2PGE2 activates EP4 in subchondral bone osteoclasts to regulate osteoarthritis显示文摘Prostaglandin E2(PGE2), a major cyclooxygenase-2(COX-2) product, is highly secreted by the osteoblast lineage in the subchondral bone tissue of osteoarthritis(OA) patients. However, NSAIDs, including COX-2 inhibitors, have severe side effects during OA treatment. Therefore, the identification of novel drug targets of PGE2 signaling in OA progression is urgently needed. Osteoclasts play a critical role in subchondral bone homeostasis and OA-related pain. However, the mechanisms by which PGE2 regulates osteoclast function and subsequently subchondral bone homeostasis are largely unknown. Here, we show that PGE2 acts via EP4 receptors on osteoclasts during the progression of OA and OA-related pain. Our data show that while PGE2 mediates migration and osteoclastogenesis via its EP2 and EP4 receptors, tissue-specific knockout of only the EP4 receptor in osteoclasts(EP4 Lys M) reduced disease progression and osteophyte formation in a murine model of OA. Furthermore, OA-related pain was alleviated in the EP4 Lys M mice, with reduced Netrin-1 secretion and CGRP-positive sensory innervation of the subchondral bone. The expression of plateletderived growth factor-BB(PDGF-BB) was also lower in the EP4 Lys Mmice, which resulted in reduced type H blood vessel formation in subchondral bone. Importantly, we identified a novel potent EP4 antagonist, HL-43, which showed in vitro and in vivo effects consistent with those observed in the EP4 Lys Mmice. Finally, we showed that the Gαs/PI3 K/AKT/MAPK signaling pathway is downstream of EP4 activation via PGE2 in osteoclasts. 3Preparation of CdTe nanocrystal-polymer composite microspheres in aqueous solution by dispersing method显示文摘Highly fluorescent CdTe nanocrystals were synthesized in aqueous solution, and then processible CdTe nanocrystal-polymer composites were fabricated by coating the aqueous nanocrystals with copolymers of styrene and octadecyl-p-vinyl-benzyldimethylammonium chloride (SOV- DAC) directly. A dichloromethane solution of CdTe nano- crystal-polymer composites was dispersed in the aqueous solution of poly (vinyl alcohol) (PVA) generating highly fluo- rescent microspheres. 4The application of biomacromolecules to improve oral absorption by enhanced intestinal permeability:A mini-review显示文摘Oral administration has been widely regarded as the most conve nient,quick and safe approach compared to other routes of drug delivery.However,oral absorption of drugs is often limited due to rigorous environments and complex obstacles in gastrointestinal tract.Having received considerable attention,biomacromolecules have been applied for oral drug delivery to improve the bioavailability,which could be attributed to its stability and unique bioactivities,including intestinal adhesion,opening of epithelial tight junctions,inhibiting cell efflux and regulating relative protein expression.Specifically,enhancing intestinal permeability has been regarded as a promising strategy for improving bioavailability of oral drug delivery.In this review,a series of biomacromolecules and the related mechanisms of increasing intestinal permeability for enhanced oral bioavailability are comprehensively classified and elucidated.In addition,recent advances in biomacromolecules based oral delivery and related future directions are mentioned and predicted in this review article.Min Sun Hankun Hu Leming Sun Zhen Fan 2020Chinese Chemical Letters2020,31,7:3
5Textbook Outcome as a measure of surgical quality assessment and prognosis in gastric neuroendocrine carcinoma:A large multicenter sample analysis显示文摘Objective:Quality assurance is crucial for oncological surgical treatment assessment.For rare diseases,singlequality indicators are not enough.We aim to develop a comprehensive and reproducible measurement,called the'Textbook Outcome'(TO),to assess the quality of surgical treatment and prognosis of gastric neuroendocrine carcinoma(G-NEC)patients.Methods:Data from patients with primary diagnosed G-NEC included in 24 high-volume Chinese hospitals from October 2005 to September 2018 were analyzed.TO included receiving a curative resection,≥15 lymph nodes examined,no severe postoperative complications,hospital stay≤21 d,and no hospital readmission≤30 d after discharge.Hospital variation in TO was analyzed using a case mix-adjusted funnel plot.Prognostic factors of survival and risk factors for non-Textbook Outcome(non-TO)were analyzed using Cox and logistic models,respectively.Results:TO was achieved in 56.6%of 860 G-NEC patients.TO patients had better overall survival(OS),disease-free survival(DFS),and recurrence-free survival(RFS)than non-TO patients(P<0.05).Moreover,TO patients accounted for 60.3%of patients without recurrence.Multivariate Cox analysis revealed non-TO as an independent risk factor for OS,DFS,and RFS of G-NEC patients(P<0.05).Increasing TO rates were associated with improved OS for G-NEC patients,but not hospital volume.Multivariate logistic regression revealed that nonlower tumors,open surgery,and>200 mL blood loss were independent risk factors for non-TO patients(P<0.05).Conclusions:TO is strongly associated with multicenter surgical quality and prognosis for G-NEC patients.Factors predicting non-TO are identified,which may help guide strategies to optimize G-NEC outcomes.Qiyue Chen Zhongliang Ning Zhiyu Liu Yanbing Zhou Qingliang He Yantao Tian Hankun Hao Wei Lin Lixin Jiang Gang Zhao Ping Li Chaohui Zheng Changming Huang on behalf of the Study Group for Gastric Neuroendocrine Tumors 2021Chinese Journal of Cancer Research2021,33,4:2

10Temperature measurement and analysis of postharvest agricultural products associated with thermal disinfestations显示文摘Hot air and hot water treatments are practical,environmentally-friendly and non-chemical heating methods,which are widely used for postharvest insect control and quality preservation in agricultural products.Taking apple and pear as representative fruits,this study mainly analyzed influences of their thermal properties,diameter,and medium speed on the heating rates of fruits through their real-time measured temperatures at surface and center.Based on the reported thermal death kinetic models of the target codling moth,the minimum heating time was estimated to achieve 100%insect mortality.The results showed that the heating rates in fruits decreased gradually with the increasing depth from the surface to the center.With increasing heating time,the heating rate became small.The apple was heated faster than the pear.Hot water was more effective than hot air in treating fruits.Increasing hot air speed increased the heating rate but increasing water circulating speed had no clear effects on the heating rate.Based on the measured temperature-time history of the fruit center,the minimum heating time could be effectively determined for codling moth control through the estimated total equivalent thermal lethal time.The results could provide reliable validation data for the computer simulation and a scientific basis to improve the hot air and hot water treatments.Yan Rongjun Huang Zhi Li Rui Zhu Hankun Ling Bo Shaojin Wang 2013International Journal of Agricultural and Biological Engineering2013,6,2:1

12Axial compressive behavior of Moso Bamboo and its components with respect to fiber-reinforced composite structure显示文摘Bamboo is a unique fiber-reinforced bio-composite with fibers embedded into a parenchyma cell matrix.We conducted axial compression tests on bamboo blocks prepared from bottom to top,and from inner to outer portions of the culm.The apparent Young’s modulus and compressive strength of whole thickness bamboo blocks exhibited slight increases with increasing height along the culm,due to slight increases of fiber volume fraction(Vf)from 28.4 to 30.4%.Other blocks showed a significant increase in apparent Young’s modulus and strength from the inner to outer part of the culm wall,mainly owing to a sharp increase of Vf from 17.1 to 59.8%.With a decrease of fiber fraction volume there was a transition from relatively brittle behavior to very ductile behavior in bamboo blocks.Results indicated that stiffness and strength of bamboo was primarily due to fiber in compression,and ductility of bamboo was provided by the parenchyma cell matrix acting as a natural fiber-reinforced composite.Xuexia Zhang Zixuan Yu Yan Yu Hankun Wang Jinghao Li 2019Journal of Forestry Research2019,30,6:1

15The Arrangement and Size of Cellulose Microfibril Aggregates in the Cell Walls of Sclerenchyma Fibers and Parenchyma Tissue in Bamboo显示文摘Understanding the assembly and spatial arrangement of bamboo cell wall components is crucial for its optimal utilization.Bamboo cell walls consist of aggregates of cellulose microfibrils and matrix.In the present study,the size and arrangement of cellulose microfibril aggregates in the cell walls of sclerenchyma fibers and parenchyma cells in moso bamboo were investigated with NMR and FE-SEM.The NMR measurement showed that the characteristic sizes of the microfibril aggregates of fibers and parenchyma cells were approximately 25.8 nm and 18.8 nm,respectively.Furthermore,high-resolution SEM showed the size of microfibril aggregates varied little across the cell wall of sclerenchyma fiber.However,there were significant size differences between the broad and narrow lamellae both in fiber and parenchyma cells,which is thought to be closely related to the orientation of microfibrils in these layers.The microfibril aggregates in the fibers mainly appear in a random arrangement,although occasionally in a radial or tangential arrangement in individual cell.Parenchyma cells have a relatively thinner cell wall layers,in which microfibril aggregates appear in a concentric lamellar arrangement.Wenting Ren Fei Guo Minghui Liu Haocheng Xu Hankun Wang Yan Yu 2021Journal of Renewable Materials2021,9,12:0
16MOF-derived spherical Ni_(x)S_(y)/carbon with B-doping enabling high supercapacitive performance显示文摘This work uses a simple Ni-metal organic framework(Ni-MOF)to generate a uniform metal-containing carbon hybrid structure of Ni/C by in-situ pyrolysis.Then,after NaBH_(4) treatment and hydrothermal vul-canization of Ni/C,multiphase B-doped Ni_(x)S_(y) nanoparticles can be obtained and uniformly anchored in the carbon skeleton,forming a highly porous flower-shaped B-Ni_(x)S_(y)/C composite.The positive role of B doping was theoretically confirmed by Density Function Theory(DFT)calculations.The MOF-derived car-bon framework has porous,conductive,and continuous features beneficial for fast charge transfer.There are also multiple Ni-sulfide phases in B-Ni_(x)S_(y)/C,dominated by hexagonal NiS,hexagonal Ni 2 S 3,and cu-bic Ni_(3)S_(4),which give rich valance state and are expected to bring active electrochemical reactions.In addition,the boronation process by the reducing agent of NaBH_(4) is also proved beneficial to bring high capacitance,possibly due to the incorporation of more active sites by B.Therefore,the B-Ni_(x)S_(y)/C compos-ite electrode delivers a high specific capacity of 1250.4 C g^(-1) at 1 A g^(-1) and excellent rate performance.The B-Ni_(x)S_(y)/C-based asymmetric supercapacitor also shows promising prospects for future energy stor-age devices,delivering high cyclability with capacitance retention of 87.6%after 7000 cycles.This work proves the efficiency of MOF-derived carbon framework and B-doping in improving metal sulfide’s electrochemical performances.Jingjing Hu Yan Shi Li Sun Feng Xie Kaiyue Gao Yaru Qu Hankun Tan Yihe Zhang 2023Journal of Materials Science & Technology2023,,22:0
17An Equivalent Strain Based Multi-Scale Damage Model of Concrete显示文摘A multi-scale damage model of concrete is proposed based on the concept of energy equivalent strain for generic two-or three-dimensional applications.Continuum damage mechanics serves as the framework to describe the basic damage variables,namely the tensile and compressive damage.The homogenized Helmholtz free energy is introduced as the bridge to link the micro-cell and macroscopic material.The crack propagation in micro-cells is modeled,and the Helmholtz free energy in the cracked micro-structure is calculated and employed to extract the damage evolution functions in the macroscopic material.Based on the damage energy release rates and damage consistent condition,the energy equivalent strain is used to expand the uniaxial damage model to the multi-dimensional damage model.Agreements with existing experimental data that include uniaxial tensile and compressive tests,biaxial compression and biaxial peak stress envelop demonstrate the capacity of the multi-scale damage model in reproducing the typical nonlinear performances of concrete specimens.The simulation of precast laminated concrete slab further demonstrates its application to concrete structures.Shixue Liang Hankun Liu 2020Computer Modeling in Engineering & Sciences2020,,3:0
18Glucose-responsive,antioxidative HA-PBA-FA/EN106 hydrogel enhanced diabetic wound healing through modulation of FEM1b-FNIP1 axis and promoting angiogenesis显示文摘The diabetic wounds remain to be unsettled clinically,with chronic wounds characterized by drug-resistant bacterial infections,compromised angiogenesis and oxidative damage to the microenvironment.To ameliorate oxidative stress and applying antioxidant treatment in the wound site,we explore the function of folliculininteracting protein 1(FNIP1),a mitochondrial gatekeeper protein works to alter mitochondrial morphology,reduce oxidative phosphorylation and protect cells from unwarranted ROS accumulation.And our in vitro experiments showed the effects of FNIP1 in ameliorating oxidative stress and rescued impaired angiogenesis of HUVECs in high glucose environment.To realize the drug delivery and local regulation of FNIP1 in diabetic wound sites,a novel designed glucose-responsive HA-PBA-FA/EN106 hydrogel is introduced for improving diabetic wound healing.Due to the dynamic phenylboronate ester structure with a phenylboronic acid group between hyaluronic acid(HA)and phenylboronic acid(PBA),the hydrogel is able to realize a glucose-responsive release of drugs.Fulvic acid(FA)is added in the hydrogel,which not only severs as crosslinking agent but also provides antibacterial and anti-inflammatory abilities.Moreover,the release of FEM1b-FNIP1 axis inhibitor EN106 ameliorated oxidative stress and stimulated angiogenesis through FEM1b-FNIP1 axis regulation.These in vivo and in vitro results demonstrated that accelerated diabetic wounds repair with the use of the HA-PBA-FA/EN106 hydrogel,which may provide a promising strategy for chronic diabetic wound repair.Wenqian Zhang Kangkang Zha Yuan Xiong Weixian Hu Lang Chen Ze Lin Chenyan Yu Wu Zhou Faqi Cao Hankun Hu Bobin Mi Guohui Liu 2023Bioactive Materials2023,,12:0
19Calcium-chelating peptides from rabbit bone collagen:characterization,identification and mechanism elucidation显示文摘This study aimed to characterize and identify calcium-chelating peptides from rabbit bone collagen and explore the underlying chelating mechanism.Collagen peptides and calcium were extracted from rabbit bone by instant ejection steam explosion(ICSE)combined with enzymatic hydrolysis,followed by chelation reaction to prepare rabbit bone peptide-calcium chelate(RBCP-Ca).The chelating sites were further analyzed by liquid chromatography-tandem mass(LC-MS/MS)spectrometry while the chelating mechanism and binding modes were investigated.The structural characterization revealed that RBCP successfully chelated with calcium ions.Furthermore,LC-MS/MS analysis indicated that the binding sites included both acidic amino acids(Asp and Glu)and basic amino acids(Lys and Arg),Interestingly,three binding modes,namely Inter-Linking,Loop-Linking and Mono-Linking were for the first time found,while Inter-Linking mode accounted for the highest proportion(75.1%),suggesting that chelation of calcium ions frequently occurred between two peptides.Overall,this study provides a theoretical basis for the elucidation of chelation mechanism of calcium-chelating peptides.Fuhuan Yuan Yu Fu Liang Ma Hankun Zhu Yong Yu Xin Feng Yi Sun Hongjie Dai Xin Liu Zhengfang Liu Yuhao Zhang 2024Food Science and Human Wellness2024,13,3:0
