1Variations of geochemical compositions and the paleoclimatic significance of a loess-soil sequence from Garzê County of western Sichuan Province,China显示文摘The West Sichuan Plateau is located in the southeast margin of the Tibetan Plateau, where the climate is mainly influenced by the Indian southwest summer monsoon and the Tibetan Plateau monsoon. In this study, detailed geochemical analysis has been carried out on Ganzisi loess-paleosol sequence in Ganzê County of western Sichuan Province. The results indicate that Ganzê loess and paleosol have experienced the incipient stage of chemical weathering in dust source regions, characterized by the decomposition of plagioclase which caused the depletion of mobile elements Na and Ca. The post-depositional chemical weathering is characterized by carbonate dissolution and oxidation of Fe2+. The variations of some geochemical indexes (such as CIA values, Na/K and Fe2+/ Fe3+ ratios) in Ganzisi loess-paleosol sequence indicate a gradually decreased chemical weathering intensity in the dust source regions and deposition areas since 1.15 Ma BP consistent with the general increase of global ice volume, reflecting that the arid trend since 1.15 Ma BP in the southeast Tibetan Plateau is a regional response to the global climate change. The geochemical indexes in this section also reveal an obvious drying step occurred at about 250 ka BP in this region. We interpret this drying step as a result of decreased influence of the Indian southwest summer monsoon. 2Neddylation of PTEN regulates its nuclear import and promotes tumor development显示文摘PTEN tumor suppressor opposes the PI3K/Akt signaling pathway in the cytoplasm and maintains chromosomal integrity in the nucleus.Nucleus–cytoplasm shuttling of PTEN is regulated by ubiquitylation,SUMOylation and phosphorylation,and nuclear PTEN has been proposed to exhibit tumor-suppressive functions.Here we show that PTEN is conjugated by Nedd8 under high glucose conditions,which induces PTEN nuclear import without effects on PTEN stability.PTEN neddylation is promoted by the XIAP ligase and removed by the NEDP1 deneddylase.We identify Lys197 and Lys402 as major neddylation sites on PTEN.Neddylated PTEN accumulates predominantly in the nucleus and promotes rather than suppresses cell proliferation and metabolism.The nuclear neddylated PTEN dephosphorylates the fatty acid synthase(FASN)protein,inhibits the TRIM21-mediated ubiquitylation and degradation of FASN,and then promotes de novo fatty acid synthesis.In human breast cancer tissues,neddylated PTEN correlates with tumor progression and poor prognosis.Therefore,we demonstrate a previously unidentified pool of nuclear PTEN in the Nedd8-conjugated form and an unexpected tumor-promoting role of neddylated PTEN.Ping Xie Zhiqiang Peng Yujiao Chen Hongchang Li Mengge Du Yawen Tan Xin Zhang Zhe Lu Chun-Ping Cui Cui Hua Liu Fuchu He Lingqiang Zhang 2021Cell Research2021,31,3:11
3A Preliminary Study on the Soft–Sediment Deformation Structures in the Late Quaternary Lacustrine Sediments at Tashkorgan, Northeastern Pamir, China显示文摘This study identified soft–sediment deformation structures(SSDS) of seismic origin from lacustrine sediments in the late Quaternary paleo–dammed lake at Tashkorgan, northeastern Pamir. The observed deformation structures include sand dykes, liquefied diapir and convolute structures, gravity induced SSDS, and thixotropic pillar and tabular structures. We conducted a preliminary study on the morphology, formation and trigger mechanisms of pillar and tabular structures formed by liquefaction of underlying coarse sand and thixotropy of the upper silty clay. The regional tectonic setting and distribution of lacustrine strata indicate that the most probable trigger for the SSDS in lacustrine sediments was seismic activity, with an approximate earthquake magnitude of M>6.0; the potential seismogenic fault is the southern part of the Kongur normal fault extensional system. AMS 14 C dating results indicate that the SSDS were formed by seismic events occurring between 26050±100 yr BP and 22710±80 yr BP, implying intense fault activity in this region during the late Pleistocene. 5LSECtin on tumor-associated macrophages enhances breast cancer sternness via interaction with its receptor BTN3A3显示文摘Macrophages have bee n suggested to con tribute to con structing a cancer stem cell(CSC)niche.However,whether and how macrophages regulate the activity of CSCs through juxtacrine signaling are poorly understood.Here we report LSECtin,a transmembrane protein highly expressed on tumor-associated macrophages(TAMs),enhances sternness of breast cancer cells(BCCs).We identified BTN3A3,a B7 family member with previously unknown functions as the receptor for LSECtin on BCCs responsible for stemness-promoting effect of LSECtin.In mice bearing human tumor xenografts,either macrophage-specific ablation of LSECtin or silencing of BTN3A3 in BCCs decreased CSC frequency and tumor growth.Admixture of LSECtin-positive macrophages increased the tumorigenic activity of BCCs dependent on BTN3A3.Disruption of the LSECtin-BTN3A3 axis with BTN3A3-Fc or anti-BTN3A3 mAb has a therapeutic effect on breast cancer.These findings define a juxtacrine signaling mechanism by which TAMs promote cancer stemness.Targeting this axis in the CSC niche may provide potential therapies to breast cancer.Di Liu Qian Lu Xing Wang Jun Wang Ning Lu Zefei Jiang Xiaopeng Hao Jianbin Li Jing Liu Pengbo Cao Guilin Peng Yuandong Tao Dianyuan Zhao Fuchu He Li Tang 2019Cell Research2019,29,5:8
6Magnetostratigraphic and paleoclimatic studies on the Red Earth Formation from the Chengdu Plain in Sichuan Province,China显示文摘Two red earth sections from the Chengdu Plain in Sichuan Province are dated using geomagnetic methods. The Brunhes/Matuyama (B/M) reversal boundary is recognized within the upper parts of the two sections,and the Jaramillo normal subzone occurs in the middle-lower parts of the vermiculated red soil (VRS),which indicate that the parent material for the VRS deposited in the middle Quaternary. Sedimentologic and geochemical studies suggest that the Chengdu Clay and part of the parent material for the VRS are of aeolian origin. Pollen analysis indicates that the Chengdu Plain was mainly covered by needle-leaved forest with a cool to cool temperate humid climate when the parent material for the VRS deposited. 8Nusap1 is essential for neural crest cell migration in zebrafish显示文摘Microtubules play important roles in mitotic spindle assembly and chromosome segregation to maintain normal cell cycle progression.A number of microtubule-associated proteins have been identified in epithelial and neural cell cultures;however,their physiological significance is not well characterized due to the lack of appropriate in vivo animal models.Nucleolar spindleassociated protein(NuSAP)is a microtubule-binding protein and is reported to be involved in mitosis by cell culture studies.In this report,we identified the zebrafish homologue of human NuSAP and investigated its expression profile and functions.Using in situ hybridization,we demonstrated that transcripts of zebrafish nusap1 are specifically expressed in the retina,forebrain,hindbrain and neural crest.When the in vivo expression of nusap1 was knocked down through antisense oligonucleotide morpholino technology,the morphants of nusap1 showed impaired morphogenesis in the trunk and yolk extension,implying the involvement of Nusap1 in cell migration.Mechanistic studies revealed that nusap1 morphants have an altered expression pattern of neural crest markers crestin and sox9b,but normal expression of blood vessel and notochord markers gata1 and shh.In addition,nusap1 mRNA injection caused serious apoptosis in retina and hindbrain tissue,and these phenotypes can be rescued by co-injection of morpholino against nusap1.These observations not only suggest a role for Nusap1 in connecting apoptosis with cell migration,but also provide strong evidences that Nusap1 is potentially involved in morphogenesis in vertebrates.Jing Nie Hua Wang Fuchu He Huizhe Huang 2010Protein & Cell2010,1,3:7

11Antigenically dominant proteins within the human liver mitochondrial proteome identified by monoclonal antibodies显示文摘Analysis of the mitochondrial proteome would provide valuable insight into the function of this important organelle, which plays key roles in energy metabolism, apoptosis, free radical production, thermogenesis, and calcium signaling. It could also increase our understanding about the mechanisms that promote mitochondrial disease. To identify proteins that are antigenically dominant in human liver mitochondria, we generated >240 hybridoma cell lines from native mitochondrial proteins after cell fusion, screening, and cloning. Antibodies that recognized mitochondrial proteins were identified by screening human liver cDNA expression libraries. In this study, we identified 6 major antigens that were recognized by at least 2 different monoclonal antibodies (mAbs). The proteins that were antigenically dominant were: acetyl-Coenzyme A acyltransferase 2 (mitochondrial 3-oxoacyl-Coenzyme A thiolase), aldehyde dehydrogenase 1 family member A1, carbamoyl phosphate synthetase 1, dihydrolipoamide S-acetyltransferase (E2 component of pyruvate dehydrogenase complex), enoyl coenzyme A hydratase 1, and hydroxysteroid (11-beta) dehydrogenase 1. We also determined the subcellular localizations of these enzymes within the mitochondria using immunohistocytochemistry. We believe that these well-characterized antibodies will provide a valuable resource for the Human Liver Proteome Project (HLPP), and will make studies aimed at investigating liver mitochondrial function far easier to perform in future. 12Plasma biomarker screening for liver fibrosis with the N-terminal isotope tagging strategy显示文摘A non-invasive diagnostic approach is crucial for the evaluation of severity of liver disease,treatment decisions,and assessing drug efficacy.This study evaluated plasma proteomic profiling via an N-terminal isotope tagging strategy coupled with liquid chromatography/Fourier transform ion cyclotron resonance mass spectrometry measurement to detect liver fibrosis staging.Pooled plasma from different liver fibrosis stages,which were assessed in advance by the current gold-standard of liver biopsy,was quantitatively analyzed.A total of 72 plasma proteins were found to be dysregulated during the fibrogenesis process,and this finding constituted a valuable candidate plasma biomarker bank for follow-up analysis.Validation results of fibronectin by Western blotting reconfirmed the mass-based data.Ingenuity Pathways Analysis showed four types of metabolic networks for the functional effect of liver fibrosis disease in chronic hepatitis B patients.Consequently,quantitative proteomics via the N-terminal acetyl isotope labeling technique provides an effective and useful tool for screening plasma candidate biomarkers for liver fibrosis.We quantitatively monitored the fibrogenesis process in CHB patients.We discovered many new valuable candidate biomarkers for the diagnosis of liver fibrosis and also partly identified the mechanism involved in liver fibrosis disease.These results provide a clearer understanding of liver fibrosis pathophysiology and will also hopefully lead to improvement of clinical diagnosis and treatment.LI ShuLong LIU Xin WEI Lai WANG HuiFen ZHANG JiYang WEI HanDong QIAN XiaoHong JIANG Ying HE FuChu 2011Science China(Life Sciences)2011,54,5:6
13Wet reclamation of sodium silicate used sand and biological treatment of its wastewater by Nitzschia palea显示文摘The massive amount of sodium silicate in the used sand was a pollution source,especially in the waste water from the wet reclamation of used sand.A new process of wet reclamation by biologically treating the waste water produced during the wet reclamation process of used sand was studied in the paper.In the work,the pre-cultivation of N.palea was performed firstly,and three different scrubbing solutions:(1) tap water,(2) modified medium for N.palea,and (3) filtrate of the broth treated by N.palea for 15 days,were used.The results of the primary investigation show that a de-skinning ratio of 90% is obtained when using the scrubbing solution containing modified medium for N.palea at the ratio 1:2 of sand and scrubbing solution,and the maximal concentrations of Na+ and SiO32are 1.49 g·L-1 and 0.51 g·L-1,respectively.The results of the optimal biomass,pH value decrease and Na+ and SiO32consumption indicate the optimal incubation conditions are at the irradiance of 5,000 lux and 25 oC.Using the filtrate of the broth treated by N.palea for 15 days as the scrubbing solution directly,a de-skinning ratio of 93% is the highest compared to the results of the tap water and the modified medium for N.palea.In the biological process using N.palea,less water is used and little wastewater is produced,which is advantageous to the purpose of green manufacturing and environmental protection.Wang Huafang Fan Zitian Yu Shaoqiang Liu Fuchu Li Xuejie 2012China Foundry2012,9,1:5
14A new low-cost method of reclaiming mixed foundry waste sand based on wet-thermal composite reclamation显示文摘A lot of mixed clay-resin waste sand from large-scale iron foundries is discharged every day; so mixed waste sand reclamation in low cost and high quality has a great realistic significance. In the study to investigate the possibility of reusing two types of waste foundry sands, resin bonded sand and clay bonded sand which came from a Chinese casting factory, a new low-cost reclamation method of the mixed foundry waste sand based on the wet-thermal composite reclamation was proposed. The waste resin bonded sand was first reclaimed by a thermal method and the waste clay bonded sand was reclaimed by a wet method. Then, hot thermal reclaimed sand and the dehydrated wet reclaimed sand were mixed in certain proportions so that the hot thermal reclaimed sand dried the wet reclaimed sand leaving some water. The thermal reclamation efficiency of the waste resin bonded sand was researched at different heat levels. The optimized wet reclamation process of the waste clay bonded sand was achieved by investigating the effects of wet reclamation times, sand-water ratio and pH value on the reclaimed sand characteristics. The composite reclamation cost also was calculated. The research results showed that the properties of the mixed reclaimed sand can satisfy the application requirements of foundries; in which the temperature of the thermal reclamation waste resin bonded sand needs to be about 800 oC, the number of cycles of wet reclamation waste clay bonded sand should reach four to five, the optimal sand-water ratio of wet reclamation is around 1:1.5, and the pH value should be adjusted by adding acid. 15Failure Mechanism and Stability Analysis of an Active Landslide in the Xiangjiaba Reservoir Area,Southwest China显示文摘Dam construction for hydropower development is a very important subject all around the world, especially in developing countries due to energy crises. Filling of reservoir lakes may trigger or reactivate landslides in reservoir area. Active landslides in populated districts in the Xiangjiaba Reservoir area have become a striking problem for residents, local government and construction engineers. The key objective of this study is to analyze the role of reservoir filling and fluctuation in the activation/reactivation of the landslide as well as the mechanism of landslide from microstructures of pre-sheared slip surface. A large active landslide with a total volume of 1.25 Mm^3 at the left margin of Jinsha River in Pingshan County of Sichuan Province, Southwest China, was selected as a case study. Field investigation, field monitoring and laboratory tests were carried out to find out the failure mechanism and the stability of the active landslide. The shear strength test and stability analysis confirm that the water level fluctuations have an adverse effect on slope stability. 17A Modified DRASTIC Approach to Shallow Groundwater Vulnerability in the West Lake Watershed in Hangzhou, China显示文摘The quality of shallow groundwater in the West Lake watershed was investigated from March to July 2000. Integrating with Blackland GRASS GIS system, the DRASTIC model was used to compile the groud-water vulnerability map. A land use factor was added to the DRASTIC model and the modified model (LDRASTIC) increased the accuracy of prediction from 26.9% to 51.3%. The vulnerability map showed that the lowly, moderately and highly susceptible area predicted occupied about 11.6%, 70.9% and 17.5% of the whole watershed, respectively. Compared with the observed values of nitrate and electric conductivity, the LDRASTIC index improved the Pearson correlation coefficients from -0.010 to 0.237 and 0.380 to 0.503 both the improved coefficients were significant at the 0.01 level. 18Aberrant seed development in Litchi chinensis is associated with the impaired expression of cell wall invertase genes显示文摘Cell wall invertase(CWIN)are known to play important roles in seed development.However,most reports to date have focused on a single gene family member,and have mainly investigated CWIN functions during the filling stage of seed development.In this study,we found significant lower levels of CWIN protein and activity associated with seed abortion in the Litchi chinensis cultivar“Nuomici.”We identified five litchi CWIN genes and observed that the expression of LcCWIN5 was limited to the flower tissues and decreased sharply with fruit development.Silencing of LcCWIN5 expression before 28 DAA(cell division stage)resulted in perturbed liquid endosperm development,smaller seeds,and higher seed abortion rate,while silencing after 28 DAA(filling stage)had no effect on seed development.In contrast,LcCWIN2 was mostly expressed in the funicle and seed coat,and increased with fruit development.Decreased LcCWIN2 expression and CWIN activity during early seed filling coincided with smaller seeds in the cultivar“Feizixiao.”Silencing of LcCWIN2 caused a reduction in the seed size without inducing seed abortion.We propose that CWIN activity in seed maternal tissues during cell division stage is likely due to LcCWIN5 expression,which regulates early seed development.On the other hand,CWIN activity during the filling stage is due to the expression of LcCWIN2,which may promote carbon import by creating a sucrose gradient.Comparable LcCWIN5 expression,but much lower CWIN activity,detected in the funicle of“Nuomici”is consistent with post-translational regulation.Jieqiong Zhang Zichen Wu Fuchu Hu Lian Liu Xuming Huang Jietang Zhao Huicong Wang 2018Horticulture Research2018,5,1:4

20Geochemical Characteristics of Eolian Deposits in the Chengdu Plain of Sichuan Province and the Implications for Provenance显示文摘In this study Chengdu Red Earth(CRE)from the Chengdu Plain(CP),Sichuan province,was analyzed for its elemental(major and trace elements)and isotopic(Sm-Nd)geochemistry and compared with Pleistocene loess and paleosol samples from the Chinese Loess Plateau(CLP)in Northern China.The geochemical composition of CRE is similar to north China loess,and also resembles the average UCC.This indicates that CRE,as loess deposits in Northern China,was derived from well-mixed sedimentary protoliths that have undergone numerous upper crustal recycling processes.However,obvious differences in the geochemical characteristics of CRE and the north China loess are also revealed in our results.For chemically stable elements,CRE has higher Ti,Zr,Hf and lowerΣREE,Ba contents in comparison with loess samples from the CLP.Further analysis shows that CRE has higher TiO2/Al2O3,SiO2/Al2O3,Ba/Rb and lower Ce/Yb,Eu/Yb,LaN/YbN andΣLREE/ΣHREE ratios.In Sm-Nd isotopic geochemistry,Sm and Nd content and theεNd(0)value in CRE are significantly higher than those in north China loess.The higher TiO2content in CRE coincided with a high background concentration of Ti in the Sichuan Basin and the surrounding regions.The lowerΣREE and higher Sm,Nd,εNd(0)values are related to the wide distribution of basalt in the southwest Sichuan Basin.The elemental and isotopic geochemistry of CRE indicates that eolian materials in the CP predominantly come from the Sichuan Basin and the surrounding regions,which differs from loess deposits in the CLP.TAN Yuanlong QIAO Yansong ZHAO Zhizhong WANG Yan QI Lin FU Jianli LIU Zongxiu YAO Haitao WANG Shubing JIANG Fuchu 2013Acta Geologica Sinica(English Edition)2013,87,6:3
