2Overexpression of cyclooxygenase-2 in human HepG2, Bel-7402 and SMMC-7721 hepatoma cell lines and mechanism of cyclooxygenase-2 selective inhibitor celecoxib-induced cell growth inhibition and apoptosis显示文摘AIM: To investigate the cyclooxygenase-2 (COX-2)expression level in human HepG2, Bel-7402 and SMMC-7721hepatoma cell lines and the molecular mechanism of COX-2 selective inhibitor celecoxib-induced cell growth inhibition and cell apoptosis.METHODS: Hepatoma cells were cultured and treated with celecoxib. Cell in situ hybridization (ISH) and immunocytochemistry were used to detect COX-2 mRNA and protein expression. Proliferating cell nuclear antigen and phosphorylated Akt were also detected by immunocytochemistry assay. Cell growth rates were assessed by 3-(4, 5-dimethylthiazol-2-yl-2, 5-diphenyltetrazolium (MTT) bromide colorimetric assay. Celecoxibinduced cell apoptosis was measured by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) and flow cytometry (FCM). The phosphorylated Akt and activated fragments of caspase-9, caspase-3 were examined by Western blotting analysis.RESULTS: Increased COX-2 mRNA and protein expression were detected in all three hepatoma cell lines. Celecoxib could significantly inhibit cell growth and the inhibitory effect was in a dose- and time-dependent manner evidenced by MTr assays and morphological changes.The apoptotic index measured by TUNEL increased correspondingly with the increased concentration of celecoxib and the reaction time. With 50 μmol/L celecoxib treatment for 24 h, the apoptotic index of HepG2, BEL-7402and SMMC-7721 cells was 25.01±3.08%, 26.40±3.05%,and 30.60±2.89%, respectively. Western blotting analysis showed remarkable activation of caspase-9, caspase-3and dephosphorylation of Akt (Thr308). Immunocytochemistry also showed the reduction of PCNA expression and phosphorylation Akt (Thr308) after treatment with celecoxib.CONCLUSION: COX-2 mRNA and protein overexpression in HepG2, Bel-7402 and SMMC-7721 cell lines correlate with the increased cell growth rate. 3Three-dimensional-printed individualized porous implants:A new“implant-bone”interface fusion concept for large bone defect treatment显示文摘Bone defect repairs are based on bone graft fusion or replacement.Current large bone defect treatments are inadequate and lack of reliable technology.Therefore,we aimed to investigate a simple technique using three-dimensional(3D)-printed individualized porous implants without any bone grafts,osteoinductive agents,or surface biofunctionalization to treat large bone defects,and systematically study its long-term therapeutic effects and osseointegration characteristics.Twenty-six patients with large bone defects caused by tumor,infection,or trauma received treatment with individualized porous implants;among them,three typical cases underwent a detailed study.Additionally,a large segmental femur defect sheep model was used to study the osseointegration characteristics.Immediate and long-term biomechanical stability was achieved,and the animal study revealed that the bone grew into the pores with gradual remodeling,resulting in a long-term mechanically stable implant-bone complex.Advantages of 3D-printed microporous implants for the repair of bone defects included 1)that the stabilization devices were immediately designed and constructed to achieve early postoperative mobility,and 2)that osseointegration between the host bone and implants was achieved without bone grafting.Our osseointegration method,in which the“implant-bone”interface fusion concept was used instead of“bone-bone”fusion,subverts the traditional idea of osseointegration.Teng Zhang Qingguang Wei Hua Zhou Zehao Jing Xiaoguang Liu Yufeng Zheng Hong Cai Feng Wei Liang Jiang Miao Yu Yan Cheng Daoyang Fan Wenhao Zhou Xinhong Lin Huijie Leng Jian Li Xinyu Li Caimei Wang Yun Tian Zhongjun Liu 2021Bioactive Materials2021,6,11:9
4Sustainable Practice in Pavement Engineering through Value-Added Collective Recycling of Waste Plastic and Waste Tyre Rubber显示文摘Waste plastics,such as waste polyethylene terephthalate(PET)beverage bottles and waste rubber tyres are major municipal solid wastes,which may lead to various environmental problems if they are not appropriately recycled.In this study,the feasibility of collectively recycling the two types of waste into performance-increasing modifiers for asphalt pavements was analyzed.This study aimed to investigate the recycling mechanisms of waste PET-derived additives under the treatment of two amines,triethylenetetramine(TETA)and ethanolamine(EA),and characterize the performances of these additives in modifying rubberized bitumen,a bitumen modified by waste tyre rubber.To this end,infrared spectroscopy and thermal analyses were carried out on the two PET-derived additives(PET–TETA and PET–EA).In addition,infrared spectroscopy,viscosity,dynamic shear rheology,and multiple stress creep recovery tests were performed on the rubberized bitumen samples modified by the two PET-derived additives.We concluded that waste PET can be chemically upcycled into functional additives,which can increase the overall performance of the rubberized bitumen.The recycling method developed in this study not only helps alleviate the landfilling problems of both waste PET plastic and scrap tyres,but also turns these wastes into value-added new materials for building durable pavements.Xiong Xu Zhen Leng Jingting Lan Wei Wang Jiangmiao Yu Yawei Bai Anand Sreeram Jing Hu 2021Engineering2021,7,6:7
5Ecological provinces of spring phytoplankton in the Yellow Sea:species composition显示文摘Phytoplanktonic ecological provinces of the Yellow Sea(31.20°–39.23°N, 121.00°–125.16°E) is derived in terms of species composition and hydrological factors(temperature and salinity). 173 samples were collected from 40 stations from April 28 to May 18, 2014, and a total of 185 phytoplanktonic algal species belonging to 81 genera of 7phyla were identified by Uterm?hl method. Phytoplankton abundance in surface waters is concentrated in the west coast of Korean Peninsula and Korea Bay, and communities in those areas are mainly composed of diatoms and cyanobacteria with dominant species of Cylindrotheca closterium, Synechocystis pevalekii, Chroomonas acuta,Paralia sulcata, Thalassiosira pacifica and Karenia mikimotoi, etc. The first ten dominant species of the investigation area are analyzed by multidimensional scaling(MDS) and cluster analysis, then the Yellow Sea is divided into five provinces from Province I(P-I) to Province V(P-V). P-I includes the coastal areas near southern Liaodong Peninsula, with phytoplankton abundance of 35 420×10~3–36 163×10~3 cells/L and an average of 35 791×10~3 cells/L, and 99.84% of biomass is contributed by cyanobacteria. P-II is from Shandong Peninsula to Subei coastal area. Phytoplankton abundance is in a range of 2×10~3–48×10~3 cells/L with an average of 24×10~3cells/L, and 63.69% of biomass is contributed by diatoms. P-III represents the Changjiang(Yangtze River) Diluted Water. Phytoplankton abundance is 10×10~3–37×10~3 cells/L with an average of 24×10~3 cells/L, and 73.14% of biomass is contributed by diatoms. P-IV represents the area affected by the Yellow Sea Warm Current.Phytoplankton abundance ranges from 6×10~3 to 82×10~3 cells/L with an average of 28×10~3 cells/L, and 64.17% of biomass is contributed by diatoms. P-V represents the cold water mass of northern Yellow Sea. Phytoplankton abundance is in a range of 41×10~3–8 912×10~3 cells/L with an average of 1 763×10~3 cells/L, and 89.96% of biomass is contributed by diatoms. Overall, structures of phytoplankton community in spring are quite heterogeneous in different provinces. 6Application of endoscopic hemoclips for nonvariceal bleeding in the upper gastrointestinal tract显示文摘AIM:To investigate acute nonvariceal bleeding in the upper gastrointestinal(GI)tract and evaluate the effects of endoscopic hemoclipping. METHODS:Sixty-eight cases of acute nonvariceal bleeding in the upper GI tract were given endoscopic treatment with hemoclip application.Clinical data, endoscopic findings,and the effects of the therapy were evaluated. RESULTS:The 68 cases(male:female=42:26,age from 9 to 70 years,average 54.4)presented with hematemesis in 26 cases(38.2%),melena in nine cases(13.3%),and both in 33 cases(48.5%).The causes of the bleeding included gastric ulcer(29 cases),duodenal ulcer(11 cases),Dieulafoy's lesion (11 cases),Mallory-Weiss syndrome(six cases),post- operative(three cases),post-polypectomy bleeding (five cases),and post-sphincterotomy bleeding(three cases);42 cases had active bleeding.The mean number of hemoclips applied was four.Permanent hemostasis was obtained by hemoclip application in 59 cases;6 cases required emergent surgery(three cases had peptic ulcers,one had Dieulafoy's lesion,and two were caused by sphincterotomy);three patients died(two had Dieulafoy's lesion and one was caused by sphincterotomy);and one had recurrent bleeding with Dieulafoy's lesion 10 mo later,but in a different location. 7MATRIEX imaging:multiarea two-photon real-time in vivo explorer显示文摘Two-photon laser scanning microscopy has been extensively applied to study in vivo neuronal activity at cellular and subcellular resolutions in mammalian brains.However,the extent of such studies is typically confined to a single functional region of the brain.Here,we demonstrate a novel technique,termed the multiarea two-photon real-time in vivo explorer(MATRIEX),that allows the user to target multiple functional brain regions distributed within a zone of up to 12mm in diameter,each with a field of view(FOV)of ~200μm in diameter,thus performing two-photon Ca2+imaging with single-cell resolution in all of the regions simultaneously.For example,we demonstrate real-time functional imaging of single-neuron activities in the primary visual cortex,primary motor cortex and hippocampal CA1 region of mice in both anesthetized and awake states.A unique advantage of the MATRIEX technique is the configuration of multiple microscopic FOVs that are distributed in three-dimensional space over macroscopic distances(>1 mm)both laterally and axially but that are imaged by a single conventional laser scanning device.In particular,the MATRIEX technique can be effectively implemented as an add-on optical module for an existing conventional single-beam-scanning two-photon microscope without requiring any modification to the microscope itself.Thus,the MATRIEX technique can be readily applied to substantially facilitate the exploration of multiarea neuronal activity in vivo for studies of brain-wide neural circuit function with single-cell resolution.Mengke Yang Zhenqiao Zhou Jianxiong Zhang Shanshan Jia Tong Li Jiangheng Guan Xiang Liao Bing Leng Jing Lyu Kuan Zhang Min Li Yan Gong Zhiming Zhu Junan Yan Yi Zhou Jian K Liu Zsuzsanna Varga Arthur Konnerth Yuguo Tang Jinsong Gao Xiaowei Chen Hongbo Jia 2019Light(Science & Applications)2019,8,1:4
8The ethyl acetate extraction of Pileostegia tomentella(ZLTE) exerts anti-cancer effects on H1299 cells via ROS-induced canonical apoptosis显示文摘Lung cancer is the leading cause of cancer death and the most common malignant tumor, the long-term survival of which has stagnated in the past several decades.Pileostegia tomentella Hand.Mazz is a traditional Chinese medicine called 'Zhongliuteng'(ZLT) in the pharmacopeia, which has been proved to possess a potent anti-tumor effect on various cancers.In this study, the effects of ZLT N-butanol extraction(ZLTN) and ZLT ethyl acetate extraction(ZLTE) on the viability of non-small cell lung cancer cell(NSCLC) lines H1299 and A549 were evaluated.Here, we firstly reported that ZLTE significantly inhibited H1299 cells growth without affecting the release of lactate dehydrogenase(LDH).In addition, ZLTE induced caspase-dependent apoptosis in a concentration-dependent manner and increased the expression cleaved-PARP and decreased pro-caspase-3, pro-caspase-7, pro-caspase-8, and pro-caspase-9.Moreover, ZLTE increased the level of cellular reactive oxygen species(ROS) in H1299 cells to lead to apoptosis,which was reversed by N-acetyl-cysteine(NAC).Taken together, our results revealed that ZLTE induced caspase-dependent apoptosis via ROS generation, suggesting that ZLTE is a promising herbal medicine for the treatment of NSCLC.FAN Qiu-Mei ZHAO Wen-Tong YUAN Renyikun WANG Qin-Qin ZHANG Li-Feng GAO Hong-Wei LENG Jing YANG Shi-Lin 2020Chinese Journal of Natural Medicines2020,18,7:4
9Analysis of floral scent emitted from Syringa plants显示文摘Syringa is an important aromatic woody angiosperm that is widely planted in gardens. Its dry flowers are traditionally used to manufacture infusions and spices in China. In this study, the floral volatiles emitted from nine different Syringa species and varieties, viz.S. chinensis, S. protolaciniata, S. oblata, S. oblata var.giraldii, S. oblata var. plena, S. vulgaris ‘Mrs Harry Bickle', S. vulgaris ‘Bright Centennial', S. vulgaris ‘White Spires', and S. vulgaris ‘President Lincoln' were collected by the dynamic headspace technique, and then identified by automated thermal desorption–gas chromatography/mass spectrometry. There were significant differences incomponents and corresponding contents of floral scent emitted from the nine Syringa species and varieties.Among the detected components, b-ocimene exhibited the highest content, accounting for more than 70 % of the floral scent of S. protolaciniata, S. oblata var. giraldii, and S. vulgaris ‘Bright Centennial'. A significant daytime variation of floral scent emitted from S. pekinensis was recorded, especially for benzaldehyde, whose content first increased and then decreased. We studied the scent emitted from the in vitro flowers of S. vulgaris ‘Pres Lincoln'during the process of natural drying. Alcohol compounds were released in the greatest content, and the content of 2-ethyl-1-hexanol accounted for about 40 % of the total release amount. 11Screening and characterization of a novel ruminal cellulase gene(Umcel-1) from a metagenomic library of gayal(Bos frontalis)显示文摘Gayal is a rare semi-wild bovine species found in the Indo-China.They can graze grasses,including bamboo leaves,as well as reeds and other plant species,and grow to higher mature live weights than Yunnan Yellow cattle maintained in similar harsh environments.The aim of this study was to identify specific cellulase in the gayal rumen.A metagenomic fosmid library was constructed using genomic DNA isolated from the ruminal contents of four adult gayals.This library contained38400 clones with an average insert size of 35.5 kb.The Umcel-1 gene was isolated from this library.Investigation of the cellulase activity of 24 random clones led to the identification of the Umcel-1 gene,which exhibited the most potent cellulase activity.Sequencing the Umcel-1 gene revealed that it contained an open reading frame of 942 base pairs that encoded a product of 313 amino acids.The putative gene Umcel-1 product belonged to the glycosyl hydrolase family 5 and showed the highest homology to the cellulase(GenBank accession no.YP_004310852.1)from Clostridium lentocellum DSM 5427,with 44%identity and 62%similarity.The Umcel-1 gene was heterologously expressed in Escherichia coli BL21,and recombinant Umcel-1 was purified.The activity of purified recombinant Umcel-1 was assessed,and the results revealed that it hydrolyzed carboxymethyl cellulose with optimal activity at pH 5.5 and 45℃.To our knowledge,this study provides the first evidence for a cellulase produced by bacteria in gayal rumen.LI Bi-feng ZHU Ya-xin GU Zhao-bing CHEN Yuan LENG Jing GOU Xiao FENG Li LI Qing XI Dong-mei MAO Hua-ming YANG Shu-Li 2016Journal of Integrative Agriculture2016,15,4:2
1220(R)-ginsenoside Rg3,a product of high-efficiency thermal deglycosylation of ginsenoside Rd,exerts protective effects against scrotal heat-induced spermatogenic damage in mice显示文摘Heat stress(HS)reaction can lead to serious physiological dysfunction associated with cardiovascular and various organ diseases.Ginsenoside Rg3(G-Rg3)is a representative component of ginseng rare saponin and can protect against multiple organs,also used as functional food to adjust the balance of the human body,but the therapeutic effect and molecular mechanism of G-Rg3 on male diseases under HS are underexplored.The aim of the present study,G-Rg3 was prepared through the efficient conversion of ginsenoside Rd and investigate the contribution of G-Rg3 to testicular injury induced exposure to HS.All mice were divided into four groups as follows:normal group,HS group,and HS+G-Rg3(5 and 10 mg/kg)groups.G-Rg3 was administered orally for 14 days,then exposed to a single scrotal heat treatment(43°C,18min)on the 7th day.After HS treatment,the morphology of testis and epididymis changes,and caused a significant loss of multinucleated giant cells,desquamation of germ cells in destructive seminiferous tubules,and degenerative Leydig cells,further destroying the production of sperm.After administration G-Rg3(5 and 10 mg/kg/day)for 2 weeks,the spermatogenic-related indexes of testosterone levels and superoxide dismutase(SOD)activity,glutathione(GSH)content significantly(p<0.01)increase compared with the HS group.Moreover,G-Rg3 treatment effectively ameliorated the production of malondialdehyde(MDA)(p<0.05 or p<0.01).Importantly,G-Rg3 exhibited the protective potential against HS-induced injury not only suppressing the protein levels of heme oxygenase-1(HO-1),hypoxia-inducible factor-1α(HIF-1α),and heat shock protein 70(HSP70)but also modulating the Bcl-2 family(p<0.01 or p<0.001)and activation of mitogen-activated protein kinase(MAPK)signaling pathways(p<0.01).For most of the parameters tested,the HS+G-Rg3(10 mg/kg)group exhibited potent effects compared with those exhibited by the low dose(5 mg/kg)group.In conclusion,the present study demonstrated that G-Rg3 exerted protective effects against HS-induced testicular dysfunction via inhibiting the MAPK-mediated oxidative stress and apoptosis in mice.WEI LIU ZI WANG JING LENG HENG WEI SHEN REN XIAOJIE GONG CHEN CHEN YINGPING WANG RUI ZHANG WEI LI 2020BIOCELL2020,44,4:2

14A 0.19 ppm/°C bandgap reference circuit with high-PSRR显示文摘A high-order curvature-compensated CMOS bandgap reference(BGR) topology with a low temperature coefficient(TC) over a wide temperature range and a high power supply reject ratio(PSRR) is presented.High-order correction is realized by incorporating a nonlinear current INL, which is generated by ?V_(GS) across resistor into current generated by a conventional first-order current-mode BGR circuit. In order to achieve a high PSRR over a broad frequency range, a voltage pre-regulating technique is applied. The circuit was implemented in CSMC 0.5 μm 600 V BCD process. The experimental results indicate that the proposed topology achieves TC of0.19 ppm/°C over the temperature range of 165 °C(-40 to 125 °C), PSRR of-123 d B @ DC and-56 d B @ 100 k Hz. 15Cloning and expression analysis of the nuclear factor erythroid 2-related factor 2(Nrf2)gene of grass carp(Ctenopharyngodon idellus)and the dietary effect of Eucommia ulmoides on gene expression显示文摘Nuclear factor erythroid 2-related factor 2(Nrf2)is an important transcription factor in the regulation of redox homeostasis,which plays a vital role in metabolism,the cell cycle,and cell protection.In this study,the complete cDNA sequence of Nrf2 gene of grass carp was obtained by rapid amplification of cDNA ends-polymerase chain reaction(RACE-PCR),and the characteristics of Nrf2 proteins predicted from gene sequencing were analyzed by bioinformatics methods.Tissue differential expression patterns were also detected by Real-time Fluorescence quantitative PCR(qRT-PCR).Then,Eucommia ulmoides(EU),a traditional Chinese medicine,was added into diet with an inclusion of 2%to feed grass carp for 3 months to investigate the effects on the expression of Nrf2 gene in tissues.The results showed that the full-length of Nrf2 gene of grass carp is 2760 bp,encoding 593 amino acids,and the predicted Nrf2 protein is a hydrophilic and non-secretory protein.Grass carp Nrf2 gene showed higher expression in kidney,gill,liver,spleen,and blood tissues than in other tissues.In the feeding trial,dietary EU significantly promoted the Nrf2 mRNA expression in kidney,gill,and liver tissues after three months feeding.The results suggest that Nrf2 plays an important role in the anti-oxidative capacity of grass carp and its expression is enhanced by dietary EU.Junpeng Wang Jun Xie Zhihan Tu Jing Wei Wenqian Pan Jing Hu Yi Zhang Xiangjun Leng Xiaoqin Li 2018Aquaculture and Fisheries2018,3,5:2
