| 1 | The versatile functions of Sox9 in development,stem cells,and human diseases显示文摘The transcription factor Sox9 was first discovered in patients with campomelic dysplasia,a haploinsufficiency disorder with skeletal deformities caused by dysregulation of Sox9 expression during chondrogenesis.Since then,its role as a cell fate determiner during embryonic development has been well characterized;Sox9 expression differentiates cells derived from all three germ layers into a large variety of specialized tissues and organs.However,recent data has shown that ectoderm-and endoderm-derived tissues continue to express Sox9 in mature organs and stem cell pools,suggesting its role in cell maintenance and specification during adult life.The versatility of Sox9 may be explained by a combination of posttranscriptional modifications,binding partners,and the tissue type in which it is expressed.Considering its importance during both development and adult life,it follows that dysregulation of Sox9 has been implicated in various congenital and acquired diseases,including fibrosis and cancer.This review provides a summary of the various roles of Sox9 in cell fate specification,stem cell biology,and related human diseases.Ultimately,understanding the mechanisms that regulate Sox9 will be crucial for developing effective therapies to treat disease caused by stem cell dysregulation or even reverse organ damage. | Alice Jo Sahitya Denduluri Bosi Zhang Zhongliang Wang Liangjun Yin Zhengjian Yan Richard Kang Lewis L.Shi James Mok Michael J.Lee Rex C.Haydon | 2014 | Genes & Diseases2014,1,2: | 16 |
| 2 | Hydrogen sulfide suppresses transforming growth factor-β1-induced differentiation of human cardiac fibroblasts into myofibroblasts显示文摘In heart disease, transforming growth factor-β1(TGF-β1) converts fibroblasts into myofibroblasts, which synthesize and secrete fibrillar type I and III collagens. The purpose of the present study was to investigate how hydrogen sulfide(H2S) suppresses TGF-β1-induced differentiation of human cardiac fibroblasts to myofibroblasts. Human cardiac fibroblasts were serum-starved in fibroblast medium for 16 h before exposure to TGF-β1(10 ng m L-1) for 24 h with or without sodium hydrosulfide(Na HS, 100 μmol L-1, 30 min pretreatment) treatment. Na HS, an exogenous H2 S donor, potently inhibited the proliferation and migration of TGF-β1-induced human cardiac fibroblasts and regulated their cell cycle progression. Furthermore, Na HS treatment led to suppression of fibroblast differentiation into myofibroblasts, and reduced the levels of collagen, TGF-β1, and activated Smad3 in TGF-β1-induced human cardiac fibroblasts in vitro. We therefore conclude that H2 S suppresses TGF-β1-stimulated conversion of fibroblasts to myofibroblasts by inhibiting the TGF-β1/Smad3 signaling pathway, as well as by inhibiting the proliferation, migration, and cell cycle progression of human cardiac myofibroblasts. | 4 | Soil hydraulic conductivity as affected by vegetation restoration age on the Loess Plateau,China显示文摘The Loess Plateau of China has experienced extensive vegetation restoration in the past several decades,which leads to great changes in soil properties such as soil bulk,porosity,and organic matter with the vegetation restoration age.And these soil properties have great effect on the soil infiltration and soil hydraulic conductivity.However,the potential changes in soil hydraulic conductivity caused by vegetation restoration age have not been well understood.This study was conducted to investigate the changes in soil hydraulic conductivity under five grasslands with different vegetation restoration ages(3,10,18,28 and 37 years)compared to a slope farmland,and further to identify the factors responsible for these changes on the Loess Plateau of China.At each site,accumulative infiltration amount and soil hydraulic conductivity were determined using a disc permeameter with a water supply pressure of –20 mm.Soil properties were measured for analyzing their potential factors influencing soil hydraulic conductivity.The results showed that the soil bulk had no significant changes over the initial 20 years of restoration(P>0.05);the total porosity,capillary porosity and field capacity decreased significantly in the grass land with 28 and 37 restoration ages compared to the slope farmland;accumulative infiltration amount and soil hydraulic conductivity were significantly enhanced after 18 years of vegetation restoration.However,accumulative infiltration amount and soil hydraulic conductivity fluctuated over the initial 10 years of restoration.The increase in soil hydraulic conductivity with vegetation restoration was closely related to the changes in soil texture and structure.Soil sand and clay contents were the most influential factors on soil hydraulic conductivity,followed by bulk density,soil porosity,root density and crust thickness.The Pearson correlation coefficients indicated that the soil hydraulic conductivity was affected by multiply factors.These results are helpful to understand the changes in hydrological and erosion processes response to vegetation succession on the Loess Plateau. | REN Zongping ZHU Liangjun WANG Bing CHENG Shengdong | 2016 | Journal of Arid Land2016,8,4: | 9 |

The paper first builds a stochastic mean-reversion model for fuel oil prices and natural gas prices respectively, then estimates and tests the parameters using the fuel oil and natural gas prices data from the U.S. market. This paper also conducts a sensitivity analysis with important parameters. | 7 | Fenitized Wall Rock Geochemistry of the First Carbonatite Dyke at Bayan Obo, Inner Mongolia, China显示文摘The first carbonatite dyke at Bayan Obo is well exposed on the surface for a length and width of approximately 60 m and 1.1–1.5 m, respectively. Along its strike, the fenitized H1(Qs) and H2(Cs) quartzite is replaced by Na-amphiboles, aegirines, and alkali-feldspars, intermittently stretching as far away as 800 m in length. Based on petrographical characteristics, the dyke's fenitized wall rocks are divisible into different zones:(1) outer,(2) middle, and(3) inner. The outer zone is 5–17 m from the NW margin of the dyke. The middle zone is located at 3.5–5 m from the NW margin of the dyke. The inner contact zone is located between direct contact with the dyke and 3.5 m from the dyke. In the outer zone, upon visual examination, no evidence of outcrop fenitization was found and the major elemental rock composition is nearly identical to the unaltered H1 and H2 lithologies. In the thin sections, however, small amounts of Na-amphibole and phlogopite are present. Despite relatively poor development throughout the 5 m of fenitization, the wall rocks have retained at least a small geochemical signature comparable to the original sedimentary protolith. The fenites occurring in the inner zone exhibit distinct variations, not only for the sharp contact at the outcrop scale, but also for variations in major, rare earth elements(REE), and trace elements and Sm-Nd isotope composition. The wall rocks within 3.5 m have undergone strong fenitization, inheriting the geochemical signature derived from the carbonatite dyke. Fenitization in the middle zone was not as strong, at least compared to the inner zone, but was stronger than the outer zone. Compared to some trace elements and REEs, the major elements are relatively immobile during fenitization. The Sm-Nd isotope data for the carbonatite dyke and the adjacent fenitized wall rocks, where the Sm and Nd originate solely from the dyke, plots as a six-point isochron with an age of 1308±56 Ma. This age is identical to that of ore-bearing dolomite carbonatite and the related ore-forming events, indicating that there may be a petrogenetic link between the two. | 11 | Efficacy and safety of chemotherapy combined with bevacizumab in Chinese patients with metastatic colorectal cancer:A prospective,multicenter,observational,non-interventional phaseⅣtrial显示文摘Objective:Bevacizumab has an important and evolving role in improving outcomes in patients with metastatic colorectal cancer(mCRC)worldwide and was approved in China in 2010.However,there are limited real-world data on the efficacy and safety of chemotherapy regimens combined with bevacizumab in Chinese patients with mCRC.This observational,phase IV trial study aimed to obtain more experience on the efficacy and safety of bevacizumab combined with chemotherapy in Chinese mCRC patients.Methods:Between September 2013 and November 2016,patients with histologically confirmed mCRC were enrolled in a prospective,multicenter,observational,non-interventional phase IV trial at 26 centers across China.Eligible patients received different chemotherapeutic regimens combined with bevacizumab.The efficacy and safety data in the intention-to-treat study population were analyzed.Results:A total of 611 patients were included in the efficacy analysis.The median overall survival and median progression-free survival was 18.00 and 10.05 months,respectively.The objective response rate was 21.00%and disease control rate was 89.40%.In subgroup analyses,the survival differences were observed according to metastatic status,duration of treatment and elevation in blood pressure.A total of 613 patients were evaluable for safety assessments.And 569(92.82%)patients reported at least one adverse event(AE),and 151(24.63%)experienced grade 3 or higher AEs.The incidence of bevacizumab-associated AEs of special interest was reported in 31(5.06%)patients with hypertension(n=12),abscesses and fistulae(n=7),bleeding(n=6),proteinuria(n=3),gastrointestinal perforation(n=2)and venous thrombotic events(n=1).Conclusions:This observational phase IV trial broadens our experience and knowledge of bevacizumab in the Chinese population and provides a good indication of its overall efficacy and safety.Bevacizumab in combination with chemotherapy offers clinical benefits to Chinese patients with mCRC and has an acceptable and manageable safety profile. | Fenghua Wang Guanghai Dai Yanhong Deng Yong Tang Wei Wang Zuoxing Niu Feng Bi Liangjun Zhu Zengqing Guo Jin Yan Bing Hu Min Tao Shujun Yang Suzhan Zhang Lu Wen Ruihua Xu | 2021 | Chinese Journal of Cancer Research2021,33,4: | 2 |

| 15 | Inhibition of aberrant Hif1αactivation delays intervertebral disc degeneration in adult mice显示文摘The intervertebral disc(IVD) is the largest avascular tissue. Hypoxia-inducible factors(HIFs) play essential roles in regulating cellular adaptation in the IVD under physiological conditions. Disc degeneration disease(DDD) is one of the leading causes of disability, and current therapies are ineffective. This study sought to explore the role of HIFs in DDD pathogenesis in mice. The findings of this study showed that among HIF family members, Hif1α was significantly upregulated in cartilaginous endplate(EP) and annulus fibrosus(AF) tissues from human DDD patients and two mouse models of DDD compared with controls. Conditional deletion of the E3 ubiquitin ligase Vhl in EP and AF tissues of adult mice resulted in upregulated Hif1α expression and age-dependent IVD degeneration. Aberrant Hif1α activation enhanced glycolytic metabolism and suppressed mitochondrial function. On the other hand, genetic ablation of the Hif1α gene delayed DDD pathogenesis in Vhl-deficient mice. Administration of 2-methoxyestradiol(2ME2), a selective Hif1α inhibitor, attenuated experimental IVD degeneration in mice. | 20 | Synthesis of AgInS_2 QDs in droplet microreactors:Online fluorescence regulating through temperature control显示文摘For the synthesis of AgInS_2 quantum dots(QDs), a suitable temperature is extremely important for control of the size, shape and fluorescence properties of QDs. Most of synthesis methods for AgInS_2 QDs are based on batch reactors, which bring uneven distribution of temperature, affecting their fluorescence properties and size uniformity. Here we designed a droplet microreactor with a temperature-controllable region, and successfully synthesized water-soluble AgInS_2 QDs. By accurately controlling temperature,we also studied how the reaction temperature affected the fluorescence properties of AgInS_2 QDs. The results showed that with the increasing of reaction temperature, the QDs size increased and the fluorescence peak constantly red-shifted along with enhanced fluorescence intensity.
