| 2 | Chronic Hepatitis B Infection with Low Level Viremia Correlates with the Progression of the Liver Disease显示文摘Background and Aims:Currently,insufficient clinical data are available to address whether low-level viremia(LLV)observed during antiviral treatment will adversely affect the clinical outcome or whether treatment strategies should be altered if LLV occurs.This study compared the clinical out-comes of patients with a maintained virological response(MVR)and patients who experienced LLV and their treatment strategies.Methods:A retrospective cohort of 674 patients with chronic hepatitis B virus(HBV)infection who received antiviral treatment for more than 12 months was analyzed for the development of end-stage liver disease and treatment strategies during the follow-up period.End-stage liver disease included decompensated liver cirrhosis and hepatocellular carcinoma(HCC).Results:During a median 42-month follow-up,end-stage liver disease developed more frequently in patients who experienced LLV than in those who experienced MVR(7.73%and 15.85%vs.0.77%and 5.52%at 5 and 10 years,respectively;p=0.000).The trend was consistent after propensity score matching.In the high-risk group of four HCC risk models,LLV patients had a higher risk of HCC development(p<0.05).By Cox proportional hazard model analysis,LLV was an independent risk factor for end-stage liver disease and HCC(hazard ratio[HR]=6.280,confidence interval[CI]=2.081-18.951,p=0.001;HR=5.108,CI=1.392-18.737,respectively;p=0.014).Patients achieved a lower rate of end-stage liver disease by adjusting treatment compared to continuing the original treatment once LLV occurred(p<0.05).Conclusions:LLV is an independent risk factor for end-stage liver disease and HCC,and treatment adjustments can be considered. | Qian Zhang Hong Peng Xiaoqing Liu Huimin Wang Jinjie Du Xinhua Luo Hong Ren Peng Hu | 2021 | Journal of Clinical and Translational Hepatology2021,9,6: | 25 |

| 4 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 5 | Efficiency and Inheritance of Targeted Mutagenesis in Maize Using CRISPR-Cas9显示文摘CRISPR/Cas(clustered regularly interspaced short palindromic repeats/CRISPR-associated proteins) is an adaptive immune system in bacteria and archaea to defend against invasion from foreign DNA fragments. Recently, it has been developed as a powerful targeted genome editing tool for a wide variety of species. However, its application in maize has only been tested with transiently expressed somatic cells or with a limited number of stable transgenic T0 plants. The exact efficiency and specificity of the CRISPR/Cas system in the highly complex maize genome has not been documented yet. Here we report an extensive study of the well-studied type II CRISPR-Cas9 system for targeted genome editing in maize, with the codon-optimized Cas9 protein and the short non-coding guide RNA generated through a functional maize U6 sn RNA promoter. Targeted gene mutagenesis was detected for 90 loci by maize protoplast assay, with an average cleavage efficiency of 10.67%. Stable knockout transformants for maize phytoene synthase gene(PSY1) were obtained. Mutations occurred in germ cells can be stably inherited to the next generation. Moreover, no off-target effect was detected at the computationally predicted putative off-target loci. No significant difference between the transcriptomes of the Cas9 expressed and non-expressed lines was detected. | 7 | Precise base editing of non-allelic acetolactate synthase genes confers sulfonylurea herbicide resistance in maize显示文摘Single-nucleotide polymorphisms contribute to phenotypic diversity in maize. Creation and functional annotation of point mutations has been limited by the low efficiency of conventional methods based on random mutation. An efficient tool for generating targeted single-base mutations is desirable for both functional genomics and precise genetic improvement. The objective of this study was to test the efficiency of targeted C-to-T base editing of two non-allelic acetolactate synthase(ALS) in generating sulfonylurea herbicide-resistant mutants. A CRISPR/Cas9 nickase-cytidine deaminase fused with uracil DNA glycosylase inhibitor(UGI) was employed to achieve targeted conversion of cytosine to thymine in ZmALS1 and ZmALS2. Both protoplasts and recovered mutant plants showed the activity of the cytosine base editor, with an in vivo efficiency of up to 13.8%. Transgene-free edited plants harboring a homozygous ZmALS1 mutation or a ZmALS1 and ZmALS2 double mutation were tested for their resistance at a dose of up to 15-fold the recommended limit of chlorsulfuron, a sulfonylurea herbicide widely used in agriculture. | 10 | Jerk-limited feedrate scheduling and optimization for five-axis machining using new piecewise linear programming approach显示文摘In this paper, a new computation method and an optimization algorithm are presented for feedrate scheduling of five-axis machining in compliance with both machine drive limits and process limits. Five-axis machine tool with its ability of controlling tool orientation to follow the sculptured surface contour has been widely used in modern manufacturing industry. Feedrate scheduling serving as a kernel of CNC control system plays a critical role to ensure the required machining accuracy and reliability for five-axis machining. Due to the nonlinear coupling effects of all involved drive axes and the saturation limit of servo motors, the feedrate scheduling for multi-axis machining has long been recognized and remains as a critical challenge for achieving five-axis machine tools’ full capacity and advantage. To solve the nonlinearity nature of the five-axis feedrate scheduling problems, a relaxation mathematical process is presented for relaxing both the drive motors’ physical limitations and the kinematic constraints of five-axis tool motions. Based on the primary optimization variable of feedrate, the presented method analytically linearizes the machining-related constraints, in terms of the machines’ axis velocities, axis accelerations and axis jerks. The nonlinear multi-constrained feedrate scheduling problem is transformed into a manageable linear programming problem. An optimization algorithm is presented to find the optimal feedrate scheduling solution for the five-axis machining problems. Both computer implementation and laboratorial experiment testing by actual machine cutting were conducted and presented in this paper. The experiment results demonstrate that the proposed method can effectively generate efficient feedrate scheduling for five-axis machining with constraints of the machine tool physical constraints and limits. Then using the theory of fuzzy mathematics (FUZZY) and fuzzy relationship synthesis theory, we establish the fuzzy evaluation model of comprehensive benefits of land consolidation; using analytic hierarchy process (AHP), we determine the weight of each indicator, and conduct fuzzy comprehensive evaluation from various levels. By the weighted average synthesis algorithm, we calculate the evaluation indicators and finally summarize the overall evaluation results for comprehensive evaluation. | 16 | Thermal–structure coupling analysis and multi-objective optimization of motor rotor in MSPMSM显示文摘With the high speed, the rotor of magnetically suspended permanent magnet synchronous motor(MSPMSM) suffers great thermal stress and mechanical stress resulting from the temperature rise problem caused by rotor losses, which leads to instability and inefficiency.In this paper, the mechanical–temperature field coupling analysis is conducted to analyze the relationship between the temperature field and structure, and multi-objective optimization of a rotor is performed to improve the design reliability and efficiency. Firstly, the temperature field is calculated by the 2 D finite element model of MSPMSM and the method of applying the 2 D temperature result to the 3 D finite element model of the motor rotor equivalently is proposed. Then the thermal–structure coupling analysis is processed through mathematic method and finite element method(FEM),in which the 3 D finite element model is established precisely in a way and approaches the practical operation state further. Moreover, the impact produced by the temperature and structure on the mechanical strength is analyzed in detail. Finally, the optimization mathematical model of the motor rotor is established with Sequential Quadratic Programming-NLPQL selected in the optimization scheme. The intended mutation activity was achieved by in vivo CRISPR/Cas9 machinery involving desired-target mutation of the Wx locus in the ZC01 background,abbreviated as ZC01-DTM^(wx). Triple selection was applied to segregants to obtain high genome background recovery with transgene-free wx mutations. The targeted mutation was identified, yielding six types of mutations among progeny crossed with ZC01-DTM^(wx).The amylopectin contents of the endosperm starch in mutant lines and hybrids averaged94.9%, while those of the wild-type controls were significantly(P < 0.01) lower, with an average of 76.9%. Double selection in transgene-free lines was applied using the Bar strip test and Cas9 PCR screening. The genome background recovery ratios of the lines were determined using genome-wide SNP data. That of lines used as male parents was as high as98.19% and that of lines used as female parents was as high as 86.78%. Conversion hybrids and both parental lines showed agronomic performance similar to that of their wild-type counterparts. | 18 | Fine mapping of the awn gene on chromosome 4 in rice by association and linkage analyses显示文摘Awnness is a key trait in rice domestication, yet no studies have been conducted on fine mapping or association mapping of the rice awn gene. In this study, we investigated the awnness and genotype of a core collection of 303 cultivated rice varieties and a BC5F2 segregating population of 200 individuals. Combining association and linkage analyses, we mapped the awnness related genes to chromosome 4. Primary association analysis using 24 SSR markers revealed five loci significantly associated with awnness on chromosome 4. The associated markers cover previously identified regions. Fine association mapping was conducted using another 29 markers within a 4-Mb region, covering the associated marker in34, which is close to the awn gene Awn4.1. Seven associated markers were revealed, distributed over an 870-kb region. Combining the fine association mapping and linkage analysis of awnness in the 200 BC5F2 segregating population, we finally identified a 330-kb region as the candidate region for Awn4.1. Therefore,it is very important to monitor its running state. At present,a small number of reciprocating compressors have been installed on-line monitoring and diagnosis system,most of which can only monitor a single vertical subsidence of piston rod and it can't fully represent the running state of piston rod. Therefore,a method of monitoring the vertical and horizontal displacement of piston rod axis orbit is simultaneously used. In view of the characteristics that the piston rod axis orbit is disordered and difficult to extract features,purification of the axis orbit is carried out based on harmonic wavelet and then features are extracted such as vibration energy,natural frequency and the axis orbit envelope area. After that,a nonlinear local tangent space manifold learning algorithm is used to reduce the dimension of the features and obtain sensitive features. By analyzing the practical cases,the effectiveness of the method for fault monitoring and diagnosis of reciprocating compressor piston rod assembly has been verified.
