| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Predicting Cadmium Safety Thresholds in Soils Based on Cadmium Uptake by Chinese Cabbage显示文摘Cadmium(Cd), a common toxic heavy metal in soil, has relatively high bioavailability, which seriously threatens agricultural products. In this study, 8 different soils with contrasting soil properties were collected from different regions in China to investigate the Cd transfer coefficient from soil to Chinese cabbage(Brassica chinensis L.) and the threshold levels of Cd in soils for production of Chinese cabbage according to the food safety standard for Cd. Exogenous Cd(0–4 mg kg^(-1)) was added to the soils and equilibrated for 2 weeks before Chinese cabbage was grown under greenhouse conditions. The influence of soil properties on the relationship between soil and cabbage Cd concentrations was investigated. The results showed that Cd concentration in the edible part of Chinese cabbage increased linearly with soil Cd concentration in 5 soils, but showed a curvilinear pattern with a plateau at the highest dose of exogenous Cd in the other 3 soils. The Cd transfer coefficient from soil to plant varied significantly among the different soils and decreased with increasing soil p H from 4.7 to 7.5. However, further increase in soil pH to > 8.0 resulted in a significant decrease in the Cd transfer coefficient. According to the measured Cd transfer coefficient and by reference to the National Food Safety Standards of China, the safety threshold of Cd concentration in soil was predicted to be between 0.12 and 1.7 mg kg^(-1) for the tested soils. The predicted threshold values were higher than the current soil quality standard for Cd in 5 soils, but lower than the standard in the other 3 soils. | 3 | Protonation induced high-T_c phases in iron-based superconductors evidenced by NMR and magnetization measurements显示文摘Chemical substitution during growth is a well-established method to manipulate electronic states of quantum materials, and leads to rich spectra of phase diagrams in cuprate and iron-based superconductors. Here we report a novel and generic strategy to achieve nonvolatile electron doping in series of(i.e.11 and 122 structures) Fe-based superconductors by ionic liquid gating induced protonation at room temperature. Accumulation of protons in bulk compounds induces superconductivity in the parent compounds, and enhances the Tclargely in some superconducting ones. Furthermore, the existence of proton in the lattice enables the first proton nuclear magnetic resonance(NMR) study to probe directly superconductivity. Using Fe S as a model system, our NMR study reveals an emergent high-Tcphase with no coherence peak which is hard to measure by NMR with other isotopes. | 5 | Dihydromyricetin promotes apoptosis, suppresses proliferation and tumor necrosis factor-α-mediated nuclear factor kappa-B activation in nasopharyngeal carcinoma CNE-2 cells显示文摘OBJECTIVE: To investigate the efficacy of dihydromyricetin(DMY) on nasopharyngeal carcinoma(NPC) cell proliferation, apoptosis and to reveal the underlying mechanism in vitro experiments.METHODS: The CNE-2 cell line was treated with different concentrations of DMY and the effects of DMY on cell viability and proliferation were evaluated using cell counting kit-8(CCK-8) assay and plate colony formation assay. Cellular apoptosis was detected by flow cytometry following Annexin V fluorescein isothiocyanate/propidine iodide staining.Nuclei morphology was observed under a fluorescence microscope following Hoechst 333258 staining. The expression of phosphorylated inhibitor of nuclear factor kappa-B kinase subunit beta(p-IKKβ), phosphorylated inhibitor of nuclear factor kappa-B kinase subunit alpha(p-IKKα), inhibitor of nuclear factor kappa-B alpha(IκB-α), nuclear factor kappa-B(NF-κB)/p65 was examined by Western blot analysis and the nuclear translocation of NF-κB/p65 was observed using a confocal laser scanning microscopy.RESULTS: DMY inhibited the proliferative capability and colony formation of NPC CNE-2 cells. Meanwhile, DMY induced apoptosis of CNE-2 cells in a dose and time-dependent manner via upregulating B-cell lymphoma-2 associated X, but downregulating B-cell lymphoma-2 and pro-caspase-3. Importantly, we found that DMY suppressed tumor necrosis factor alpha(TNF-α)-mediated NF-κB activation via inhibiting p-IKKβ, p-IKKα and blocking NF-κB subunit p65.CONCLUSION: Our experiments demonstrated that DMY had significant antiproliferative and apoptosisinducing effects on CNE-2 cells. Additionally, DMY promoted inactivation of p-IKKβ, p-IKKα,and blocked the nuclear translocation of NF-κB subunit p65. | 7 | mRNA expression of Rho GTPase-related signaling molecules during rat hippocampal development显示文摘BACKGROUND:Rho GTPase family members have been shown to participate in neurite growth by regulating the neuronal cytoskeleton.However,there are very few reports of developmental roles of signaling molecules related to Rho GTPases. OBJECTIVE:To investigate messenger ribonucleic acid mRNA expression of signaling molecules associated with Rho GTPases,including Rho-A,Rac-1,collapsin response mediator protein 1(CRMP-1),and tubulin(53(Tubβ3) during rat hippocampus development. DESIGN,TIME AND SETTING:A non-randomized,controlled,animal experiment,based on different developmental stages of the rat hippocampus,was performed at the Guangdong Key Laboratory of Tissue Construction and Detection,Institute of Clinical Anatomy,Southern Medical University between December 2005 and July 2007. MATERIALS:Trizol reagent was purchased from Invitrogen,USA.RNA PCR kit(AMV) Ver 3.0 and 150 bp DNA Ladder Marker were purchased from TaKaRa,Japan.Unless otherwise specified,all other reagents were purchased from Sigma-Aldrich,USA. METHODS:Twenty-five Sprague Dawley rats were assigned to five groups(n = 5) according to developmental stages:embryonic(embryonic 15 days),neonatal(postnatal 5 days),juvenile (postnatal 1 month),adult(postnatal 3 months),and senile(postnatal 18 months). MAIN OUTCOME MEASURES:Detection of mRNA expression of Rho-A,Rac-1,CRMP-1,and Tubβ3 during various hippocampal developmental stages by reverse-transcription polymerase chain reaction. RESULTS:Hippocampal mRNA expression of Rho-A,as well as Rac-1,reached peak levels at embryonic,juvenile,and senile stages,and was relatively less during neonatal and adult stages. mRNA expression of Rac-1 was greater than Rho-A during each hippocampal developmental stage. CRMP-1 mRNA expression levels were as follows:embryonic>neonatal>juvenile>adultneonatal>juvenile>adult = senile. | 9 | Determination of optimal dietary selenium levels by full expression of selenoproteins in various tissues of broilers from 1 to 21 d of age显示文摘The current NRC dietary selenium(Se)requirement(0.15 mg/kg)of broilers is primarily based on growth performance data reported in 1986.Our study aimed to determine optimal dietary Se levels of broilers fed a practical corn-soybean meal diet for the full expression of selenoproteins in various tissues.A total of 384 one-d-old male broilers(n=8 replicates/diet)were fed a basal corn-soybean meal diet or the basal diet supplemented with 0.1,0.2,0.3,0.4 or 0.5 mg Se/kg in the form of Na_(2)SeO_(3) for 21 d.Regression analysis was conducted to evaluate the optimal dietary Se levels using broken-line,quadratic or asymptotic models.The activity of glutathione peroxidase(GPX)in the plasma,liver,kidney and pancreas,iodothyronine deiodinase(DIO)in the plasma,liver and pancreas,and thioredoxin reductase(Txnrd)in the liver and pancreas,the mRNA levels of Gpx1,Gpx4,Dio1,selenoprotein(Seleno)h,Selenop and Selenou in the liver,Gpx4,Dio1,Txnrd1,Txnrd2,Selenoh,Selenop and Selenou in the kidney,and Gpx1,Gpx4,Selenoh and Selenou in the pancreas,and the protein levels of GPX4 in the liver and kidney of broilers were influenced(P<0.05)by added Se levels,and increased quadratically(P<0.05)with the increase of added Se levels.The estimates of optimal dietary Se levels were 0.07 to 0.36 mg/kg based on the fitted broken-line,quadratic or asymptotic models(P<0.001)of the aforementioned selenoprotein expression in the plasma,liver and kidney,and 0.09 to 0.46 mg/kg based on the fitted broken-line models(P<0.001)of the aforementioned selenoprotein expression in the pancreas.The results indicate that the optimal dietary Se levels would be 0.36 mg/kg to support the full expression of selenoproteins in the plasma,liver and kidney,and 0.46 mg/kg to support the full expression of selenoproteins in the pancreas of broilers fed a practical corn-soybean meal diet from 1 to 21 d of age. | Xiudong Liao Guoqing Liu Guangming Sun Xiaoming Sun Tao Liu Lin Lu Liyang Zhang Minhong Zhang Yanli Guo Xugang Luo | 2021 | Animal Nutrition2021,,4: | 2 |

| 11 | Three-dimensional characteristics of mesoscale eddies simulated by a regional model in the northwestern Pacific Ocean during 2000–2008显示文摘Mesoscale eddies play vital roles in ocean processes.Although previous studies focused on eddy surface features and individual three-dimensional(3D)eddy cases in the northwestern Pacific Ocean,the analysis of unique eddy3D regional characteristics is still lacking.A 3D eddy detection scheme is applied to 9 years(2000-2008)of eddyresolving Regional Ocean Modeling System(ROMS)output to obtain a 3D eddy dataset from the surface to a depth of 1000 m in the northwestern Pacific Ocean(15°-35°N,120°-145°E).The 3D characteristics of mesoscale eddies are analyzed in two regions,namely,Box1(Subtropical Countercurrent,15°-25°N,120°-145°E)and Box2(Southern Kuroshio Extension,25°-35°N,120°-145°E).In Box1,the current is characterized by strong vertical shear and weak horizontal shear.In Box2,the current is characterized by the strong Kuroshio,topographic effect,and the westward propagation of Rossby waves.The results indicate the importance of baroclinic instability in Box1,whereas in Box2,both the barotropic and baroclinic instability are important.Moreover,the mesoscale eddies’properties in Box1 and Box2 are distinct.The eddies in Box1 have larger number and radius but a shorter lifetime.By contrast,Box2 has fewer eddies,which have smaller radius but longer lifetime.Vertically,more eddies are detected at the subsurface than at the surface in both regions;the depth of 650 m is the turning point in Box1.Above this depth,the number of cyclonic eddies(CEs)is larger than that of anticyclonic eddies(AEs).In Box2,the number of CEs is dominant vertically.Eddy kinetic energy(EKE)and mean normalized relative vorticity in Box2are significantly higher than those in Box1.With increasing depth,the attenuation trend of EKE and relative vorticity of Box1 become greater than those of Box2.Furthermore,the upper ocean(about 300 m in depth)contains 68.6%of the eddies(instantaneous eddy).Only 16.6%of the eddies extend to 1000 m.In addition,about87%of the eddies are bowl-shaped eddies in the two regions.Only about 3%are cone-shaped eddies.With increasing depth of the eddies,the proportion of bowl-shaped eddies gradually decreases.Conversely,the coneand lens-shaped eddies are equal in number at 700-1000 m,accounting for about 30%each.Studying the 3D characteristics of eddies in two different regions of the northwestern Pacific Ocean is an important stepping stone for discussing the different eddy generation mechanisms. | Guijing Yang Xiayan Lin Guoqing Han Yu Liu Gengxin Chen Jianhui Wang | 2022 | Acta Oceanologica Sinica2022,41,10: | 1 |

| 18 | Biological distribution of ^(131)I-labeled anti-nucleus antigen monoclonal antibody chTNT in patients with pulmonary metastases from differentiated thyroid carcinoma显示文摘This work is to study the in vivo biological distribution of 131I-labeled mouse/human chimeric monoclonal antibody (131I-chTNT) in patients with pulmonary metastases from differentiated thyroid carcinoma. Ten patients with differentiated thyroid carcinoma were injected intravenously with a single dose of 131I-chTNT (5 MBq·kg-1 body weight). Radioactivity of blood and urine samples was measured at different time points. The in vivo stability and the metabolic status of 131I-chTNT were detected with supersaturated trichloroacetic acid. Continuous imaging was performed to outline the region of interest (ROI) and estimate the intake level on the whole body,major organs and tumor lesions at different time points. The serum time-radioactivity curve of 131I-chTNT accorded with the two-compartment model after a single intravenous injection: T1/2(h)=65.28±14.83,AUC0-t(MBq·h·mL-1)=8.93±1.32,AUC0-∞(MBq·h·mL-1)=10.58±2.19,and CL(mL·min-1·kg-1)=1635±359. The time-radioactivity percentage curve of 131I-chTNT urine excretion accorded with the one-compartment model after a single intravenous injection: T1/2(h)=99±10,and accumulative (31±9) % radioactivity of the injected dose was excreted in urine in one week. The percentages of serum 131I-chTNT in radioactive components at 24,48 and 72 h were over 95% and it was still (88±7) % at 168 h. As for chemical composition of radioactive substances in urine,radioactivity in urine samples originated from free 131I by 100%. Radioactivity of 131I-chTNT after intravenous administration was mainly concentrated in the lung and liver,least in the brain. Radioactivity of tumor tissues reached the maximum at 24 h and the tumor/normal tissue (T/N) ratio reached the maximum (1.28~3.83) during 3~7 d. | 20 | Triphenyltin inhibition of the proteasome activity and its influence on substrate protein levels in nerve cells显示文摘Triphenyltin (TPT) widely exists as environmental pollutant, but its toxicity towards nerve cells and the related mechanism remain unclear. In this research, SHSY-5Y cells were exposed to TPT, and the cellular proteasome activity, Iκb proteins, Bax and α-synuclein levels were investigated. The results show that TPT can inhibit the cellular proteasome activity, and result in the accumulation of ubiquitinized proteins. TPT exposure can change the protein levels of IκB, Bax, and α-synuclein, affect NFκB pathway, activate the process of cell death, and induce α-synuclein aggregation.
