3Mucus production stimulated by IFN-AhR signaling triggers hypoxia of COVID-19显示文摘Silent hypoxia has emerged as a unique feature of coronavirus disease 2019(COVID-19).In this study,we show that mucins are accumulated in the bronchoalveolar lavage fluid(BALF)of COVID-19 patients and are upregulated in the lungs of severe respiratory syndrome coronavirus 2(SARS-CoV-2)-infected mice and macaques.We find that induction of either interferon(IFN)-P or IFN-y upon SARS-CoV-2 infection results in activation of aryl hydrocarbon receptor(AhR)signaling through an IDO-Kyn-dependent pathway,leading to transcriptional upregulation of the expression of mucins,both the secreted and membrane-bound,in alveolar epithelial cells.Consequently,accumulated alveolar mucus affects the blood-gas barrier,thus inducing hypoxia and diminishing lung capacity,which can be reversed by blocking AhR activity.These findings potentially explain the silent hypoxia formation in COVID-19 patients,and suggest a possible intervention strategy by targeting the AhR pathway.Yuying Liu Jiadi Lv Jiangning Liu Man Li Jing Xie Qi Lv Wei Deng Nannan Zhou Yabo Zhou Jiangping Song Peng Wang Chuan Qin Wei-Min Tong Bo Huang 2020Cell Research2020,30,12:5

8SARS-CoV-2 treatment effects induced by ACE2-expressing microparticles are explained by the oxidized cholesterolincreased endosomal pH of alveolar macrophages显示文摘Exploring the cross-talk between the immune system and advanced biomaterials to treat SARS-CoV-2 infection is a promising strategy.Here,we show that ACE2-overexpressing A549 cell-derived microparticles(AO-MPs)are a potential therapeutic agent against SARS-CoV-2 infection.Intranasally administered AO-MPs dexterously navigate the anatomical and biological features of the lungs to enter the alveoli and are taken up by alveolar macrophages(AMs).Then,AO-MPs increase the endosomal pH but decrease the lysosomal pH in AMs,thus escorting bound SARS-CoV-2 from phago-endosomes to lysosomes for degradation.This pH regulation is attributable to oxidized cholesterol,which is enriched in AO-MPs and translocated to endosomal membranes,thus interfering with proton pumps and impairing endosomal acidification.In addition to promoting viral degradation,AO-MPs also inhibit the proinflammatory phenotype of AMs,leading to increased treatment efficacy in a SARS-CoV-2-infected mouse model without side effects.These findings highlight the potential use of AO-MPs to treat SARS-CoV-2-infected patients and showcase the feasibility of MP therapies for combatting emerging respiratory viruses in the future.Zhenfeng Wang Jiadi Lv Pin Yu Yajin Qu Yabo Zhou Li Zhou Qiangqiang Zhu Shunshun Li Jiangping Song Wei Deng Ran Gao Yuying Liu Jiangning Liu Wei-Min Tong Chuan Qin Bo Huang 2022Cellular & Molecular Immunology2022,19,2:1

10DNMT3A inhibits E2F1-induced arterial marker expression and impairs angiogenesis in human umbilical artery endothelial cells显示文摘Arterial marker genes EphrinB2 and HEY2 are essential for cardiovascular development and post-natal neovascularization.Our previous study confirmed.that E2F1 could activate the transcription of EphrinB2 and HEY2 in human mesenchymal stem cells;however,the detailed mechanism has not been resolved yet.In this study,we focused on the interaction between E2F1 and DNMT3A,a de novo DNA methyltransferase,on regulating the expression of EphrinB2 and HEY2,and explored the potential mechanisms.Gain-and loss-of-function experiments implicated the positive effect of E2F1 on the expression of EphrinB2 and HEY2 and tube formation in human umbilical artery endothelial cells.Accumulation of DNMT3A decreased the levels of EphrinB2 and HEY2,and impaired tube formation induced by E2F1,while inhibiting DNMT3A by RNA interference aug-mented their expression and angiogenesis in E2F 1-trasfected cells.We then asked whether the low expressions of EphrinB2 and HEY2 induced by DNMT3A are related to the methylation status of their promoters.Surprisingly,the methylation status of the CpG islands in the promoter region was not significantly affected by overexpression of exogenous DNMT3A.Furthermore,the inter-action between E2F1 and DNMT3A was confirmed by co-immunoprecipitation.DNMT3A could inhibit the transcription of EphrinB2 and HEY2 promoters by affecting the binding of E2F1 to its recognition sequences as revealed by luciferase reporter assay and chromatin immunoprecipita-tion.These results identified a novel mechanism underlying the cooperation of DNMT3A with E2F1 on regulating target gene expression,and revealed their roles in the angiogenic process.Kaiyue Su Ningning Lin Shouqiang Xie Yabo Han Zaiming Yang Hongmin Zhang Hongpeng He Sa Zhou Wenjian Ma Tongcun Zhang Nan Wang 2020Acta Biochimica et Biophysica Sinica2020,52,11:1
11Establishment of image-guided radiotherapy of orthotopic hepatocellular carcinoma mouse model显示文摘Background:Hepatocellular carcinoma(HCC)is the most common type of liver cancer.Recently,developments in radiotherapy technology have led to radiotherapy becoming one of the main therapeutics of HCC.Therefore,a suitable animal model for radiotherapy of the orthotopic HCC mouse model is urgently needed.Methods:In the present study,Hepa1-6 cells were injected into the liver of C57BL/6 mice in situ to mimic the pathological characteristics of the original HCC.Tumor formation was monitored by applying magnetic resonance imaging techniques and verified by H&E histopathological staining,AFP staining,and Ki67 staining.A single dose of 10 Gy X-ray was applied to simulate clinical radiotherapy plans using image-guided radiotherapy(IGRT)equipment.The efficiency of radiotherapy was then assessed by examining tumor size and weight one week after radiation.Cleaved-caspase3 staining and TUNEL were used to assess apoptosis in tumor tissues.Results:Intrahepatic tumor development was detected in the liver according using MRI.A high-density shadow could be seen 10 days after cell injection,which indicated the formation of HCC in vivo.The tumors grew steadily bigger,and underwent precision radiotherapy 20 days after injection.The typical pathological characteristics of HCC,such as large,deeply stained nuclei and irregular cell size,were visible with H&E staining.After radiotherapy,significantly higher expression of the immunohistochemical markers Ki67 and AFP were detected in tumor tissue than in the nearby normal tissue.Compared with the control group,the tumor volume(p=0.05)and weight(p<0.05)of the irradiated group were significantly reduced.In addition,a higher frequency of apoptosis was identified in irradiated HCC tumor tissue using the TUNEL and cleaved-caspase3 staining assay.Conclusions:In a well-established orthotopic HCC model,MRI was utilized to monitor the formation of tumors,and IGRT was used to simulate clinical radiotherapy.The present study could provide a suitable preclinical system for HCC radiotherapyrelated studies.Kaixiao Zhou Yabo Jiang Shuang Feng Wei Mo Jing Nie Jianping Cao Yang Jiao 2023Animal Models and Experimental Medicine2023,6,5:0

13An efficient approach for the treatment of radioactive waste perfluoropolyether lubricants via a synergistic effect of thermal catalysis and immobilization显示文摘Perfluoropolyether(PFPE)lubricants are a kind of high-molecular polymer with many excellent properties.However,the use of PFPEs in the nuclear industry can lead to partial decomposition and carrying radionuclides,resulting in a large amount of radioactive waste PFPE lubricants annually.Moreover,radioactive waste PFPE lubricants are difficult to be effectively treated due to their high stability,the risk of possible leakage of radionuclides,and hypertoxic fluorine-containing by-products.In this study,without any precedent,a strategy of Mn O_(2)-catalyzed decomposition and Na_(2)CO_(3)-immobilized conversion was proposed for PFPE lubricant decomposition and fluorine immobilization simultaneously based on the Lewis acid-base and oxygen vacancies concept.A high fluorine conversion efficiency of 95.4%was achieved.Meanwhile,the mechanism of decomposition suggested that Mn O_(2)mainly provided Lewis acid sites and attacked the(basic)fluorine or oxygen atoms in PFPE molecules.The decomposition of PFPE chains was proceed down and volatile fluorine-containing gas was released by partial electron transfer,intramolecular disproportionation reaction,and unzipping fashion.Subsequently,gas by-products could be further oxidized and then immobilized into fluoride salts by carbonate solid absorbents.Overall,this study provides a simple,safe,and potentially practical strategy for the harmless conversion of refractory fluorinated organic wastes,especially perfluoropolymers.Xinhang Du Wenhua Tong Xinyu Zhou Jia Luo Yuqiu Liu Yabo Wang Panyu Li Yongkui Zhang 2024Journal of Environmental Sciences2024,,2:0

17Research on damage failure mechanism and dynamic mechanical behavior of layered shale with different angles under confining pressure显示文摘The hydrostatic or confining pressure of deep rocks has a significant impact on the mechanical behavior of brittle materials.Especially when confining pressure is applied,the mechanical properties of rock materials will undergo significant changes.Considering that the process of shale sample subjected to impact load is in a closed container in the dynamic triaxial SHPB test,the failure process of the sample cannot be observed.Meanwhile,the activation volume of the shale sample would be large and local failure would occur in the test under the high strain rate loading.Therefore,thefinite element model of shale considering the bedding effect under confining pressure was established in this study.Taking shale materials with different bedding dip angles as simulation objects,the dynamic failure characteristics of shale were studied using the dynamic analysis software ANSYS/LS‐DYNA from three aspects:stress‐strain curve,failure growth process,and failure morphology.The research results obtained can serve as the key technical parameters for deep resource extraction.Ning Luo Haohao Zhang Yabo Chai Penglong Li Cheng Zhai Jianan Zhou Tianran Ma 2023Deep Underground Science and Engineering2023,2,4:0
