| 1 | Targeting inflammation in diabetes: Newer therapeutic options显示文摘Inflammation has been recognised to both decrease beta cell insulin secretion and increase insulin resis-tance. Circulating cytokines can affect beta cell function directly leading to secretory dysfunction and increased apoptosis. These cytokines can also indirectly affect beta cell function by increasing adipocyte inflamma-tion.The resulting glucotoxicity and lipotoxicity further enhance the inflammatory process resulting in a vicious cycle. Weight reduction and drugs such as metformin have been shown to decrease the levels of C-Reactive Protein by 31% and 13%, respectively. Pioglitazone, insulin and statins have anti-inflammatory effects. In-terleukin 1 and tumor necrosis factor-α antagonists are in trials and NSAIDs such as salsalate have shown an improvement in insulin sensitivity. Inhibition of 12-lipo-oxygenase, histone de-acetylases, and activation of sirtuin-1 are upcoming molecular targets to reduce in-flammation. These therapies have also been shown to decrease the conversion of pre-diabetes state to diabe-tes. Drugs like glicazide, troglitazone, N-acetylcysteine and selective COX-2 inhibitors have shown benefit in diabetic neuropathy by decreasing inflammatory mark-ers. Retinopathy drugs are used to target vascular en-dothelial growth factor, angiopoietin-2, various protein-ases and chemokines. Drugs targeting the proteinases and various chemokines are pentoxifylline, inhibitors of nuclear factor-kappa B and mammalian target of rapa-mycin and are in clinical trials for diabetic nephropathy. Commonly used drugs such as insulin, metformin, per-oxisome proliferator-activated receptors, glucagon like peptide-1 agonists and dipeptidyl peptidase-4 inhibitors also decrease inflammation. While crowdsourcing is widely used in crowd management, conventional solutions, such as Upwork and Amazon Mechanical Turk, generally depend on a trusted third-party platform. There exist several potential security concerns(e.g., sensitive leakage, single point of failure and unfair judgment) in such a centralized paradigm. Hence, a recent trend in crowdsourcing is to leverage blockchain(a decentralized ledger technology) to address some of the existing limitations. A small number of blockchain-based crowdsourcing systems(BCSs) with incentive mechanisms have been proposed in the literature, but they are generally not designed with security in mind. However, achieving desirable performance is difficult owing to the irregular, nonstationary, nonlinear, and noisy nature of the observed data. Therefore, a new attention-based encoderdecoder model is proposed, called empirical mode decomposition-attention-long short-term memory(EMD-Att-LSTM).EMD is a data-driven technique used for the decomposition of complex series into subsequent simpler series. It explores the inherent properties of data to obtain the components such as trend and seasonality. Neural network architecture driven by deep learning uses the idea of a fine-grained attention mechanism, that is, considering the hidden state instead of the hidden state vectors, which can help reflect the significance and contributions of each hidden state dimension. In addition, it is useful for locating and concentrating the relevant temporary data,leading to a distinctly interpretable network. To evaluate the proposed model, we use the repository dataset of Australian energy market operator(AEMO). The proposed architecture provides superior empirical results compared with other advanced models. | 8 | Synthesis,isolation,identification and characterization of new process-related impurity in isoproterenol hydrochloride by HPLC,LC/ESI-MS and NMR显示文摘One unknown impurity(Imp-Ⅱ) during the analysis of laboratory batches of isoproterenol hydrochloride was detected in the level ranging from 0.04% to 0.12% by high performance liquid chromatography with UV detection. The unknown impurity structure was proposed as 4-[2-(propan-2-ylamino)ethyl]benzene-1,2-diol(Imp-Ⅱ) using the liquid chromatography–mass spectrophotometry(LC–MS) analysis. Imp-Ⅱ was isolated by semi-preparative liquid chromatography from the impurity-enriched reaction crude sample. Its proposed structure was confirmed by nuclear magnetic spectroscopy such as ~1H,^(13)C, DEPT(1D NMR), HSQC(2D NMR)and infrared spectroscopy(IR), and retention time and purity with HPLC followed by the chemical synthesis.Due to less removable nature of Imp-Ⅱ during the purification, the synthetic process was optimized proficiently to control the formation of Imp-Ⅱ below to the limit < 0.12% in the course of reaction.
