1ZIKA virus isolated from mosquitoes:a field and laboratory investigation in China,2016显示文摘A field investigation of arboviruses was conducted in Dejiang, Guizhou Province in the summer of 2016. A total of 8,795 mosquitoes, belonging to four species of three genera, and 1,300 midges were collected. The mosquito samples were identified on site according to their morphology, and the pooled samples were ground and centrifuged in the laboratory. The supernatant was incubated with mosquito tissue culture cells(C6/36) and mammalian cells(BHK-21) for virus isolation. The results indicated that 40%(3,540/8,795) were Anopheles sinensis, 30%(2,700/8,795) were Culex pipiens quinquefasciatus, and 29%(2,530/8,795)were Armigeres subbalbeatus. Furthermore, a total of eight virus isolates were obtained, and genome sequencing revealed two Zika viruses(ZIKVs) isolated from Culex pipiens quinquefasciatus and Armigeres subbalbeatus, respectively; three Japanese encephalitis viruses(JEVs) isolated from Culex pipiens quinquefasciatus; two Banna viruses(BAVs) isolated from Culex pipiens quinquefasciatus and Anopheles sinensis, respectively; and one densovirus(DNV) isolated from Culex pipiens quinquefasciatus.The ZIKVs isolated from the Culex pipiens quinquefasciatus and Armigeres subbalbeatus mosquitoes represent the first ZIKV isolates in China's Mainland. 2Whole-Genome Resequencing of a Worldwide Collection of Rapeseed Accessions Reveals the Genetic Basis of Ecotype Divergence显示文摘Rapeseed (Brassica napus),an important oilseed crop,has adapted to diverse climate zones and latitudes by forming three main ecotype groups,namely winter,semiwinter,and spring types. However,genetic variations underlying the divergence of these ecotypes are largely unknown. Here,we report the global pattern of genetic polymorphisms in rapeseed determined by resequencing a worldwide collection of 991 germplasm accessions.A total of 5.56 and 5.53 million singlenucleotide polymorphisms (SNPs)as Well as 1.86 and 1.92 million InDels were identified by mapping reads to the reference genomes of 'Darmor-bzh'and 'Tapidor,'respectively.We generated a map of allelic drift paths that shows splits and mixtures of the main populations,and revealed an asymmetric evolution of the two subgenomes of B.napus by calculating the genetic diversity and linkage disequilibrium parameters.Selective-sweep analysis revealed genetic changes in genes orthologous to those regulating various aspects of plant development and response to stresses.A genome-wide association study identified SNPs in the promoter regions of FLOWERING LOCUS T and FLOWERING LOCUS C orthologs that corresponded to the different rapeseed ecotype groups. 3Spin probes for electron paramagnetic resonance imaging显示文摘Electron paramagnetic resonance imaging (EPRI) is a relatively recent imaging technique, which provides potentially multidimensional imaging of the spatial distribution of paramagnetic species. Thanks to the use of stable spin probes, low frequency EPR imaging has recently allowed the use of large tissue samples or whole animals in vivo in the field of biology and medicine. It is normally necessary to introduce prior intravenous or intramuscular infusion of stable or slowly metabolizable non-toxic water-soluble paramagnetic materials, or stable implantable particulate materials as spin probes into the system. The classification and research progress of spin probes at present were described briefly. 4Correlation of Gut Microbiome Between ASD Children and Mothers and Potential Biomarkers for Risk Assessment显示文摘Variation of maternal gut microbiota may increase the risk of autism spectrum disorders(ASDs) in offspring. Animal studies have indicated that maternal gut microbiota is related to neurodevelopmental abnormalities in mouse offspring, while it is unclear whether there is a correlation between gut microbiota of ASD children and their mothers. We examined the relationships between gut microbiome profiles of ASD children and those of their mothers, and evaluated the clinical discriminatory power of discovered bacterial biomarkers. Gut microbiome was profiled and evaluated by 16S ribosomal RNA gene sequencing in stool samples of 59 mother–child pairs of ASD children and 30 matched mother–child pairs of healthy children. Significant differences were observed in the gut microbiome composition between ASD and healthy children in our Chinese cohort. Several unique bacterial biomarkers, such as Alcaligenaceae and Acinetobacter, were identified. Mothers of ASD children had more Proteobacteria, Alphaproteobacteria, Moraxellaceae, and Acinetobacter than mothers of healthy children. There was a clear correlation between gut microbiome profiles of children and their mothers; however, children with ASD still had unique bacterial biomarkers, such as Alcaligenaceae, Enterobacteriaceae, and Clostridium. Candidate biomarkers discovered in this study had remarkable discriminatory power. 6A botanical medicine dragon’s blood exhibited clinical antithrombosis efficacy similar to low molecular weight heparin显示文摘Deep vein thrombosis(DVT)is a common complication following traumatic fracture with a 0.5%–1%annual incidence.Low molecular weight heparin(LMWH)is the most commonly used anticoagulation drug for DVT prevention,but treatment with LMWH is invasive.Our aim is to compare the antithrombotic effect of dragon’s blood,an oral botanical anticoagulant medicine approved by the Chinese FDA,with LMWH in patients undergoing hip fracture surgery and to explore the molecular mechanisms of anticoagulation treatment.Our study recruited patients and divided them into LMWH and dragon’s blood treatment group.Coagulation index tests,Doppler ultrasound and mRNA sequencing were performed before and after anticoagulation therapy.There was no significant difference in postoperative DVT incidence between the two groups(23.1%versus 15.4%,P=0.694).D-dimer(D-D)and fibrinogen degradation product(FDP)showed significant reductions in both groups after anticoagulation treatments.We identified SLC4A1,PROS1,PRKAR2B and seven other genes as being differentially expressed during anticoagulation therapy in both groups.Genes correlated with coagulation indexes were also identified.Dragon’s blood and LMWH showed similar effects on DVT and produced similar gene expression changes in patients undergoing hip fracture surgery,indicating that dragon’s blood is a more convenient antithrombosis medicine(oral)than LMWH(hypodermic injection).Jiangbin Liang Song Mei Xiangyu Qiao Wei Pan Yan Zhao Shaohui Shi Yaling Zhai Haizhao Wen Guoping Wu Chengyu Jiang 2021Science China(Life Sciences)2021,64,10:4
7Analysis of rare-earth elements in fluid inclusions by inductively coupled plasma-mass spectrometry (ICP-MS)显示文摘Inductively coupled plasma_mass spectrometry (ICP_MS) is used to determine rare_earth elements abundances in fluid inclusions. Quartz separates from the Lannigou and Yata Carlin_type gold deposits, southwestern China, were prepared by the decrepitation_leach method. Leachates were analyzed by ICP_MS, using conventional sample introduction. The results demonstrate that the decrepitation_leach method, in combination with ICP_MS analysis of leachates, is effective for REE determination. 9Effects of Industrial Relocation on Chinese Regional Economic Growth Disparities: Based on System Dynamics Modeling显示文摘The economic growth of China has led to increasing growth disparities between regions. Such disparities are uncontrolled and are severely negative symptoms in the process of economic development. On the basis of system dynamics(SD) modeling and the relationship between industrial relocation and regional economic growth, we construct a model of the interrelationship between the two aforementioned phenomena. The model is an effective and creative exploration for examining effects of industrial relocation on Chinese regional economic growth disparities. The SD model is employed in this study to build an inter-regional labor migration SD model, an inter-regional capital migration SD model, an intra-industry SD model, an intra-regional population SD model, and an intra-regional SD model which are based on realities in labor and capital flow from the view of industrial relocation. VENSIM software is utilized to perform a system simulation based on the data of the eastern, middle, and western regions from 2000 to 2010. Results show that industrial relocation gradually narrows the relative disparity in GDP among the three regions. Moreover, the absolute one is enlarged continuously. 10Inhibition of ASCT2 induces hepatic stellate cell senescence with modified proinflammatory secretome through an IL-1α/NF-κB feedback pathway to inhibit liver fibrosis显示文摘Senescence of activated hepatic stellate cells(aHSCs) is a stable growth arrest that is implicated in liver fibrosis regression.Senescent cells often accompanied by a multi-faceted senescence-associated secretory phenotype(SASP).But little is known about how alanine-serine-cysteine transporter type-2(ASCT2),a high affinity glutamine transporter,affects HSC senescence and SASP during liver fibrosis.Here,we identified ASCT2 is mainly elevated in aHSCs and positively correlated with liver fibrosis in human and mouse fibrotic livers.We first discovered ASCT2 inhibition induced HSCs to senescence in vitro and in vivo.The proinflammatory SASP were restricted by ASCT2 inhibition at senescence initiation to prevent paracrine migration.Mechanically,ASCT2 was a direct target of glutaminolysisdependent proinflammatory SASP,interfering IL-1α/NF-κB feedback loop via interacting with precursor IL-1α at Lys82.From a translational perspective,atractylenolide Ⅲ is identified as ASCT2 inhibitor through directly bound to Asn230 of ASCT2.The presence of -OH group in atractylenolide Ⅲ is suggested to be favorable for the inhibition of ASCT2.Importantly,atractylenolide Ⅲ could be utilized to treat liver fibrosis mice.Taken together,ASCT2 controlled HSC senescence while modifying the proinflammatory SASP.Targeting ASCT2 by atractylenolide Ⅲ could be a therapeutic candidate for liver fibrosis.Feixia Wang Zhanghao Li Li Chen Ting Yang Baoyu Liang Zili Zhang Jiangjuan Shao Xuefen Xu Guoping Yin Shijun Wang Hai Ding Feng Zhang Shizhong Zheng 2022Acta Pharmaceutica Sinica B2022,12,9:2

18Influencing factors of colostrum exposure to low-level lead and their relationship with early neural development of infants显示文摘BACKGROUND: Exposure to low-level lead has a toxic effect on the development of neonates, which has attracted wide attention. Colostrum lead level can be used as the indication of lead exposure. OBJECTIVE: To observe the relationship of colostrum lead level and the neurobehavioral development of infants. DESIGN: A prospective control observation. SETTING: Center for Maternal and Child Health, Shanxi Provincial Children's Hospital. PARTICIPANTS: Totally 128 neonates of full-term normal delivery, 76 male and 52 female, from Shanxi Provincial Maternal and Child Health Center and Jiexiu Maternal and Child Health Center were involved in this study. All the involved neonates had no peripartal ischemic/hypoxic history or fetus intrauterine developmental lag. Pregnant women had no various acute and chronic diseases in pregnancy, family history of neurological disease or occupational lead exposure. 128 portions of colostrum sample of full-term normal delivery were collected. Informed consents of detected items were obtained from the puerperants and their relatives. METHODS: ① Experimental grouping: Lead level in the colostrum was determined by atomic absorption spectrometry. According to lead level in the colostrum, the neonates were classified into two exposure groups of greater than or equal to 0.24 μmol/L in a high-level lead group and less than 0.24 μ mol/L in a low-level lead group. ② Experimental evaluation: Mental developmental index (MDI) and psychological developmental index (PDI) of 3-month-old infants were evaluated with Bayley Scales of Infant Development (BSID). The relationships of MDI, PDI and colostrum lead level were performed correlation regression analysis; The relationship of colostrum lead level and development was performed multi-factor analysis with family environment and health questionnaires. MAIN OUTCOME MEASURES: ① Evaluation results of MDI and PDI. ② Multi-factor analysis results. RESULTS: Totally 128 neonates were involved in the study. Ten and eleven neonates were lost due to emigration in the high-level lead group and low-level lead group respectively, and the other 107 neonates participated in the final analysis. ① MDI and PDI in the high-level lead group were significantly lower than those in the low-level lead group, respectively (P < 0.01); Regression analysis results showed that two developmental indexes were statistically negatively correlated with colostrum lead level (regression equation y = 1.9+0.01x1,-0.04x2,+0.04x3,+0.03x4). ② Four variables of the factors included by family environment and health questionnaires were taken into equation. Large maternal age, irrational dietary pattern in pregnancy and pollution degree of habitation environment in pregnancy were the risk factors of colostrum lead level (partial regression coefficien t =0.598 4,0.426 8,0.306 7,P < 0.05-0.01), and calcium supplementation in pregnancy was a protective factor (partial regression coefficien t =-0.455 8, P < 0.01). 20Comparison of the Arctic upper-air temperatures from radiosonde and radio occultation observations显示文摘The air temperature is one of the most important parameters used for monitoring the Arctic climate change. The constellation observing system for meteorology, ionosphere, and climate and Formosa Satellite Mission 3(COSMIC/FORMOSAT-3) radio occultation(RO) 'wet' temperature product(i.e., 'wet Prf') is used to analyze the Arctic air temperature profiles at 925–200 hPa in 2007–2012. The 'wet' temperatures are further compared with radiosonde(RS) observations. The results from the spatially and temporally synchronized RS and COSMIC observations show that their temperatures agree well with each other, especially at 400 hPa. Comparisons of seasonal temperatures and anomalies from the COSMIC and homogenized RS observations suggest that the limited number of COSMIC observations during the spatial matchup may be insufficient to describe the smallscale spatial structure of temperature variations. Furthermore, comparisons of the seasonal temperature anomalies from the RS and 5°×5° gridded COSMIC observations at 400 hPa during the sea ice minimum(SIM) of2007 and 2012 are also made. The results reveal that similar Arctic temperature variation patterns can be obtained from both RS and COSMIC observations over the land area, while extra information can be further provided from the densely distributed COSMIC observations.
