| 3 | Mutation analysis of APC gene in gastric cancer with microsatellite instability显示文摘AIM: To evaluate the role of APC mutation in gastric carcinogenesis and to correlate APC mutation with microsatellite instability (MSI) in gastric carcinomas. METHODS: APC mutation was measured with multiplex PCR, denaturing gradient gel electrophoresis and DNA sequencing; and MSI was analyzed by PCR-based methods.RESULTS: Sixty-eight cases of sporadic gastric carcinoma were studied for APC mutation at exon 15 and MSI. APC mutaions were detected in 15(22.1%) gastric cancers. | 8 | Contrast-enhanced ultrasound:Improving the preoperative staging of hepatocellular carcinoma and guiding individual treatment显示文摘AIM:To investigate the clinical role of contrast-enhanced ultrasound(CEUS) combined with contrast-enhanced computed tomography(CE-CT) or magnetic resonance imaging to improve the preoperative staging of hepatocellular carcinoma(HCC) and guide surgical decisionmaking.METHODS:Sixty-nine patients who underwent liver resection for HCC in our center were enrolled prospectively in the study.CEUS and CE-CT/MRI were performed before surgery.Intraoperative ultrasound(IOUS)wascarried out after liver mobilization.Lesions depicted by each imaging modality were counted and mapped.To investigate the impact of tumor size on the study,we divided the patients into two groups,the'Smaller group'(S-group,≤5 cm in diameter)and the'Largergroup'(L-group,>5 cm in diameter).The sensitivity,specificity,accuracy,positive predictive value,and negative predictive value of CE-CT/MRI,CEUS,IOUS,CEUS+CE-CT/MRI and the tumor node metastasis staging of tumors were calculated and compared.Changes in the surgical strategy as a result of CEUS and IOUS were analyzed.RESULTS:One hundred and twenty-seven nodules,comprising 94 HCCs confirmed by histopathology and33 benign lesions confirmed by histopathology and follow-up,were identified in 69 patients.The overall diagnostic sensitivity rates of CE-CT/MRI,CEUS,IOUS and CEUS+CE-CT/MRI were 78.7%,89.4%,89.4%and 89.4%,respectively.There was a significant difference between CEUS+CE-CT/MRI and CE-CT/MRI(P=0.046).Combining CEUS with CT or MRI increased,the diagnostic specificity compared with CT/MRI,CEUS and IOUS,and this difference was statistically significant(100%,72.7%,97.0%,and 69.7%,P=0.004,P=0.002,P=0.002,respectively).The diagnostic accuracy was significantly higher for CEUS+CT/MRI compared with CT/MRI(92.1%vs 77.2%,P=0.001).The TNM staging of tumors based on CEUS+CE-CT/MRI approximated to the final pathological TNM staging(P=0.977).There was a significant difference in the accuracy of TNM staging when comparing CEUS+CECT/MRI with CE-CT/MRI(P=0.002).Before surgery,strategies were changed in 15.9%(11/69)of patients as a result of CEUS.Finally,only 5.7%(4/69)of surgical strategies were changed because of IOUS findings.In the S-group,CEUS revealed 12 false positive lesions,including seven false positive lesions that were diagnosedby preoperative imaging examinations and five by IOUS.In contrast,in the L-group,IUOS revealed eight new malignant lesions;six of these lesions were true HCCs that were also identified by preoperative CEUS.CONCLUSION:CEUS combined with CT or MRI improves the accuracy of preoperative staging for hepatocellular carcinoma and may help to guide individualized treatment for patients with HCC.CEUS may better identify non-malignant lesions in patients with small tumors and discover new malignant lesions in patients with large tumors. | Xiao-Yun Zhang Yan Luo Tian-Fu Wen Li Jiang Chuan Li Xiao-Fei Zhong Jing-Yi Zhang Wen-Wu Ling Lu-Nan Yan Yong Zeng Hong Wu | 2014 | World Journal of Gastroenterology2014,20,35: | 16 |

Identifying animal species susceptible to infection with the SARS-CoV-2/HCoV-19 pathogen is essential for controlling the outbreak and for testing valid prophylactics or therapeutics based on animal model studies. Here,different aged Chinese tree shrews(adult group, 1 year old;old group, 5–6 years old), which are close relatives to primates, were infected with SARS-CoV-2. X-ray, viral shedding, laboratory, and histological analyses were performed on different days postinoculation(dpi). Results showed that Chinese tree shrews could be infected by SARS-CoV-2. Lung infiltrates were visible in X-ray radiographs in most infected animals. Viral RNA was consistently detected in lung tissues from infected animals at 3,5, and 7 dpi, along with alterations in related parameters from routine blood tests and serum biochemistry, including increased levels of aspartate aminotransferase(AST) and blood urea nitrogen(BUN). Histological analysis of lung tissues from animals at 3 dpi(adult group) and 7 dpi(old group) showed thickened alveolar septa and interstitial hemorrhage. Several differences were found between the two different aged groups in regard to viral shedding peak. | 16 | The emergence, development and regional differences of mixed farming of rice and millet in the upper and middle Huai River Valley, China显示文摘Mixed farming of rice and millet is one of the basic agricultural modes in the upper and middle Huai River Valley(HRV). According to the latest data, this agricultural mode appeared during the middle and late Peiligang Culture(7.8–7.0 ka BP) in the upper HRV, and then became a common subsistence economy in the end of the Neolithic(5.0–4.0 ka BP) in both the upper and middle HRV. However, it is still not clear how this mixed farming developed in the upper HRV after its occurrence, nor are the regional differences in the development of mixed farming between the upper and middle HRV during the Neolithic completely understood. In this paper, flotation and starch analyses were conducted on samples from eight archaeological sites in the upper and middle HRV. The results indicate that the mixed farming of rice and millet first appeared in the later phase of the middle Neolithic in the regions of the Peiligang Culture, then developed quite rapidly in the late Neolithic(6.8–5.0 ka BP), finally becoming the main subsistence economy at the end of the Neolithic in the upper HRV. However, there are obvious differences in the emergence and development of agriculture between the middle and upper HRV. Rice farming was the only agricultural system during the middle Neolithic, lasting until the end of the Neolithic, when mixed farming appeared in the middle HRV. Furthermore, although mixed farming appeared in both the upper and middle HRV during the end of the Neolithic, the roles of rice, foxtail millet and broomcorn millet in the subsistence economy were not the same. | 17 | Clinical Value of Whole-body Magnetic Resonance Diffusion Weighted Imaging on Detection of Malignant Metastases显示文摘Objective To evaluate the value of whole-body diffusion weighted imaging(WB-DWI) on detection of malignant metastasis.Methods Forty-six patients with malignant tumors underwent WB-DWI examinations between April 2007 and August 2007 in our hospital.Before WB-DWI examination,the primary cancers of all the patients were confirmed by pathology,and the TNM-stage was assessed with conventional magnetic resonance imaging(MRI) or computed tomography(CT).WB-DWI was performed using short TI inversion recovery echo-planar imaging(STIR-EPI) sequence.Abnormal high signal intensities on WB-DWI were considered as metastases.The results of WB-DWI were compared with other imaging modalities.For the assessment of the diagnostic capability of WB-DWI,WB-DWI were compared with CT for demonstrating mediastinal lymph node metastases and lung metastases,and with conventional MRI for demonstrating metastases in other locations.Results WB-DWI demonstrated 143 focuses,14 of which were diagnosed to be benign lesions in routine imaging.The number of bone metastases depicted on WB-DWI and routine imaging was 85 and 86;lymph node metastases was 17 and 18;liver metastases was 14 and 14;lung metastases was 4 and 8;and brain metastases was 6 and 8,respectively.WB-DWI failed to detect 12 metastatic lesions including 3 osteoplastic bone metastases,4 lung metastases,3 mediastinal lymph node metastases,and 2 brain metastases.Four metastatic lesions including 2 deltopectoral lymph nodes and 2 rib metastases were detected with WB-DWI alone,all of which evolved greatly during clinical follow-up for more than 6 months.WB-DWI had higher detection rates for metastatic lesions in liver,bone,and lymph nodes than those in lung and brain(χ2=30,P<0.001).Conclusions WB-DWI could detect most of metastatic lesions that were diagnosed with conventional MRI and CT.The limitations of WB-DWI might be had high false-positive rate and low efficiency in detecting mediastinal lymph node,brain,and lung metastases. | Cheng Li Zhen-sheng Liu Xian-mao Du Ling He Jian Chen Wei Wang Fei Sun Fang Du Zhi-gang Luo Zhen-long Xue Yi Zhao Chang-wu Zhou | 2009 | Chinese Medical Sciences Journal2009,24,2: | 11 |
