2Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 3Experimental study of rotating gliding arc discharge plasma-assisted combustion in an aero-engine combustion chamber显示文摘The combustion chamber is the core component of an aero-engine, and affects its reliability and security operation, even the performance of the aircraft. In this work, a Plasma-Assisted Combustion(PAC) test platform was developed to validate the feasibility of using PAC actuators to enhance annular combustor performance. Two plans of PAC(rotating gliding arc discharge plasma) were designed, Assisted Combustion from Primary Holes(ACPH) and Assisted Combustion from Dilution Holes(ACDH). Comparative experiments and analysis between conventional combustion and PAC were conducted to study the effects of ACPH and ACDH on the performances including average outlet temperature, combustion efficiency, pattern factor under four different excessive air coefficients(0.8, 1, 2, and 4), and lean blowout performance at different inlet airflow velocities. Experimental results show that the combustion efficiency is improved after PAC compared with that in normal conditions, and the combustion efficiency of ACPH increases2.45%, 1.49%, 1.04%, and 0.47%, while it increases 2.75%, 1.67%, 1.36%, and 0.36% under ACDH conditions. The uniformity of the outlet temperature field and the lean blowout performance are improved after PAC. Especially for ACPH, the widening of the lean blowout limit is8.3%, 12.4%, 12.8%, and 25% respectively when the inlet velocity ranges from 60 m/s to120 m/s. 4Genomic insights into the fast growth of paulownias and the formation of Paulownia witches' broom显示文摘Paulownias are among the fastest growing trees in the world,but they often suffer tremendous loss of wood production due to infection by Paulownia witches'broom(PaWB)phytoplasmas.In this study,we have sequenced and assembled a high-quality nuclear genome of Paulownia fortunei,a commonly cultivated paulownia species.The assembled genome of P.fortunei is 511.6 Mb in size,with 93.2%of its sequences anchored to 20 pseudo-chromosomes,and it contains 31985 protein-coding genes.Phylogenomic analyses show that the family Paulowniaceae is sister to a clade composed of Phrymaceae and Orobanchaceae.Higher photosynthetic efficiency is achieved by integrating C3 photosynthesis and the crassulacean acid metabolism pathway,which may contribute to the extremely fast growth habit of paulownia trees.Comparative transcriptome analyses reveal modules related to cambial growth and development,photosynthesis,and defense responses.Additional genome sequencing of PaWB phytoplasma,combined with functional analyses,indicates that the effector PaWB-SAP54 interacts directly with Paulownia PfSPLa,which in turn causes the degradation of PfSPLa by the ubiquitin-mediated pathway and leads to the formation of witches'broom.Taken together,these results provide significant insights into the biology of paulownias and the regulatory mechanism for the formation of PaWB.Yabing Cao Guiling Sun Xiaoqiao Zhai Pingluo Xu Liming Ma Minjie Deng Zhenli Zhao Haibo Yang Yanpeng Dong Zhonghai Shang Yujie Lv Lijun Yan Haifang Liu Xibing Cao Bingbing Li Zhe Wang Xiaogai Zhao Haiyan Yu Fan Wang Wen Ma Jinling Huang Guoqiang Fan 2021Molecular Plant2021,14,10:7
5How is mRNA expression predictive for protein expression? 7Size-tunable synthesis of gold nanorods using pyrogallol as a reducing agent显示文摘A novel and facile seed-mediated method for the preparation of monodispersed gold nanorods(GNRs) is presented by introducing pyrogallol as a reductant. Fast Fourier transformation of high-resolution transmission electron microscopy reveals that the synthesized GNRs are single crystalline. The longitudinal surface plasmon resonance of GNRs can be finely tuned by varying silver ion concentrations or seed amounts. Also, both thick(diameter >30 nm) and thin(diameter <10 nm) GNRs with exceptional monodispersity can be well prepared by this method. 8Effects of Dielectric Barrier Discharge Plasma on the Combustion Performances of Reverse-Flow Combustor in an Aero-Engine显示文摘In order to apply plasma assisted combustion(PAC) into a reverse-flow aero-engine and verify the improvement of combustion performance, a feasible approach was proposed in this work. In this approach, based on the structure characteristics of the reverse-flow combustor, a parallel plate double dielectric barrier discharge(DBD) PAC actuator was designed to generate plasma. It was installed at the front of combustor. When the actuator is driven, the original air flow is not disturbed for the device’s structure and installation. Using aviation kerosene as fuel, the effects of plasma on ignition boundary and outlet temperature of the combustor were experimentally investigated at atmosphere pressures. Through the dual high voltage differential power supply, the large gap, large area and uniform plasma discharge was achieved. The results of PAC actuator discharge indicate that inlet air temperature has a small increase of 4–9 K. After PAC is applied, the combustion performances of reverse-flow combustor in an aero-engine are remarkably improved. Experimental results indicate that ignition boundary is widened by 3.7%–12.5% because of the impact of plasma. Outlet highest temperature of combustor is raised by 19–75 K;outlet temperature distribution coefficient is reduced by 11.1%–26.6%. 10Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 11Disruption of crosstalk between LX-2 and liver cancer stem-like cells from MHCC97H cells by DFOG via inhibiting FOXM1显示文摘Hepatic stellate cell(HSC)line LX-2 is activated by liver cancer stem-like cells(LCSLCs)and produces various cytokines that make up most of the hepatocellular carcinoma(HCC)microenvironment.The new genistein derivative,7-difluoromethoxyl-5,4-dirboctylgenistein(DFOG),shows anticancer effects in multiple malignancies by controlling forkhead box M1(FOXM1).In this study,we aimed to assess whether DFOG disrupts the crosstalk between human HSC LX-2 cells and LCSLCs.Distinct gen erations of MHCC97H-derived spheres were obtained with the sec ond generation considered as LCSLCs which displayed enhanced self-renewal ability and elevated expression levels of CD133,CD44,and EpCAM proteins,as well as tumorigenicity,as revealed by colony formation assay in vitro and tumorigenicity assay in vivo.LX-2 and MHCC97H cells were co-cultured with/without DFOG(1,5,and 10 pM,respectively)using the transwell system.FOXM1 overexpression and/or knockdown were employed for mechanistic investigations.Our results suggested that Co-CM promoted LX-2 cell transformation into liver cancer-associated HSCs.Meanwhile,FOXM1 was up-regulated and the level of hepatocyte growth factor(HGF)was in creased in LX-2 cells and in the super nata nt after Co-CM stimulati on.Sphere and colony formation abilities in MHCC97H cells,and protein levels of CD133r CD44,and EpCAM,were also markedly elevated.DFOG dose-dependently inhibited the above effects,similar to FOXM1 knockdown in LX-2 cells.FOXM1 overexpression reversed the inhibitory effects of DFOG or FOXM1 knockdown or both on LX-2 cell activation and LCSLC feature induction in MHCC97H cells by LCSLC/LX-2 co-culture.This study demonstrated that DFOG disrupts the crosstalk between HSCs and LCSLCs to suppress LCSLC features via dowrrregulating FOXM1 expression and reducing HGF secretion in HSCs.A Chen Chang Xu Yimin Luo Lihua Liu Kun Song Guangqi Deng Mengjie Yang Jianguo Cao Liming Yuan Xiang Li 2019Acta Biochimica et Biophysica Sinica2019,51,12:2

13A new modified canine model of portal vein thrombosis显示文摘Portal vein thrombosis (PVT) is a common complication of liver cirrhosis. Gao et al. [1] reported that the prevalence of PVT in patients with cirrhosis is 8.82%. With the rapid developments in image-based diagnostic technology, the diagnosis rate of PVT is gradually increasing [2]. A follow-up study of 1243 patients with cirrhosis revealed that the cumulative incidences of PVT at 1, 3, and 5 years are 4.6%, 8.2%, and 10.7%, respectively [3]. PVT is a life-threatening factor for patients with cirrhosis, leading to portal hypertension and aggravating gastrointestinal hemorrhage [4]. The prevention of PVT has become a focus of clinical concern nowadays. It is well known that the establishment of animal models is the basis of scientific research. However, it is currently difficult to establish PVT models in large animals. Thereby, the establishment of a stable PVT animal model to study PVT is urgent and significant. We successfully established a new canine model of PVT involving the injection of an autologous thrombus through an indwelling catheter in the portal vein. 14Design and Experiment of a Deformable Bird-inspired UAV Perching Mechanism显示文摘Energy consumption and acoustic noise can be significantly reduced through perching in the sustained flights of small Unmanned Aerial Vehicles(UAVs).However,the existing flying perching robots lack good adaptability or loading capacity in unstructured environments.Aiming at solving these problems,a deformable UAV perching mechanism with strong adaptability and high loading capacity,which is inspired by the structure and movements of birds'feet,is presented in this paper.Three elastic toes,an inverted crank slider mechanism used to realize the opening and closing movements,and a gear mechanism used to deform between two configurations are included in this mechanism.With experiments on its performance towards different objects,Results show that it can perch on various objects reliably,and its payload is more than 15 times its weight.By integrating it with a quadcopter,it can perch on different types of targets in outdoor environments,such as tree branches,cables,eaves,and spherical lamps.In addition,the energy consumption of the UAV perching system when perching on objects can be reduced to 0.015 times that of hovering.Long Bai Hao Wang Xiaohong Chen Jia Zheng Liming Xin Yupeng Deng Yuanxi Sun 2021Journal of Bionic Engineering2021,18,6:1

16Bronchial Fistula: Rare Complication of Treatment with Anlotinib显示文摘Background and objective Anlotinib is a newly developed small molecule multiple receptor tyrosine kinase(RTK) inhibitor that was approved for the treatment of patients with lung cancer in China. We aim to report 3 cases of rare complication of anlotinib-bronchial fistula(BF) during the treatment of lung cancer patients and summarize the possible causes.Methods We collected three patients who developed BF due to anlotinib treatment, and conducted a search of Medline and Pub Med for medical literature published between 2018 and 2020 using the following search terms: 'anlotinib,' 'lung cancer,' and 'fistula.'Results Our literature search produced two case reports(three patients) which, in addition to our three patients. We collated the patients’ clinical characteristics including demographic information, cancer type, imaging features, treatment received, risk factors for anlotinib related BF, and treatment-related outcomes. The six patients shared some common characteristics: advanced age, male, concurrent infection symptoms, diabetes mellitus(DM), advanced squamous cell and small cell lung cancers, centrally located tumors, tumor measuring ≥5 cm in longest diameter, and newly formed tumor cavitation after multi-line treatment especially after receiving radiotherapy. Fistula types included broncho-pericardial fistula, broncho-pleural fistula, and esophagotracheobronchial fistula. Six patients all died within 6 months.Conclusion Although anlotinib is relatively safe, it is still necessary to pay attention to the occurrence of BF, a rare treatment side effect that threatens the quality of life and overall survival of patients. 17Characterizing the tumor microenvironment at the single-cell level reveals a novel immune evasion mechanism in osteosarcoma显示文摘The immune microenvironment extensively participates in tumorigenesis as well as progression in osteosarcoma(OS).However,the landscape and dynamics of immune cells in OS are poorly characterized.By analyzing single-cell RNA sequencing(sc RNA-seq)data,which characterize the transcription state at single-cell resolution,we produced an atlas of the immune microenvironment in OS.The results suggested that a cluster of regulatory dendritic cells(DCs)might shape the immunosuppressive microenvironment in OS by recruiting regulatory T cells.We also found that major histocompatibility complex class I(MHC-I)molecules were downregulated in cancer cells.The findings indicated a reduction in tumor immunogenicity in OS,which can be a potential mechanism of tumor immune escape.Of note,CD24 was identified as a novel“don’t eat me”signal that contributed to the immune evasion of OS cells.Altogether,our findings provide insights into the immune landscape of OS,suggesting that myeloid-targeted immunotherapy could be a promising approach to treat OS.Weijian Liu Hongzhi Hu Zengwu Shao Xiao Lv Zhicai Zhang Xiangtian Deng Qingcheng Song Yong Han Tao Guo Liming Xiong Baichuan Wang Yingze Zhang 2023Bone Research2023,11,1:1
18Traumatic brain injury stimulates sympathetic tone-mediated bone marrow myelopoiesis to favor fracture healing显示文摘Traumatic brain injury(TBI)accelerates fracture healing,but the underlying mechanism remains largely unknown.Accumulating evidence indicates that the central nervous system(CNS)plays a pivotal role in regulating immune system and skeletal homeostasis.However,the impact of CNS injury on hematopoiesis commitment was overlooked.Here,we found that the dramatically elevated sympathetic tone accompanied with TBI-accelerated fracture healing;chemical sympathectomy blocks TBIinduced fracture healing.TBI-induced hypersensitivity of adrenergic signaling promotes the proliferation of bone marrow hematopoietic stem cells(HSCs)and swiftly skews HSCs toward anti-inflammation myeloid cells within 14 days,which favor fracture healing.Knockout ofβ3-orβ2-adrenergic receptor(AR)eliminate TBI-mediated anti-inflammation macrophage expansion and TBIaccelerated fracture healing.RNA sequencing of bone marrow cells revealed that Adrb2 and Adrb3 maintain proliferation and commitment of immune cells.Importantly,flow cytometry confirmed that deletion ofβ2-AR inhibits M2 polarization of macrophages at 7th day and 14th day;and TBI-induced HSCs proliferation was impaired inβ3-AR knockout mice.Moreover,β3-andβ2-AR agonists synergistically promote infiltration of M2 macrophages in callus and accelerate bone healing process.Thus,we conclude that TBI accelerates bone formation during early stage of fracture healing process by shaping the anti-inflammation environment in the bone marrow.These results implicate that the adrenergic signals could serve as potential targets for fracture management.Weijian Liu Wei Chen Mao Xie Chao Chen Zengwu Shao Yiran Zhang Haiyue Zhao Qingcheng Song Hongzhi Hu Xin Xing Xianyi Cai Xiangtian Deng Xinyan Li Peng Wang Guohui Liu Liming Xiong Xiao Lv Yingze Zhang 2023Signal Transduction and Targeted Therapy2023,8,8:1
19Genetic susceptibility loci of lung cancer are associated with malignant risk of pulmonary nodules and improve malignancy diagnosis based on CEA levels显示文摘Objective:The heightened prevalence of pulmonary nodules(PN)has escalated its significance as a public health concern.While the precise identification of high-risk PN carriers for malignancy remains an ongoing challenge,genetic variants hold potentials as determinants of disease susceptibility that can aid in diagnosis.Yet,current understanding of the genetic loci associated with malignant PN(MPN)risk is limited.Methods:A frequency-matched case-control study was performed,comprising 247 MPN cases and 412 benign NP(BNP)controls.We genotyped 11 established susceptibility loci for lung cancer in a Chinese cohort.Loci associated with MPN risk were utilized to compute a polygenic risk score(PRS).This PRS was subsequently incorporated into the diagnostic evaluation of MPNs,with emphasis on serum tumor biomarkers.Results:Loci rs10429489G>A,rs17038564A>G,and rs12265047A>G were identified as being associated with an increased risk of MPNs.The PRS,formulated from the cumulative risk effects of these loci,correlated with the malignant risk of PNs in a dose-dependent fashion.A high PRS was found to amplify the MPN risk by 156%in comparison to a low PRS[odds ratio(OR)=2.56,95%confidence interval(95%CI),1.40−4.67].Notably,the PRS was observed to enhance the diagnostic accuracy of serum carcinoembryonic antigen(CEA)in distinguishing MPNs from BPNs,with diagnostic values rising from 0.716 to 0.861 across low-to high-PRS categories.Further bioinformatics investigations pinpointed rs10429489G>A as an expression quantitative trait locus.Conclusions:Loci rs10429489G>A,rs17038564A>G,and rs12265047A>G contribute to MPN risk and augment the diagnostic precision for MPNs based on serum CEA concentrations.Zhi Li Liming Lu Yibin Deng Amei Zhuo Fengling Hu Wanwen Sun Guitian Huang Linyuan Liu Boqi Rao Jiachun Lu Lei Yang 2023Chinese Journal of Cancer Research2023,35,5:0
20Dynamic changes of beta-amyloid protein deposition in hippocampus of female ovariectomized rats显示文摘BACKGROUND: To evaluate and summarize the effects of cerebral perfusion and vascular reserve on the treatment of SICAS. Recently, research on β-amyloid protein has focused on the regulatory effects of es- trogen or phytoestrogen on its deposition. However, there have been only a few reports on dynamic changes of β-amyloid protein deposition in hippocampus of ovariectomized rats. OBJECTIVE: To measureβ-amyloid protein deposition in the hippocampal formation of ovariectomized rats by using immunohistochemistry; to observe time-dependent dynamic changes. DESIGN: Randomized controlled animal study. SETTING: Third Xiangya Hospital of Central South University. MATERIALS: The experiment was carried out in the Central Laboratory of the Third Xiangya Hospital of Central South University from November 2005 to December 2006. Fifty healthy female Sprague Dawley (SD) rats, weighing (293 ± 10) g, were provided by the Animal Laboratory of Xiangya Medical College, Central South University. All rats had neither a childbearing history nor hepatic or renal disease, or skeletal deformity. β-amyloid protein immunohistochemical kit was provided by Wuhan Boster Company. The ex- periment was in accordance with animal ethics standards. METHODS: All rats were randomly divided into five groups, including normal control group (n = 10), sham operation group (n = 10), and ovariectomized group (n = 30). After anesthesia in the ovariectomized group, the bilateral ovaries were separated and resected. The same volume of fat was resected in the sham operation group. Rats from the normal control group, however, did not receive any surgical treatments. Rats in the normal control group and sham operation group were sacrificed by anesthesia 7 weeks after surgery. Every ten rats from the ovariectomized group was respectively sacrificed at 7, 15, and 30 weeks after surgery. Immunohistochemistry was used to detectβ-amyloid protein deposition in hippocampal sections. Cell counting and gray value measurements served to record the dynamic changes in β-amyloid protein deposi- tion. MAIN OUTCOME MEASURES: ① Morphological changes. ② Positive cell counts from β-amyloid protein stainings and gray value measurements. RESULTS: All 50 rats were involved in the final analysis. ① Morphological changes. β-amyloid-positive cells were detected in the hippocampus of all rats. Biebrich scarlet stained neurites with a swollen cytoplasm. A few β-amyloid-positive cells were observed in all groups 7 weeks after surgery, and plasma and neurites were slightly stained. By 15 weeks after surgery, a number of β-amyloid-positive cells were observed in the ovariectomized group, and plasma and neurites were also slightly stained. By 30 weeks after surgery, how- ever, many β-amyloid-positive cells were observed in the ovariectomized group. These cells were partially aggregated and darkly stained. ② Positive cell counts and gray value of β-amyloid protein in hippocam- pus. At 7 weeks after surgery, cell counts and gray value measurements were not significantly different in the ovariectomized group compared to the sham operation group and normal control group (P > 0.05). Cell counts and gray value measurements were higher in the ovariectomized group by 15 weeks compared to those by 7 weeks in the normal control group, sham operation group and ovariectomized group (P < 0.05). At 30 weeks after surgery, cell counts and gray value measurements were higher in the ovariectomized group compared to the normal control group. In addition, there were significant differences between sham opera- tion group and ovariectomized group at 7 and 15 weeks after operation (P < 0.05-0.01). Cell counts and gray value measurements increased in all groups over time.
