| 1 | Verification and fine-mapping of QTLs conferring days to flowering in soybean using residual heterozygous lines显示文摘The results of QTL mapping based on a primary mapping population should be further verified and refined for its real utilization in marker-assisted selection or map-based cloning.The primary mapping population contains 114 BC1F1 plants of the backcross between Essex (maturity group V,MG V) as the donor parent and ZDD2315 (MG II) as the recurrent parent.In this study,a genetic linkage map with 250 SSR markers spanning a total length of 2963.5 cM on 25 linkage groups (LG) was constructed using software MAPMAKER3.0.Six kinds of genetic statistical models of 4 softwares,i.e.WinQTL Cartographer Version 2.5,IciMapping Version 2.0,MapQTL Version 5.0 and QTLnetwork Version 2.0,were used to map QTLs conferring days to flowering of the BC1F3 lines.Nine QTLs were mapped on 6 different linkage groups (LG).Of those,6 QTLs were detected by at least two different genetic statistical models,while the other three were detected by only one procedure.Among the three QTLs,Flwdt7 was mapped between Sat_213 and Satt643 on LG C2 with only 11.0% contribution rate.For confirmation of Flwdt7,5 RHL populations were developed through selfing eight BC1F5 plants heterozygous at seven markers around the locus.The RHL populations with the same segregating loci were bulked and used to construct a secondary linkage map of the specific segment using software JoinMap 3.0.The genetic distances among the markers on the specific segment became shorter than those of the whole genome map.On the secondary map,Flwdt7 was mapped between Satt277 and Satt489,next to its primary interval Sat_213-Satt643,with distance 1.40 cM to Satt277 and 0.45 cM to Satt489,confidence interval narrowed to 2.7 cM,and contribution rate increased to 36.8%.The results were confirmed with significance analysis among marker genotypes on individual loci and comparison analysis of target marker intervals among near isogenic lines (plants).Thus the strategy by using residual heterozygous lines for QTL fine-mapping on target segments based on primary whole genome scanning with multiple mapping models was demonstrated to be effective. | SU ChengFu LU WeiGuo ZHAO TuanJie GAI JunYi | 2010 | Chinese Science Bulletin2010,55,6: | 6 |

| 3 | Exploration of presence/absence variation and corresponding polymorphic markers in soybean genome显示文摘This study was designed to reveal the genome‐wide distribution of presence/absence variation(PAV) and to establish a database of polymorphic PAV markers in soybean. The 33 soybean whole‐genome sequences were compared to each other with that of Williams 82 as a reference genome. A total of 33,127 PAVs were detected and 28,912 PAV markers with their primer sequences were designed as the database NJAUSoyPAV_1.0. The PAVs scattered on whole genome while only 518(1.8%) overlapped with simple sequence repeats(SSRs) in BARCSOYSSR_1.0database. In a random sample of 800 PAVs, 713(89.13%) showed polymorphism among the 12 differential genotypes. Using 126 PAVs and 108 SSRs to test a Chinese soybean germplasm collection composed of 828 Glycine soja Sieb. et Zucc. and Glycine max(L.) Merr. accessions, the per locus allele number and its variation appeared less in PAVs than in SSRs. The distinctness among alleles/bands of PCR(polymerase chain reaction) products showed better in PAVs than in SSRs, potential in accurate marker‐assisted allele selection. | 8 | Brilliant blue G attenuates lipopolysaccharide-mediated microglial activation and inflammation显示文摘Previous studies have confirmed that oxidized adenosine triphosphate, a P2X7 receptor antagonist, attenuates lipopolysaccharide-mediated microglial activation and inflammatory expression following neuronal damage in rat brain. NaCl and temperature may affect the potency of oxidized adenosine triphosphate. Brilliant blue G is a derivative of a widely used food additive and has little toxicity. This study explored the effects of brilliant blue G, a selective P2X7 receptor antagonist, on microglial activation and inflammation. Results demonstrated that brilliant blue G inhibited the release of cyclooxygenase-2 and interleukin-6 in BV2 cells. Immunofluorescence displayed that brilliant blue G could suppress lipopolysaccharide-induced microglial activation. This study used RNA interference to block P2X7 receptor expression and found that small interfering RNA also suppressed the release of cyclooxygenase-2 and interleukin-6 in BV2 cells. | 14 | Country-level land carbon sink and its causing components by the middle of the twenty-first century显示文摘Background:Countries have long been making efforts by reducing greenhouse-gas emissions to mitigate climate change.In the agreements of the United Nations Framework Convention on Climate Change,involved countries have committed to reduction targets.However,carbon(C)sink and its involving processes by natural ecosystems remain difficult to quantify.Methods:Using a transient traceability framework,we estimated country-level land C sink and its causing components by 2050 simulated by 12 Earth System Models involved in the Coupled Model Intercomparison Project Phase 5(CMIP5)under RCP8.5.Results:The top 20 countries with highest C sink have the potential to sequester 62 Pg C in total,among which,Russia,Canada,USA,China,and Brazil sequester the most.This C sink consists of four components:productiondriven change,turnover-driven change,change in instantaneous C storage potential,and interaction between production-driven change and turnover-driven change.The four components account for 49.5%,28.1%,14.5%,and 7.9%of the land C sink,respectively.Conclusion:The model-based estimates highlight that land C sink potentially offsets a substantial proportion of greenhouse-gas emissions,especially for countries where net primary production(NPP)likely increases substantially and inherent residence time elongates. | Lifen Jiang Junyi Liang Xingjie Lu Enqing Hou Forrest MHoffman Yiqi Luo | 2021 | Ecological Processes2021,10,1: | 0 |

| 17 | A novel nine gene signature integrates stemness characteristics associated with prognosis in hepatocellular carcinoma显示文摘Cancer stem cells(CSCs)are heterogeneous with self-renewal and differentiation ability.The mRNA expression-based stemness index(mRNAsi)described the similarity between tumor cells and CSCs,which is positively associated with the poor prognosis of cancer patients.However,the key prognostic genes related to mRNAsi in hepatocellular carcinoma(HCC)remains unclear.A 9-gene signature related to mRNAsi and HCC prognosis including PSMG3,SNRPD1,DTYMK,PIGU,NME1,TXNL4A,IPO4,PES1,and REXO4 was obtained.High expression of this signature indicates poor prognosis of HCC.PIGU was an independent prognostic factor of HCC,which was significantly associated with progression of HCC.Among them,DTYMK and NME1 enriched in pyrimidine metabolism,SNRPD1 and TXNL4A enriched in spliceosome and PIGU enriched in glycosyl phosphatidylinositol(GPI)-anchor biosynthesis pathways.High levels of IPO4,NME1,PES1,PIGU and SNRPD1 were closely associated with metastasis of HCC,and low levels of IPO4,PIGU and REOX4 were significantly associated with sorafenib resistance of HCC.High expression of the 9-gene signature was negatively correlated with the stromal cell infiltration,and positively correlated with specific immune subtypes-related to angiogenesis,M1/M2 macrophage polarization,and M2 response.The 9-gene signature was negatively correlated with the stroma,and SNRPD1 and TXNL4 were positively correlated with immune infiltrate.NME1 was negatively correlated with tumor purity.Therefore,a 9-gene signature related to mRNAsi and poor prognosis in HCC were identified,which can be used as biomarkers for the diagnosis of HCC and functional mechanism exploration of CSCs in HCC.These genes such as IPO4 and PIGU might drive the transition of tumor cells into CSCs which possibly controls the balance between metastasis and drug resistance in HCC.The challenge on balance between metastasis and drug resistance for tumor therapy was firstly reported by the present study. | XINGHONG YAO CHENGQUAN LU JUNYI SHEN WENLI JIANG YAN QIU YE ZENG LIANG LI | 2021 | BIOCELL2021,45,6: | 0 |
| 18 | Fabrication of highly luminescent TiO_(2):Eu^(3+) thin film with low annealing temperature requirement by co-doping with Sn^(4+) ions显示文摘Rare earth(RE)-doped TiO_(2)thin films possess important applications in modern optoelectronic devices.However,the high annealing temperature requirement remains a critical restriction in device fabrications.In this work,TiO_(2)thin films doped with trivalent europium(Eu^(3+))ions were fabricated by a convenient sol-gel approach and subsequent annealing treatment.Interestingly,it is found that the optimal post-annealing temperature of TiO_(2):Eu^(3+)thin film is dramatically reduced from 800 to 300℃by incorporating 10 at%Sn^(4+)into the sol precursor.The Sn-incorporated film annealed at 300℃shows a 4-fold enhancement in photoluminescence(PL)intensity related to Eu^(3+)ions to the optimized Sn-free film.Systematic analysis reveals that this enhancement is attributed to Sn element promoting the crystallization of both anatase TiO_(2)and rutile SnO_(2)clusters at 300℃,which provides sufficient SnO_(2)/TiO_(2)crystal boundary facilitating Eu^(3+)to occupy and sensitized by SnO_(2)crystals.These results provide valuable insights into fabricating highly luminescent RE-doped TiO_(2)thin films with low annealing temperature requirement,which is essential for designing optoelectronic devices based on RE materials more freely in the future. | Lu Zhang Bolin Zheng Junyi Zhai Tao Lin | 2023 | Journal of Rare Earths2023,41,10: | 0 |
