| 2 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 3 | Integrated transcriptomic and metabolomic analyses of a wax deficient citrus mutant exhibiting jasmonic acid-mediated defense against fungal pathogens显示文摘Naturally,resistant crop germplasms are important resources for managing the issues of agricultural product safety and environment deterioration.We found a spontaneous mutant of‘Newhall’navel orange(Citrus sinensis Osbeck)(MT)with broad-spectrum protections against fungal pathogens in the orchard,postharvest-storage,and artificial inoculation conditions.To understand the defense mechanism of MT fruit,we constructed a genome-scale metabolic network that integrated metabolome and transcriptome datasets.The coordinated transcriptomic and metabolic data were enriched in two sub-networks,showing the decrease in very long chain fatty acid(by 41.53%)and cuticular wax synthesis(by 81.34%),and increase in the synthesis of jasmonic acid(JA)(by 95.23%)and JA-induced metabolites such as 5-dimethylnobietin(by 28.37%)in MT.Furthermore,cytological and biochemical analyses confirmed that the response to fungal infection in MT was independent of wax deficiency and was correlated with the levels of jasmonates,and the expression of plant defensin gene PDF1.2.Results of exogenous application of MeJA and JA inhibitors such as propyl gallate proved that JA-mediated defense contributes to the strong tolerance against pathogens in MT.Our results indicated that jasmonate biosynthesis and signaling are stimulated by the fatty acid redirection of MT,and participate in the tolerance of pathogenic fungi. | Yizhong He Jingwen Han Runsheng Liu Yuduan Ding Jinqiu Wang Li Sun Xiaoming Yang Yunliu Zeng Weiwei Wen Juan Xu Hongming Zhang Xiang Yan Zhaoxing Chen Zuliang Gu Hong Chen Huanqing Tang Xiuxin Deng Yunjiang Cheng | 2018 | Horticulture Research2018,5,1: | 8 |

| 8 | Detection of Citrus yellow vein clearing virus by Quantitative Real-time RT-PCR显示文摘To develop a rapid and reliable detection method for Citrus yellow vein clearing virus(CYVCV), a quantitative real-time reverse transcriptionpolymerase chain reaction(q RT-PCR) system based on SYBR Green I was established by using a pair of specific primers designed from its conserved coat protein gene. The sensitivity, specificity, and applicability of the system were evaluated accordingly. The results showed that amplicons were produced from CYVCV isolates, whereas no amplicons from non-CYVCV citrus virus samples, including Citrus tristeza virus(CTV) and Citrus tatter leaf virus(CTLV), were obtained. The sensitivity of the q RT-PCR was 100-fold higher than that of conventional RT-PCR. An excellent linear correlation(R2= 0.999) was obtained from two standard curves of c RNA, and the amplification efficiency was 102%. | 11 | Holocene Dune Mobility and Forcing Mechanisms at the Northern Margin of the East Asian Monsoon显示文摘Dune fields at the northern margin of the East Asian monsoon (EAM), are mosaics of mobile and vegetation-stabilized aeolian dunes. These sand dunes are highly sensitive to environmental change, thus the distribution and the timing of their development may provide important clues to past environmental dynamics. Due to the strong wind erosion and dune migration, long and continuous stratigraphic records are seldom preserved. Synthesizing a large body of events, ultimately producing a relatively complete and high-resolution record, may be a proper method to investigate the dune development history and climate change. In this study, we synthesized a large body of luminescence ages for aeolian deposits from the Mu Us, Otindag, Horqin dune fields at the northern margin of the EAM. The results show that these dune fields, as a whole experienced a most extensive mobility during the early Holocene, followed by a widespread shift toward limited mobility and soil development in the mid-Holocene, and widespread reactivation occurred during late Holocene. The dune developments are directly linked to the effective moisture change controlled by the EAM changes, which respond to the low latitude summer insolation variation. The increased subsidence at the margin contrary to the core EAM, the delay from the feedback of the soil-vegetation-air coupled system, the increased evaporation due to the high temperature all play partial role in the lag of the margin EAM effective moisture change to the low latitude summer insolation. The asynchronous end of the wetter mid-Holocene mainly responds to the southeastwardly shift of the precipitation belt, while the regional sensitivity, response speed and internal feedback also contributed. | 17 | Geochronology and Petrogenesis of Mafic-Intermediate Intrusions on the Northern Margin of the Central Tianshan(NW China): Implications for Tectonic Evolution显示文摘Late Paleozoic mafic-intermediate intrusions are widespread in the central Tianshan belt, but their tectonic settings remain controversial. Petrogenesis of these intrusions can provide insights into the tectonic evolution of the belt and its adjacent areas. This study presents new whole-rock geochemical and zircon U-Pb geochronology and Hf isotopic data for the Yaxi diorite and Qianzishan gabbro on the northern margin of the central Tianshan(NW China). Zircon U-Pb dating yielded the magma crystallization ages of 313±4 and 295±4 Ma for the Yaxi diorite and Qianzishan gabbro, respectively. They have lower Mg# values, Ni and Cr contents than typical mantle-derived primary melt, with negative correlations between MgO, TFeO and SiO2 contents, indicating clinopyroxene and olivine fractionation during magma evolution. They are characterized by enrichment of large ion lithophile elements(e.g., Rb, Ba and Sr) and depletion of high field strength elements(e.g., Nb, Ta and Ti) with high Ba/Th and Rb/Y, suggesting that their mantle sources had been metasomatized by slab-derived fluids. In addition, the Qianzishan gabbro has high Al2 O3 contents(19.54 wt.%–20.88 wt.%) and positive Eu anomalies(Eu/Eu*=1.09–1.42), which can be attributed to accumulation of plagioclase. Geochemical and zircon Hf isotopic compositions reveal that both the Yaxi diorite and Qianzishan gabbro were derived from depleted lithospheric mantle in the spinel stability field with insignificant crustal contamination. During Plasma-MIG hybrid arc welding process, the interface behavior of copper and steel dissimilar alloy is investigated. The results show that electromagnetic stirring effect decreases and the heat input increases with the increase of outer plasma current in the hybrid arc welding process. The interface diffusion and interface thickness is controlled by the tradeoff of electromagnetic stirring effect degradation and heat input increase. | 20 | Cagelike mesoporous silica encapsulated with microcapsules for immobilized laccase and 2,4-DCP degradation显示文摘In this study,cage-like mesoporous silica was used as the carrier to immobilize laccase by a physical approach,followed by encapsulating with chitosan/alginate microcapsule membranes to form microcapsules of immobilized laccase based on layer-by-layer technology.The relationship between laccase activity recovery/leakage rate and the coating thickness was simultaneously investigated.Because the microcapsule layers have a substantial network of pores,they act as semipermeable membranes,while the laccase immobilized inside the microcapsules acts as a processing plant for degradation of2,4-dichlorophenol.The microcapsules of immobilized laccase were able to degrade 2,4-dichlorophenol within a wide range of 2,4-dichlorophenol concentration,temperature and p H,with mean degradation rate around 62%.Under the optimal conditions,the thermal stability and reusability of immobilized laccase were shown to be improved significantly,as the removal rate and degradation rate remained over 40.2% and 33.8% respectively after 6 cycles of operation.Using mass spectrometry(MS) and nuclear magnetic resonance(NMR),diisobutyl phthalate and dibutyl phthalate were identified as the products of 2,4-dichlorophenol degradation by the microcapsules of immobilized laccase and laccase immobilized by a physical approach,respectively,further demonstrating the degradation mechanism of 2,4-dichlorophenol by microcapsule-immobilized laccase. | Junya Yang Yan Huang Yuxiang Yang Hongming Yuan Xiangnong Liu | 2015 | Journal of Environmental Sciences2015,27,12: | 2 |
