2Combined detection of serum tumor markers for differential diagnosis of solid lesions located at the pancreatic head显示文摘BACKGROUND: The differential diagnosis of solid lesions located at the pancreatic head is very important for choosing therapies and setting up surgical tactics. This study was designed to evaluate the clinical significance of combined measurement of multiple serum tumor markers and the application of the receiver-operating characteristic (ROC) curves in the differential diagnosis of solid lesions located at the pancreatic head. METHODS: The serum levels of CA19-9, CA242, CA50 and carcinoembryonic antigen (CEA) in 112 patients with carcinoma of the pancreatic head and 38 patients with focal chronic pancreatitis in the pancreatic head were measured with ELISA. The sensitivity, specificity, positive likelihood ratio (PLR) and negative likelihood ratio (NLR) of the four serum tumor markers were calculated. The ROC curves for the four serum tumor markers were constructed and the area under the curve (AUC) was calculated. RESULTS: The AUCs of CA19-9, CA242, CA50 and CEA were 0.805, 0.749, 0.738 and 0.705; the PLRs were 1.91, 3.43, 5.09 and 5.46; and the NLRs were 0.41, 0.56, 0.59 and 0.71, respectively. Combined measurements increased the diagnostic specificity, and parallel combined testing increased the diagnostic sensitivity. CONCLUSIONS: Combined measurement of serum tumor markers CA19-9, CA242, CA50 and CEA is valuable in differential diagnosis of solid lesions located at the pancreatic head, and CA19-9 has the best diagnostic ability. Combined measurements can increase the specificity ofdiagnosis. 6GPER agonist G1 suppresses neuronal apoptosis mediated by endoplasmic reticulum stress after cerebral ischemia/reperfusion injury显示文摘Studies have confirmed a strong association between activation of the endoplasmic reticulum stress pathway and cerebral ischemia/reperfusion(I/R) injury.In this study,three key proteins in the endoplasmic reticulum stress pathway(glucose-regulated protein 78,caspase-12,and C/EBP homologous protein) were selected to examine the potential mechanism of endoplasmic reticulum stress in the neuroprotective effect of G protein-coupled estrogen receptor.Female Sprague-Dawley rats received ovariectomy(OVX),and then cerebral I/R rat models(OVX+ I/R) were established by middle cerebral artery occlusion.Immediately after I/R,rat models were injected with 100 μg/kg E2(OVX + I/R +E2),or 100 μg/kg G protein-coupled estrogen receptor agonist G1(OVX + I/R + G1) in the lateral ventricle.Longa scoring was used to detect neurobehavioral changes in each group.Infarct volumes were measured by 2,3,5-triphenyltetrazolium chloride staining.Morphological changes in neurons were observed by Nissl staining.Terminal dexynucleotidyl transferase-mediated nick end-labeling staining revealed that compared with the OVX + I/R group,neurological function was remarkably improved,infarct volume was reduced,number of normal Nissl bodies was dramatically increased,and number of apoptotic neurons in the hippocampus was decreased after E2 and G1 intervention.To detect the expression and distribution of endoplasmic reticulum stress-related proteins in the endoplasmic reticulum,caspase-12 distribution and expression were detected by immunofluorescence,and mRNA and protein levels of glucose-regulated protein 78,caspase-12,and C/EBP homologous protein were determined by polymerase chain reaction and western blot assay.The results showed that compared with the OVX+ I/R group,E2 and G1 treatment obviously decreased mRNA and protein expression levels of glucose-regulated protein 78,C/EBP homologous protein,and caspase-12.However,the G protein-coupled estrogen receptor antagonist G15(OVX + I/R + E2 + G15) could eliminate the effect of E2 on cerebral I/R injury.These results confirm that E2 and G protein-coupled estrogen receptor can inhibit the expression of endoplasmic reticulum stress-related proteins and neuronal apoptosis in the hippocampus,thereby improving dysfunction caused by cerebral I/R injury.Every experimental protocol was approved by the Institutional Ethics Review Board at the First Affiliated Hospital of Shihezi University School of Medicine,China(approval No.SHZ A2017-171) on February 27,2017.Zi-Wei Han Yue-Chen Chang Ying Zhou Hang Zhang Long Chen Yang Zhang Jun-Qiang Si Li Li 2019Neural Regeneration Research2019,14,7:16

8Response-surface-based structural reliability analysis with random and interval mixed uncertainties显示文摘Traditional reliability analysis requires probability distributions of all the uncertain parameters.However,in many practical applications,the variation bounds can be only determined for the parameters with limited information.A complex hybrid reliability problem then will be caused when the random and interval variables coexist in a same structure.In this paper,by introducing the response surface technique,we develop a new hybrid reliability method to efficiently compute the interval of the failure probability of the structure due to the probability-interval hybrid uncertainty.The present method consists of a sequence of iterations.At each step,a response surface model is constructed for the limit-state function by using a quadratic polynomial and a modified axial experimental design method.An approximate hybrid reliability problem is created based on the response surface model,which is subsequently solved by an efficient decoupling approach.An updating strategy is suggested to improve the quality of the response surface and whereby ensure the reliability analysis precision.A computational procedure is then summarized for the whole iterations.Four numerical examples and also a practical application are provided to demonstrate the effectiveness of the present method.HAN Xu JIANG Chao LIU LiXin LIU Jie LONG XiangYun 2014Science China(Technological Sciences)2014,57,7:14

10Performance analysis of ghost imaging lidar in background light environment显示文摘The effect of background light on the imaging quality of three typical ghost imaging(GI) lidar systems(namely narrow pulsed GI lidar, heterodyne GI lidar, and pulse-compression GI lidar via coherent detection) is investigated. By computing the signal-to-noise ratio(SNR) of fluctuation-correlation GI, our analytical results, which are backed up by numerical simulations, demonstrate that pulse-compression GI lidar via coherent detection has the strongest capacity against background light, whereas the reconstruction quality of narrow pulsed GI lidar is the most vulnerable to background light. 11The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 12The Coix Genome Provides Insights into Panicoideae Evolution and Papery Hull Domestication显示文摘Coix is a grass crop domesticated as early as the Neolithic era.It is still widely cultivated for both highly nutritional food and medicinal use.However,the genetic study and breeding of this crop are hindered by the lack of a sequenced genome.Here,we report de novo sequencing and assembly of the 1619-Mb genome of Coix,and annotation of 75.39%repeats and 39629 protein-coding genes.Comparative genomics analysis showed that Coix is more closely related to sorghum than maize,but intriguingly only Coix and maize had a recent genome duplication event,which was not detected in sorghum.We further constructed a genetic map and mapped several important traits,especially the strength of hull.Selection of papery hull(thin:easy dehulling)from the stony hull(thick:difficult dehulling)in wild progenitors was a key step in Coix domestication.The papery hull makes seed easier to process and germinate.Anatomic and global transcriptome analysis revealed that the papery hull is a result of inhibition of cell division and wall biogenesis.We also successfully demonstrated that seed hull pressure resistance is controlled by two major quantitative trait loci(QTLs),which are associated with hull thickness and color,respectively.The two QTLs were further fine mapped within intervals of 250 kb and 146 kb,respectively.These resources provide a platform for evolutionary studies and will facilitate molecular breeding of this important crop.Chao Guo Yanan Wang Aiguo Yang Jun He Chaowen Xiao Shanhua Lv Fengming Han Yibing Yuan Yuan Yuan Xiaolong Dong Juan Guo YawenYang-Hailan Liu Ningzhi Zuo Yaxi Hu Kangxu Zhao Zhengbo Jiang Xing Wang Tingting Jiang Yaou Sherf Moju Cao Yuan Wang Zhaobo Long Tingzhao Rong Luqi Huang Shufeng Zhou 2020Molecular Plant2020,13,2:11
13Facile synthesis of ultrathin metal-organic framework nanosheets for Lewis acid catalysis显示文摘Ultrathin metalorganic framework (MOF) nanosheets are attracting great interest in catalysis due to their unique and intriguing two-dime nsional (2D) features. Although many progresses have been achieved, it is still highly desirable to develop novel strategies for controllable synthesis of the well-defined ultrathin MOF nanosheets. Herein we report a polyvinylpyrrolidone (PVP)-assisted route to synthesize the ultrathin Ni-MOF nanosheets characteristic of 1.5 nm in thickness, in which PVP is reacted with 2-aminoterephthalic acid (H2BDC-NH2) via formation of C=N bon d, followed by coord inatio n with Ni2+ io ns to form the ultrathi n MOF n anosheets. 14Super-resolution dipole orientation mapping via polarization demodulation显示文摘Fluorescence polarization microscopy(FPM)aims to detect the dipole orientation of fluorophores and to resolve structural information for labeled organelles via wide-field or confocal microscopy.Conventional FPM often suffers from the presence of a large number of molecules within the diffraction-limited volume,with averaged fluorescence polarization collected from a group of dipoles with different orientations.Here,we apply sparse deconvolution and least-squares estimation to fluorescence polarization modulation data and demonstrate a super-resolution dipole orientation mapping(SDOM)method that resolves the effective dipole orientation from a much smaller number of fluorescent molecules within a sub-diffraction focal area.We further apply this method to resolve structural details in both fixed and live cells.For the first time,we show that different borders of a dendritic spine neck exhibit a heterogeneous distribution of dipole orientation.Furthermore,we illustrate that the dipole is always perpendicular to the direction of actin filaments in mammalian kidney cells and radially distributed in the hourglass structure of the septin protein under specific labelling.The accuracy of the dipole orientation can be further mapped using the orientation uniform factor,which shows the superiority of SDOM compared with its wide-field counterpart as the number of molecules is decreased within the smaller focal area.Using the inherent feature of the orientation dipole,the SDOM technique,with its fast imaging speed(at sub-second scale),can be applied to a broad range of fluorescently labeled biological systems to simultaneously resolve the valuable dipole orientation information with super-resolution imaging.Karl Zhanghao Long Chen Xu-San Yang Miao-Yan Wang Zhen-Li Jing Hong-Bin Han Michael Q Zhang Dayong Jin Jun-Tao Gao Peng Xi 2016Light(Science & Applications)2016,5,1:11

16Combination of mild therapeutic hypothermia and adipose-derived stem cells for ischemic brain injury显示文摘Mild therapeutic hypothermia has been shown to mitigate cerebral ischemia, reduce cerebral edema, and improve the prognosis of patients with cerebral ischemia. Adipose-derived stem cell-based therapy can decrease neuronal death and infiltration of inflammatory cells, exerting a neuroprotective effect. We hypothesized that the combination of mild therapeutic hypothermia and adipose-derived stem cells would be neuroprotective for treatment of stroke. A rat model of transient middle cerebral artery occlusion was established using the nylon monofilament method. Mild therapeutic hypothermia(33°C) was induced after 2 hours of ischemia. Adipose-derived stem cells were administered through the femoral vein during reperfusion. The severity of neurological dysfunction was measured by a modified Neurological Severity Score Scaling System. The area of the infarct lesion was determined by 2,3,5-triphenyltetrazolium chloride staining. Apoptotic neurons were detected by terminal deoxynucleotidyl transferase-mediated d UTP-biotin nick end labeling(TUNEL) staining. The regeneration of microvessels and changes in the glial scar were detected by immunofluorescence staining. The inflammatory responses after ischemic brain injury were evaluated by in situ staining using markers of inflammatory cells. The expression of inflammatory cytokines was measured by reverse transcription-polymerase chain reaction. Compared with mild therapeutic hypothermia or adipose-derived stem cell treatment alone, their combination substantially improved neurological deficits and decreased infarct size. They synergistically reduced the number of TUNEL-positive cells and glial fibrillary acidic protein expression, increased vascular endothelial growth factor levels, effectively reduced inflammatory cell infiltration and down-regulated the m RNA expression of the proinflammatory cytokines interleukin-1β, tumor necrosis factor-α and interleukin-6. 17Is Chinese Medicine Injection Applicable for Treating Acute Lung Injury and Acute Respiratory Distress Syndrome?A Systematic Review and Meta-analysis of Randomized Controlled Trials显示文摘Objective To assess the efficacy and safety of Chinese medicine injection(CMI)for treating acute lung injury/acute respiratory distress syndrome(ALI/ARDS).Methods Randomized controlled trials(RCTs)were identified by searching 3 English databases and 4 Chinese databases from their inceptions until February 2019.The Cochrane Handbook was used to evaluate risk of bias in the included studies.Data analysis was conducted using RevMan 5.3.3 software.Results A total of 19 eligible RCTs involving 1,334 participants was included in this systematic review and meta-analysis.The main meta-analysis showed that CMI combined with conventional therapy(CT)was more effective than CT alone in reducing the acute physiology and chronic health evaluation(APACHE)H score[mean difference(MD):−1.74 points,95%confidence interval(CI):−2.77 to−0.71,I^2=0]and increasing the total effective rate[relative risk(RR):1.35,95%CI:1.17 to 1.56,I^2=37%].Compared with CT,CMI combined with CT showed improvements in the arterial partial pressure of oxygen(PaO2,MD:9.25 mm Hg,95%CI:0.87 to 17.63,I^2=98%)and oxygenation index[arterial partial pressure of oxygen(PaO2)/fraction of inspired oxygen(FiO2),MD:50.75 mm Hg,95%CI:35.18 to 66.31,I^2=94%].CMI plus CT was superior to CT in reducing the systemic inflammatory response syndrome(SIRS)score(MD:−0.84 points,95%CI:−1.26 to−0.42,I^2=65%),length of hospital stay(MD:−4.22 days,95%CI:−6.49 to−1.95,I^2=92%),and duration of mechanical ventilation(MD:−2.94 days,95%CI:−4.68 to−1.21,I^2=89%).Only 1 study reported adverse events.Conclusions CMI as an adjuvant therapy showed great potential benefits for the treatment of ALI/ARDS.However,we could not make a definite conclusion due to low quality of included studies and uncertain security.Future studies should focus on improving research design,especially in blindness and placebo.The reporting of adverse events was also needed.CHEN Yuan-bin LIU Qiao XIE Han YIN Shuo-miao WU Lei YU Xu-hua FAN Long LIN Lin 2020Chinese Journal of Integrative Medicine2020,26,11:9

19Simulation of Morphological Development of Soil Cracks in Yuanmou Dry-hot Valley Region, Southwest China显示文摘Soil cracking is an important process influencing water and solutes transport in the Yuanmou Dry-hot Valley region of Southwest China. Studying the morphological development of soil cracks helps to further reveal the close relationship between the soil cracking process and water movement in such semi-arid regions. Here we report regular changes on surface morphology of soil cracks with decreasing water in four different soils (Typ-Ustic Ferrisols,Ver-Ustic Ferrisols,Tru-Ustic Vertisols and Typ-Ustic Vertisols) through simulation experiments. Our results indicate the following: 1) Different soils ultimately have different development degrees of soil cracks,according to their various values of crack area density. Soil cracks in Typ-Ustic Ferrisols can only develop to the feeble degree,while those in the other three soils are capable of developing into the intensive degree,and even into the extremely intensive degree. 2) Soil crack complexity,as expressed by the value of the area-weighted mean of crack fractal dimension (AWMFRAC),is found to continuously decrease as a whole through the whole cracking process in all the studied soils. 3) Soil crack connectivity shows a uniform trend in the studied soils,that is to say,connectivity gradually increases with soil crack development.XIONG Donghong YAN Dongchun LONG Yi LU Xiaoning HAN Jianning HAN Xueqin SHI Liangtao 2010Chinese Geographical Science2010,20,2:8
20Development of a large-scale isolation chamber system for the safe and humane care of medium-sized laboratory animals harboring infectious diseases显示文摘The close phylogenetic relationship between humans and non-human primates makes non-human primates an irreplaceable model for the study of human infectious diseases.In this study,we describe the development of a large-scale automatic multi-functional isolation chamber for use with medium-sized laboratory animals carrying infectious diseases.The isolation chamber,including the transfer chain,disinfection chain,negative air pressure isolation system,animal welfare system,and the automated system,is designed to meet all biological safety standards.To create an internal chamber environment that is completely isolated from the exterior,variable frequency drive blowers are used in the air-intake and air-exhaust system,precisely controlling the filtered air flow and providing an air-barrier protection.A double door transfer port is used to transfer material between the interior of the isolation chamber and the outside.A peracetic acid sterilizer and its associated pipeline allow for complete disinfection of the isolation chamber.All of the isolation chamber parameters can be automatically controlled by a programmable computerized menu,allowing for work with different animals in different-sized cages depending on the research project.The large-scale multi-functional isolation chamber provides a useful and safe system for working with infectious medium-sized laboratory animals in high-level bio-safety laboratories.Xin PAN Jian-cheng QI Ming LONG Hao LIANG Xiao CHEN Han LI Guang-bo LI Hao ZHENG 2010Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2010,11,10:8
