1The timing of tectonic transition from compression to extension in Dabieshan: Evidence from Mesozoic granites显示文摘With the methods of single grain zircon U-Pb and hornblende-biotite Ar-Ar dating, a U-Pb zircon age (135.4(2.7) Ma of Liujiawa high Sr/Y granite and 40Ar-39Ar plateau ages (139.0(1.0) Ma and (125.3(0.2) Ma of hornblende and biotite respectively of Fenliupu high Sr/Y granite in the Dabieshan high- and ultrahigh-pressure metamorphic belt are yielded. Following the emplacement of the high Sr/Y granites, extension-related mafic intrusions and late granitoids with negative Sr and Eu anomalies and isotopic ages of 105-130 Ma were emplaced. Geochemical comparison between the two types of granitoids shows that, from early to late, the alkalinities, the K2O contents and the Sr/Y ratios as well as FeO/(FeO+MgO) ratios of the rocks increase, while the abundance of Sr reduces, suggesting that as time goes on the depth of crustal melting decreases. Rheological stratification of the crust due to early Cretaceous partial melting of crustal rocks resulted in crustal extension and deep-buried rock exhumation. 2Discussion on Induan-Olenekian Boundary in Chaohu, Anhui Province, China显示文摘This paper proposes a scheme for the definition of the Lower Triassic Induan Olenekian boundary (IOB) based on investigation of sections in Chaohu, Anhui Province, China as well as data accumulated from other studies elsewhere. The conodont Neospathodus waageni is suggested as the index fossil of the boundary. 3Genetically modified CD7-targeting allogeneic CAR-T cell therapy with enhanced efficacy for relapsed/refractory CD7-positive hematological malignancies:a phase I clinical study显示文摘Chimeric antigen receptor(CAR)-T cell therapy against T cell malignancies faces major challenges including fratricide between CAR-T cells and product contamination from the blasts.Allogeneic CAR-T cells,generated from healthy donor T cells,can provide ready-to-use,blast-free therapeutic products,but their application could be complicated by graft-versus-host disease(GvHD)and host rejection.Here we developed healthy donor-derived,CD7-targeting CAR-T cells(RD13-01)with genetic modifications to resist fratricide,GvHD and allogeneic rejection,as well as to potentiate antitumor function.A phase I clinical trial(NCT04538599)was conducted with twelve patients recruited(eleven with T cell leukemia/lymphoma,and one with CD7-expressing acute myeloid leukemia).All patients achieved pre-set end points and eleven proceeded to efficacy evaluation.No dose-limiting toxicity,GvHD,immune effector cell-associated neurotoxicity or severe cytokine release syndrome(grade≥3)were observed.28 days post infusion,81.8%of patients(9/11)showed objective responses and the complete response rate was 63.6%(7/11,including the patient with AML).3 of the responding patients were bridged to allogeneic hematopoietic stem cell transplantation.With a median follow-up of 10.5 months,4 patients remained in complete remission.Cytomegalovirus(CMV)and/or Epstein-Barr virus(EBV)reactivation was observed in several patients,and one died from EBV-associated diffuse large B-cell lymphoma(DLBCL).Expansion of CD7-negative normal T cells was detected post infusion.In summary,we present the first report of a Phase I clinical trial using healthy donor-derived CD7-targeting allogeneic CAR-T cells to treat CD7^(+)hematological malignancies.Our results demonstrated the encouraging safety and efficacy profiles of the RD13-01 allogeneic CAR-T cells for CD7^(+)tumors.Yongxian Hu Yali Zhou Mingming Zhang Houli Zhao Guoqing Wei Wengang Ge Qu Cui Qitian Mu Gong Chen Lu Han Tingting Guo Jiazhen Cui Xiaoyan Jiang Xiujun Zheng Shuhui Yu Xiaolong Li Xingwang Zhang Mingxi Chen Xiuju Li Ming Gao Kang Wang Cheng Zu Hao Zhang Xiaohong He Yanbin Wang Dongrui Wang Jiangtao Ren He Huang 2022Cell Research2022,32,11:2
4Dynamic-Response Analysis of the Branch System of a Utility Tunnel Subjected to Near-Fault and Far-Field Ground Motions in Different Input Mechanisms显示文摘There are few studies on the dynamic-response mechanism of near-fault and far-field ground motions for large underground structures,especially for the branch joint of a utility tunnel(UT)and its internal pipeline.Based on the theory of a 3D viscous-spring artificial boundary,this paper deduced the equivalent nodal force when a P wave and an SV wave were vertically incident at the same time and transformed the ground motion into an equivalent nodal force using a self-developed MATLAB program,which was applied to an ABAQUS finite element model.Based on near-fault and far-field groundmotions obtained fromtheNGA-WEST2 database,the dynamic responses of a utility tunnel and its internal pipeline in different inputmechanisms of near-fault and far-field groundmotions were compared according to bidirectional input and tridirectional input,respectively.Generally,the damage to the utility tunnel caused by the near-fault ground motion was stronger than that caused by the far-field ground motion,and the vertical ground motion of near-fault ground motion aggravated the damage to the utility tunnel.In addition,the joint dislocation of the upper and lower three-way joints of the pipeline in the branch systemunder the seismic action led to local stress concentrations.In general,the branch system of the utility tunnel had good seismic performance to resist the designed earthquake action and protect the internal pipeline fromdamage during the rare earthquake.Yuhang Jin Endong Guo Houli Wu Peilei Yan 2022Computer Modeling in Engineering & Sciences2022,,1:2

19Natural killer cell-derived extracellular vesicle significantly enhanced adoptive T cell therapy against solid tumors via versatilely immunomodulatory coordination显示文摘Insufficient tumor tropism,MHC classⅠmolecules(MHC-I)defects of tumor cells,and immunosuppressive tumor microenvironment(TME)seriously imperil the efficacy of adoptive T cell therapy on solid tumors.Here,natural killer cell-derived extracellular vesicle(Nev)is used as a versatile toolkit to synergistically improve adoptive T cell therapy for solid tumors.Specifically,Nev is modified with dibenzocyclooctynes(DBCO)linked with p H-sensitive benzoic-imine bonds;meanwhile,cytotoxic T lymphocyte(CTL)is decorated with azide groups.Then CTL is armed with Nev(CTL-Nev)through the click chemistry reaction.After systematic administration,Nev obviously promotes the tumor-targeting accumulation of CTL coming from its native tumor-tropism capability.Then,the cleavage of benzoic-imine bonds in the slightly acidic TME leads to the release of Nev,which not only directly induces tumor apoptosis but also promotes the action of CTL via multiplex pathways,such as up-regulating the MHC-I expression on tumor cells,reprogramming tumor-associated macrophages from pro-tumoral M2 phenotypes to tumoricidal M1 phenotypes.The all-around coordination of Nev with CTL results in potent tumor repression.Weidong Nie Wenlin Fan Anqi Jiang Guanghao Wu Houli Liu Li-Li Huang Hai-Yan Xie 2021Science China Chemistry2021,64,11:1
