1Bmi-1:At the crossroads of physiological and pathological biology显示文摘Bmi-1 is a member of the Polycomb repressor complex 1 that mediates gene silencing by regulating chromatin structure and is indispensable for self-renewal of both normal and cancer stem cells.Despite three decades of research that have elucidated the transcriptional regulation,post-translational modifications and functions of Bmi-1 in regulating the DNA damage response,cellular bioenergetics,and pathologies,the entire potential of a protein with such varied functions remains to be realized.This review attempts to synthesize the current knowledge on Bmi-1 with an emphasis on its role in both normal physiology and cancer.Additionally,since cancer stem cells are emerging as a new paradigm for therapy resistance,the role of Bmi-1 in this perspective is also highlighted.The wide spectrum of malignancies that implicate Bmi-1 as a signature for stemness and oncogenesis also make it a suitable candidate for therapy.Nonetheless,new approaches are vitally needed to further characterize physiological roles of Bmi-1 with the long-term goal of using Bmi-1 as a prognostic marker and a therapeutic target.Resham Bhattacharya Soumyajit Banerjee Mustafi Mark Street Anindya Dey Shailendra Kumar Dhar Dwivedi 2015Genes & Diseases2015,2,3:15
2Biomedical Perspective of Electrochemical Nanobiosensor显示文摘Electrochemical biosensor holds great promise in the biomedical area due to its enhanced specificity, sensitivity, label-free nature and cost effectiveness for rapid point-of-care detection of diseases at bedside. In this review, we are focusing on the working principle of electrochemical biosensor and how it can be employed in detecting biomarkers of fatal diseases like cancer, AIDS, hepatitis and cardiovascular diseases. Recent advances in the development of implantable biosensors and exploration of nanomaterials in fabrication of electrodes with increasing the sensitivity of biosensor for quick and easy detection of biomolecules have been elucidated in detail. Electrochemical-based detection of heavy metal ions which cause harmful effect on human health has been discussed. 3Treatment of wheat straw using tannase and white-rot fungus to improve feed utilization by ruminants显示文摘Background: Current research to enrich cattle feed has primarily focused on treatment using white rot fungi, while there are scarce reports using the enzyme tannase, which is discussed only in reviews or in the form of a hypothesis. In this context, the aim of the present study was to evaluate the effect of tannase on wheat straw(WS)and also the effect of lyophilized tannase at concentrations of 0.1%, 0.2%, and 0.3%(w/w) on WS followed by fermentation with Ganoderma sp. for 10 d and compared in relation to biochemical parameters, crude protein(CP)content, and nutritional value by calculating the C/N ratio in order to improve the nutritional value of cattle feed.Results: Penicillium chorlesii, a tannase-producing microorganism, produced 61.4 lU/mL of tannase in 54 h when 2%(w/v) tannic acid(TA) was initially used as a substrate in medium containing(% w/v) sucrose(1.0), NaN03(1.0), and MgS〇4(0.08 pH, 5.0) in a 300-L fermentor(working volume 220 L), and concomitantly fed with 1.0%(w/v) TA after24 h. The yield of partially purified and lyophilized tannase was 5.8 lU/mg. The tannin-free myco-straw at 0.1%(w/w) tannase showed 37.8%(w/w) lignin degradation with only a 20.4%(w/w) decrease in cellulose content and the in vitro feed digestibility was 32.2%. An increase in CP content(up to 1.28-fold) along with a lower C/N ratio of25.0%, as compared to myco-straw, was obtained.Conclusions: The use of tannin-free myco-straw has potential to improve the nutritional content of cattle feed.This biological treatment process was safe, eco-friendly, easy to perform, and was less expensive as compared to other treatment methods.Shailendra Raghuwanshi Swati Misra Rajendra Kumar Saxena 2014Journal of Animal Science and Biotechnology2014,5,2:5

6Synthesis,isolation,identification and characterization of new process-related impurity in isoproterenol hydrochloride by HPLC,LC/ESI-MS and NMR显示文摘One unknown impurity(Imp-Ⅱ) during the analysis of laboratory batches of isoproterenol hydrochloride was detected in the level ranging from 0.04% to 0.12% by high performance liquid chromatography with UV detection. The unknown impurity structure was proposed as 4-[2-(propan-2-ylamino)ethyl]benzene-1,2-diol(Imp-Ⅱ) using the liquid chromatography–mass spectrophotometry(LC–MS) analysis. Imp-Ⅱ was isolated by semi-preparative liquid chromatography from the impurity-enriched reaction crude sample. Its proposed structure was confirmed by nuclear magnetic spectroscopy such as ~1H,^(13)C, DEPT(1D NMR), HSQC(2D NMR)and infrared spectroscopy(IR), and retention time and purity with HPLC followed by the chemical synthesis.Due to less removable nature of Imp-Ⅱ during the purification, the synthetic process was optimized proficiently to control the formation of Imp-Ⅱ below to the limit < 0.12% in the course of reaction. 7GDF15 promotes prostate cancer bone metastasis and colonization through osteoblastic CCL2 and RANKL activation显示文摘Bone metastases occur in patients with advanced-stage prostate cancer(PCa). The cell-cell interaction between PCa and the bone microenvironment forms a vicious cycle that modulates the bone microenvironment, increases bone deformities, and drives tumor growth in the bone. However, the molecular mechanisms of PCa-mediated modulation of the bone microenvironment are complex and remain poorly defined. Here, we evaluated growth differentiation factor-15(GDF15) function using in vivo preclinical PCa-bone metastasis mouse models and an in vitro bone cell coculture system. Our results suggest that PCa-secreted GDF15 promotes bone metastases and induces bone microarchitectural alterations in a preclinical xenograft model. Mechanistic studies revealed that GDF15 increases osteoblast function and facilitates the growth of PCa in bone by activating osteoclastogenesis through osteoblastic production of CCL2 and RANKL and recruitment of osteomacs.
