| 3 | Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. | 4 | Restudy of the storage and migration model of the Quaternary groundwater in Beijing Plain area显示文摘Through a wide-spread groundwater sampling mainly at nested monitoring wells in the Beijing Plain area,combined with data collection,the study analyzes water level,hydrochemistry,temperature,tritium,deuterium,18O,14C and other aspects,and indicates that there exits an obvious stratification phenomenon of the groundwater in the alluvial plain area down in the middle part of alluvial-pluvial fans,and the hydraulic connection between layers is very weak.The analysis of the tritium concentration suggests that the average influence depth of the modern precipitation is approximately 120m.Based on the analysis of the 22 paleowater samples through the D18O method and 14C isotopic dating,the deep groundwater circulation in the fine sediments area of Beijing Plain down in the fringe part of alluvial-pluvial fans is relatively slow.On average the paleowater point occurs approximately below the 180m at these points.The main reasons for the formation of stratification and paleowater points are the water-blocking effect of the clay type strata and the compaction effect during the sedimentary process,especially the differential compaction,which could form closed or relatively closed stagnant aquifers.Groundwater in this kind of stagnant aquifers to some degree belongs to non-renewable resource,and should be reconsidered and re-arranged in the water resources management strategy. | LIU YuanZhang WU Qiang LIN Pei LIU JiuRong XING LiTing GAO ZhiHui | 2012 | Science China Earth Sciences2012,55,7: | 7 |
| 5 | Self‑Assembly MXene‑rGO/CoNi Film with Massive Continuous Heterointerfaces and Enhanced Magnetic Coupling for Superior Microwave Absorber显示文摘MXene, as a rising star of two-dimensional(2 D) materials, has been widely applied in fields of microwave absorption and electromagnetic shielding to cope with the arrival of the 5 G era. However, challenges arise due to the excessively high permittivity and the difficulty of surface modification of few-layered MXenes severely, which infect the microwave absorption performance. Herein, for the first time, a carefully designed and optimized electrostatic selfassembly strategy to fabricate magnetized MXene-r GO/Co Ni film was reported. Inside the synthesized composite film, r GO nanosheets decorated with highly dispersed Co Ni nanoparticles are interclacted into MXene layers, which effectively suppresses the originally self-restacked of MXene nanosheets, resulting in a reduction of high permittivity. In addition, owing to the strong magnetic coupling between the magnetic Fe Co alloy nanoparticles on the r GO substrate, the entire MXener GO/Co Ni film exhibits a strong magnetic loss capability. Moreover, the local dielectric polarized fields exist at the continuous heterointerfaces between 2 D MXene and r GO further improve the capacity of microwave loss. Hence, the synthesized composite film exhibits excellent microwave absorption property with a maximum reflection loss value of-54.1 d B at 13.28 GHz. | 6 | Cell Membrane-camouflaged Multi-functional Dendritic Large Pore Mesoporous Silica Nanoparticles for Combined Photothermal Therapy and Radiotherapy of Cancer显示文摘Combining photothermal therapy and radiotherapy(PTT-RT) with reducing tumor hypoxia acts as an important antitumor modality. However, it is a great challenge to realize photothermal therapy, radiotherapy and exogenous oxygen supply in one nanosystem. To realize a combination of the three functions, we fabricated a red blood cell membrane(RBCm)-camouflaged, red blood cell content(RBCc) and the copper sulfide(CuS) co-loaded dendritic large pore mesoporous silica nanoparticle(DLMSN/CuS/RBCc/ RBCm). The cell membrane coating endowed the nanoparticles with good stability in the physiological environment, and CuS allowed the nanoparticle exhibiting good photothermal and radiosensitization properties. RBCc loaded nanoparticle DLMSN/CuS/RBCc enhanced superior anti-tumor effect than DLMSN/CuS during combined PTT-RT therapy because the introduction of RBCc increased the exogenous oxygen supply. The in vitro study further demonstrated that the combination of photothermal therapy and radiotherapy induced superior antitumor efficacy than single therapy. | 7 | Building the Chordata Olfactory Receptor Database using more than 400,000 receptors annotated by Genome2OR显示文摘Olfactory receptors are poorly annotated for most genome-sequenced chordates.To address this deficiency,we developed a nhmmer-based olfactory receptor annotation tool Genome2OR(http://gffzz188fe103f8f1460as9bbqb9nwwuun6u9b.ffgz.tsg.suse.edu.cn/To Hanwei/Genome2OR.git),and used it to process 1,695 sequenced chordate genomes in the NCBI Assembly database as of January,2021.In total,765,248 olfactory receptor genes were annotated,with 404,426 functional genes and 360,822 pseudogenes,which represents a four-fold increase in the number of annotated olfactory receptors.Based on the annotation data,we built a database called Chordata Olfactory Receptor Database(CORD,http://gffzz608a37c433994787s9bbqb9nwwuun6u9b.ffgz.tsg.suse.edu.cn)for archiving,analysing and disseminating the data.Beyond the primary data,we offer derivative information,including pictures of species,cross references to public databases,structural models,sequence similarity networks and sequence profiles in the CORD.Furthermore,we did brief analyses on these receptors,including building a huge protein sequence similarity network covering all receptors in the database,and clustering them into 20 communities,classifying the 20 communities into three categories based on their presences/absences in ray-finned fish and/or lobe-finned fish.We infer that olfactory receptors should have unique activation and desensitization mechanisms by analysing their sequences and structural models.We believe the CORD can benefit the researchers and the general public who are interested in olfaction. | Wei Han Yiran Wu Liting Zeng Suwen Zhao | 2022 | Science China(Life Sciences)2022,65,12: | 1 |

In this paper, a nanostructured nanocomposite with graphene supported CoFe_2O_4 nanoparticles(NPs) was prepared via simple hydrothermal reaction. The uniform CoFe_2O_4 NPs were anchored on graphene sheets, which brought a good performance on cyclability. Combined with the optimization of graphene content, the anode delivered a better capacity retention of 944 m A h g^(-1)over 50 cycles at current density of 100 m A g^(-1)and the good reversible capacity as 990 m A h g^(-1)when the rate returned from 5 A g^(-1)to 0.1 A g^(-1)after 60 cycles. | 19 | Amyloid-beta peptide neurotoxicity in human neuronal cells is associated with modulation of insulin-like growth factor transport, lysosomal machinery and extracellular matrix receptor interactions显示文摘Extracellular deposits of the amyloid-beta peptide(Aβ) are known as the main pathological hallmark of Alzheimer's disease. In Alzheimer's disease, neurons are injured and die throughout the brain, a process in which Aβ neurotoxicity is considered to play an important role. However, the molecular mechanisms underlying Aβ toxicity that lead to neurodegeneration are not clearly established. Here we have elucidated the molecular pathways and networks which are impacted by Aβ in neurons using SH-SY5Y human neuroblastoma cells as a model. These cells were treated with Aβ_(1–42) peptides to study changes in biochemical networks using tandem mass tag labeled quantitative proteomic technique followed by computational analysis of the data. The molecular impacts of Aβ on cells were evident in a time-and dose-dependent manner, albeit the duration of treatment induced greater differential changes in cellular proteome compared to the effects of concentration. Aβ induced early changes in proteins associated with lysosomes, collagen chain trimerization and extracellular matrix receptor interaction, complement and coagulation cascade, oxidative stress induced senescence, ribosome biogenesis, regulation of insulin-like growth factor transport and uptake by insulin-like growth factor-binding protein.
