper,a new human–earth system model,BNUHESM1.0,constructed by combining the economic and climate damage components of the Dynamic Integrated Model of Climate Change and Economy to the BNU-ESM model,is introduced. The ability of BNU-HESM1.0 in simulating the global CO2 concentration and surface temperature is also evaluated. We find that,compared to observation,BNU-HESM1.0underestimates the global CO2 concentration and its rising trend during 1965–2005,due to the uncertainty in the economic components. However,the surface temperature simulated by BNU-HESM1.0 is much closer to observation,resulting from the overestimates of surface temperature by the original BNU-ESM model. The uncertainty of BNU-ESM falls within the range of present earth system uncertainty,so it is the economic and climate damage component of BNU-HESM1.0 that needs to be improved through further study. However,the main purpose of this paper is to introduce a new approach to investigate the complex relationship between human activity and the earth system. 7Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic,chondrogenic and adipogenic lineages.We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood.Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors.We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers,matrix mineralization and ectopic bone formation from MSCs.Thus,our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling.Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained,and thus may be used as an effective delivery vehicle of siRNA therapeutics.Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng 2018Genes & Diseases2018,5,2:8
8Load-sharing Characteristic of Multiple Pinions Driving in Tunneling Boring Machine显示文摘The failure of the key parts, such as gears, in cutter head driving system of tunneling boring machine has not been properly solved under the interaction of driving motors asynchronously and wave tunneling torque load. A dynamic model of multi-gear driving system is established considering the inertia effects of driving mechanism and cutter head as well as the bending-torsional coupling. By taking into account the nonlinear coupling factors between ring gear and multiple pinions, the influence for meshing angle by bending-torsional coupling and the dynamic load-sharing characteristic of multiple pinions driving are analyzed. Load-sharing coefficients at different rotating cutter head speeds and input torques are presented. Numerical results indicate that the load-sharing coefficients can reach up to 1.2-1.3. A simulated experimental platform of the multiple pinions driving is carried out and the torque distributions under the step load in driving shaft of pinions are measured. The imbalance of torque distribution of pinions is verified and the load-sharing coefficients in each pinion can reach 1.262. The results of simulation and test are similar, which shows the correctness of theoretical model. A loop coupling control method is put forward based on current torque master slave control method. The imbalance of the multiple pinions driving in cutter head driving system of tunneling boring machine can be greatly decreased and the load-sharing coefficients can be reduced to 1.051 by using the loop coupling control method. 9Influence of the carbon cycle on the attribution of responsibility for climate change显示文摘The carbon cycle is one of the fundamental climate change issues.Its long-term evolution largely affects the amplitude and trend of human-induced climate change,as well as the formulation and implementation of emission reduction policy and technology for stabilizing the atmospheric CO2concentration.Two earth system models incorporating the global carbon cycle,the Community Earth System Model and the Beijing Normal University-Earth System Model,were used to investigate the effect of the carbon cycle on the attribution of the historical responsibility for climate change.The simulations show that when compared with the criterion based on cumulative emissions,the developed(developing)countries’responsibility is reduced(increased)by 6%–10%using atmospheric CO2concentration as the criterion.This discrepancy is attributed to the fact that the developed world contributed approximately61%–68%(61%–64%)to the change in global oceanic(terrestrial)carbon sequestration for the period from 1850 to2005,whereas the developing world contributed approximately 32%–49%(36%–39%).Under a developed world emissions scenario,the relatively larger uptake of global carbon sinks reduced the developed countries’responsibility for carbon emissions but increased their responsibility for global ocean acidification(68%).In addition,the large emissions from the developed world reduced the efficiency of the global carbon sinks,which may affect the long-term carbon sequestration and exacerbate global warming in the future.Therefore,it is necessary to further consider the interaction between carbon emissions and the carbon cycle when formulating emission reduction policy.Ting Wei Wenjie Dong Wenping Yuan Xiaodong Yan Yan Guo 2014Chinese Science Bulletin2014,59,19:6
10Thermodynamic modelling of the phengite geobarometry显示文摘Using the updated models of activity-composition relations for solid solutions,P-T pseudosections and Si isopleths of phengite in the system KFMASH (K2O-FeO-MgO-Al2O3-SiO2-H2O) are calculated with the software THERMOCALC 323 for three bulk compositions of the average,Al-rich and K-rich metapelites. The Si isopleths of phengite in the calculated pseudosections are highly dependent on mineral as-semblages. In the chlorite-and biotite-bearing di-and trivariant fields,and in the garnet + kyanite-bearing trivariant fields,the phengite can be used as a good geobarometer. But in the carpho-lite-present and chlorite-absent fields,the Si contents of phengite mainly indicate variance of tem-perature. For a given Si value,the phengite in kyanite-bearing assemblages shows higher pressures than in the K-feldspar-bearing assemblages. Addition of Na2O into the KFMASH leads to reduction of stability of the AFM phases. The Si isopleths of phengite in the mineral assemblages containing only one Na-phase are similar to those in the corresponding KFMASH assemblages. 11Establishment and functional characterization of the reversibly immortalized mouse glomerular podocytes(imPODs)显示文摘Glomerular podocytes are highly specialized epithelial cells and play an essential role in establishing the selective permeability of the glomerular filtration barrier of kidney.Maintaining the viability and structural integrity of podocytes is critical to the clinical management of glomerular diseases,which requires a thorough understanding of podocyte cell biology.As mature podocytes lose proliferative capacity,a conditionally SV40 mutant tsA58-immortalized mouse podocyte line(designated as tsPC)was established from the Immortomouse over 20 years ago.However,the utility of the tsPC cells is hampered by the practical inconvenience of culturing these cells.In this study,we establish a user-friendly and reversibly-immortalized mouse podocyte line(designated as imPOD),on the basis of the tsPC cells by stably expressing the wildtype SV40 T-antigen,which is flanked with FRT sites.We show the imPOD cells exhibit long-term high proliferative activity,which can be effectively reversed by FLP recombinase.The imPOD cells express most podocyte-related markers,including WT-1,Nephrin,Tubulin and Vinculin,but not differentiation marker Synaptopodin.The imPOD cells do not form tumor-like masses in vivo.We further demonstrate that TGFb1 induces a podocyte injury-like response in the FLP-reverted imPOD cells by suppressing the expression of slit diaphragm-associated proteins P-Cadherin and ZO-1 and upregulating the expression of mesenchymal markers,a-SMA,Vimentin and Nestin,as well as fibrogenic factors CTGF and Col1a1.Collectively,our results strongly demonstrate that the newly engineered im-POD cells should be a valuable tool to study podocyte biology both under normal and under pathological conditions.Xinyi Yu Liqun Chen Ke Wu Shujuan Yan Ruyi Zhang Chen Zhao Zongyue Zeng Yi Shu Shifeng Huang Jiayan Lei Xiaojuan Ji Chengfu Yuan Linghuan Zhang Yixiao Feng Wei Liu Bo Huang Bo Zhang Wenping Luo Xi Wang Bo Liu Rex C.Haydon Hue H.Luu Tong-Chuan He Hua Gan 2018Genes & Diseases2018,5,2:5

14Enhanced Hole-Injection Property in an OLED with a Self-assembled Monolayer of Hole-Transporting TPD on Thin Au as the Anode显示文摘A thioester-functionalized triphenylamine hole-transporting molecule(TPD-SAc) was synthesized and self-assembled to form a monolayer on an ultra-thin Au film supported on indium-tin oxide glass. The modified surface was characterized by aqueous contact angle, ellipsometer, atomic force microscopy, X-ray photoelectron spectroscopy, and ultraviolet photoelectron spectrometer to substantiate the formation of compact and pinhole-free monolayers. The modified organic light emitting diode device [indium-tin oxide/Au(5 nm)/self-assembled monolayers(SAM)/TPD(50 nm)/Alq_3(40 nm)/TPBI(15 nm)/LiF(1 nm)/Al(100 nm)] showed a luminance of 7303.90 cd/m^2 and a current efficiency of 8.49 cd/A with 1.78 and 2.29-fold increase, respectively, compared to the control device without SAM. 15A Survey on Quantum Cryptography显示文摘Quantum cryptography(QC) as an important technology to protect the security of the future network communication, has gained extensive attention from both academia and industry in recent years. This paper provides a concise up-to-date review of QC, including Quantum key distribution(QKD), Quantum authentication, Quantum public-key cryptography(QPKC), and so on. Our aim is to pave a comprehensive and solid starting ground for interested readers. We firstly clarify the definition of QC. 16HB-PLS:A statistical method for identifying biological process or pathway regulators by integrating Huber loss and Berhu penalty with partial least squares regression显示文摘Gene expression data features high dimensionality,multicollinearity,and non-Gaussian distribution noise,posing hurdles for identification of true regulatory genes controlling a biological process or pathway.In this study,we integrated the Huber loss function and the Berhu penalty(HB)into partial least squares(PLS)framework to deal with the high dimension and multicollinearity property of gene expression data,and developed a new method called HB-PLS regression to model the relationships between regulatory genes and pathway genes.To solve the Huber-Berhu optimization problem,an accelerated proximal gradient descent algorithm with at least 10 times faster than the general convex optimization solver(CVX),was developed.Application of HB-PLS to recognize pathway regulators of lignin biosynthesis and photosynthesis in Arabidopsis thaliana led to the identification of many known positive pathway regulators that had previously been experimentally validated.As compared to sparse partial least squares(SPLS)regression,an efficient method for variable selection and dimension reduction in handling multicollinearity,HB-PLS has higher efficacy in identifying more positive known regulators,a much higher but slightly less sensitivity/(1-specificity)in ranking the true positive known regulators to the top of the output regulatory gene lists for the two aforementioned pathways.In addition,each method could identify some unique regulators that cannot be identified by the other methods.Our results showed that the overall performance of HB-PLS slightly exceeds that of SPLS but both methods are instrumental for identifying real pathway regulators from high-throughput gene expression data,suggesting that integration of statistics,machine leaning and convex optimization can result in a method with high efficacy and is worth further exploration.Wenping Deng Kui Zhang Cheng He Sanzhen Liu Hairong Wei 2021Forestry Research2021,1,1:1

19Numerical simulation and data assimilation of the water-energy cycle over semiarid northeastern China显示文摘The default fractional vegetation cover and terrain height were replaced by the estimated fractional vegetation cover, which was calculated by the Normalized Difference Vegetation Index(NDVI) of Earth Observing System Moderate-Resolution Imaging Spectroradiometer(EOS-MODIS) and the Digital Elevation Model of the Shuttle Radar Topography Mission(SRTM) system. The near-surface meteorological elements over northeastern China were assimilated into the three-dimensional variational data assimilation system(3DVar) module in the Weather Research and Forecasting(WRF) model. The structure and daily variations of air temperature, humidity, wind and energy fields over northeastern China were simulated using the WRF model. Four groups of numerical experiments were performed, and the simulation results were analyzed of latent heat flux, sensible heat flux, and their relationships with changes in the surface energy flux due to soil moisture and precipitation over different surfaces. The simulations were compared with observations of the stations Tongyu, Naiman, Jinzhou, and Miyun from June to August, 2009. The results showed that the WRF model achieves high-quality simulations of the diurnal characteristics of the surface layer temperature, wind direction, net radiation, sensible heat flux, and latent heat flux over semiarid northeastern China in the summer. The simulated near-surface temperature, relative humidity, and wind speed were improved in the data assimilation case(Case 2) compared with control case(Case 1). The simulated sensible heat fluxes and surface heat fluxes were improved by the land surface parameterization case(Case 3) and the combined case(Case 4). The simulated temporal variations in soil moisture over the northeastern arid areas agree well with observations in Case 4, but the simulated precipitation should be improved in the WRF model. This study could improve the land surface parameters by utilizing remote sensing data and could further improve atmospheric elements with a data assimilation system. This work provides an effective attempt at combining multi-source data with different spatial and temporal scales into numerical simulations.
