1Liraglutide alleviates cardiac fibrosis through inhibiting P4hα-1 expression in STZ-induced diabetic cardiomyopathy显示文摘Diabetic cardiomyopathy is an important contributor to morbidity and mortality of diabetic patients by causing heart failure.Interstitial and perivascular fibrosis plays a crucial role in diabetic cardiomyopathy.However,there is a lack of effective specific treatments available for diabetic cardiomyopathy.In the present study,we aim to explore the effects of Liraglutide,a GLP-1 analogue,on diabetic cardiomyopathy in STZ-induced diabetic rats fed with high-fat diet.A total of 60 male Wistar rats were randomly assigned to three groups,i.e.normal group,model group,and Liraglutide group,with 20 rats in each group.Serum levels of TC,TG,LDL-C,NEFA,and hydroxyproline were measured using commercial kits.Cardiac function was evaluated by QRS waves,LVEDd,LVESd,and LVEF.Myocardial fibrosis was measured by immunohistochemistry.Our results demonstrated that chronic administration of Liraglutide decreased the level of blood glucose and significantly alleviated lipid metabolic disturbance compared with the model group.Furthermore,Liraglutide was found to improve the damaged cardiac function.In line with this,we also found that the alleviation of cardiac dysfunction was associated with the decreased fibrosis in diabetic myocardial tissues,which was reflected by the decreased expressions of P4hα-1,COL-1,COL-3,MMP-1,and MMP-9.Our results thus suggest that Liraglutide might have a myocardial protective effect by inhibiting P4hα-1-mediated myocardial fibrosis.Tong Zhao Huiqiang Chen Fei Xu Juan Wang Yusheng Liu Xiaowei Xing Linlin Guo Mingxiang Zhang Qinghua Lu 2019Acta Biochimica et Biophysica Sinica2019,51,3:9
2Excessive gestational weight gain in early pregnancy and insufficient gestational weight gain in middle pregnancy increased risk of gestational diabetes mellitus显示文摘Background:Gestational weight gain(GWG)is associated with the risk of gestational diabetes mellitus(GDM).However,the effect of weight gain in different trimesters on the risk of GDM is unclear.This study aimed to evaluate the effect of GWG on GDM during different trimesters.Methods:A birth cohort study was conducted from 2017 to 2020 in Shenzhen,China.In total,51,205 participants were included comprising two models(early pregnancy model and middle pregnancy model).Gestational weight(kg)was measured at each prenatal clinical visit using a standardized weight scale.Logistic regression analysis was used to assess the risk of GDM.Interaction analysis and mediation effect analysis were performed in the middle pregnancy model.Results:In the early pregnancy model,the risk of GDM was 0.858 times lower(95%confidence interval[CI]:0.786,0.937)with insufficient GWG(iGWG)and 1.201 times higher(95%CI:1.097,1.316)with excessive GWG after adjustment.In the middle pregnancy model,the risk of GDM associated with iGWG increased 1.595 times(95%CI:1.418,1.794)after adjustment;for excessive GWG,no significant difference was found(P=0.223).Interaction analysis showed no interaction between GWG in early pregnancy(GWG-E)and GWG in middle pregnancy(GWG-M)(F=1.268;P=0.280).The mediation effect analysis indicated that GWG-M plays a partial mediating role,with an effect proportion of 14.9%.Conclusions:eGWG-E and iGWG-M are associated with an increased risk of GDM.Strict control of weight gain in early pregnancy is needed,and sufficient nutrition should be provided in middle pregnancy.Aiqi Yin Fuying Tian Xiaoxia Wu Yixuan Chen Kan Liu Jianing Tong Xiaonian Guan Huafan Zhang Linlin Wu Jianmin Niu 2022Chinese Medical Journal2022,,9:5
3Surface Oxygen Injection in Tin Disulfide Nanosheets for Efficient CO_(2) Electroreduction to Formate and Syngas显示文摘Surface chemistry modification represents a promising strategy to tailor the adsorption and activation of reaction intermediates for enhancing activity.Herein,we designed a surface oxygen-injection strategy to tune the electronic structure of SnS_(2) nanosheets,which showed effectively enhanced electrocatalytic activity and selectivity of CO_(2) reduction to formate and syngas(CO and H_(2)).The oxygen-injection SnS_(2) nanosheets exhibit a remarkable Faradaic efficiency of 91.6%for carbonaceous products with a current density of 24.1 mA cm^(−2) at−0.9 V vs RHE,including 83.2%for formate production and 16.5%for syngas with the CO/H_(2) ratio of 1:1.By operando X-ray absorption spectroscopy,we unravel the in situ surface oxygen doping into the matrix during reaction,thereby optimizing the Sn local electronic states.Operando synchrotron radiation infrared spectroscopy along with theoretical calculations further reveals that the surface oxygen doping facilitated the CO_(2) activation and enhanced the affinity for HCOO*species.This result demonstrates the potential strategy of surface oxygen injection for the rational design of advanced catalysts for CO_(2) electroreduction.Tao Chen Tong Liu Tao Ding Beibei Pang Lan Wang Xiaokang Liu Xinyi Shen Sicong Wang Dan Wu Dong Liu Linlin Cao Qiquan Luo Wei Zhang Wenkun Zhu Tao Yao 2021Nano-Micro Letters2021,13,11:3
4SARS-CoV-2 crosses the blood-brain barrier accompanied with basement membrane disruption without tight junctions alteration显示文摘SARS-CoV-2 has been reported to show a capacity for invading the brains of humans and model animals.However,it remains undear whether and how SARS-CoV-2 crosses the blood-brain barrier(BBB).Herein,SARS-CoV-2 RNA was occasionally detected in the vascular wall and perivascular space,as well as in brain microvascular endothelial cells(BMECs)in the infected K18-hACE2 transgenic mice.Moreover,the permeability of the infected vessel was in creased.Furthermore,disin tegrity of BBB was discovered in the infected hamsters by administration of Evans blue.Interestingly,the expression of claudin5,ZO-1,occludin and the ultrastructure of tight junctions(TJs)showed unchanged,whereas,the basement membrane was disrupted in the infected animals.Using an in vitro BBB model that comprises primary BMECs with astrocytes,SARS-CoV-2 was found to infect and cross through the BMECs.Consistent with in vivo experiments,the expression of MMP9 was increased and collagen IV was decreased while the markers for TJs were not altered in the SARS-CoV-2-infected BMECs.Besides,inflammatory responses including vasculitis,glial activation,and upregulated inflammatory factors occurred after SARS-CoV-2 infection.Overall,our results provide evidence supporting that SARS-CoV-2 can cross the BBB in a transcellular pathway accompanied with basement membrane disrupted without obvious alteration of TJs.Ling Zhang Li Zhou Linlin Bao Jiangning Liu Hua Zhu Qi Lv Ruixue Liu Wei Chen Wei Tong Qiang Wei Yanfeng Xu Wei Deng-Hong Gao Jing Xue Zhiqi Song Pin Yu Yunlin Han Yu Zhang Xiuping Sun Xuan Yu Chuan Qin 2021Signal Transduction and Targeted Therapy2021,6,10:2
5PD1^+CCR2^+CD8^+T Cells Infiltrate the Central Nervous System during Acute Japanese Encephalitis Virus Infection显示文摘Japanese encephalitis(JE)is a viral encephalitis disease caused by Japanese encephalitis virus(JEV)infection.Uncontrolled inflammatory responses in the central nervous system(CNS)are a hallmark of severe JE.Although the CCR2-CCL2 axis is important for monocytes trafficking during JEV infection,little is known about its role in CNS trafficking of CD8^+T cells.Here,we characterized a mouse model of JEV infection,induced via intravenous injection(i.v.)and delineated the chemokines and infiltrating peripheral immune cells in the brains of infected mice.The CNS expression of chemokines,Ccl2,Ccl3,and Ccl5,and their receptors,Ccr2 or Ccr5.was significantly up-regulated after JEV infection and was associated with the degree of JE pathogenesis.Moreover,JEV infection resulted in the migration of a large number of CD8^+T cells into the CNS.In the brains of JEV-infected mice,infiltrating CD8^+T cells expressed CCR2 and CCR5 and were found to comprise mainly effector T cells(CD44^+CD62L_).JEV infection dramatically enhanced the expression of programmed death 1(PD-1)on infiltrating CD8^+T cells in the brain,as compared to that on peripheral CD8^+T cells in the spleen.This effect was more pronounced on infiltrating CCR2^+CD8^+T cells than on CCR2-CD8^+T cells.In conclusion,we identified a new subset of CD8^+T cells(PD1^+CCR2^+CD8^+T cells)present in the CNS of mice during acute JEV infection.These CD8^+T cells might play a role in JE pathogenesis.Fang Zhang Linlin Qi Tong Li Xiaojing Li Dan Yang Shengbo Cao Jing Ye Bin Wei 2019Virologica Sinica2019,34,5:2

13A neural network-based production process modeling and variable importance analysis approach in corn to sugar factory显示文摘Corn to sugar process has long faced the risks of high energy consumption and thin profits.However,it’s hard to upgrade or optimize the process based on mechanism unit operation models due to the high complexity of the related processes.Big data technology provides a promising solution as its ability to turn huge amounts of data into insights for operational decisions.In this paper,a neural network-based production process modeling and variable importance analysis approach is proposed for corn to sugar processes,which contains data preprocessing,dimensionality reduction,multilayer perceptron/convolutional neural network/recurrent neural network based modeling and extended weights connection method.In the established model,dextrose equivalent value is selected as the output,and 654 sites from the DCS system are selected as the inputs.LASSO analysis is first applied to reduce the data dimension to 155,then the inputs are dimensionalized to 50 by means of genetic algorithm optimization.Ultimately,variable importance analysis is carried out by the extended weight connection method,and 20 of the most important sites are selected for each neural network.The results indicate that the multilayer perceptron and recurrent neural network models have a relative error of less than 0.1%,which have a better prediction result than other models,and the 20 most important sites selected have better explicable performance.The major contributions derived from this work are of significant aid in process simulation model with high accuracy and process optimization based on the selected most important sites to maintain high quality and stable production for corn to sugar processes.Yi Tong Mou Shu Mingxin Li Yingwei Liu Ran Tao Congcong Zhou You Zhao Guoxing Zhao Yi Li Yachao Dong Lei Zhang Linlin Liu Jian Du 2023Frontiers of Chemical Science and Engineering2023,17,3:1

16Interface behavior of chalcopyrite during flotation from cyanide tailings显示文摘The interface characteristics of cyanide tailings are different from those of the raw ore. In this study, valuable elements could not be thoroughly recovered via the flotation of cyanide tailings from Shandong, China. The interface and floatability of these tailings were investig- ated by phase analysis and flotation tests. The chalcopyrite in the cyanide tailings was fine and had a porous surface. The floatability of 68% chalcopyrite was similar to that of galena in the presence of a collector. A layer of fine galena particles compactly wrapped the chalcopyrite. The chalcopyrite recovery sharply decreased as the nonpolar oil residue in cyanide tailings was extracted using alcohol;however, this removal had no effect on the galena. 18Freezing-directed construction of enzyme/nano interfaces:Reagentless conjugation,superior activity,and better stability显示文摘Immobilizing enzyme to nano interfaces has demonstrated to be a favorable strategy for prompting the industrialized application of enzyme.Despite tremendous endeavor has been devoted to using gold nanoparticles(AuNPs)as conjugation matrix due to its fascinating physico-chemical properties,maintaining enzymatic activity while circumventing cumbersome modification remains a formidable challenge.Herein,the freezing-directed conjugation of enzyme/nano interfaces was constructed without extra reagent.As the proof of concept,glucose oxidase(GOx)was chosen as model enzyme.The one-pot conjugation process can be facilely completed at−20°C under aqueous solution.Moreover,with the loading of GOx on AuNP at freezing,the enzyme exhibited superior catalytic activity and stability upon thermal and pH perturbation.The mechanism of boosted activity was then discussed in detail.It was found that higher loading density under freezing condition and more enzyme tending to bind AuNPs via Au-S bond were the main factors for the superior activity.More importantly,this methodology was universal and can also be applied to other enzyme which contains natural cysteine,such as horseradish peroxidase(HRP)and papain.This facile conjugation strategy accompanied by remarkable bioactivity expand the possibilities for enzymatic biosensing,microdevice and even drug delivery.Ke Quan Jiajie Tong Lifang Chen Shuyao Fang Mengjiao Li Linlin Wu Zhihe Qing 2024Chinese Chemical Letters2024,35,1:0
