1Isolation and characterization of H7N9 viruses from live poultry markets--Implication of the source of current H7N9 infection in humans显示文摘On March 31, 2013, the National Health and Family Planning Commission announced that human infections with a previously undescribed influenza A (H7N9) virus had occurred in Shanghai and Anhui Province, China. To investigate the possible origins of the H7N9 viruses causing these human infections, we collected 970 samples, including drinking water, soil, and cloacal and tracheal swabs of poultry from live poultry markets and poultry farms in Shanghai and Anhui Province. Twenty samples were positive for the H7N9 influenza virus. Notably, all 20 viruses were isolated from samples collected from live poultry markets in Shanghai. Phylogenetic analyses showed that the six internal genes of these novel human H7N9 viruses were derived from avian H9N2 viruses, but the ancestor of their HA and NA genes is uncertain. When we examined the phylogenetic relationship between the H7N9 isolates from live poultry markets and the viruses that caused the human infections, we found that they shared high homology across all eight gene segments. We thus identified the direct avian origin of the H7N9 influenza viruses that caused the human infections. Importantly, we observed that the H7N9 viruses isolated from humans had acquired critical mutations that made them more 'human-like'. 3The altered PD-1/PD-L1 pathway delivers the ‘one-two punch’ effects to promote the Treg/Th17 imbalance in pre-eclampsia显示文摘The programmed cell death-1(PD-1)/PD-ligand 1(PD-L1)pathway is critical for normal pregnancy by promoting regulatory T(Treg)cell development and inhibiting the Th17 response.However,the relationship between the PD-1/PD-L1 pathway and the Treg/Th17 imbalance in pre-eclampsia(PE)is an enigma.In this study,decreased PD-1 and PD-L1 expression and a Treg/Th17 imbalance were observed at the maternal-fetal interface in PE.The regulatory effects of the PD-1/PD-L1 pathway on the Treg and Th17 cell quantities were determined in vitro by targeting T-cell proliferation,differentiation and transdifferentiation.First,decreased PD-1 expression might contribute to a higher Th17 cell frequency by promoting proliferation in PE.Second,the percentages of Treg but not Th17 cells differentiated from peripheral naive CD4+T cells were increased by PD-L1 Fc administration.This effect was accompanied by decreased PI3K/AKT/m-TOR and increased PTEN mRNA expression and was completely reversed by PD-1 blockade.Finally,the percentage of IL-17-producing Treg cells increased and was positively associated with the Th17 cell frequency in PE.Increased RORγt and IL-17 but not Foxp3 and IL-10 mRNA expression by Treg cells was observed with PD-1 blockade.Similar findings occurred when Treg cells were exposed to IL-6/IL-23/IL-1βand were reversed by PD-L1 Fc.Taken together,our findings indicate that the PD-1/PD-L1 pathway contributes to the Treg/Th17 imbalance via‘one-two punch’approaches:(i)promoting Th17 cell proliferation,(ii)inhibiting Treg cell differentiation and(iii)enhancing Treg cell plasticity into Th17 cells in PE.The therapeutic value of PD-L1 Fc for PE treatment will be explored in the future.Yonghong Zhang Zhaozhao Liu Mei Tian Xiaohui Hu Liling Wang Jinlu Ji Aihua Liao 2018Cellular & Molecular Immunology2018,15,7:21
4Soil Biological Changes for a Natural Forest and Two Plantations in Subtropical China显示文摘Conversion of natural forests into pure plantation forests is a common management practice in subtropical China.To evaluate the effects of forest conversion on soil fertility, microbe numbers and enzyme activities in topsoils (0-10 cm)were quantified in two 33-year-old monoculture plantations of Castanopsis kawakamii Hayata (CK) and Cunninghamia lanceolata Lamb. (Chinese fir) (CF), and compared to a neighboring relict natural C. kawakamii forest (NF), in Sanming,Fujian. Five soil samples were collected once each in January, April, July, September and November in 2000 in each forest for laboratory analysis. Over the sampling year, there were significant differences for bacteria, fungi and actinomycetes between forests and between seasons (P < 0.05). The largest bacteria and fungi populations were in NF, while CF contained the greatest number of actinomycetes. There were also significant differences (P < 0.05) with microbial respiration for forests and seasons. Additionally, compared with NF, urease and acid phosphatase were significantly lower (P < 0.05)in CK and CF. Also, the correlations of soil hydrolysable N and available P to soil microbial and enzymatic activities were highly significant (P < 0.01). 5Comparison of breast-conserving surgery and mastectomy in early breast cancer using observational data revisited:a propensity score-matched analysis显示文摘Recent observational studies showed that breast-conserving surgery(BCS) resulted in superior survival compared to mastectomy in breast cancer patients. This study compared the clinical outcomes of BCS and mastectomy using propensity score(PS)matching analysis, which had advantages over conventional methods in reducing bias. Nonmetastatic breast cancer patients who underwent BCS and mastectomy were matched 1:1 based on their PS. We used the Kaplan-Meier method and Cox-regression model to estimate the treatment effects. A total of 2,866 patients with a median follow-up time of 67 months were included in the original study population. Although the mastectomy cohort(N=1,219) had more advanced disease compared to the BCS cohort(N=1,647), LRFS was similar between the two groups(93.8% vs. 92.4%, P>0.05). BCS(vs. mastectomy) was associated with improved DFS(73.8% vs. 58.7%, P<0.01) and CSS(91% vs. 78.2%, P<0.01) in the original population. In the PS-matched population(N=1,668), clinicopathological features were equally distributed between the two cohorts. BCS(vs. mastectomy) was not associated with improved DFS(70.7% vs. 66.9%, P>0.05) or CSS(87.5% vs. 84.9%, P>0.05). We found that PS methods reduce bias when estimating treatment effects using observational data. 6Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. 8Pollution characteristics of the rivers in suburban Shanghai显示文摘Forty middle and small rivers in three towns of Fengjing, Songjiang and Zhujiajiao of suburban Shanghai were chosen as sampling sites for water quality research. Measurement results of DO, CODCr, TP, TN and so on show that the rivers are under heavy eutropic conditions, which are several times greater than the critical values of the worst level (Type V) of water. Water pollution situation has different temporal and spatial characteristics. 10Physiological responses of osteoblasts to cyclic stretching and the change of intracellular calcium concentration显示文摘The development of bone tissues is regulated by mechanical stimulation. Cyclic stretching was applied to the osteoblasts that were delivered from rat calvarie. The results showed that stretching at 500 me increased the cell prolifera-tion while loading at 1000 me and 1500 me inhabited cell growth. Loading also increased the adhesive force between cells and substrate as well as spreading areas of osteobalsts. Furthermore, the fluorescence probe Fluo-3/AM was used to investigate the effect of stretching stimulation on the intra-cellular calcium concentration of osteoblasts. The intracellu-lar calcium concentration of osteoblasts that were stretched at 500 me for 5 min was 92.9% higher than the control. After being treated with the panax ontoginseng saponins, the stretched osteoblasts still expressed 28.6% higher intracellu-lar calcium concentration than that of the control, which proved that both the influx of extracellular calcium and the release of intracellular calcium store were involved in the increase of intracellular calcium concentration when os-teoblasts responded to the cyclic stretching. 11Preliminary studies on the early quality identification of A uric ularia auricular显示文摘In production industry of Auricularia auricular, the varieties quality is most important impact factor on output. For evaluating the early quality of the edible fungus, 9 varieties of Auricularia auricular (Au9, CF09, CF05, 29, 916, chang10, chang7, Au.Japanese and 8808) were cultured on the medium consists of potato dextrose agar (PDA) and sawdust to test their mycelium growth rate, endurance to high temperature, resistance to mildew, assimilation of nutriment, and resistance to drought. The result showed that the mycelium of Chang 10, CF09, 29 and Au.Japanese varieties had the eminent characteristics such as short lifespan, stronger assimilation of nutriment, and endurance to high temperature and steady growth. 13Comparison of chicoric acid,and its metabolites caffeic acid and caftaric acid:In vitro protection of biological macromolecules and inflammatory responses in BV2 microglial cells显示文摘Chicoric acid(CA),a natural phenolic acid,has been used as a nutraceutical food ingredient due to its powerful antioxidant,anti-HIV and anti-diabetic bioactivities.CA could be partly metabolized into caffeic acid(CFA)and caftaric acid(CTA)on cytochrome P450s in rat liver microsomes.To compare the protective effects of CA and its metabolites on biomolecules and inflammatory responses,oxidative damage induced by free radicals in vitro and microglial inflammation triggered by lipopolysaccharide in BV2 cells were constructed.Results showed that CA,CTA and CFA all significantly inhibited protein degradation and carbonylation induced by hydroxyl radicals and alcoxyl radicals,and suppressed hemin/nitrite/H2O2 triggered-nitration.Moreover,CA,CTA and CFA all exerted remarkable inhibition capacities on linoleic acid and soybean lecithin liposomes peroxidation in a dose-dependent manner,and restrained the oxidation of herring sperm DNA,as well as the breakage of pBR322 plasmid DNA.Furthermore,CA and its metabolites suppressed lipopolysaccharide-induced decline of BV2 cell viability and the production of NO and ROS.However,bioactivities of CA were significantly stronger than those of its metabolites within a certain concentration range.This study provides scientific basis for the application of CA and its metabolites as nutrition and natural antioxidants.Qian Liu Fan Liu Liling Zhang Yajie Niu Zhigang Liu Xuebo Liu 2017Food Science and Human Wellness2017,6,4:5
14Competing endogenous RNAs in lung cancer显示文摘Competing endogenous RNAs(ce RNAs)containing micro RNA response elements can competitively interact with micro RNA via mi RNA response elements,which can combine non-coding RNAs with protein-coding RNAs through complex ce RNA networks.Ce RNAs include non-coding RNAs(long non-coding RNAs,circular RNAs,and transcribed pseudogenes)and protein-coding RNAs(m RNAs).Molecular interactions in ce RNA networks can coordinate many biological processes;however,they may also lead to ce RNA network imbalance and thus contribute to cancer occurrence when disturbed.Recent studies indicate that many dysregulated RNAs derived from lung cancer may function as ce RNAs to regulate multitudinous biological functions for lung cancer,including tumor cell proliferation,apoptosis,growth,invasion,migration,and metastasis.This study therefore reviewed the research progress in the field of non-coding and protein-coding RNAs as ce RNAs in lung cancer,and highlighted validated ce RNAs involved in biological lung cancer functions.Furthermore,the roles of ce RNAs as novel prognostic and diagnostic biomarkers were also discussed.Interpreting the involvement of ce RNAs networks in lung cancer will provide new insight into cancer pathogenesis and treatment strategies.Meilian Zhao Jianguo Feng Liling Tang 2021Cancer Biology & Medicine2021,18,1:5
15Aquaporin 5 is degraded by autophagy in diabetic submandibular gland显示文摘Autophagy is a catabolic process which is involved in the development of many diseases including diabetes mellitus and its complications. Hyposalivation is a common complication of diabetes mellitus, whereas its mechanism remains unclear. Here, we observed that the stimulated salivary flow rate of SMG was significantly decreased in db/db mice, a diabetic mice model. The expressions of aquaporin 5(AQP5), a water channel protein, were decreased, whereas the m RNA level of AQP5 was increased in SMGs of both diabetic patients and mice. Under transmission electron microcope, more autophagosomes were detected in diabetic SMGs. Expressions of autophagy related proteins LC3 II, Beclin-1 and ATG5 were increased, meanwhile autophagy substrate p62 was decreased in SMGs of diabetic patients and mice, indicating that autophagy was activated in diabetic SMG.Double immunofluorescence staining showed that the colocalization of AQP5 and LC3 was increased in SMGs of diabetic mice.In cultured SMG-C6 cells, high glucose(HG), but not high osmotic pressure, reduced AQP5 protein expression and induced autophagy. Moreover, inhibition of autophagy by 3-methyladenin, an autophagy inhibitor, or by autophagy-related gene 5 siRNA, decreased HG-induced AQP5 reduction in SMG-C6 cells. Additionally, the expression of p-p85, p-Akt and p-mTOR were decreased in HG-treated SMG-C6 cells. Pretreatment with 740 Y-P, a PI3 K agonist, significantly suppressed HG-induced autophagy and AQP5 degradation. Taken together, these results indicate that autophagy plays a crucial role in AQP5 degradation in diabetic SMG via PI3 K/Akt/mTOR signaling pathway, which contributes to the dysfunction of diabetic SMG. 19Tracking of time-evolving sound speed profiles with an auto-regressive state-space model显示文摘An approach for time-evolving sound speed profiles tracking in shallow water is discussed.The inversion of time-evolving sound speed profiles is modeled as a state-space estimation problem,which includes a state equation for predicting the time-evolving sound speed profile and a measurement equation for incorporating local acoustic measurements.In the paper,auto-regression(AR) method is introduced to obtain a high-order AR evolution model of the sound speed field time variations,and the ensemble Kalman filter is utilized to track the sound speed field.To validate the approach,the accuracy in sound speed estimation is analyzed via a numerical implementation using the ASIAEX experimental environment and the sound velocity measurement data.Compared with traditional approaches based on the state evolution represented as a random walk,simulation results show the proposed AR method can effectively reduce the tracking errors of sound speed,and still keep good tracking performance at low signal-to-noise ratios.JIN Liling LI Jianlong XU Wen 2017Chinese Journal of Acoustics2017,36,3:4
