| 4 | A Blockchain-Based Flexible Data Auditing Scheme for the Cloud Service显示文摘Nowdays, cloud storage technology has become a hot topic, and an increasing number of users are concerned with the security of their data in the cloud.Many auditing schemes on the cloud are proposed and the introduction of a third-party auditor to assist users in verifying the integrity of cloud data. As a centralized node, the third-party auditor has to communicate with all cloud users and cloud service providers, which becomes the bottleneck of the whole scheme. To solve this problem,we design a blockchain-based flexible cloud data auditing scheme. In our scheme, a decentralized auditing framework is proposed to eliminate the dependency on the thirdparty auditor, which increases the stability, security and performance of the whole scheme. Since the cloud service provider can automatically generates auditing proofs, our scheme can relieve the communication burdens of the cloud service provider. | 5 | Methane hydrate formation and dissociation behaviors in montmorillonite显示文摘The methane hydrate formation and the methane hydrate dissociation behaviors in montmorillonite are experimentally studied. Through the analyses of the microstructure characteristic, the study obtains the porous characteristic of montmorillonite. It is indicated that methane hydrate in montmorillonite forms the structure I (si) crystal. Meanwhile, molecular dynamics simulation is carried out to study the processes of the methane hydrate formation and the methane hydrate dissociation in montmorillonite. The microstructure and microscopic properties are analyzed. The methane hydrate formation and methane hydrate dissociation mechanisms in the montmorillonite nanopore and on the montmorillonite surface are expounded. Combining the experimental and simulating analyses, the results indicate the methane hydrate formation and methane hydrate dissociation processes have little influence upon the crystal structure of porous media from either micro- or macro-analysis. | 9 | Junction plakoglobin regulates and destabilizes HIF2α to inhibit tumorigenesis of renal cell carcinoma显示文摘Background:Increased hypoxia-inducible factor 2α(HIF2α)activation is a common event in clear cell renal cell carcinoma(ccRCC)progression.However,the function and underlying mechanism of HIF2α in ccRCC remains uninvestigated.We conducted this study to access the potential link between junction plakoglobin(JUP)and HIF2αin ccRCC.Methods:Affinity purification and mass spectrometry(AP-MS)screening,glutathione-s-transferase(GST)pull-down and co-immunoprecipitation(Co-IP)assays were performed to detect the interacting proteins of HIF2α.Quantitative PCR(qPCR)and Western blotting were used to detect the expression of JUP in human ccRCC samples.Luciferase reporter assays,chromatin immunoprecipitation(ChIP),cycloheximide chase assays,and ubiquitination assays were conducted to explore the regulation of JUP on the activity of HIF2α.Cell Counting Kit-8(CCK-8)assays,colony formation assays,transwell assays,and xenograft tumor assays were performed to investigate the effect of JUP knockdown or overexpression on the tumorigenicity of renal cancer cells.Results:We identified JUP as a novel HIF2α-binding partner and revealed an important role of JUP in recruiting von Hippel-Lindau(VHL)and histone deacetylases 1/2(HDAC1/2)to HIF2α to regulate its stability and transactivation.JUP knockdown promoted and overexpression suppressed the tumorigenicity of renal cell carcinoma in vitro and in vivo.Importantly,the low expression of JUP was found in clinical ccRCC samples and correlated with enhanced hypoxia scores and poor treatment outcomes.Conclusion:Taken together,these data support a role of JUP in modulating HIF2α signaling during ccRCC progression and identify JUP as a potential therapeutic target. | Ke Chen Jin Zeng Yi Sun Wei Ouyang Gan Yu Hui Zhou Yangjun Zhang Weimin Yao Wei Xiao Junhui Hu Jinchun Xing Kefeng Xiao Lily Wu Zhiqiang Chen Zhangqun Ye Hua Xu | 2021 | Cancer Communications2021,41,4: | 1 |
