1Rupture segmentation and slip partitioning of the mid-eastern part of the Kunlun Fault,north Tibetan Plateau显示文摘The Kunlun Fault,an active fault on the border between the Bayan Har and Kunlun-Qaidam blocks,is one of the major left lateral strike-slip faults in the Tibetan Plateau.Previous research has not reached a consensus on agreeable slip rates along much of its length and the slip rate gradient along the eastern part,both of which play critical roles in a range of models for the eastward extrusion and thickened crust of the Tibetan Plateau.New slip rates have been determined at sites along the eastern part of the Kunlun Fault by dating deposits and measuring atop displaced fluvial terrace risers.Field investigations and interpretation of satellite images reveal geometrical features of the fault and the late Quaternary offset,new earthquake ruptures and surface-rupturing segmentation,from which long-term slip rates and earthquake recurrence intervals on the fault are estimated.The tectonic geomorphology method has determined that the long-term horizontal slip rates on the Tuosuohu,Maqin and Maqu segments from west to east are 11.2±1,9.3±2,and 4.9±1.3 mm/a while their vertical slip rates are 1.2±0.2,0.7±0.1,and 0.3 mm/a in the late Quaternary.Results indicate that the slip rates regularly decrease along the eastern ~300 km of the fault from >10 to <5 mm/a.This is consistent with the decrease in the gradient such that at the slip rate break point is at the triple point intersection with the transverse fault,which in turn is transformed to the Awancang Fault.The vector decomposition for this tectonic transformation shows that the western and eastern branches of the Awancang Fault fit the slip-partitioning mode.The slip rate of the southwestern wall is 4.6 mm/a relative to the northeastern wall and the slip direction is 112.1°.The mid-eastern part of the Kunlun Fault can be divided into three independent segments by the A'nyêmaqên double restraining bend and the Xigongzhou intersection zone,which compose the surface rupture segmentation indicators for themselves as well as the ending point of the 1937 M7.5 Tuosuohu earthquake.The average recurrence interval of the characteristic earthquakes are estimated to be 500-1000 a,respectively.The latest earthquake ruptures occurred in AD 1937 on the western Tuosuohu segment,as compared to ~514-534 a BP on the Maqin segment,and ~1055 to 1524 a BP on the Maqu segment.This may indicate a unidirectional migration for surface rupturing earthquakes along the mid-eastern Kunlun Fault related to stress triggered between these segments.Meanwhile,the long-term slip rate is obtained through the single event offset and the recurrence interval,which turn out to be the same results as those determined by the offset tectonic geomorphology method,i.e.,the decreasing gradient corresponds to the geometrical bending and the fault's intersection with the transverse fault.Therefore,the falling slip rate gradient of the mid-eastern Kunlun Fault is mainly caused by eastward extension of the fault and its intersection with the transverse fault.LI ChenXia XU XiWei WEN XueZe ZHENG RongZhang CHEN GuiHua YANG Hu AN YanFen GAO Xiang 2011Science China Earth Sciences2011,54,11:33
2Degradation of organic pollutants by a Co_3O_4 -graphite composite electrode in an electro-Fenton-like system显示文摘A novel composite electrode was constructed by pressing together Co3O4 and graphite and it was used as the cathode in an electro-Fenton-like (EFL) system. The poor electron transport characteristic of Co3O4 was overcome by incorporating graphite. In situ electro-catalytic generation of hydroxyl radicals (·OH) occurred at high current efficiencies from pH 2-10, extending the traditional Fenton reaction pH range. Cyclic voltammetry and AC impedance spectrometry were used to characterize the composite electrode. The ability of the EFL system to degrade organic compounds was investigated using sulforhodamine B (SRB) and 2,4-dichlorophenol (2,4-DCP) as probes. Decoloration of SRB (1.0×10-5 mol/L) was complete (100%) in 150 min and SRB was effectively degraded from pH 2-10. The decomposition of SRB was studied using Fourier transform infrared spectroscopy (FT-IR) and total organic carbon (TOC) analysis and results indicated that the final degradation products were carbon dioxide, carboxylic acids and amines. The EFL system also decomposed 2,4-DCP and the degradation was 98.6% in 240 min. Electro-catalytic degradation of SRB occurs by a ·OH mechanism. After 5 times reused, the degradation rate of SRB did not significantly slow down. 3Characteristics of the microbial communities in the integrated vertical-flow constructed wetlands显示文摘Microorganisms play an important role in removing pollutants from constructed wetlands.We investigated the microbial characteristics in a novel integrated vertical-flow constructed wetland(IVCW) ,which has been in operation in Wuhan,China since 1998.We used phospholipid fatty acid(PLFA) and amoA gene to analyze the structure and diversity of the microbial community within the IVCW.PLFA results suggested that the amount of bacterial PLFA was significantly higher than that of fungal PLFA,but the total microbial biomass represented by PLFA index was low in the system.Microbial spatial distribution showed significantly higher bacterial(both G+and G-) and fungal biomass in the surface than in the subsurface layers.The ratios of monounsaturated to branched PLFA demonstrated that an anaerobic layer sandwiched by two aerobic layers existed in the IVCW,consistent with the redox potential results.Analysis of the amoA revealed the presence of Nitrosomonas-like sequences in the surface substrate of the downflow chamber and apparent diversities of ammonia-oxidizing bacteria in the system.These results suggest that microorganisms,despite their relatively low biomass,have inhabited the IVCW,and the results will offer some valuable information on microbe to system designers and managers.ZHOU Qiaohong,HE Feng,Zhang Liping,WANG Yanfen,WU Zhenbin State Key Laboratory of Freshwater Ecology and Biotechnology,Institute of Hydrobiology,Chinese Academy of Sciences,Wuhan 430072,China Department of Biology and Chemistry,City University of Hong Kong 2009Journal of Environmental Sciences2009,21,9:14
4Fission track dating of the Cenozoic uplift in Mabian area, southern Sichuan Province, China显示文摘The apparent ages of samples are obtained from fission track dating of apatite samples collected from the fault zones in Mabian area, southern Sichuan Province. In addition, thermal history is simulated from the obtained data by applying AFT Solve Program, to acquire the thermal evolution history of the samples. The result shows that tectonically the Mabian area was relatively stable between 25 and 3 Ma, compared to the inner parts and other marginal areas of the Tibetan Plateau. The studied area had little response to the rapid uplift events that occurred for several times in the Tibetan Plateau during 25-3 Ma. The latest thermal event related to the activity of the Lidian fault zone (about 8 Ma ) is later than that of the Ebian fault zone (18-15 Ma ) to the west, indicating to some extent that the evolution of fault activity in the Mabian area has migrated from west to east. The latest extensive tectonic uplift occurred since about 3 Ma. As compared with the Xianshuihe fault zone, the Mabian area is closer to the east- ern margin of the plateau, while the time of fast cooling event in this area is later than that in the southeast segment of the Xianshuihe fault zone (3.6-3.46 Ma ). 5Visible light-induced degradation of organic pollutants using Fe(II) supported on silica gel as an effective catalyst显示文摘Silica gel-supported Fe(II) (SiOFe) was prepared and used for heterogeneous degradation of sulforho-damine B (SRB) and 2,4-dichlorophenol (DCP) under visible irradiation (λ > 420 nm) as an effective catalyst. UVvisible spectra, and infrared Spectrophotometry (IR), fluorescence, total organic carbon (TOC) and electron spin resonance (ESR) measurements were employed to analyze the photoreaction products. The results showed that SRB could be efficiently degraded by SiOFe/H2O2 system under visible irradiation with 100% decolorization and 72.3% TOC removal after 180 min illumination. 6Activity of the Lenglongling fault system and seismotectonics of the 2016 MS6.4 Menyuan earthquake显示文摘The MS6.4 Menyuan earthquake occurred on the northern side of the Lenglongling fault(LLLF) in the mid-western of the Qilian-Haiyuan fault zone on January 21, 2016. The earthquake epicenter was distant from the Minle-Damaying and Huangcheng-Shuangta faults, eastern of the Northern Qilian Shan fault zone. A near northwest-striking rupture plane intersects the two faults at a certain angle. The focal mechanism solution shows that this was a thrust-type earthquake, slightly different from the strike-slip movement with a thrust component of the LLLF. Field geological mapping, tectonic geomorphology analysis, trench excavation and 14 C dating reveal that(1) the LLLF has been obviously active since the Holocene, and may behave with characteristic slip behavior and produce M_W7.3–7.5 earthquakes;(2) the LLLF appears as a flower structure in terms of structure style, and dips NNE at a steep angle; and(3) the most recent earthquake event occurred after 1815–1065 a BP. An associated fault, the Northern Lenglongling fault(NLLLF), is located at the northwestern end of the LLLF. Consequently, the NLLLF was continually subject to tectonic pushing effects from the left-lateral shear at the end of the LLLF, and, accordingly, it bent and rotated outward tectonically.Subsequently, the fault deviated from the dominant rupture azimuth and activity weakened. In the late Quaternary, it behaved as a thrust fault with no obvious deformation at the surface. This is indicated by the arc shape, with a micro-protrusion northeastward,and no geologic or geomorphic signs of surface rupturing since the late Quaternary. However, such faults could still rupture at depth, producing moderate-strong earthquakes. The geometric and kinematic properties of the NLLLF are in good agreement with the occurrence and kinematic properties of nodal plane 2, and with the distribution characteristics of the aftershocks and seismic intensity. Therefore, the NLLLF is a more suitable seismogenic structure for the MS 6.4 Menyuan earthquake. In addition, the thrust movement of the NLLLF accommodates subsequent movement of the LLLF. 7Visible light-induced N-doped TiO_2 nanoparticles for the degradation of microcystin-LR显示文摘N-doped nano-crystalline TiO2 powders have been synthesized by the sol-gel method.The shape and crystal structure of the resulting N-doped TiO2 were investigated by X-ray Photoelectron Spectroscopy (XPS),X-ray spectroscopy (XRD),Transmission Electron Microscopy (TEM) and UV-vis reflection spectrum.The results showed that doping TiO2 with nitrogen can lower its band gap and apparently shift its optical response to the visible region.Under the visible light (λ> 420 nm) irradiation,the MC-LR was degraded by the synthesized N-TiO2 nano-material.The variation of MC-LR amount and its intermediates were detected by high performance liquid chromatography (HPLC) and LC-MS,respectively.The mineralization of MC-LR was determined by total organic carbon (TOC) analysis.Simultaneously,transient oxidative species generated during photocatalysis were tracked by electron spin resonance (ESR) and Peroxidase method.All these results indicated that visible-light excited N-TiO2 can activate molecular oxygen and thereby achieve degradation of MC-LR completely within 14 h.The removal of 59% of TOC was achieved after 20 h irradiation.The major oxidative species in the system were hydroxyl radical (·OH) and H2O2.13 Kinds of intermediates were primarily identified in the process.Based on these results,a reasonable conclusion was drawn for the degradation of MC-LR wherein its four positions are easy to be attacked by the photo-generated OH radical followed by the hydrolyzation of peptides.YANG Jing,CHEN DengXia,DENG AnPing,FANG YanFen,LUO GuangFu,LI DeJiang,LI RuiPing & HUANG YingPing Alan G.MacDiarmid Research Institute of Renewable Energy,China Three Gorges University,Yichang 443000,China 2010Science China Chemistry2010,53,8:12
8The calcium-dependent kinase OsCPK24 functions in cold stress responses in rice显示文摘Calcium-dependent protein kinases(CPKs)are serine/threonine protein kinases that function in plant stress responses. Although CPKs are recognized as key messengers in signal transduction, the specific roles of CPKs and the molecular mechanisms underlying their activity remain largely unknown. Here, we characterized the function of Os CPK_(24), a cytosol-localized calciumdependent protein kinase in rice. Os CPK_(24) was universally and highly expressed in rice plants and was induced by cold treatment. Whereas Os CPK_(24) knockdown plants exhibited increased sensitivity to cold compared to wild type(WT), Os CPK_(24)-overexpressing plants exhibited increased cold tolerance. Plants overexpressing Os CPK_(24) exhibited increased accumulation of proline(an osmoprotectant) and glutathione(an antioxidant) and maintained a higher GSH/GSSG(reduced glutathione to oxidized glutathione) ratio during cold stress compared to WT. In addition to these effects in response to cold stress, we observed the kinase activity of Os CPK_(24) varied under different calcium concentrations. Further,Os CPK_(24) phosphorylated Os Grx_(10), a glutathionedependent thioltransferase, at rates modulated by changes in calcium concentration. 9Arabidopsis co-chaperonin CPN20 antagonizes Mg-chelatase H subunit to derepress ABA-responsive WRKY40 transcription repressor显示文摘Our previous study demonstrated that a chloroplast co-chaperonin 20(CPN20),one of the interaction partners of the magnesium-protoporphyrin IX chelatase H subunit(CHLH/ABAR),negatively regulates ABA signaling at the same node with ABAR but upstream of WRKY40 transcription repressor in Arabidopsis thaliana.In the present experiment,we showed that ABA directly inhibits the ABAR-CPN20 interaction,and also represses expression of CPN20,which depends on ABAR.CPN20 inhibits ABAR-WRKY40 interaction by competitively binding to ABAR.ABAR downregulates,but CPN20 upregulates,WRKY40 expression.The cpn20-1 mutation induces downregulation of WRKY40,and suppresses the upregulated level of WRKY40 due to the cch mutation in the ABAR gene.ABA-induced repressive effect of the WRKY40 gene is strengthened by downregulation of CPN20 but reduced by upregulation of CPN20.Together with our previously reported genetic data,we provide evidence that CPN20 functions through antagonizing the ABAR-WRKY40 coupled pathway,and ABA relieves this pathway of repression by inhibiting the ABAR-CPN20 interaction to activate ABAR-WRKY40 interaction.ZHANG XiaoFeng JIANG Tao YU YongTao WU Zhen JIANG ShangChuan LU Kai FENG XiuJing LIANG Shan LU YanFen WANG XiaoFang ZHANG DaPeng 2014Science China(Life Sciences)2014,57,1:11
10SHP2 promotes proliferation of breast cancer cells through regulating Cyclin D1 stabilityviathe PI3K/AKT/GSK3β signaling pathway显示文摘Objective:The tyrosine phosphatase SHP2 has a dual role in cancer initiation and progression in a tissue type-dependent manner.Several studies have linked SHP2 to the aggressive behavior of breast cancer cells and poorer outcomes in people with cancer.Nevertheless,the mechanistic details of how SHP2 promotes breast cancer progression remain largely undefined.Methods:The relationship between SHP2 expression and the prognosis of patients with breast cancer was investigated by using the TCGA and GEO databases.The expression of SHP2 in breast cancer tissues was analyzed by immunohistochemistry.CRISPR/Cas9 technology was used to generate SHP2-knockout breast cancer cells.Cell-counting kit-8,colony formation,cell cycle,and EdU incorporation assays,as well as a tumor xenograft model were used to examine the function of SHP2 in breast cancer proliferation.Quantitative RT-PCR,western blotting,immunofluorescence staining,and ubiquitination assays were used to explore the molecular mechanism through which SHP2 regulates breast cancer proliferation.Results:High SHP2 expression is correlated with poor prognosis in patients with breast cancer.SHP2 is required for the proliferation of breast cancer cellsin vitro and tumor growthin vivo through regulation of Cyclin D1 abundance,thereby accelerating cell cycle progression.Notably,SHP2 modulates the ubiquitin–proteasome-dependent degradation of Cyclin D1viathe PI3K/AKT/GSK3βsignaling pathway.SHP2 knockout attenuates the activation of PI3K/AKT signaling and causes the dephosphorylation and resultant activation of GSK3β.GSK3βthen mediates phosphorylation of Cyclin D1 at threonine 286,thereby promoting the translocation of Cyclin D1 from the nucleus to the cytoplasm and facilitating Cyclin D1 degradation through the ubiquitin–proteasome system.Conclusions:Our study uncovered the mechanism through which SHP2 regulates breast cancer proliferation.SHP2 may therefore potentially serve as a therapeutic target for breast cancer.Yue Yuan Yanling Fan Zicong Gao Xuan Sun He Zhang Zhiyong Wang Yanfen Cui Weijie Song Zhaosong Wang Fei Zhang Ruifang Niu 2020Cancer Biology & Medicine2020,17,3:11
11CD30 expression in extranodal natural killer/T-cell lymphoma,nasal type among 622 cases of mature T-cell and natural killer-cell lymphoma at a single institution in South China显示文摘Background:Mature T-cell and natural killer(NK)-cell lymphomas compose a heterogeneous group of non-Hodgkin lymphomas,and extranodal NK/T-cell lymphoma,nasal type(ENKTL) is an aggressive subtype with sporadic CD30 expression.However,the significance of CD30 expression in ENKTL is controversial.We aimed to classify a large cohort of patients with mature T-cell and NK-cell lymphomas according to the 2016 World Health Organization(WHO) classification guidelines and to study the association between CD30 expression and prognosis of patients with ENKTL.Methods:We selected consecutive patients with mature T-cell and NK-cell lymphomas who attended our institution between September 1,2009 and August 31,2013.We classified the lymphomas according to the 2016 revision of the WHO classification of lymphoid neoplasms,analyzed the associations between CD30 expression and clinicopathologic features of ENKTL patients,and evaluated the prognostic implications of CD30 expression.Results:We identified 622 consecutive patients with mature T-cell and NK-cell lymphomas,including 317(51.0%)patients with ENKTL.In addition,CD30 expression was detected in 43(47.3%) of a subset of 91 patients with ENKTL.No clinicopathologic features were associated with CD30 expression,and CD30 positivity showed no prognostic significance in patients with ENKTL.Conclusions:ENKTL is the most common type of mature T-cell and NK-cell lymphoma diagnosed at our institution.CD30 is frequently expressed in ENKTL and represents a therapeutic target;however,it may not be a prognostic marker.Yanfen Feng Huilan Rao Yiyan Lei Yuhua Huang Fang Wang Yu Zhang Shaoyan Xi Qiuliang Wu Jianyong Shao 2017Chinese Journal of Cancer2017,36,6:10
12A three-dimensional semi-implicit unstructured grid finite volume ocean model显示文摘A new three-dimensional semi-implicit finite-volume ocean model has been developed for simulating the coastal ocean circulation, which is based on the staggered C -unstructured non-orthogonal grid in the horizontal direction and z -level grid in the vertical direction. The three-dimensional model is discretized by the semi-implicit finite-volume method, in that the free-surface and the vertical diffusion are semi-implicit, thereby removing stability limitations associated with the surface gravity wave and vertical diffusion terms. The remaining terms in the momentum equations are discretized explicitly by an integral method. The partial cell method is used for resolving topography, which enables the model to better represent irregular topography. The model has been tested against analytical cases for wind and tidal oscillation circulation, and is applied to simulating the tidal flow in the Bohai Sea. 13Noxious invasive Eupatorium adenophorum may be a moving target:Implications of the finding of a native natural enemy,Dorylus orientalis显示文摘Eupatorium adenophorum is a noxious invasive weed that is distributed throughout subtropical areas worldwide.Enemy release may be at least one of the reasons underlying its success as an invader.In this study,we observed damage to the epidermis,cortex,phloem,cambium,and xylem tissues of roots and stems around the root collar of E.adenophorum growing in an experimental field in Yunnan Province,southwest China.The damage was caused by Dorylus orientalis,a polyphagous ant feeding on the weed and resulted in the death of the plants by interrupting nutrient exchange between shoots and roots.This ant showed selectivity for the invader to some extent.The finding of D.orientalis and other native enemies indicate that E.adenophorum may gradually naturalize in introduced landscapes.In addition,this ant may have potential for use as a biocontrol agent against the invader.NIU YanFen FENG YuLong XIE JianLei LUO FuCheng 2010Chinese Science Bulletin2010,55,33:10

15Inflammasome activation in mouse inner ear in response to MCMV induced hearing loss显示文摘Objective: To identify presence of inflammasome activated in mouse cochlea with sensorineural hearing loss(SNHL) caused by cytomegalovirus(CMV) infection.Method: MCMV was injected into the right cerebral hemisphere in neonatal BALB/c mice at 2000 pfu virus titers. Auditory brainstem responses(ABRs) were tested to evaluate hearing at 21 days. Histopathological studies were conducted to confirm localizations of MCMV infected cells in the inner ear. Expression of inflammasome related factors was assessed by immunofluorescence, Quantitative real-time PCR and Western blotting.Results: In the mouse model of CMV induced SNHL, inflammasome related kinase Caspase-1 and downstream inflammatory factor IL-1b and IL-18 were found increased and activated after CMV infection in the cochlea. These factors could further up-regulate expression of IL-6 and TNFa. These inflammatory factors are neurotoxicity and may contribute to hearing impairment. Furthermore, we also detected significantly increased AIM2 protein that accumulated in the SGN of cochleae with CMV infection.Significance: We have shown that inflammasome as a novel inherent immunity mechanism may contribute to hearing impairment.Conclusion: Our data indicate that imflammasome assemble in mouse inner ear in response to CMV infection. We have revealed a novel pathology event in CMV induced SNHL involving activation of inflammasome in mouse cochlea. 16Comparative study on skin temperature response to menstruation at acupuncture points in healthy volunteers and primary dysmenorrhea patients显示文摘OBJECTIVE:To assess skin temperature response to menstruation at acupuncture points in primary dysmenorrhea(PD) patients and healthy volunteers so as to explore acupuncture point specificity in reflecting diseases in the light of skin temperature.METHODS:Fifty-two PD patients and 49 healthy volunteers were recruited.Skin temperature measurements were performed with a skin temperature assessment device at 10 points.Absolute difference between skin temperature of the same point on the left and right side is used as main outcome measure.RESULTS:On the first day of menstruation, when menstrual pain attacking in PD patients, a significant increase in skin temperature difference was detected at Taixi(KI 3) compared with the healthy group(P < 0.01).A significant reduction in skin temperature difference was detected at Taixi(KI 3) in the first day of menstruation compared with those values in the third day after menstruation(P < 0.01)in the healthy group.On the third day after menstruation, a significant reduction in skin temperature difference was found at Zhongdu(LR 6) in PD group compared with the healthy group(P < 0.05).No significant differences of skin temperature were detected at other points(P > 0.05).CONCLUSION:The skin temperature difference at menstruation-relevant points in PD patients did not all change significantly more than those in women without PD.Significant difference was only found in Taixi(KI 3), the Yuan-source point of Kidney meridian.She Yanfen Ma Liangxiao Zhu Jiang Qi Conghui Wang Yanxia Tang Ling Li Chunhua Yuan Hongwen Liu Yuqi Song Jiashan 2017Journal of Traditional Chinese Medicine2017,37,2:8
17Seismotectonics of the 26 November 2005 Jiujiang-Ruichang,Jiangxi,Ms 5.7 Earthquake显示文摘The 26 November 2005 Jiujiang-Ruichang,Jiangxi,Ms 5.7 earthquake occurred in a seismotectonic setting of moderate earthquake.The northwest-trending Xiangfan-Guangji fault(XFG) does not enter into the epicenter vicinity,but the northeast-trending Ruichang-Wuning fault(RWF) as a regional fault extends to the epicenter nearby,appearing as the Ruichang basin and its marginal faults.Tilting of the Ruichang Basin(RCB) in the Quaternary was controlled by the RCB southeast-marginal,buried fault(RSMBF).Shallow geophysical survey reveals that the RSMBF caused an offset of the reflection layers.Drill hole columnar section demonstrates that there are about 10-12 m displacement in the lower section of the middle-Pleistocene Series along the RSMBF,but no disruption is found in the upper section of the middle-Pleistocene Series.The RSMBF not only has activity in the Quaternary,but also coincides with the nodal plane I from the focal mechanism of the Jiujiang-Ruichang Ms 5.7 earthquake.This evidence,including aftershock distribution and isoseismic lines,strongly suggests that the RSMBF might be the seismogenic tectonics.The RWF is discontinuous at the surface,and consists of three en echelon Quaternary basins,which are the Ruichang,Fanzhen and Wuning basins.Three moderate earthquakes,the Fanzhen ML 4.9 earthquake,the Yejiapu ML 4.1 earthquake and the Jiujiang-Ruichang Ms 5.7 earthquake,have happened in the basins since 1995.The seismogenic tectonics of the Jiujiang-Ruichang Ms 5.7 earthquake is not isolated,but may be controlled by the RWF at depth,the slip of which causes the accumulation of energy for earthquake occurrence.HAN Zhujun LUFushui JI Fengju ZENG Xinfu An Yanfen 2012Acta Geologica Sinica(English Edition)2012,86,2:6
18Determination of PCDD/Fs and dioxin-like PCBs in food and feed using gas chromatography-triple quadrupole mass spectrometry显示文摘A method was developed on a gas chromatograph coupled to a triple quadrupole mass spectrometer(GC-MS/MS) for trace level determination of polychlorinated dibenzo-p-dioxins/dibenzofurans(PCDD/Fs) and dioxin-like polychlorinated biphenyls(DL-PCBs) in food and feed. The results demonstrated good sensitivity and repeatability for PCDD/Fs and DL-PCBs at an extremely low level(10 pg mL^(-1) for 2,3,7,8-TCDD/F), as well as wide linear response of over 3 or 4 orders of magnitude in concentration ranges; 0.5–200 ng mL^(-1) for PeCDD/F and 0.2–2000 ng mL^(-1) for DL-PCBs. The method detection limits for PCDD/Fs and DL-PCBs were in the range from 0.018–0.17 pg g^(-1) to 0.13–0.36 pg g^(-1), respectively. The performance of the GC-MS/MS for food and feed sample analysis showed high precision and accuracy compared to the high resolution gas chromatograph/high resolution mass spectrometer. 19PKM2 promotes reductive glutamine metabolism显示文摘Objective:Pyruvate kinases M(PKM),including the PKM1 and PKM2 isoforms,are critical factors in glucose metabolism.PKM2promotes aerobic glycolysis,a phenomenon known as'the Warburg effect'.The purpose of this study was to identify the roles of PKM2 in regulating cellular metabolism.Methods:The CRISPR/Cas9 system was used to generate the PKM-knockout cell model to evaluate the role of PKM in cellular metabolism.Lactate levels were measured by the Vitros LAC slide method on an autoanalyzer and glucose levels were measured by the autoanalyzer AU5800.The metabolism of ^(13)C_6-glucose or ^(13)C_5-glutamine was evaluated by liquid chromatography/mass spectrometry analyses.The effects of PKM on tumor growth were detected in vivo in a tumor-bearing mouse model.Results:We found that both PKM1 and PKM2 enabled aerobic glycolysis,but PKM2 converted glucose to lactate much more efficiently than PKM1.As a result,PKM2 reduced glucose levels reserved for intracellular utilization,particularly for the production of citrate,and thus increased theα-ketoglutarate/citrate ratio to promote the generation of glutamine-derived acetylcoenzyme A through the reductive pathway.Furthermore,reductive glutamine metabolism facilitated cell proliferation under hypoxia conditions,which supports in vivo tumor growth.In addition,PKM-deletion induced a reverse Warburg effect in tumorassociated stromal cells.Conclusions:PKM2 plays a critical role in promoting reductive glutamine metabolism and maintaining proton homeostasis.This study is helpful to increase the understanding of the physiological role of PKM2 in cancer cells.Miao Liu Yuanyuan Wang Yuxia Ruan Changsen Bai Li Qiu Yanfen Cui Guoguang Ying Binghui Li 2018Cancer Biology & Medicine2018,15,4:6
20Immobilization of acetylcholinesterase on one-dimensional gold nanoparticles for detection of organophosphorous insecticides显示文摘This paper reports a simple method for immobilization of acetylcholinesterase (AChE) on one-dimensional (1D) gold (Au) nanoparticles for detection of organophosphorous (OP) insecticides. 1D Au nanoparticles were prepared by electrodeposition in the pores of an alumina template which was subsequently removed by 2.0 M NaOH solution. They were characterized by XRD and FESEM. The immobilized AChE retained its biological activity and catalyzed the hydrolysis of acetylthiocholine to form thiocholine, which was subsequently oxidized to produce detectable signals. Based on the inhibition toward the enzymatic activity of AChE by OP insecticides, sensitive detection of methamidophos (an OP insecticide) was performed. Under optimal conditions, the sensors could be used for the determination of methamidophos ranging from 0.004 to 24 μg/mL with the detection limit of 0.001 μg/mL. The developed OP insecticide biosensors exhibited satisfactory stability and reproducibility.
