| 1 | Physical-biogeochemical interactions and potential effects on phytoplankton and Ulva prolifera in the coastal waters off Qingdao(Yellow Sea, China)显示文摘In recent years, the spectacular massive green tide of Ulva prolifera has become a recurrent phenomenon appearing every summer in the coastal waters off Qingdao(Yellow Sea, China), attracting the attention of scientists and local government. Based on multidisciplinary data collected during summer and winter, this study focuses on the hydrological characteristics and regional biogeochemical processes in coastal waters off Qingdao.The results show that the boundary of the Yellow Sea Cold Water Mass(YSCWM) can reach the Qingdao coastal region in summer and is locally raised to the upper layers to form coastal upwelling beyond tidal mixing and favorable wind. The regional summer upwelling off the Qingdao coast effectively enriches the nutrient concentrations in the upper water column and thus promotes growth of phytoplankton but reduces the dissolved oxygen(DO) concentration and pH value in the bottom. The regional summer upwelling off Qingdao coast may facilitate the growth and regional blooming of the U. prolifera that migrate to this region with the southerly wind.Additionally, the effects of the front on the aggregation of U. prolifera may be significant. In winter, the Yellow Sea Warm Current(YSWC) extends and spreads along the offshore region off the Subei Shoal towards the Qingdao coastal sea. This tongue-shaped warm water meets the cold coastal water off Qingdao, which leads to the formation of a physical front. As a consequence, remarkable fronts of nutrient and chlorophyll a(Chl a) also form between the shoreward warm water and the cold coastal water. | 4 | Expression Profiles of SIRT1 and APP Genes in Human Neuroblastoma SK-N-SH Cells Treated with Two Epigenetic Agents显示文摘In our previous studies, significant hypermethylation of the sirtuin 1(SIRT1) gene and demethylation of the b-amyloid precursor protein(APP) gene were found in patients with Alzheimer's disease(AD) compared with the normal population. Moreover, the expression of SIRT1 was significantly decreased while that of APP was increased in AD patients. These results indicated a correlation of DNA methylation with gene expression levels in AD patients. To further investigate the epigenetic mechanism of gene modulation in AD, we used two epigenetic drugs, the DNA methylation inhibitor 5-aza-20-deoxycytidine(DAC) and the histone deacetylase inhibitor trichostatin A(TSA), to treat human neuroblastoma SK-N-SH cells in the presence of amyloid b-peptide Ab25–35(Ab25–35). We found that DAC and TSA had different effects on the expression trends of SIRT1 and APP in the cell model of amyloid toxicity. Although other genes, such as microtubule-associated protein s, presenilin 1, presenilin 2, and apolipoprotein E, were up-regulated after Ab25–35treatment, no significant differences were found after DAC and/or TSA treatment. | 11 | Spatiotemporal variations of ambient air pollutants and meteorological influences over typical urban agglomerations in China during the COVID-19 lockdown显示文摘To investigate the air quality change during the COVID-19 pandemic,we analyzed spatiotemporal variations of six criteria pollutants in nine typical urban agglomerations in China using ground-based data and examined meteorological influences through correlation analysis and backward trajectory analysis under different responses.Concentrations of PM2.5,PM10,NO2,SO2 and CO in urban agglomerations respectively decreased by 18%–45%(30%–62%),17%–53%(22%–39%),47%-64%(14%–41%),9%–34%(0%–53%)and 16%-52%(23%–56%)during Lockdown(Post-lockdown)period relative to Pre-lockdown period.PM2.5 pollution events occurred during Lockdown in Beijing-Tianjin-Hebe(BTH)and Middle and South Liaoning(MSL),and daily O3 concentration rose to gradeⅡstandard in Post-lockdown period.Distinct from the nationwide slump of NO2 during Lockdown period,a rebound(~40%)in Post-lockdown period was observed in Cheng-Yu(CY),Yangtze River Middle-Reach(YRMR),Yangtze River Delta(YRD)and Pearl River Delta(PRD).With slightly higher wind speed compared with 2019,the reduction of PM2.5(51%–62%)in Post-lockdown period is more than2019(15%–46%)in HC(Harbin-Changchun),MSL,BTH,CP(Central Plain)and SP(ShandongPeninsula),suggesting lockdown measures are effective to PM2.5 alleviation.Although O3 concentrations generally increased during the lockdown,its increment rate declined compared with 2019 under similar sunlight duration and temperature.Additionally,unlike HC,MSL and BTH,which suffered from additional(>30%)air masses from surrounding areas after the lockdown,the polluted air masses reaching YRD and PRD mostly originated from the long-distance transport,highlighting the importance of joint regional governance. | Linping Fan Shuang Fu Xin Wang Qingyan Fu Haohao Jia Hao Xu Guimei Qin Xue Hu Jinping Cheng | 2021 | Journal of Environmental Sciences2021,33,8: | 0 |
| 12 | Effects of electrostimulation and administration of succinylcholine on the expression of Fos protein in mesencephalic periaqueductal gray matter of rats after simulated weightlessness显示文摘BACKGROUND: Expression of Fos in neurons of periaqueductal gray (PAG) is used to reflect the excitability. However, changes of expression of Fos in neurons of PAG are caused by injured electrostimulation after simulated weightlessness, and the relationship between pretreatment and injection of succinylcholine has not been determined yet. OBJECTIVE: To investigate the changes of expression of Fos in PAG induced by injured electrostimulation, pretreatment and injection of succinylcholine at 2 weeks after simulated weightlessness. DESIGN: Observational and controlled animal study. SETTING: Department of Physiology, Medical School, Xi’an Jiaotong University; Key Laboratory of Environment and Genes Related to Diseases, Ministry of Education. MATERIALS: A total of 24 adult female SD rats, of clean grade and weighing 180-220 g, were selected in this study. METHODS: The experiment was completed in the Experimental Animal Center of Xi’an Jiaotong University. ① All rats were randomly divided into 2 groups according to body mass: simulated weightlessness group and control group with 12 in each group. And then, each group was also divided into 3 subgroups: electrostimulation group, succinylcholine-pretreatment group and succinylcholine-injection group with 4 in each subgroup. ② The model of weightlessness was simulated by tail-suspended female rats, which were described and modified by Cheng Jie. Rats in normal control group were given the same interventions as simulated weightlessness group except for tail-suspended. ③ Experimental method: The rats in electrostimulation group were given nociceptive stimulus by a pair of subcutaneous electrodes inserted into 1 and 5 claw of left hindlimb. The stimulus (current: 10 mA; duration: 1 ms; interval: 1 s) lasted for 30 minutes. The rats in succinylcholine-pretreatment group received stimulus after intravenous administration of succinylcholine, rats in succinylcholine-injection group were not given stimulus, just received succinylcholine. ④ All rats were perfused and fixed after 2 hours from the end of stimulation. The brains were removed, and serial frozen sections of midbrain were stained using immunocytochemical method, observed and taken photos under light-microscope. The number and morphological characters of Fos-immunoreactive (Fos-IR) neurons in ventrolateral part of PAG were investigated. MAIN OUTCOME MEASURES: The alterations in number and morphological characters of Fos-IR neurons in ventrolateral PAG of all rats. RESULTS: A total of 24 rats were involved in the final analysis. ① The morphological changes of Fos-IR neurons: The expressions of Fos in ventrolateral part of PAG were observed in both control and simulated weightlessness groups rats after being given nociceptive stimulus. As compared with control group, Fos-IR neurons in simulated weightlessness group were dyed lightly, cellular integrity was impaired, and cellular verge was unclear. ② The numbers of Fos-IR neurons: In control group, the numbers of Fos-IR neurons in ventrolateral part of PAG in simulated weightlessness group were obviously lower than succinylcholine-pretreatment group, but obviously higher than succinylcholine-injection group (46.94±3.38, 71.06±8.96 and 35.04±4.62, respectively, P < 0.05). In 14-day simulated weightlessness group, the numbers of Fos-IR neurons in electrostimulation group were also obviously lower than succinylcholine-pretreatment group, and obviously higher than succinylcholine-injection group (27.77±3.27, 32.91±2.99 and 11.75±1.00, respectively, P < 0.05). The numbers of Fos-IR neurons in all subgroups in control group were obviously higher than those subgroups in simulated weightlessness group. Compared with electrostimulation group, the percentage of expression of Fos in ventrolateral part of PAG responsed to nociceptive stimulus after administration of succinylcholine (SCH) was increased to 51.83% in control group and 18.51% in simulated weightlessness group.
