2Gender-related differences in irritable bowel syndrome: Potential mechanisms of sex hormones显示文摘According to epidemiological studies,twice as many women as men are affected by irritable bowel syndrome(IBS)in western countries,suggesting a role for sex hormones in IBS pathophysiology.Despite growing evidence about the implications of sex hormones in IBS symptom modulation,data on mechanisms by which they influence disease development are sparse.This review aims to determine the state of knowledge about the role of sex hormones in sensorimotor dysfunctions and to address the possible interplay of sex hormones with common risk factors associated with IBS.The scientific bibliography was searched using the following keywords:irritable bowel syndrome,sex,gender,ovarian hormone,estradiol,progesterone,testosterone,symptoms,pain,sensitivity,motility,permeability,stress,immune system,brain activity,spinal,supraspinal,imaging.Ovarian hormones variations along themenstrual cycle affect sensorimotor gastrointestinal function in both healthy and IBS populations.They can modulate pain processing by interacting with neuromodulator systems and the emotional system responsible for visceral pain perception.These hormones can also modulate the susceptibility to stress,which is a pivotal factor in IBS occurrence and symptom severity.For instance,estrogen-dependent hyper-responsiveness to stress can promote immune activation or impairments of gut barrier function.In conclusion,whereas it is important to keep in mind that ovarian hormones cannot be considered as a causal factor of IBS,they arguably modulate IBS onset and symptomatology.However,our understanding of the underlying mechanisms remains limited and studies assessing the link between IBS symptoms and ovarian hormone levels are needed to improve our knowledge of the disease evolution with regard to gender.Further studies assessing the role of male hormones are also needed to understand fully the role of sex hormones in IBS.Finally,investigation of brain-gut interactions is critical to decipher how stress,ovarian hormones,and female brain processing of pain can translate into gut dysfunctions.Mathieu Meleine Julien Matricon 2014World Journal of Gastroenterology2014,20,22:14

4Pioneer of prostate cancer: past, present and the future of FOXA1显示文摘Prostate cancer is the most commonly diagnosed noncutaneous cancers in North American men.While androgen deprivation has remained as the cornerstone of prostate cancer treatment,resistance ensues leading to lethal disease.Forkhead box A1(FOXA1)encodes a pioneer factor that induces open chromatin conformation to allow the binding of other transcription factors.Through direct interactions with the Androgen Receptor(AR),FOXA1 helps to shape AR signaling that drives the growth and survival of normal prostate and prostate cancer cells.FOXA1 also possesses an AR-independent role of regulating epithelial-to-mesenchymal transition(EMT).In prostate cancer,mutations converge onto the coding sequence and c/s-regulatory elements(CREs)of FOXA1,leading to functional alterations.In addition,FOXA1 activity in prostate cancer can be modulated post-translationally through various mechanisms such as LSD1-mediated protein demethylation.In this review,we describe the latest discoveries related to the function and regulation of FOXA1 in prostate cancer,pointing to their relevance to guide future clinical interventions.Mona Teng Stanley Zhou Changmeng Cai Mathieu Lupien Housheng Hansen He 2021Protein & Cell2021,12,1:12

7interrelationships between ghrelin, insulin and glucose homeostasis: physiological relevance显示文摘Ghrelin is a 28 amino acid peptide mainly derived from the oxyntic gland of the stomach. Both acylated(AG) and unacylated(UAG) forms of ghrelin are found in the circulation. Initially, AG was considered as the only bioactive form of ghrelin. However, recent advances indicate that both AG and UAG exert distinct and common effects in organisms. Soon after its discovery, ghrelin was shown to promote appetite and adiposity in animal and human models. In response to these anabolic effects, an impressive number of elements have suggested the influence of ghrelin on the regulation of metabolic functions and the development of obesityrelated disorders. However, due to the complexity ofits biochemical nature and the physiological processes it governs, some of the effects of ghrelin are still debated in the literature. Evidence suggests that ghrelin influences glucose homeostasis through the modulation of insulin secretion and insulin receptor signaling. On the other hand, insulin was also shown to influence circulating levels of ghrelin. 8不同饵料对二次抱卵期间中华绒螯蟹生殖性能和HUFA组成的影响(英文)显示文摘中华绒螯蟹(Eriocheir sinensis)经一次受精便可多次产卵。与其他的虾类一样,在卵巢发育过程中,中华绒螯蟹的肝胰脏需从外界汲取许多能量和营养物质,通过血淋巴转移到卵巢以供胚胎发育和幼体孵化。亲蟹的营养需求可以通过第一次和第二次卵巢发育过程加以研究。第一次抱卵之后亲蟹消耗了大量的营养和能量物质,需要在二次抱卵前的短时间内得到及时补充。通常来说,二次抱卵蟹的产卵力低下,幼体质量差。因此研究饵料对二次抱卵期问中华绒螯蟹生殖性能和幼体质量的影响是很有意义的。本实验分4组,分别以3种天然饵料缢蛏(TC)、沙蚕(TS)、杂鱼(TT)和一种人工饵料(TA)投喂中华绒螯蟹亲蟹,投喂周期为2个月(从初始一次抱卵至二次抱卵结束)。天然饵料和人工饵料的日投喂量分别为亲蟹体重的10%和1%~3%,每日下午4点投喂。用PVC板将8m^2的玻璃纤维槽隔成4个单元作为试验槽。每个单元底面积为2m2,水深40cm。同一槽中的3个单元用做一个试验组的3个重复,每个重复为10只蟹。海水盐度为20g·L^-1,pH8.20~8.30,溶解氧为8.40~8.90mg·L^-1,依据水中氨氮和亚硝酸氮的浓度(分别小于1mg·L^-1和0.2mg·L^-1),每2至3天换一次水,水温为15~17℃。实验结束后,对二次抱卵亲蟹的生殖性能(相对饱卵量、卵径和亲蟹成活率)、孵化出的幼体质量(蚤Ⅰ幼体在饥饿状态下的成活率)以及卵、卵巢和肝胰脏的脂肪酸组成进行了测定。结果表明,卵、卵巢和肝胰脏的高级不饱和脂肪酸(HUFA)组成与试验饵料的组成具有很好的相关性,其中卵的DHA与EPA水平、肝胰脏DHA水平与饵料中相应的HUFA水平成正相关(R^2〉0.90),说明外界营养物质的吸收和转移对卵巢的二次发育相当重要。统计学分析表明,投喂杂鱼的亲蟹(TT)孵小的蚤Ⅰ幼体对饥饿的耐受力以及投喂缢蛏的亲蟹(TC)的卵径显著高于其他组,各组之间其他指标(相对抱卵量和成活率等)均无显著差异。二次抱卵的中华绒螯蟹的卵、卵巢和肝胰脏中HUFA的水平与试验饵料中HUFA水平具有很好的相关性,说明无论在第一次卵巢发育过程中积聚了多少营养物质,第一次抱卵后消耗殆尽的亲蟹需要从外界食物中汲取大量的能量和营养物质,通过肝胰脏转运到卵巢供二次发育之用。本研究结论与其他作者对其他甲壳类的研究结果是一致的。此外组织器官的HUFA水平不只局限于饵料HUFA水平,还受器官本身对这些脂肪酸特定需求的影响。肝胰脏作为甲壳类动物的脂肪存储器,在饵料中的脂类物质被吸收和利用之前首先将其存储起来。甲壳类动物具有合成更长链的脂肪酸能力,虽然这种能力很有限,但仍能重新合成一定数量的脂肪酸。由于卵巢和卵中的n-3 HUFA,尤其是EPA和DHA含量高于肝胰脏,因此有理由相信它们在中华绒螯蟹繁殖过程中起着非常重要的作用。用人工饵料投喂对中国对虾的研究表明,n-3 HUFA的水平与亲本的生殖性能有很大关系,其中EPA与产卵量有很大的关系,而DHA在胚胎发育早期和幼体孵化起着很大的作用。本实验结果可以推论,EPA和DHA时中华绒螯蟹的卵巢的发育和生殖性能具有相当重要的作用,其中EPA的作用更加明显。遗憾的是本实验没有得出非常明显的相关性结论。应该指出的是本实验所用的不同饵料,不但HUFA含量不同,而且在蛋白质、总脂和微量营养素(维他命、矿物质、胆固醇、磷脂、钙盐等)的组成上差异很大,因此不宜下确定的结论。计阳 隋丽英 吴旭干 成永旭 Wille Mathieu Sorgeloos Patrick 2006中国水产科学2006,13,1:8

11INT-767 improves histopathological features in a dietinduced ob/ob mouse model of biopsy-confirmed nonalcoholic steatohepatitis显示文摘AIM To characterize the efficacy of the dual FXR/TGR5 receptor agonist INT-767 upon histological endpoints in a rodent model of diet-induced and biopsy-confirmed non-alcoholic steatohepatitis(NASH).METHODS The effects of INT-767 on histological features of NASH were assessed in two studies using Lep^(ob/ob)(ob/ob) NASH mice fed the AMLN diet(high fat with transfat, cholesterol and fructose). In a proof-of-conceptstudy, Lep^(ob/ob)(ob/ob) NASH mice were first dosed with INT-767(3 or 10 mg/kg for 8 wk). A second ob/ob NASH study compared INT-767(3 and 10 mg/kg) to obeticholic acid(OCA)(10 or 30 mg/kg; 16 wk). Primary histological endpoints included qualitative and quantitative assessments of NASH. Other metabolic and plasma endpoints were also assessed. A comparative assessment of INT-767 and OCA effects on drug distribution and hepatic gene expression was performed in C57 Bl/6 mice on standard chow. C57 Bl/6 mice were orally dosed with INT-767 or OCA(1-30 mg/kg) for 2 wk, and expression levels of candidate genes were assessed by RNA sequencing and tissue drug levels were measured by liquid chromatography tandem-mass spectrometry.RESULTS INT-767 dose-dependently(3 and 10 mg/kg, PO, QD, 8 wk) improved qualitative morphometric scores on steatohepatitis severity, inflammatory infiltrates and fibrosis stage. Quantitative morphometric analyses revealed that INT-767 reduced parenchymal collagen area, collagen fiber density, inflammation(assessed by Galectin-3 immunohistochemistry) and hepatocyte lipid droplet area following INT-767 treatment. In a comparative study(16 wk), the FXR agonists OCA(10 and 30 mg/kg) and INT-767(3 and 10 mg/kg) both improved NASH histopathology, with INT-767 exerting greater therapeutic potency and efficacy than OCA. Mechanistic studies suggest that both drugs accumulate similarly within the liver and ileum, however, the effects of INT-767 may be driven by enhanced hepatic, but not ileal, FXR function. 14Galectin-1-mediated biochemical controls of melanoma and glioma aggressive behavior显示文摘Gliomas and melanomas are associated with dismal prognosis because of their marked intrinsic resistance to proapoptotic stimuli,such as conventional chemotherapy and radiotherapy,as well as their ability to escape immune cell attacks.In addition,gliomas and melanomas display pronounced neoangiogenesis.Galectin-1 is a hypoxia-sensitive protein,which is abundantly secreted by glioma and melanoma cells,which displays marked proangiogenic effects.It also provides immune tolerogenic environments to melanoma and glioma cells through the killing of activated T cells that attack these tumor cells.Galectin-1 protects glioma and melanoma cells against cytotoxic insults(including chemotherapy and radiotherapy) through a direct role in the unfolded protein response.Altogether,these facts clearly point to galectin-1 as an important target to be combated in gliomas and melanomas in order to:(1) weaken the defenses of these two types of cancers against radiotherapy,chemotherapy and immunotherapy/vaccine therapy;and(2) reinforce antiangiogenic therapies.In the present article,we review the biochemical and molecular biology-related pathways controlled by galectin-1,which are actually beneficial for melanoma and glioma cells,and therefore detrimental for melanoma and glioma patients.Florence Lefranc Véronique Mathieu Robert Kiss 2011World Journal of Biological Chemistry2011,2,9:6
15Attributable Causes of Cancer in China:Fruit and Vegetable显示文摘Objective:To provide an evidence-based and consistent assessment of the burden of cancer attributable to inadequate fruit and vegetable intake in China in 2005.Methods:The proportions of cancers attributable to low consumption of vegetable and fruit were calculated separately to estimate the burden of related cancers for the year 2005 in China.Data on the prevalence of exposure were derived from a Chinese nutrition and health survey.Data on relative risks were mainly derived from meta-analysis.Attributable fractions were calculated based on the counterfactual scenario which was a shift in the exposure distribution.Results:The total cancer burden attributable to inadequate consumption of fruit was up to 233,000 deaths (13.0% of all cancers) and 300,000 cases (11.6% of all cancers) in 2005.Increasing consumption of vegetable to the highest quintile could avoid total cancer deaths and cases by 3.6% (64,000 persons) and 3.4% (88,000 persons).The contributions to cancer burden were higher in rural areas than in urban areas.They have greater influence on men than on women.The largest proportions of cancer burden attributable to low fruit and vegetable intake were for oral and pharyngeal cancers.Conclusion:This study showed that inadequate intake of fruit and vegetable makes a significant contribution to the cancer burden.Increasing consumption of fruit and vegetable could prevent many cancer deaths and save many lives.Promoting the consumption of fruit and vegetable is an important component in diet-based strategies for preventing cancer.Hui-juan Xiao Hao Liang Jian-bing Wang Cheng-Yu Huang Wen-qiang Wei Mathieu Boniol You-lin Qiao Paolo Boffetta 2011Chinese Journal of Cancer Research2011,23,3:5

17PCR方法诊断家犬感染细粒棘球绦虫的特异性及在临床诊断中的应用价值显示文摘目的验证PCR方法检测终宿主家犬感染细粒棘球绦虫的特异性及其在临床诊断中的应用价值。方法取感染细粒棘球绦虫家犬粪便,显微镜下计数细粒棘球绦虫虫卵,稀释后PCR检测其敏感性。同时取家犬小肠内水泡带绦虫、多头绦虫、羊绦虫,将其DNA与细粒棘球绦虫DNA一起进行PCR扩增,观察其特异性。对133 bp的目标条带进行序列测定,对比分析。结果PCR检测终宿主家犬感染细粒棘球绦虫具有高度的灵敏性,即使粪便中有1个虫卵(8pg的DNA),也能测出,但扩增后特异性较差,无法利用扩增带型进行诊断。4种寄生虫均具有相同的133 bp DNA重复序列,在400 bp均可检测出相同条带。结论PCR方法检测终宿主感染细粒棘球绦虫DNA重复序列尽管有优良的灵敏性,因其特异性差,不适合用于临床诊断和推广。张亚楼 Jean Mathieu Bart 温浩 马旭东 苗玉清 陈雪鸿 2006中国地方病学杂志2006,25,5:5
