5Correlation between tongue manifestations and glucose, total cholesterol, and high-density lipoprotein cholesterol in patients with acute cerebral infarction显示文摘OBJECTIVE: To analyze the association between tongue manifestations and the levels of glucose (GLU), total cholesterol(TCH), and high-density lipoprotein cholesterol(HDL-C) in subjects with acute cerebral infarction.METHODS: Hospitalized patients with first unilateral cerebral infarction in the Neurological Department of Xuanwu Hospital were included and the correlation between tongue fur color, fur nature,and the levels of GLU,TCH, HDL-C were analyzed.RESULTS: HDL level in the thin fur group was higher than that in the thick fur group(P=0.02). The difference in the levels of GLU,TCH, and HDL-C among the groups was significant(P<0.05), classified in terms of slippery, moist, and dry fur. Further comparison between the groups by Student-Newman-Keuls test showed that GLU level in the dry fur group was the highest. 6Prognostic evaluation of postoperative adjuvant therapy for operable cervical cancer:10 years'experience of National Cancer Center in China显示文摘Objective: The aim of this study was to investigate the prognostic factors and to evaluate the impact of adjuvant therapy on clinical outcome for early-stage cervical cancer.Methods: The clinical-pathological data of all 1,335 patients with the International Federation of Gynecology and Obstetrics(FIGO) Ib–IIa cervical cancer treated with primary radical surgery at the Chinese National Cancer Center between May 2007 and Dec 2013 were retrospectively reviewed.The median follow-up was 70 months.Results: Of all the patients,61.6% of the cases received adjuvant therapy,with 5-year disease-free survival(DFS)of 92.1% and 5-year overall survival(OS) of 95.0%.In multivariate analysis,differentiation of G3(P<0.05),lymph node metastasis(LNM,P<0.05) and lymphovascular space invasion(LVSI,P<0.05) were independent predictors for OS,while LNM(P<0.05),deep stroma invasion(DSI,P<0.05) and LVSI(P<0.05) were independent factors for DFS.The samples were stratified by histologic type,and cervical squamous cell carcinoma(SCC) was found to share the same independent factors except for differentiation of OS.As to patients with cervical adenocarcinoma/adenosquamous carcinoma(AC/ASC),differentiation was the independent predictor of OS(P<0.05); and LVSI of DFS(P<0.05).Of 236 patients with high-risk factors,there was no significant difference in survival between concurrent chemoradiotherapy(CCRT,n=195),radiotherapy(RT,n=24),and chemotherapy(CT,n=17).Among the 190 patients with LNM who underwent CCRT,124 cases showed improved DFS after sequential CT(P=0.118),with a recurrence rate decrease of 14%,though the difference was not statistically significant.Patients with single intermediate-risk factors like DSI or LVSI were found to partially benefit from adjuvant therapy,but the difference was not statistically significant.Conclusions: LNM,LVSI,DSI and differentiation were found to be independent prognostic factors for operable cervical cancer.Aggressive postoperative adjuvant therapy based on single risk factors in Chinese National Cancer Center could benefit survival.CCRT+CT outperformed CCRT in high-risk patients.For patients with single non-high-risk factor,the role of adjuvant therapy needs to be further discussed.Tong Shu Dan Zhao Bin Li Yating Wang Shuanghuan Liu Pingping Li Jing Zuo Ping Ba i RongZhang Lingying Wu 2017Chinese Journal of Cancer Research2017,29,6:12
7Influence of coupled boundary layer suction and bowed blade on flow field and performance of a diffusion cascade显示文摘Based on the investigation of mid-span local boundary layer suction and positive bowed cascade, a coupled local tailored boundary layer suction and positive bowed blade method is developed to improve the performance of a highly loaded diffusion cascade with less suction slot. The effectiveness of the coupled method under different inlet boundary layers is also investigated.Results show that mid-span local boundary layer suction can effectively remove trailing edge separation, but deteriorate the flow fields near the endwall. The positive bowed cascade is beneficial for reducing open corner separation, but is detrimental to mid-span flow fields. The coupled method can further improve the performance and flow field of the cascade. The mid-span trailing edge separation and open corner separation are eliminated. Compared with linear cascade with suction, the coupled method reduces overall loss of the cascade by 31.4% at most. The mid-span loss of the cascade decreases as the suction coefficient increases, but increases as bow angle increases. The endwall loss increases as the suction coefficient increases. By contrast, the endwall loss decreases significantly as the bow angle increases. The endwall loss of coupled controlled cascade is higher than that of bowed cascade with the same bow angle because of the spanwise inverse ‘‘C' shaped static pressure distribution. 8Intestinal lysozyme liberates Nod1 ligands from microbes to direct insulin trafficking in pancreatic beta cells显示文摘Long-range communication between intestinal symbiotic bacteria and extra-intestinal organs can occur through circulating bacterial signal molecules, through neural circuits, or through cytokines or hormones from host cells. Here we report that Nod1 ligands derived from intestinal bacteria act as signal molecules and directly modulate insulin trafficking in pancreatic beta cells. The cytosolic peptidoglycan receptor Nod1 and its downstream adapter Rip2 are required for insulin trafficking in beta cells in a cell-autonomous manner. Mechanistically, upon recognizing cognate ligands, Nod1 and Rip2 localize to insulin vesicles, recruiting Rab1a to direct insulin trafficking through the cytoplasm. Importantly, intestinal lysozyme liberates Nod1 ligands into the circulation, thus enabling long-range communication between intestinal microbes and islets. The intestine-islet crosstalk bridged by Nod1 ligands modulates host glucose tolerance. 9Gut microbiota drives macrophage-dependent self-renewal of intestinal stem cells via niche enteric serotonergic neurons显示文摘Lgr5+intestinal stem cells(ISCs)reside within specialized niches at the crypt base and harbor self-renewal and differentiation capacities.ISCs in the crypt base are sustained by their surrounding niche for precise modulation of self-renewal and differentiation.However,how intestinal cells in the crypt niche and microbiota in enteric cavity coordinately regulate ISC stemness remains unclear.Here,we show that ISCs are regulated by microbiota and niche enteric serotonergic neurons.The gut microbiota metabolite valeric acid promotes Tph2 expression in enteric serotonergic neurons via blocking the recruitment of the NuRD complex onto Tph2 promoter.5-hydroxytryptamine(5-HT)in turn activates PGE2 production in a PGE2+macrophage subset through its receptors HTR2A/3 A;and PGE2 via binding its receptors EP1/EP4,promotes Wnt/β-catenin signaling in ISCs to promote their self-renewal.Our findings illustrate a complex crosstalk among microbiota,intestinal nerve cells,intestinal immune cells and ISCs,revealing a new layer of ISC regulation by niche cells and microbiota.Pingping Zhu Tiankun Lu Jiayi Wu Dongdong Fan Benyu Liu Xiaoxiao Zhu Hui Guo Ying Du Feng Liu Yong Tian Zusen Fan 2022Cell Research2022,32,6:8
10Quantitative trait loci for Fusarium head blight resistance in wheat cultivars Yangmai 158 and Zhengmai 9023显示文摘Fusarium head blight(FHB)is one of the prevalent fungal diseases of wheat worldwide.Exploring new FHB resistance quantitative trait loci(QTL)in adapted wheat cultivars is a critical step for breeding new FHB-resistant cultivars.In this study,we developed a population of 236 F5:7 recombinant inbred lines(RILs)using two popular Chinese wheat cultivars,Yangmai 158 and Zhengmai 9023,with moderate FHB resistance to identify the QTL for FHB type II resistance.This population was evaluated for percentage of symptomatic spikelets per spike(PSS)using single floret injection in repeated greenhouse experiments.Mean PSSs were 33.2%for Yangmai 158 and 30.3%for Zhengmai 9023.A genetic linkage map of 1002 single nucleotide polymorphisms(SNPs)generated by genotyping-by-sequencing(GBS)was constructed for the RIL population.Six QTL were identified for FHB resistance,and three of them were repeatable in the both experiments.Zhengmai 9023 contributed the resistance allele at one repeatable QTL,designated as Qfhb.7D,whereas Yangmai 158 contributed the resistance alleles at the other two repeatable QTL,Qfhb.3AL and Qfhb.2DS.The additional QTL,Qfhb.4AS was significant in the mean PSS,and Qfhb.2DL and Qfhb.7AS were significant in only one experiment.Replacement of each allele individually at the three repeatable QTL significantly changed PSSs.Qfhb.3AL,Qfhb.2DS,and Qfhb.7D explained 8.35%to 9.89%,5.13%to 7.43%,and 6.15%to 9.32%of the phenotypic variations,respectively.The three repeatable QTL contributed by the two parents were additive and stacking the resistance alleles from all the three repeatable QTL showed the highest level of resistance in the current RIL population.Ten SNPs in the QTL regions of Qfhb.3AL,Qfhb.2DS,and Qfhb.7D were converted into KBioscience competitive allele-specific PCR(KASP)assays.One KASP marker for Qfhb.3AL was validated in a panel of wheat cultivars from China.Some of these KASP markers could be useful for marker-assisted selection to stack these QTL.Pingping Zhang Chenjin Guo Zhao Liu Amy Bernardo Hongxiang Maa Hongxiang Ma Peng Jiang Guicheng Song Guihua Bai 2021The Crop Journal2021,9,1:8
11Tumor-expressed B7-H3 mediates the inhibition of antitumor T-cell functions in ovarian cancer insensitive to PD-1 blockade therapy显示文摘Although PD-L1/PD-1 blockade therapy has been approved to treat many types of cancers,the majority of patients with solid tumors do not respond well,but the underlying reason remains unclear.Here,we studied ovarian cancer(OvCa),a tumor type generally resistant to current immunotherapies,to investigate PD-1-independent immunosuppression.We found that PD-L1 was not highly expressed in the tumor microenvironment(TME)of human OvCa.Instead,B7-H3,another checkpoint molecule,was highly expressed by both tumor cells and tumor-infiltrating antigen-presenting cells(APCs),which correlated with T-cell exhaustion in patients.Using ID8 OvCa mouse models,we found that B7-H3 expressed on tumor cells,but not host cells,had a dominant role in suppressing antitumor immunity.Therapeutically,B7-H3 blockade,but not PD-1 blockade,prolonged the survival of ID8 tumorbearing mice.Collectively,our results demonstrate that tumor-expressed B7-H3 inhibits the function of CD8^(+)T cells and suggest that B7-H3 may be a target in patients who are not responsive to PD-L1/PD-1 inhibition,particularly OvCa patients.Dongli Cai Jiaming Li Dingfeng Liu Shanjuan Hong Qin Qiao Qinli Sun Pingping Li Nanan Lyu Tiantian Sun Shan Xie Li Guo Ling Ni Liping Jin Chen Dong 2020Cellular & Molecular Immunology2020,17,3:8
12Anti-parkinsonian effects of octacosanol in 1-methyl-4-phenyl-1,2,3,6 tetrahydropyridine-treated mice显示文摘Our previous research showed that octacosanol exerted its protective effects in 6-hydroxydopamine-induced Parkinsonian rats. The goal of this study was to investigate whether octacosanol would attenuate neurotoxicity in 1-methyl-4-phenyl-1,2,3,6 tetrahydropyridine (MPTP)-treated C57BL/6N mice and its potential mechanism. Behavioral tests, tyrosine hydroxylase immunohistochemistry and western blot were used to investigate the effects of octacosanol in a mouse model of Parkinson's disease. Oral administration of octacosanol (100 mg/kg) significantly improved behavioral impairments in mice treated by MPTP and markedly ameliorated morphological appearances of tyrosine hydroxylase-positive neuronal cells in the substantia nigra. Furthermore, octacosanol blocked MPTP-induced phosphorylation of p38MAPK and JNK, but not ERK1/2. These findings implicated that the protective effects afforded by octacosanol might be mediated by blocking the phosphorylation of p38MAPK and JNK on the signal transduction in vivo. 14Effect of monovalent anions on cationic Gemini micro-emulsion显示文摘Micro-emulsion usually consists of water, oil, surfactants and co-surfactants, and each component has an effect on the phase behavior and solubilization of the micro-emulsion. When the surfactant in the micro-emulsion system is quaternary ammonium cationic Gemini surfactant, the surfactant mainly combines with the anions in the salt. With the increase of salt concentration, the phase transformation of Winsor I → Winsor III → Winsor II occurred, but the optimum salinity and salt width are different because of the type of salt. The effects of 5 different kinds of monovalent anions, including C_6H_5SO_3^-, I-, Br-, NO_3^- and Cl-, on the phase behavior and solubilization of quaternary ammonium cationic Gemini micro-emulsion are researched by Winsor phase diagram. It is found that the effects of organic anions C_6H_5SO_3-and I-on the phase behavior and solubilization of quaternary ammonium cationic Gemini micro-emulsion are most significant, and the effects of Br-, NO_3^- and Cl-are less significant. 16Efficient Image Transmission Schemes over Zigbee-Based Image Sensor Networks显示文摘Image transmission is one of the biggest challenges in wireless sensor networks because of the limited resource on sensor nodes. We proposed two image transmission schemes driven by reliability and real time considerations in order to transfer JPEG images over Zigbee-based sensor networks. By adding two bytes counter in the header of data packet, we can easily solve the repeated data reception problem caused by retransmission mechanism in traditional Zigbees network layer. We proposed an efficient retransmission and acknowledgment mechanism in Zigbees application layer. By classifying different data reception response events, we can provide data packets with differential responses and ensure that image packets can be transferred quickly even with large maximum number of retransmission. 17Nano-hydroxyapatite accelerates vascular calcification via lysosome impairment and autophagy dysfunction in smooth muscle cells显示文摘Vascular calcification(VC)is a common characteristic of aging,diabetes,chronic renal failure,and atherosclerosis.The basic component of VC is hydroxyapatite(HAp).Nano-sized HAp(nHAp)has been identified to play an essential role in the development of pathological calcification of vasculature.However,whether nHAp can induce calcification in vivo and the mechanism of nHAp in the progression of VC remains unclear.We discovered that nHAp existed both in vascular smooth muscle cells(VSMCs)and their extracellular matrix(ECM)in the calcified arteries from patients.Synthetic nHAp had similar morphological and chemical properties as natural nHAp recovered from calcified artery.nHAp stimulated osteogenic differentiation and accelerated mineralization of VSMCs in vitro.Synthetic nHAp could also directly induce VC in vivo.Mechanistically,nHAp was internalized into lysosome,which impaired lysosome vacuolar H+-ATPase for its acidification,therefore blocked autophagic flux in VSMCs.Lysosomal re-acidification by cyclic-3′,5′-adenosine monophosphate(cAMP)significantly enhanced autophagic degradation and attenuated nHAp-induced calcification.The accumulated autophagosomes and autolysosomes were converted into calcium-containing exosomes which were secreted into ECM and accelerated vascular calcium deposit.Inhibition of exosome release in VSMCs decreased calcium deposition.Altogether,our results demonstrated a repressive effect of nHAp on lysosomal acidification,which inhibited autophagic degradation and promoted a conversion of the accumulated autophagic vacuoles into exosomes that were loaded with undissolved nHAp,Ca^(2+),Pi and ALP.These exosomes bud off the plasma membrane,deposit within ECM,and form calcium nodules.Vascular calcification was thus accelerated by nHAP through blockage of autophagic flux in VSMCs.Qi Liu Yi Luo Yun Zhao Pingping Xiang Jinyun Zhu Wangwei Jing Wenjing Jin Mingyao Chen Ruikang Tang Hong Yu 2022Bioactive Materials2022,7,2:5

19S-Allylmercaptocysteine improves alcoholic liver disease partly through a direct modulation of insulin receptor signaling显示文摘Alcoholic liver disease(ALD) causes insulin resistance, lipid metabolism dysfunction, and inflammation. We investigated the protective effects and direct regulating target of S-allylmercaptocysteine(SAMC) from aged garlic on liver cell injury. A chronic ethanol-fed ALD in vivo model(the NIAAA model) was used to test the protective functions of SAMC. It was observed that SAMC(300 mg/kg, by gavage method) effectively ameliorated ALD-induced body weight reduction, steatosis,insulin resistance, and inflammation without affecting the health status of the control mice, as demonstrated by histological, biochemical, and molecular biology assays. By using biophysical assays and molecular docking, we demonstrated that SAMC directly targeted insulin receptor(INSR) protein on the cell membrane and then restored downstream IRS-1/AKT/GSK3 b signaling. Liver-specific knock-down in mice and siRNA-mediated knock-down in AML-12 cells of Insr significantly impaired SAMC(250 mmol/L in cells)-mediated protection. Restoration of the IRS-1/AKT signaling partly recovered hepatic injury and further contributed to SAMC’s beneficial effects. Continuous administration of AKT agonist and recombinant IGF-1 in combination with SAMC showed hepato-protection in the mice model.Long-term(90-day) administration of SAMC had no obvious adverse effect on healthy mice. 20Impact of friction stir welding on recrystallization of oxide dispersion strengthened ferritic steel显示文摘Oxide dispersion strengthened(ODS) steels can be used as the structural materials in the future fusion reactors and the fuel cladding materials in the advanced fission reactors. However, the weldability of ODS steels is a severe problem. In the present study, defect-free joints of the 15Cr-ODS ferritic steel were achieved by friction stir welding at different rotation speeds. The recrystallization, hardness and tensile properties are highly related with the rotation speed of the stir tool. The higher rotation speed results in coarser grains in the top SZ, while the grain size exhibits more complicated relation with the rotation speed in the SZ center.
