4Upreguiation of human telomerase reverse transcriptase mRNA expression by in vitro transfection of hepatitis B virus X gene into human hepatocarcinoma and cholangiocarcinoma cells显示文摘AIM: To study the changes of human telomerase reverse transcriptase (hTERT) mRNA expression in human hepatocarcinoma cell lines (HepG2) and cholangiocarcinoma cell lines (QBC939) after HBx gene transfection and to illustrate the significance of transcriptional regulation of hTERT gene by HBx gene in the carcinogenesis.METHODS: HepG2 and QBC939 cell lines were cultured and co-transfected with eukaryotic expression vector containing the HBx coding region and cloning vector containing enhanced green fluorescent protein (EGFP) coding sequence using lipid-mediated gene transduction technique. Thirty-six hours after transfection, EGFP expression in cells was used as the indicator of successful transfection. Flow cytometry was performed to determine the transfection efficiency.Cells were harvested and total RNA was extracted using TRIzol() reagent. The expression of hTERT mRNA in HepG2and QBC939 cell lines was assayed by reverse transcriptionpolymerase chain reaction. The expression of HBx protein in both cell lines was detected by immunocytochemical staining and Western blotting.RESULTS: Flow cytometry showed that the transfection efficiency was 46.4% in HepG2 cells and 29.6% in QBC939cells for both HBx gene expression vector and blank vector. The expression of hTERT mRNA was meaningfully increased in HepG2 and QBC939 cell lines when transfected with HBx gene expression vector compared to those transfected with OPTI-MEM medium and blank vector.Immunocytochemical staining and Western blotting revealed HBx protein expression in HepG2 and QBC939cells only when transfected with HBx gene.CONCLUSION: HBx gene transfection can upregulate the transcriptional expression of hTERT mRNA. 5Effect of Combining Acupuncture and Functional Training on Post-stroke Functional Impairment of Hand显示文摘Objective:To observe the clinical effect of combining acupuncture and functional training on post-stroke functional impairment of hand.Methods:A total of 165 cases who met the inclusion criteria were randomized into a treatment group(85 cases)and a control group(80cases).Cases in the treatment group were treated with routine acupuncture points plus Shixuan(EX-UE 11)and Xiaohai(SI 8),whereas cases in the control group were treated with routine acupuncture points alone.After acupuncture,cases in both groups conducted functional training in upper limbs.The finger grip strength and hand function were assessed before and after 3-course treatment.Results:Cases in the treatment group obtained better finger grip strength and hand function than those in the control group(both P<0.05).Conclusion:In addition to functional training,adding Shixuan(EX-UE 11)and Xiaohai(SI 8)to traditional acupuncture formulae works better for post-stroke functional impairment of hand.Ni Huan-huan Cui Xiao Hu Yong-shan Wu Yi Huang De-quan Qu Pei-yu Wang Jun Wu Ji Shi Jun-chao Han Chou-ping 2013Journal of Acupuncture and Tuina Science2013,11,6:16
6Development and application of mechanized maize harvesters显示文摘Maize harvesting is one of the most important filed operations of maize production.As the accelerating development of maize industry,mechanized maize harvesting is widely accepted and used by farmers in the world.According to the harvesting methods,maize harvesters could be classified into two types,one is maize-for-grain harvesters,including pickers and grain harvesters,the other is whole plant harvesters,including forage harvesters and combined grain-stover harvesters.Structure characteristics,appropriate areas and relative technologies of those harvesters are described in this paper,i.e.,pickers are suitable for multi-crop areas,Grain harvesters are mainly for one-crop areas when grain moisture content is lower than 25%,combined grain-stover harvesters are applicable in agro-pastoral ecotone,and forage harvesters are capable in large-scaled animal husbandry farming areas where large amount of silage are required.Meanwhile,the future development trend of the different harvesters is predicted.Big horsepower engines,enhanced working efficiency,automation and intelligence are required by large scaled farms.As maize breeding development,more varieties that are suitable for grain harvesting are put into practice,so grain combine harvesters will be popular in future.Yang Li Cui Tao Qu Zhe Li Kehong Yin Xiaowei Han Dandan Yan Bingxin Zhao Dongyue Zhang Dongxing 2016International Journal of Agricultural and Biological Engineering2016,9,3:13
7Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. 8Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 9The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 10A 3-year clinical trial of lamivudine in treatment of patients with chronic hepatitis B显示文摘BACKGROUND: Lamivudine was approved for the treat- ment of chronic hepatitis B in China in 1999; however the long-term result has not yet been reported in detail. This clinical trial was to evaluate the long-term efficacy and safe- ty of 3-year lamivudine treatment for chronic hepatitis B and the impact of emergence of YMDD mutation of hepa- titis B virus (HBV). METHODS: This multi-center, randomized, double-blind, placebo controlled trial began from 1996 to 1999. A total of 429 patients with serum HBsAg, HBeAg and HBV DNA positive were randomized to receive either lamivudine 100 mg daily (322 patients) or placebo (107) for the first 12 weeks. All patients were given subsequently open labelled lamivudine 100 mg/d for a total of 156 weeks. RESULTS: After 12-week lamivudine therapy, the levels of serum HBV DNA decreased rapidly. The negativity of HBV DNA (<1.6 pg/ml) at week 12 was 92.2% in the lamivudine group, whereas it was only 14.1% in the place- bo group (P<0.01). After 1-year lamivudine treatment, 72.7% of the patients showed undetectable serum HBV DNA (<1.6 pg/ml). At the end of 3 years, serum HBV DNA continued to be substantially suppressed with a me- dian level below a detectable level in patients with non-YM- DD variant HBV, which was increased to 86 mEq/ml (bD- NA method, equivalent hybridization method 10 pg/ml) in patients with YMDD mutation. At the end of 1, 2 and 3 years, the rates of HBeAg loss were 9.5%, 16.8% and 20.0% respectively and the rates of HBeAg/anti-HBe sero- conversion were 8.3%, 11.5% and 17.3%. The rates of HBeAg loss and seroconversion were correlated with the baseline level of ALT. In patients with a baseline level of alanine transaminase (ALT)>2 × upper limit of normal (ULN) and ALT >5×ULN, the rates of HBeAg loss were 42.2% and 66.7%, and the rates of seroconversion were34.4% and 61.1% respectively (P<0.01) at the end of year 3. The levels of ALT at year 3 remained normal in 58.8% of patients whose baseline level of ALT was elevated, and in 79.1% of patients whose level of ALT was normal before treatment. YMDD mutations occurred in 12.1%, 49.7% and 70.5% of patients respectively at year 1, 2 and 3. In pa- tients with YMDD mutation, the levels of HBV DNA were increased slightly with mild to moderate elevation of ALT level. HBeAg loss and seroconversion were 20.0% and 15.1% in patients with YMDD mutation at the end of year 3, which were lower than those in non-variant patients (P<0.01). Adverse drug reactions or events varied gene- rally from mild to moderate. In 2 patients serious adverse events (fatigue and abdominal distension) were related to medication. ALT flares (ALT>5×ULN) occurred in 17 patients: 10 were YMDD mutants and 7 were non-mutants; all of them were relieved. No death occurred in the period of 3 years. 11Conceptual design and aerodynamic evaluation of hypersonic airplane with double flanking air inlets显示文摘To aim at design requirements of high lift-to-drag ratio as well as high volumetric efficiency of next generation hypersonic airplanes,a body-wing-blending configuration with double flanking air inlets layout is presented.Moreover,a novel forebody design methodology which by rotating and assembling two waverider-based surfaces is firstly introduced in this paper.Some typical configurations are designed and their aerodynamic performances are evaluated by computational fluid dynamics.The results for forebodies analysis show that large volumetric efficiency,high lift-to-drag ratio,and uniformly distributed flowfield at the inlet cross section can be assured simultaneously.Furthermore,results of numerical simulation of four integrated configurations with various leading edge shapes,including three power-law curves and a cosine curve clearly show the advantage of high lift-to-drag ratio.Besides,the high pressure generated by the side wall of the airframe can be partly captured by the reasonably designed wings in the condition of small flight attack angle.Then the order of lift-to-drag ratio of four configurations at 0 degree flight attack angle is completely different from the condition of 4-degree flight attack angle.This result demonstrates that the curve shape of the leading edge is very important for the lift-to-drag ratio of the aircraft,and it should be further optimized under the cruising attack angle in future work.CUI Kai HU ShouChao LI GuangLi QU ZhiPeng SITU Ming 2013Science China(Technological Sciences)2013,56,8:10

14Long interspersed nuclear element ORF-1 protein promotes proliferation and resistance to chemotherapy in hepatocellular carcinoma显示文摘AIM:To clarify the specific roles and mechanisms of long interspersed nuclear element-1 ORF-1 protein [human long interspersed nuclear element-1(LINE-1),ORF-1p] in chemotherapeutic drug resistance and cell proliferation regulation in hepatocellular carcinoma(HCC) cells.METHODS:MTT assays were performed to identify the effect of the chemotherapeutic drug toxicity on HepG2 cells.Cell proliferation inhibition and the IC 50 were calculated by the Origin 8.0 software.Western blotting assays were performed to investigate whether LINE-1 ORF-1p modulates the expression of some important genes,including p53,p27,p15,Bcl-2,mdr,and p-gp.To corroborate the proliferation and anchor-independent growth results,the HepG2 cells were analyzed by flow cytometry to investigate the effect of LINE-1 ORF1p on the apoptosis regulation.RESULTS:LINE-1 ORF-1p contributed to the resistance to several chemotherapeutic drugs(cisplatin and epirubicin) in HepG2 cells.The IC 50 of the epirubicin and cisplatin increased from 36.04 nmol/L to 59.11 nmol/L or from 37.94 nmol/L to 119.32 nmol/L.Repression of LINE-1 ORF-1p expression by the siRNA could markedly enhance the response of HepG2 cells to the epirubicin and cisplatin.The IC 50 correspondingly decreased from 28.06 nmol/L to 3.83 nmol/L or from 32.04 nmol/L to 2.89 nmol/L.Interestingly,down-regulation of LINE-1 ORF-1p level by siRNA could promote the response of HepG2 cells to the paclitaxel.The IC 50 decreased from 35.90 nmol/L to 7.36 nmol/L.However,overexpression of LINE-1 ORF-1p did not modulate the paclitaxel toxicity in HepG2 cells.Further Western blotting revealed that LINE-1 ORF-1p enhanced mdr and p-gp gene expression.As a protein arrested in the nucleus,LINE-1 ORF-1p may function through modulating transcriptional activity of some important transcription factors.Indeed,LINE-1 ORF-1p promoted HepG2 cell proliferation,anchor-independent growth and protected the cells against apoptosis through modulating the expression of p15,p21,p53,and Bcl-2 genes.CONCLUSION:LINE-1 ORF-1p promotes HepG2 cell proliferation and plays an important role in the resistance of chemotherapeutic drugs.By establishing novel roles and defining the mechanisms of LINE-1 ORF1p in HCC chemotherapeutic drug resistance and cell proliferation regulation,this study indicates that LINE-1 ORF-1p is a potential target for overcoming HCC chemotherapeutic resistance.Fan Feng Yin-Ying Lu Fan Zhang Xu-Dong Gao Chuan-Fu Zhang Alex Meredith Zhong-Xian Xu Yu-Tao Yang Xiu-Juan Chang Hong Wang Jian-Hui Qu Zhen Zeng Jun-Lan Yang Chun-Ping Wang Yun-Feng Zhu Jia-Jun Cui Yong-Ping Yang 2013World Journal of Gastroenterology2013,19,7:8

17Flame-retardant concentrated electrolyte enabling a Li F-rich solid electrolyte interface to improve cycle performance of wide-temperature lithium–sulfur batteries显示文摘Lithium–sulfur batteries have been regarded as the most promising high-energy electrochemical energy storage device owing to the high energy density, low cost and environmental friendliness. However, traditional lithium–sulfur batteries using ether-based electrolytes often suffer from severe safety risks(i.e. combustion). Herein, we demonstrated a novel kind of flame-retardant concentrated electrolyte(6.5 M lithium bis(trifluoromethylsulphonyl)imide/fluoroethylene carbonate) for highly-safe and widetemperature lithium–sulfur batteries. It was found that such concentrated electrolyte showed superior flame retardancy, high lithium-ion transference number(0.69) and steady lithium plating/stripping behavior(2.5 m Ah cm^(-2) over 3000 h). Moreover, lithium–sulfur batteries using this flame-retardant concentrated electrolyte delivered outstanding cycle performance in a wide range of temperatures(-10 °C, 25 °C and 90 °C). This superior battery performance is mainly attributed to the LiF-rich solid electrolyte interphase formed on lithium metal anode, which can effectively suppress the continuous growth of lithium dendrites. 18Carbon and nitrogen isotopes analysis and sources of organic matter in surface sediments from the Sanggou Bay and its adjacent areas, China显示文摘Naturally existing stable carbon and nitrogen isotopes are important in the study of sedimentary organic matter sources. To identify the sources of sedimentary organic matter in Sanggou Bay and its adjacent areas, which is characterized by high-density shellfish and seaweed aquaculture, the grain size, organic carbon(OC), total nitrogen(TN), carbon and nitrogen isotopic composition(δ13C and δ15N) of organic matter in the surface sediment were determined. The results showed that, in August, sedimentary OC and TN ranged from 0.17% to 0.76% and 0.04% to 0.14%, respectively. In November, OC and TN ranged from 0.23% to 0.87% and 0.05% to 0.14%, respectively. There was a significant positive correlation between OC and TN(R=0.98, P<0.0001), indicating that OC and TN were homologous. In August, the δ13C and δ15N of organic matter varied from-23.06‰ to-21.59‰ and 5.10‰ to 6.31‰, respectively. In November, δ13C and δ15N ranged from-22.87‰ to-21.34‰ and 5.13‰ to 7.31‰, respectively. This study found that the major sources of sedimentary organic matter were marine shellfish biodeposition, seaweed farming, and soil organic matter. Using a three-end-member mixed model, we estimated that the dominant source of sedimentary organic matter was shellfish biodeposition, with an average contribution rate of 65.53% in August and 43.00% in November. 19In vivo evaluation of urokinase-loaded hollow nanogels for sonothrombolysis on suture embolization-induced acute ischemic stroke rat model显示文摘The urokinase-type plasminogen activator(uPA)loaded hollow nanogels(nUK)were synthesized by a one-step reaction of glycol chitosan and aldehyde capped poly(ethylene oxide).The resultant formulation is sensitive to diagnostic ultrasound(US)of 2 MHz.Herein,we evaluated the in vivo sonothrombolysis performance of the nUK on acute ischemic stroke rat model which was established by suture embolization of middle cerebral artery(MCA).Via intravenous(i.v.)administration,the experimental data prove a controlled release of the therapeutic protein around the clots under ultrasound stimulation,leading to enhanced thrombolysis efficiency of the nUK,evidenced from smaller infarct volume and better clinical scores when compared to the i.v.dose of free uPA no matter with or without US intervention.Meanwhile,the preservation ability of the nanogels not only prolonged the circulation duration of the protein,but also resulted in the better blood-brain barrier protection of the nUK formulation,showing no increased risk on the hemorrhagic transformation than the controls.This work suggests that the nUK is a safe sonothrombolytic formulation for the treatment of acute ischemic stroke.Yuming Teng Haiqiang Jin Ding Nan Mengnan Li Chenghe Fan Yuanyuan Liu Pu Lv Wei Cui Yongan Sun Hongjun Hao Xiaozhong Qu Zhenzhong Yang Yining Huang 2018Bioactive Materials2018,3,1:7
20Proteomic and bioinformatic analyses of possible targetrelated proteins of gambogic acid in human breast carcinoma MDA-MB-231 cells显示文摘Gambogic acid(GA) is an anticancer agent in phase Ⅱb clinical trial in China but its mechanism of action has not been fully clarified. The present study was designed to search the possible target-related proteins of GA in cancer cells using proteomic method and establish possible network using bioinformatic analysis. Cytotoxicity and anti-migration effects of GA in MDA-MB-231 cells were checked using MTT assay, flow cytometry, wound migration assay, and chamber migration assay. Possible target-related proteins of GA at early(3 h) and late stage(24 h) of treatment were searched using a proteomic technology, two-dimensional electrophoresis(2-DE). The possible network of GA was established using bioinformatic analysis. The intracellular expression levels of vimentin, keratin 18, and calumenin were determined using Western blotting. GA inhibited cell proliferation and induced cell cycle arrest at G2/M phase and apoptosis in MDA-MB-231 cells. Additionally, GA exhibited anti-migration effects at non-toxic doses. In 2-DE analysis, totally 23 possible GA targeted proteins were found, including those with functions in cytoskeleton and transport, regulation of redox state, metabolism, ubiquitin-proteasome system, transcription and translation, protein transport and modification, and cytokine. Network analysis of these proteins suggested that cytoskeleton-related proteins might play important roles in the effects of GA. Results of Western blotting confirmed the cleavage of vimentin, increase in keratin 18, and decrease in calumenin levels in GA-treated cells.
